Abstract:Host-guest chemistry involves the binding of a substrate molecule (guest) to a receptor molecule (host). Various molecules, including crown ethers, cryptands, cyclophanes, calixarenes, cyclodextrins, and so on, have been used as molecular hosts. However, only limited small molecules or simple ions can be encapsulated in these hosts. Fortunately, as a class of unique host molecules, hyperbranched polymers (HBPs) can bind to numerous guests through topological entrapment, electrostatic bonding, hydrogen bonding or hydrophobic interactions in the core, at the branching points or at the periphery. Hence, hyperbranched polymeric hosts have received increasing attention in the past few decades because of their specific and unique properties. This review briefly summarizes these unique properties related to HBPs serving as hosts. In addition, HBP-based host-guest systems will be classified according to the types of guests encapsulated. Besides, the corresponding applications will be presented as well. We hope to motivate an increased understanding of molecular recognition in HBPs, and further facilitate the optimization of future host-guest systems.
Host-Guest Supramolecular Chemistry of Dendritic Macromolecules

Abstract:Dendritic macromolecules are three-dimentional polymers with branch-on-branch topology of which dendrimers and hyperbranched polymers (HPs) are the two most researched subclasses. This paper reviews the recent progress of host-guest supramolecular chemistry in terms of various HPs and dendrimers. Because of the highly branched structure and the presence of internal space and binding sites, dendritic macromolecule can serve as host to encapsulate multiple guests, just like a highly branched "tree" accommodating multiple "bird's nests". Core-shell structure made by functionalization of dendritic polymer core with shielding shell of opposite polarity is usually needed to obtain a stable host-guest complex. Their encapsulation capacities are dependent on various factors, such as core size or molecular weight, polarity difference between core and shell, interaction strength between core and guests, shell density, and so on. Dendritic polymer-based host-guest encapsulation can be applied in many fields including phase-transfer, noncovalent functionalization, metal nanoparticle growth, and biological imaging.

Abstract:CONSPECTUS: In host-guest chemistry, a larger host molecule selectively and noncovalently binds to a smaller guest molecule or ion. Early studies of host-guest chemistry focused on the recognition of spherical metal or ammonium ions by macrocyclic hosts, such as cyclic crown ethers. In these systems, preorganization enables their binding sites to cooperatively contact and attract a guest. Although some open-chain crown ether analogues possess similar, but generally lower, binding affinities, the design of acyclic molecular recognition hosts has remained challenging. One of the most successful examples was rigid molecular tweezers, acyclic covalently bonded preorganized host molecules with open cavities that bind tightly as they stiffen. Depending on the length of the atomic backbones, hydrogen bonding-driven aromatic amide foldamers can form open or closed cavities. Through rational design of the backbones and the introduction of added functional groups, researchers can regulate the shape and size of the cavity. The directionality of hydrogen bonding and the inherent rigidity of aromatic amide units allow researchers to predict both the shape and size of the cavity of an aromatic amide foldamer. Therefore, researchers can then design guest molecules with structure that matches the cavity shape, size, and binding sites of the foldamer host. In addition, because hydrogen bonds are dynamic, researchers can design structures that can adapt to outside stimuli to produce responsive supramolecular architectures. In this Account, we discuss how aromatic amide and hydrazide foldamers induced by hydrogen bonding can produce responsive host-guest systems, based on research by our group and others. First we highlight the helical chirality induced as binding occurs in solution, which includes the induction of helicity by chiral guests in oligomeric and polymeric foldamers, the formation of diastereomeric complexes between chiral foldamer hosts and guests, and the induction of helical chirality by chiral guests into inherently flexible backbones. In addition, molecular or ion-pair guests can produce supramolecular helical chirality in the organogel state. Such structures exhibit remarkable time-dependence and a "Sergeants and Soldiers" effect that are not observed for other two-component organogels that have been reported. We further illustrate that the reversible folding behavior of an aromatic amide foldamer segment can modulate the switching behavior of donor-acceptor interaction-based [2]rotaxanes. Finally we show that a folded oligomer can induce folding in one or two attached intrinsically flexible oligomers, an example of a solvent-responsive intramolecular host-guest system.

