Abstract:Mislabeling and adulteration of tuna are common due to the diminishing of morphological characteristics during processing. The tuna authenticity is now being focused in the seafood supply chain. In this study, the lipid profiles of 3 commercial tuna species (skipjack tuna, bigeye tuna and yellowfin tuna) were investigated via ultra-high performance liquid chromatography coupled with Q-Exactive Orbitrap mass spectrometry (UPLC-Q-Exactive Orbitrap MS). A total of 439 lipid species were identified and semi-quantitated by MS-DIAL. Further biomarkers discovery was carried out by chemometrics, leading to 27 lipids being identified as potential lipid biomarkers. Comparisons to reference standards revealed that lipid biomarkers were effective for discrimination of different tuna species. Interestingly, the proposed lipid biomarkers were all glycerophospholipids, implying that they might be the focus of future study.

Abstract:Food fraud is common in the tuna industry because of the economic benefits involved. Ensuring the authenticity of tuna species is crucial for protecting both consumers and tuna stocks. In this study, GC-Q-TOF and UPLC-Q/Orbitrap mass spectrometry-based metabolomics were used to investigate the metabolite profiles of three commercial tuna species (skipjack tuna, bigeye tuna and yellowfin tuna). A total of 22 and 77 metabolites were identified with high confidence using GC-Q-TOF and UPLC-Q/Orbitrap mass spectrometry, respectively. Further screening via chemometrics revealed that 38 metabolites could potentially serve as potential biomarkers. Hierarchical cluster analysis showed that the screened metabolite biomarkers successfully distinguished the three tested tuna species. Furthermore, a total of 27 metabolic pathways were identified through enrichment analysis based on the Kyoto Encyclopaedia of Genes and Genomes (KEGG) pathways.

Abstract:The micro-flowing water system can improve the flesh quality of freshwater fish using the traditional pond farming method. However, the mechanism of this phenomenon has not yet been explored. This study intends to examine the changes of metabolites in freshwater fish after treatment with the micro-flowing purification system (MFPS). The UPLC-QTOF/MS based metabolomics method was utilized to screen the metabolites and predict the major possible metabolic pathways after MFPS treatment. There were 377 types of metabolites identified in the fish muscle, of which 54-71 represented significant different metabolites identified during different stages of MFPS treatments. The main mechanism of MFPS treatment in improving the quality of grass carp fish muscle was investigated, and the MFPS treatment was shown to improve the flesh quality and the flavor of grass carp fish muscle. This study could provide the theoretical basis for improving the quality of aquatic products.

The quality degradation and excessive fat accumulation in cultured C. idellushave long been attributed to possible alterations in the lipid metabolism of fish muscle tissues as a result of over-nutrition from artificial diets. To investigate the effects of different diets on fish muscle quality, a large-scale metabolomics study was performed on 250 tails of C. idellus. The experimental fish were divided into four groups based on sex and diets - female artificial feed (FAF), female grass feed (FGF), male artificial feed (MAF) and male grass feed (MGF). After 113 days rearing period, the AF group showed significantly higher total mass of muscle fat (P < 0.01), with the FAF group being the highest. Metabolomics profiling based on liquid chromatography-mass spectrometry (LC-MS) revealed distinctive patterns of clustering according to the four groups. Overall, artificial feeding was associated with higher concentrations of docosapentaenoic acid (DPA), dihomo-gamma-linolenic acid (DGLA) and arachidonic acid (ARA); whereas grass-feeding was associated with elevated n-3 unsaturated fatty acids (n-3 UFAs), such as eicosapentaenoic acid (EPA), alpha-linolenic acid (ALA) and gamma-linolenic acid (GLA). Artificial feeding also resulted in significant increased docosahexaenoic acid (DHA) in MAF muscle than MGF fish, whereas no significance in the comparison of female samples. Metabolic pathway analyses using both targeted and untargeted approaches consistently revealed that the arachidonic acid metabolism and steroid hormone biosynthesis pathways were significantly different between AF and GF groups. Our results suggested that grass is a better source of diet fatty acid and protein when compared to artificial feed. Grass feeding could effectively lower triglycerides in serum, reduce fat accumulation and alter lipid compositions in fish muscle by increasing the concentrations of n-3 UFAs, leading to better nutrition and health.

