Abstract:Macroalgae, the major contributor of primary productivity in coastal seas, contribute to the material cycle and energy flow in marine ecosystems. The purpose of this work was to evaluate the toxic effect of nano-TiO2 on the growth and antioxidant activity of Gracilaria lemaneiformis. An obvious inhibition of growth was observed in this study. The algae exposed to nano-TiO2 showed a negative growth rate at 20 mg L-1 and 40 mg L-1 during the 15 days exposure. The concentration of soluble protein increased slightly during the first 3 days of exposure, but it gradually diminished thereafter due to the high concentrations of nano-TiO2 and to prolonged exposure. Nano-TiO2 caused oxidative damage in G. lemaneiformis; superoxide anions accumulated, and nitrate reductase activity decreased linearly with the increase in nano-TiO2. Furthermore, extracts of G. lemaneiformis can scavenge DPPH<bold> and hydroxyl radicals for their antioxidant capacity</bold>. However, the capacity to scavenge DPPH<bold> and hydroxyl radicals in vitro decreased slightly with the increase in nano</bold>-TiO2. The results from this work imply that macroalgae can be an effective biomarker of nano-TiO2 contamination and can be useful indicators to evaluate the oxidative damage of increasing pollutants in marine ecosystems.
Adverse Effect of TiO 2 Nanoparticles on Antioxidant System and Antitumor Activities of Macroalgae Gracilaria Lemaneiformis

Abstract:Recently, the widespread use of TiO2 nanoparticles (nano-TiO2) has attracted the global concern regarding their adverse effects to aquatic organisms. Nanoparticles may get to the aquatic environments and harm the growth of algae. This paper presents the studies of nano-TiO2 concentrations influence on the photosynthetic characteristics, antioxidant system and antitumor activity of marine macroalgae Gracilaria lemaneiformis after exposed to 0, 5, 10, 20 or 40 mg L-1 of nano-TiO2. Results showed that the photosynthetic pigment content of G. lemaneiformis obviously decreased and the antioxidant defense system was significantly disturbed after exposure to nano-TiO2. Compared with control, the maximum level of malondialdehyde showed 3.3-fold increase for 15 days exposure to nano-TiO2. The activity of superoxide dismutase and catalase enhanced obviously by 1.10–2.24-fold. The aqueous extract of G. lemaneiformis (AEGL) possesses anticancer activity against HepG2 liver cancer cells proliferation and the 48 h-cell inhibition exceeds 35%. But the toxicity of nano-TiO2 would inhibit the anticancer activity of AEGL. When G. lemaneiformis was exposed to nano-TiO2, the effect of the aqueous extract of G. lemaneiformis on the cell inhibition, reactive oxygen species production, apoptosis, cycle arrest of HepG2 cells significantly lessened, and which were linearly associated with nano-TiO2 exposure concentration. Those results further confirmed that nano-TiO2 can inhibit the anticancer activity of G. lemaneiformis. The results in this work imply that macroalgae can be an effective biomarker of nano-TiO2 contaminant, and can be useful indicator to evaluate ecological effects of increasing pollutants in marine ecosystem.
Toxicity of Nano-Tio2 on Algae and the Site of Reactive Oxygen Species Production.

