Abstract:Pancreatic cancer is one of the most challenging diseases. Patients with metastatic pancreatic cancer has a poor prognosis with a 3% five-year survival rate. The high mortality rate is largely due to two major factors: the severely limited resectability and extremely high rate of metastasis. We applied laser immunotherapy (LIT) to treat a highly metastatic, poorly immunogenic pancreatic tumor model (Panc02-H7) in mice. Subcutaneous tumors were irradiated by a near-infrared laser non-invasively and orthotopic tumors were irradiated by the laser interstitially, followed by intratumoral injection of glycated chitosan, an immunoadjuvant. Long-term survival and tumor-specific immunity were observed after LIT treatment. The effectiveness of LIT in treating pancreatic tumors demonstrated in this study could lead to a feasible modality for this deadly disease.