Abstract:A H-1 NMR-based metabolomics approach was to explore the effect of ultrafiltered fish protein hydrolysate (UF) levels on the liver and muscle metabolic profile of juvenile turbot (Scophthalmus maximus L.). Fish protein hydrolysate (FPH) was produced from by-products by enzymatic treatment, and UF was obtained by diluting FPH followed filtration. Fish were fed diets containing fish meal protein, which was, respectively, replaced by UF protein 0, 50, 100, 150 and 200gkg(-1) of dietary total protein (UF-0, UF-5, UF-10, UF-15 and UF-20) for 68days. OPLS-DA of liver and muscle showed that high levels of UF in diets may lead to corresponding tissue metabolites changes. In liver tissue, changing metabolites included dimethylamine, N,N-dimethylglycine, valine, isoleucine, leucine, arginine and alanine and were mainly involved in choline metabolism and amino acid metabolism. For muscle tissue, changing metabolites included lactate, alanine, proline, fumarate, tyrosine, histidine, cystathionine and taurine. And the metabolites were mainly involved in glycolysis and gluconeogenesis, proline metabolism and taurine metabolism. H-1 NMR-based metabolomics is a useful approach to investigate different levels of UF on metabolic profile in liver and muscle tissues combined with the growth.

Abstract:This study was designed to evaluate changes in the metabolic profile of liver and muscle of turbot (Scophthalmus maximus L.) fed fishmeal-based diet, diets containing size-fractionated fish protein hydrolysate and plant protein-based diet using H-1 NMR-based metabolomics approach combined with the growth. Fish protein hydrolysate (FPH) was obtained by enzymatic treatment, permeate fraction was obtained as UF by ultrafiltered step, and retentate fraction was retained as RF. FM diet contained fish meal used as a single protein source. Four other diets (PP, UF, FPH and RF) contained 180 g kg(-1) diet fish meal. 54, 55 and 55 g kg(-1) dry diet UF, FPH and RF were supplemented to UF, FPH and RF diets. All diets were formulated to be isolipidic and isonitrogenous fed to five triplicate groups of turbot (16.05 +/- 0.03 g) for 68 days. OPLS-DA in FM versus UF, FM versus FPH, FM versus RF and FM versus PP resulted in a reliable model for muscle and liver tissue, while OPLS-DA in UF versus FPH and UF versus RF only showed metabolites changes in liver tissue. Results indicated that metabolite changes among the different treatments were consistent with the growth tendency.
Untargeted foodomics strategy using high-resolution mass spectrometry reveals potential indicators for fish freshness

Abstract:Fish among other food can easily become spoilage. However, the existing literature endeavouring into a holistic investigation of fish metabolome during storage is scarce. There is a need for a powerful tool for more in-depth determinations. The present study is leveraging high-resolution mass spectrometry (HRMS)-based untargeted foodomics in the exploration of potential indicators for fish freshness. Three batches of fish fillets were stored in the refrigerator (+4°C) for 0, 24, 48, and 72 h. Features were detected via UPLC-Q-Orbitrap HRMS and hereby undergone selection, identification, and verification. Eight sensitive indicators with significant time-dependent responses were yielded. The loss of freshness in fish is manifested in the upregulation of uracil, hypoxanthine, and inosine (involved in nucleotide changes) and α-linolenic acid, docosahexaenoic acid, arachidonic acid, and linoleic acid (involved in lipid hydrolysis) as well as in the downregulation of decanoylcarnitine (involved in fatty acid metabolism). Our work provides a promising approach to assess the quality of fish during storage and gain deeper insights into the metabolic reaction.

Abstract:Mechanism on texture change of Wuchang bream (Megalobrama amblycephala) muscle during live transportation was explored by UPLC-QTOF-MS. The shear force (hardness) of fish muscle decreased gradually as the trans-portation time was extended from 3 to 24 h, with a maximum decrease of 43.49 %. With the extension of the time, the cellular and ultrastructure of fish muscle fiber were gradually disrupted. There was a significant dif-ference between metabolites of the samples after transporting for 6, 12, 24 h versus 3 h, with organic acids and their derivatives upregulated while nucleotide metabolites downregulated. The texture deterioration might be related to metabolism of amino acids, purine, histidine, and choline. The results showed that the deterioration of Wuchang bream texture during live transportation might be mainly related to the changes of the above metabolic pathways caused by oxidative stress, which resulting in the destruction of the muscle cells and fibers.