Abstract:Given the extensive use of nanomaterials, they may enter aquatic environments and harm the growth of algae, which are primary producers in an aquatic ecosystem. Thus, the balance of an aquatic ecosystem may be destroyed. In this study, Karenia brevis and Skeletonema costatum were exposed to nano-TiO2 (anatase, average particle size of 5-10 nm, specific surface area of 210 +/- 10 m(2) g(-1)) to assess the effects of nano-TiO2 on algae. The findings of transmission electron microscopy-energy dispersive X-ray spectroscopy (TEM-EDX) and scanning electron microscopy (SEM) demonstrate aggregation of nano-TiO2 in the algal suspension. Nano-TiO2 was also found to be inside algal cells. The growth of the two species of algae was inhibited under nano-TiO2 exposure. The 72 h EC50 values of nano-TiO2 to K. brevis and S. costatum were 10.69 and 7.37 mg L-1, respectively. TEM showed that the cell membrane of K. brevis was destroyed and its organelles were almost undistinguished under nano-TiO2 exposure. The malondialdehyde (MDA) contents of K. brevis and S. costatum significantly increased compared with those of the control (p < 0.05). Meanwhile, superoxide dismutase (SOD) and catalase activities (CAT) of K. brevis and S. costatum changed in different ways. The reactive oxygen species (ROS) levels in both species were significantly higher than those of the control (p < 0.05). The site of ROS production and accumulation in K. brevis and S. costatum under nano-TiO2 exposure was explored with the addition of inhibitors of different electron transfer chains. This study indicated that nano-TiO2 in algal suspensions inhibited the growth of K. brevis and S. costatum. This effect was attributed to oxidative stress caused by ROS production inside algal cells. The levels of anti-oxidative enzymes changed, which destroyed the balance between oxidation and anti-oxidation. Thus, algae were damaged by ROS accumulation, resulting in lipid oxidation and inhibited algae growth. The inhibitors of the electron transfer chain showed that the site of ROS production and accumulation in K. brevis cells was the chloroplast. (C) 2014 Elsevier B.V. All rights reserved.
[Toxic Effects of Nano-Tio2 on Gymnodinium Breve].

Abstract:In order to reveal the toxicity and mechanism of nano-TiO2 on algae, the inhibition effect, enzyme activity, oxygen free radicals of nano-TiO2 on the growth of G. breve were investigated. The results showed that G. breve was inhibited by nano-TiO2, and the 72 h-EC50 was 9.7 mg x L(-1). With the increasing concentration of nano-titanium dioxide, the activities of SOD decrease significantly (P < 0.05). The content of hydrogen peroxide radicals and the activities of CAT increase significantly (P < 0.05), and the content of superoxide anion shows the increasing trend. The content of hydrogen peroxide radicals was 0.083 U x mL(-1) in 0 mg x L(-1) nano-TiO2 suspension while that was 1.1 U x mL(-1) in control after 48 h. Through the study of 20 mg x L(-1) nano-titanium dioxide on G. breve at different times, the activities of SOD and CAT, the content of MDA are consistent, which the highest values is achieved at the exposure time of 12 hours and the lowest value is found at the exposure time of 48 hours. The content of hydroxyl radical increased significantly at the exposure time of 48 hours. The activity of SOD was 0.14 U x (10(7) cell x min)(-1) in G. breve at 12 h which was ten times higher than that at 48 h.
Research on Toxic Effects of Nano-Tio2 on Skeletonema Costatum and Dunaliella Salina

Abstract:A large application of nano-materials increases their risk entering the marine environment.The primary purpose of this study is to explore the toxic effects and mechanisms of nano-TiO2,which is widely applied in industry and commercial products.Algal growth inhibition test was carried out,and chlorophyll concentration and lipid peroxidation level were measured using Skeletonema costatum and Dunaliella salina as model organisms.Moreover,adsorption of nano-TiO2 on microalgae was observed by SEM technique.The settlement of nano-TiO2 particles in experimental medium with 24 h was also characterized.Results showed that both growth and photosynthesis of S.costatum and D.salina were inhibited by nano-TiO2.Toxic effects of nano-TiO2 on S.costatum were similar to those of D.salina.Adsorption of nano-TiO2 and oxidative damages resulting from interactions between nano-TiO2 and microalgae could inhibit growth and photosynthesis of microalgae,which were the main reasons for toxic effects of nano-TiO2.The settlement of nano-TiO2 particles in experiment medium caused the decrease of suspension concentration which had an influence on results.

