Nucleotide Aptamers as Theranostic Biomaterials

Abstract:Polymeric aptamers, or chemical antibodies, consisting of oligonucleotides are regarded as upgraded polypeptidic antibodies. Aptamers are often sorted through systematic evolution of ligands by exponential enrichment. They specifically recognize a variety of targets, such as small ligands, macromolecules, and even cells with super high binding affinity. Since the aptamer-ligand interacts through an induced fit process, aptamers change the structure of targets, which consequently affect the functions of the target. Aptamers alone can function as therapeutics. However, their primary role is to navigate other therapeutics/vehicles to the desired targets through covalent or noncovalent conjugation to avoid off-target effects. Because of their excellent binding affinity and specificity, the combination of an aptamer with another component leads to other applications, such as signal detection and disease diagnostics. There is no doubt the development of aptamers will deepen our understanding of polynucleotides themselves and enriches our theranostic biomaterial arsenal.