Abstract:Some Eastern Asian countries deem pufferfish - especially its muscle - a culinary delight. Herein, molecular mass fingerprinting of soups prepared by Takifugu flavidus, Takifugu obscurus and Takifugu rubripes was established via matrix-assisted laser desorption/ionization - mass spectrometry (MALDI MS). Soup samples were directly analyzed by MALDI MS to collect mass spectra within 0-700 Da in a quick way, followed by principal component analysis to distinguish the different soups and to find out the distinctive compounds among the soups. High performance liquid chromatography - tandem MS (HPLC-MS/MS) was applied to identify the compounds. Nineteen compounds were identified from the HPLC-MS/MS data by using METLIN database. Through literature mining, we found that these compounds are closely related to the flavor, nutrition, and safety of pufferfish soups. This method can also be used as a facile way to distinguish between different pufferfish fillets when morphological characters have been damaged or destroyed.

Abstract:Spirotetramat, a member of tetronic and tetramic acid derivatives, is a unique insecticide and acaricide. Although the effect on zebrafish embryos lipid biosynthesis of spirotetramat has been characterized, the energy metabolism and toxic effect mechanism warrant further investigation. To investigate the toxic mechanism of spirotetramat on energy metabolism, zebrafish embryos were exposed to 100, 500 and 1000 µg/L of spirotetramat for 4 days. Untargeted metabolomics showed the synthesis and degradation of ketone pathway metabolites (R)-3-Hydroxybutyric acid and Acetoacetate significantly decreased, as well as increasing the abundance of Anti-Acetyl Coenzyme A Carboxylase protein (ACC1). Down-regulation of the genes related to ß-oxidation and the tricarboxylic acid cycle in the embryos show decreased energy metabolism. Carnitine palmitoyltransferase 1 (CPT- I) significantly decreased while citrate synthase (CS) significantly increased. Additionally, mitochondrial lesions in embryos were found using electron microscopy. Our study provides novel and robust perspectives, which show that spirotetramat treatment in embryos leads to metabolic disturbances that adversely affect cellular energy homeostasis.

Comprehensive analysis of transcriptomics and metabolomics to understand the flesh quality regulation of crucian carp (Carassius auratus) treated with short term micro-flowing water system

Abstract:The short term micro-flowing purification system (STMFPS) has been shown to improve the flesh quality of freshwater fish. However, few studies have focused on the involved underlying mechanisms. This study explored the effect of STMFPS on the flesh quality of market-size freshwater fish based on the combination of metabolomics and transcriptomics methods. The UPLC-QTOF/MS based metabolomics method was utilized to screen metabolites and predict the possible major metabolic pathways during different STMFPS treatment periods (0 d, 1 d, 5 d and 9 d). Furthermore, the transcriptomic data demonstrated that the differentially expressed genes detected in crucian carp muscle were 2915, 7852 and 7183 after 1 d, 5 d and 9 d STMFPS treatment. Results showed that the TCA cycle, ornithine cycle, purine metabolism and amino acid catabolism play important roles in improving the flesh quality of crucian carp. This study may help to understand the mechanism of improving the flesh quality of aquatic products using STMFPS.

Abstract:The intensification of aquaculture to help kerb global food security issues has led to the quest for more economical new protein-rich ingredients for the feed-based aquaculture since fishmeal (FM, the ingredient with the finest protein and lipid profile) is losing its acceptability due to high cost and demand. Although very high in protein, castor meal (CM), a by-product after oil-extraction, is disposed-off due to the high presence of toxins. Concurrently, the agro-industrial wastes’ consistent production and disposal are of utmost concern; however, having better nutritional profiles of these wastes can lead to their adoption. This study was conducted to identify potential biomarkers of CM-induced enteritis in juvenile hybrid-grouper ( Epinephelus fuscoguttatus ♀ × Epinephelus lanceolatus ♂) using ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) alongside their growth and distal intestinal (DI) health evaluation. A total of 360 fish (initial weight = 9.13 ± 0.01g) were randomly assigned into three groups, namely, fish-meal (FM) (control), 4% CM (CM4), and 20% CM (CM20). After the 56-days feeding-trial, the DI tissues of FM, CM4, and CM20 groups were collected for metabolomics analysis. Principal components analysis and partial least-squares discriminant-analysis (PLS-DA, used to differentiate the CM20 and CM4, from the FM group with satisfactory explanation and predictive ability) were used to analyze the UPLC-MS data. The results revealed a significant improvement in the growth, DI immune responses and digestive enzyme activities, and DI histological examinations in the CM4 group than the others. Nonetheless, CM20 replacement caused DI physiological damage and enteritis in grouper as shown by AB-PAS staining and scanning electron microscopy examinations, respectively. The most influential metabolites in DI contents identified as the potential biomarkers in the positive and negative modes using the metabolomics UPLC-MS profiles were 28 which included five organoheterocyclic compounds, seven lipids, and lipid-like molecules, seven organic oxygen compounds, two benzenoids, five organic acids and derivatives, one phenylpropanoids and polyketides, and one from nucleosides, nucleotides, and analogues superclass. The present study identified a broad array of DI tissue metabolites that differed between FM and CM diets, which provides a valuable reference for further managing fish intestinal health issues. A replacement level of 4% is recommended based on the growth and immunity of fish.