Abstract:The objective of the present work was to evaluate the toxic effect of nonylphenol (NP) on the antioxidant response and antitumor activity of Gracilaria lemaneiformis. An obvious oxidative damage was observed in this study. The thallus exposed to NP showed 1.2–2.0-fold increase in lipid peroxide and displayed a maximum level of 16.58 μmol g−1 Fw on 0.6 mg L−1 for 15-day exposure. The activities of antioxidant enzymes such as superoxide dismutase (SOD) and catalase (CAT) enhanced significantly by 1.1–3.2-fold and subsequently diminished at the high concentrations and prolonged exposure. The results of DNA damage in comet assay also supported that NP was obviously toxic on G. lemaneiformis with increasing the percentage of tail DNA in a dose-dependent manner. Furthermore, the ethanol extract of G. lemaneiformis (EEGL) did exhibit antitumor potential against HepG-2 cells. While decreased in cell inhibition, ROS generation, apoptosis, and caspase-3 in HepG-2 cells treated with the EEGL were observed when G. lemaneiformis was exposed to NP for 15 days, and which were related to exposure concentration of NP. These suggested that NP has strongly toxic effect on the antitumor activity of G. lemaneiformis. The results revealed in this study imply that macroalgae can be useful biomarkers to evaluate marine pollutions.
Effects Of Titanium Dioxide Nanoparticles On Photosynthetic And Antioxidative Processes Of Scenedesmus Obliquus

Abstract:The effects of the photocatalytic toxicity of titanium dioxide nanoparticle (nano-TiO2) on phytoplankton are well understood. However, as UV light intensity decreases sharply with the depth of the water column, the effects of nano-TiO2 itself on deeper water phytoplankton, such as green algae, need further research. In this research, we investigated the effects of three sizes of TiO2 (10, 50 and 200 nm) on the photosynthetic and antioxidative processes of Scenedesmus obliquus in the absence of UV light. We found that 50 nm and 10 nm TiO2 (10 mg/L) inhibited growth rates and the maximal photosystem II quantum yield compared to the control in Scenedesmus obliquus. The minimal and maximal fluorescence yields, and the contents of reactive oxygen species and lipid peroxidation, increased, indicating that photosynthetic energy/electrons transferred to oxygen and induced oxidative stress in nano-TiO2-treated samples. In addition, we found that aggregations of algae and 10 nm TiO2 were present, which could induce cell membrane disruption, and vacuoles were induced to cope with nano-TiO2 stress in Scenedesmus obliquus. These results enhance our understanding of the effects of nano-TiO2 on the photosynthetic and antioxidative processes of green algae, and provide basic information for evaluating the ecotoxicity of nano-TiO2 in freshwater ecosystems.
TiO2 nanoparticles in the marine environment: Physical effects responsible for the toxicity on algae Phaeodactylum tricornutum

Abstract:Nanoscale titanium dioxide (nTiO2) has been widely used in cosmetics, catalysts, varnishes, etc., which is raising concerns about its potential hazards to the ecosystem, including the marine environment. In this study, the toxicological effect of nTiO2 on the marine phytoplankton Phaeodactylum tricornutum was carefully investigated. The results showed that nTiO2 at concentrations ≥20mg/L could significantly inhibit P. tricornutum growth. The 5-day EC50 of nTiO2 to P. tricornutum growth is 167.71mg/L. Interestingly, nTiO2 was found to exert its most severe inhibition effects on the first day of exposure, at a lower EC50 of 12.65mg/L. During the experiment, nTiO2 aggregates were found to entrap algae cells, which is likely responsible for the observed toxic effects. Direct physical effects such as cell wall damage from the algae entrapment were confirmed by flow cytometry and TEM imaging. Moreover, low indirect effects such as shading and oxidative stress were observed, which supported the idea that direct physical effects could be the dominant factor that causes nTiO2 toxicity in P. tricornutum. Our research provides direct evidence for the toxicological impact of nTiO2 on marine microalgae, which will help us to build a good understanding of the ecological risks of nanoparticles in the marine environment.

Abstract:The increasing production of nano-TiO 2 has attracted extensive concerns about the ecological consequence and health risk of these compounds in natural ecosystem. However, little is known about its toxicity on zooplankton, especially its possibility to access to the food chain via dietary exposure. To address this concern, the toxic and cumulative effects of nano-TiO 2 on an aquatic food chain were explored through two trophic levels independently or jointly including producer and consumer. The results revealed that exposure to suspensions of nanomaterials had negative effects on both producers and consumers. Specifically, nanoparticles reduced the density of algal cells in a concentration-dependent way, and hatching life expectancy, average lifespan, net reproductive rate, and population intrinsic growth rate of rotifers decreased significantly with the concentration of nanomaterials increased ( P 1. Exposure concentration, exposure time and their interactions play a strong part in the accumulation of nanoparticles in algae and rotifers. Overall, the present findings confirmed that nano-TiO 2 was deleterious to plankton, posing a significant environmental threat to aquatic ecosystems.