Rapid target and non-target screening method for determination of emerging organic chemicals in fish

Abstract:An effective analytical method for target and non-target screening of multi-class emerging organic chem-icals in aquaculture fish muscle samples using liquid chromatography coupled with high resolution time -of-flight mass spectrometry (LC-QTOF-MS/MS) was reported. Two sample pretreatment methods (QuECh-ERS and Captiva EMR-Lipid cartridge) were compared and assessed for 151 organic compounds covering a wide range of log K-ow (-1.37-11.51) in freeze-dried fish samples. Captiva EMR-Lipid cartridge outper-formed QuEChERS by high detection frequency (> 87.5%) and effective matrix removal with satisfactory standard deviation of the compounds (< 20%), and meanwhile provided acceptable recoveries for most organic chemicals at three spiking concentration levels (10, 50, and 100 ng g(-1)). Spiking experiments suggested that the developed non-target screening workflow showed further convincible identification results with over 83.3% of the chemicals confirmed at even low spiking levels. A list of 867 organic chem-icals were tentatively identified in fish samples collected from fish ponds with 21 of them classified as high confidence levels (L1 and L2). The results showed that the simplified analytical strategy can be ap-plied for the quantification and screening analysis of a broad range of emerging organic chemicals in fish samples. Abstract:Wild large yellow croakers are generally considered superior to farmed large yellow croakers in quality and taste. The metabolites of large yellow croakers vary because of different growth environments and dietary conditions, allowing easy differentiation of wild large yellow croakers from farmed large yellow croakers. The adulteration of wild large yellow croakers using farmed large yellow croakers in the market, which is driven by economic interests, disturbs market balance and negatively affects the trade in large yellow croakers. Therefore, the quality control (QC) of wild large yellow croakers is important for the maintenance of the market and the trade development of aquatic products. Non-targeted metabolomics based on liquid chromatography–mass spectrometry was employed in this study to clarify the differences in metabolic composition between muscle tissues of wild and farmed large yellow croakers. Results showed that the partial least squares discriminant analysis model could effectively distinguish the wild and farmed large yellow croakers without overfitting. KEGG enrichment analysis showed that the main metabolism pathways were closely associated with lipid, amino acid, carbohydrate, and vitamin-related metabolism. Overall, non-targeted metabolomics can identify and differentiate wild and farmed large yellow croakers, supporting new insights to improve the QC level of aquatic products.

Abstract:A novel and accurate liquid chromatography-tandem mass spectrometry method was developed to sequentially determine three persistent herbicides (atrazine (ATZ), acetochlor (ACE), and metolachlor (MET)) and seven characteristic metabolites (desethylatrazine (DEA), deisopropylatrazine (DIA), diaminochlorotriazine (DACT), MET-oxanilic acid (MET-OA), MET-ethanesulfonic acid (MET-ESA), ACE-ESA, and ACE-OA) in fresh fish tissues from six fish species. A modified QuEChERS method was conducted to extract the target compounds from fish tissues. Matrix-matched calibrations of the target analytes were carried out at spiking levels of 1, 10, 100, and 1000 ng g(-1). The method was validated in accordance with Codex guidelines (CAC/GL 71-2009). Recoveries for the target analytes were 67-120% with relative standard deviations below 20%, and the matrix effects ranged from -58.7% to 59.3%. The limits of detection and quantitation were 0.01-1.90 and 0.02-6.35 ng g(-1), respectively. Moreover, the method was successfully applied to analyze the concentrations of the target chemicals in fresh tissue samples of six fish species (n = 67) collected from four markets in Nanning City, Guangxi Province, China. The concentrations in all samples were 1.1-140.5 ng g(-1). Interestingly, this study was the first to measure DEA and DIA in fish liver, and their highest concentrations were 10.7 and 14.2 ng g(-1), respectively. This method provides a basis for studying the pathways of biotransformation, bioaccumulation, detoxification, and exposure patterns of ACE, ATZ, MET, and their metabolites in aquatic environments.
Metaproteomic Analysis of Microbial Communities Affecting Fishery Products

Abstract:Fishery products are one of the main human nutritional sources, and due to the consumption increase, the quality of the derived products may be modified, during catching, technological processing, and storage. Detection and identification of pathogenic and spoilage microorganisms in fishery products is needed because the first may be involved in human diseases, while the second is responsible of significant economic losses. In this sense, liquid chromatography-electrospray ionization-tandem mass spectrometry (LC-ESI-MS/MS) method and computational analysis of MS data are useful tools for characterizing and identifying different microorganisms and to develop promising strategies for food science investigations. Moreover, in the past decade, metaproteomic methodologies have progressed for the study of microorganisms isolated from their natural samples and independently of the culture restrictions. Metaproteomics enables assessment of proteins and pathways from individual members of the consortium. Metaproteomics can provide a detailed understanding of which organisms occupy specific metabolic niches, how they interact, and how they utilize nutrients, and these insights can be obtained directly from environmental samples.According to that, the sample preparation of the fishery product, the LC-ESI-MS/MS dedicated method, and the MS data analysis were described in the present chapter to obtain the metaproteomic analysis of the respective microbiomes or microbial communities.
Changes in metabolites in the liver of zebrafish under sublethal halosulfuron-methyl stress

Abstract:We applied a sub-lethal dose of halosulfuron-methyl stress to zebrafish in order to study the changes in metabolites in the liver.Ultra-high performance liquid chromatography coupled with Q Exactive hybrid quadrupole-orbitrap mass spectrometry (UPLC-MS/MS) was applied to analyze the effects of the sub-lethal dose of halosulfuron-methyl on metabolites in the liver of zebrafish.Each chromatogram obtained from UPLC-MS/MS analysis was pre-processed using SIEVE software to generate discrete peak lists containing m/z,retention time,and relative peak area.Using the mzCloud and Human Metabolome Database,33 metabolites were identified including amino acids,sugars,and organic acids.The pre-processed UPLC-MS/MS data were exported to SIMICA-P 13.0 for analysis by using multivariate statistical methods.Three multivariate analysis methods were used to determine the optimal separation of clusters:principal component analysis,partial least squares-discriminant analysis,and orthogonal projection to latent structure-discriminant analysis.The results revealed significant differences (p＜0.01) between the control and treatment groups.A representative S-plot showed the variables responsible for the separation.Creatinine,taurine,dopamine,and 5b-cyprinol sulfate appeared to be the most affecting variables (p＜ 0.01).These four metabolites were identified as potential indicators of halosulfuron-methyl-induced liver abnormalities.
Comprehensive metabolomic profiling of nutrients in fish and shrimp

Abstract:Although fish and shrimp are commonly consumed in daily life and recognized as nutrient-rich species, global profiling of the endogenous nutrients in these species is lacking. Here, we optimized the sample preparation and data acquisition methods of metabolomics to comprehensively characterize the nutrients in selected fish and shrimp species and compared them with those in beef, leading to the identification of 71 differentially expressed metabolites. Of these, docosahexaenoic acid, taurine, choline and (lyso)phosphatidylcholines were found to be abundant in the examined fish species, while several nonessential amino acids were rich in the analyzed shrimp samples. Subsequently, the biological functions of the metabolites rich in shrimp were queried. Intriguingly, the examined nutrients exemplified by proline can significantly mitigate the lipopolysaccharide (LPS)-stimulated inflammatory responses in BV2 cells and RAW264.7 cells. Collectively, our findings imply that the dietary intake of certain fish and shrimp species may benefit human health through alleviating inflammatory responses.