Abstract:Due to their unique physicochemical characteristics, nanomaterials exhibit many excellent properties and functions, leading to their applications in numerous fields. The large-scale production and widespread application of nanomaterials have inevitably resulted in their release into the environment, especially the water environment. Several studies have confirmed that exposure to nanomaterials can be toxic to aquatic organisms. However, few studies have focused on the effects of nanomaterial exposure on growth and immunity in crustaceans. In the present study, juvenile Macrobrachium rosenbergii were exposed to different concentrations of titanium dioxide (TiO2)/activated carbon (AC) composite nanomaterial (0.1 and 0.5 mg/L) for 45 days. The effects of nanoparticle exposure on digestion and antioxidant-related enzyme activities, as well as the expression of growth and immunity-related genes and signaling pathway, were evaluated. Our results show that in response to low concentration of TiO2/AC nanoparticle (0.1 mg/L), most of the enzyme activities related to digestion and antioxidation (TPS, LPS, AMS, SOD, and CAT) were diminished. On the contrary, the GSH-Px activity increased under the 0.1 mg/L group of TiO2/AC nanoparticle concentration. Additionally, the level of digestive and antioxidant enzyme activities we detected was increased when exposed to 0.5 mg/L TiO2/AC nanoparticle. By comparison to the expression level of growth-related genes in the control group, MSTN, CaBP, E75, Raptor, EcR, and EGF were significantly inhibited at 0.1 and 0.5 mg/L concentrations of TiO2/AC nanoparticle, whereas the expression level of genes (TLR, JAK, STAT, PPAF, ACP, and AKP) related to immunity was increased when exposed to different concentrations of TiO2/AC nanoparticle. Compared with the control group (0 mg/L concentration), 5166 DEGs were identified in the TiO2/AC nanoparticle group, and a large number of DEGs were involved in molting, energy metabolism, stress tolerance, and germ cell development. Moreover, KEGG analysis revealed that many DEGs were assigned into signaling pathways related to metabolic growth and immune stress. These results showed that exposure to TiO2/AC nanoparticle will result in the changes of enzyme activity and routine mRNA expression, suggesting that TiO2/AC nanoparticle which existed in aquatic environment might affect the physiology of M. rosenbergii. This study will provide significant information for the evaluation of nanomaterial toxicity on aquatic crustaceans.

Abstract:The increasing use of nanotechnology highlights the need to understand and clarify the environmental impacts of nanomaterials. In this study, the acute toxicity and oxidative stress of titanium dioxide nanoparticles (nTiO(2)) in mature marine abalone (Haliotis diversicolor supertexta) were assessed. No acute effect was found in any of the treatment groups with nTiO(2) concentration gradients ranging from 0.1 to 10 mg/L. However, the activity of an antioxidant enzyme superoxide dismutase (SOD) significantly increased in the group that was exposed to 1.0 mg/L nTiO(2). The content of a non-enzymatic antioxidant, glutathione (GSH), significantly decreased in the groups with an nTiO(2) concentration 1.0 mg/L. The level of lipid peroxidation (LPO) was found to increase as the nTiO(2) dose increased. Furthermore, NO was produced in excess in abalone. These results demonstrated that, although nTiO(2) is not acutely toxic to abalone, it does exert oxidative stress on abalone. (C) 2011 Elsevier Ltd. All rights reserved.

Abstract:Nanoparticle cerium oxide (n-CeO2) has been widely used, recently, its toxicity to the aquatic environment has received increasing attention. This study aimed to explore the effects of n-CeO2 on the physiology of Chlorella pyrenoidosa. Results showed that n-CeO2 may inhibited the growth of Chlorella pyrenoidosa, and make some influence of chla and protein contents because of the ROS. The activity of SOD and MDA contents also indicated that the high concentration of n-CeO2 may beyond the range of tolerance, which means ROS content may be a key factor in the toxic effects of n-CeO2 on Chlorella pyrenoidosa.

Abstract:Titanium dioxide nanoparticles (n-TiO2) have been used in numerous applications, which results in their release into aquatic ecosystems and impact algal populations. A possible toxic mechanism of n-TiO2 on algae is via the disruption of the photosynthetic biochemical pathways, which yet remains to be demonstrated. In this study, Chlorella pyrenoidosa was exposed to different concentrations (0, 0.1, 1, 5, 10, and 20 mg/L) of a type of anatase n-TiO2, and the physiological, biochemical, and molecular responses involved in photosynthesis were investigated. The 96 h half growth inhibition concentration (IC50) of the n-TiO2 to algae was determined to be 9.1 mg/L. A variety of cellular and sub-cellular damages were observed, especially the blurry lamellar structure of thylakoids, indicating the n-TiO2 impaired the photosynthetic function of chloroplasts. Malondialdehyde (MDA) and glutathione disulfide (GSSG) significantly increased while the glutathione (GSH) content decreased. This implies the increased consumption of GSH by the increased intracellular oxidative stress upon n-TiO2 was insufficient to eliminate the lipid peroxidation. The contents of photosynthetic pigments, including chlorophyll a (Chl a) and phycobiliproteins (PBPs) in the exposed algal cells increased along with the up-regulation of genes encoding Chl a and photosystem II (PS II), which could be explained by a compensatory effect to overcome the toxicity induced by the n-TiO2. On the other hand, the photosynthetic activity was significantly inhibited, indicating the impairment on the photosynthesis via damaging the reaction center of PS II. In addition, lower productions of adenosine triphosphate (ATP) and glucose, together with the change of gene expressions suggested that the n-TiO2 disrupted the material and energy metabolisms in the photosynthesis. These findings support a paradigm shift of the toxic mechanism of n-TiO2 from physical and oxidative damages to metabolic disturbances, and emphasize the threat to the photosynthesis of algae in contaminated areas.
Complex Role of Titanium Dioxide Nanoparticles in the Trophic Transfer of Arsenic from Nannochloropsis Maritima to Artemia Salina Nauplii.

Abstract:Increasing concern has been focused on the potential risks associated with the trophic transfer to aquatic organisms of ambient contaminants in the presence of titanium dioxide nanoparticles (nano-TiO2). This study investigated the influence of nano-TiO2 on the trophic transfer of arsenic (As) from the microalgae Nannochloropsis maritima to the brine shrimp Artemia sauna nauplii. We found that nano-TiO2 could significantly facilitate As sorption on N. maritima within an exposure period of 24 h, and this sorption subsequently led to higher As trophic transfer from the algae to A. sauna according to trophic transfer factors (TTFA(AS+nano-TiO2) > TTFAs). However, after 48 h of depuration, the retention of As in A. sauna fed As-nano-TiO2-contaminated algae was even lower than that in A. sauna fed As-contaminated algae at the same exposure concentrations. This result indicates that the increased food chain transfer of As in the presence of nano-TiO2 can be explained by adsorption of As onto nano-TiO2 in contaminated food (algae), but the bioavailability of As in A. sauna is reduced after the introduction of nanoparticles. Although the stress enzyme activities of superoxide dismutase (SOD) and acetylcholinesterase (AChE) in A. sauna at a lower As concentration treatment in the presence of nano-TiO2 were not significantly changed, they increased with higher exposure concentrations of As with or without nano-TiO2. Our study highlighted the complex role of nanomaterials in the transfer of ambient contaminants via trophic chains and the potential of nano-TiO2 to reduce the bioavailability of As via trophic transfer to saltwater zoo plankton.

Abstract:The effect of anthracene on the antioxidant system of two species of marine microalgae , Nitzschia closterlum and Platymonas subcordiforming was studied. With increasing of anthracene concentration , the non enzymic component carotenoid (CAR) content and reduced glutathione (GSH ) content in the two species of marine microalgae decreased, and the enzymic component-superoxide dismutase (SOD) activity and catalase (CAT) activity decreased. When extrinsic GSH was added to the medium, the inhibition on growth of microalgae by anthracene was repaired sometimes. The analysis indicated that anthracene stress could damage the antioxidant system of marine microalgae, resulted in disequilibrium between production and scavenging of reactive oxygen spices (ROS), and further exacerbated the lipid peroxidation on algae cells. Abstract:According to the results of the experimental study, it was found that the addition of titanium dioxide nanoparticles does not lead to changes in morphological, structural, functional and fluorescent indices of the diatom Thalassiosira weissflogii and the dinophyte Prorocentrum cordatum in the concentration range of 0.5–6.2 mg L −1 . At the highest concentration of the pollutant (6.2 mg L −1 ), no decrease in the enzymatic activity (FDA fluorescence characterizes the level of metabolic activity of cells) of algae and no increase in the production of reactive oxygen (D2CFH-DA fluorescence, reflects ROS production in cells) species were observed after 72 h of cultivation. The efficiency of the cell photosynthetic apparatus (Fv/Fm) and relative electron transport rate (rETR) were maintained at a high relatively constant level for algae growing under high NPs TiO 2 concentration. Scanning electron and light microscopy micrographs also showed the absence of morphological changes in T. weissflogii and P. cordatum cells and no significant cell wall deformation when exposed to NPs TiO 2 . It is suggested that probably the absence of adsorption of titanium dioxide nanoparticles on the surface of T. weissflogii and P. cordatum cells is associated with the physicochemical properties of TiO 2 NPs, in particular, with their surface charge.

Physiological and Transcriptomic Analysis Reveals the Toxic and Protective Mechanisms of Marine Microalga Chlorella Pyrenoidosa in Response to TiO2 Nanoparticles and UV-B Radiation

Abstract:Concerns have been raised regarding the adverse effects of nanoparticles (NPs) on marine organisms, as an increasing number of NPs inevitably enter the marine environment with the development of nanotechnology. Owing to the photocatalytic properties, TiO2 NPs' toxicity may be aggravated by enhanced UV-B resulting from stratospheric ozone depletion. However, the molecular mechanisms of phytoplankton in response to TiO2 NPs under UV-B remains poorly understood. In this study, we integrated whole transcriptome analysis with physiological data to provide understanding on the toxic and protective mechanisms of marine Chlorella pyrenoidosa in response to TiO2 NPs under UV-B. The results indicated that the changes in gene expression could be related to the growth inhibition and TiO2 NP internalization in C. pyrenoidosa, and several molecular mechanisms were identified as toxicity response to TiO2 NPs and UV-B. Differential expression of genes involved in glycerophospholipids metabolism indicated that cell membrane disruption allowed TiO2 NPs to enter the algal cell under UV-B exposure, although the up-regulation of genes involved in the general secretory dependent pathway and the ATP-binding cassette transporter family drove cellular secretion of extracellular polymeric substances, acting as a barrier that prevent TiO2 NP internalization. The absence of changes in gene expression related to the antioxidant system may be responsible for the severe oxidative stress observed in algal cells following exposure to TiO2 NPs under UV-B irradiation. Moreover, differential expression of genes involved in pathways such as photosynthesis and energy metabolism were up-regulated, including the light-harvesting, photosynthetic electron transport coupled to photophosphorylation, carbon fixation, glycolysis, pentose phosphate pathway, tricarboxylic acid cycle, and oxidative phosphorylation, indicating that more energy and metabolites were supplied to cope with the toxicity of TiO2 NPs and UV-B. The obtained results provide valuable information on the molecular mechanisms of response of marine phytoplankton exposed to TiO2 NPs and UV-B.

Abstract:Metallic nanoparticles (MNPs) are commonly incorporated in products found in households, industries, and agriculture. The presence of MNPs in the aquatic environment causes damage to living organisms and pollutes the water body rendering it harmful for human consumption. Several studies have been made on the toxicity of MNPs toward microalgae. Most of these studies reported changes in the cellular structure, growth rate, pigments, proteins, and enzymatic activity of microalgae. This review paper focuses on the toxic effects of titanium, zinc, and silver nanoparticles on microalgae in the aquatic environment. A better understanding of the behavior of MNPs in the ecosystem will allow scientists to produce environmentally safe MNPs.