Abstract:Hyperbranched polymers (HBPs), an important subclass of dendritic macromolecules, are highly branched, three-dimensional globular nanopolymeric architectures. Attractive features like highly branched topological structures, adequate spatial cavities, numerous terminal functional groups and convenient synthetic procedures distinguish them from the available polymers (the linear, branched, and crosslinking polymers). Due to their unique physical/chemical properties, applications of HBPs have been explored in a large variety of fields. In particular, HBPs exhibit unique advantages in the biological and biomedical systems and devices. Firstly, the way to prepare HBPs usually only involves simple one-pot reactions and avoids the complicated synthesis and purification procedures, which makes the manufacturing process more convenient, thus reducing production costs. Secondly, the large number of end-groups of HBPs provides a platform for conjugation of the functional moieties and can also be employed to tailor-make the properties of HBPs, enhancing their versatility in biological applications. Thirdly, HBPs possess excellent biocompatibility and biodegradability, controlled responsive nature, and ability to incorporate a multiple array of guest molecules through covalent or noncovalent approaches. All of these features of HBPs are of great significance for designing and producing biomaterials. To date, significant progress has been made for the HBPs in solving some of the fundamental and technical questions toward their bioapplications. The present review highlights the contribution of HBPs to biological and biomedical fields with intent to aid the researchers in exploring HBPs for bioapplications.

Abstract:Highly stable inclusion complexes between cyclodextrin (CD) and guest molecules form due to their host-guest interaction. Polymer systems based on the complexes have been extensively reported in recent years. In this review, we highlight some recent advances from polymer systems with adamantane (Ada), azobenzene (Azo), ferrocene (Fc) and so on as guest molecules in terms of their synthesis, properties and functions. Various polymer topologies ranging from linear, branched, comb-like and hyper-branched are discussed in detail. Supra-Amphiphiles Based on Host–Guest Interactions

Abstract:The study of host–guest interactions was inspired by molecular recognition in biological processes. Since the discovery of crown ether in the 1960s, host–guest interactions have attracted much attention from chemists interested in supramolecular chemistry. Increasing numbers of macrocyclic “host” molecules have been synthesized and investigated and many self-assembled and functional materials have been constructed based on host–guest interactions. Host–guest interactions can also be used to fabricate supra-amphiphiles. This chapter gives a brief introduction to the development of macrocyclic molecules for host–guest interactions and then illustrates in detail the supra-amphiphiles fabricated on the basis of these macrocyclic molecules, including their supramolecular architectures, self-assembly, structures and functions.

Abstract:Supramolecular polymers constructed by orthogonal self-assembly based on host-guest and metal-ligand interactions are attracting increasing attention currently because of their interesting properties and potential applications. Host-guest interactions impart these polymers with good selectivity and convenient enviro-responsiveness, and metal-ligand interactions endow them with various coordination geometries, strong yet tunable coordination binding abilities, as well as magnetic, redox, photophysical, and electrochromic properties. Therefore, supramolecular polymers constructed by orthogonal host-guest and metal-ligand interactions have wide applications in the fields of soft matter, fluorescence sensing, heterocatalysis, electronics, gas storage, etc. In this critical review, we will address the recent development of supramolecular polymeric systems involving metal-ligand interactions and host-guest molecular recognition. Specifically, we classify the related supramolecular polymers depending on the types of macrocyclic hosts, and highlight their intriguing properties originating from the elegant combination of host-guest complexation and metal centers.

Abstract:A novel supramolecular hyperbranched polymer had been fabricated conveniently through host-guest recognition based on Cucurbit[8]uril in aqueous solution. The three branched monomer contained an azobenzene unit and a viologen moiety on each of its branch. The polymerization of the supramolecular polymer in water could be controlled by tuning the isomer ratio of azobenzene groups of the guest monomers upon the competitive irradiation of lights. In addition, the strictness of the structure could be adjusted by varying the molar ratio between host and guest monomers. Planar supramolecular network had been constructed by drying the solvent of the solution of supramolecular hyperbranched polymers on smooth plane. The research provided a promising way to conduct dual-responsive supramolecular polymerization.

Abstract:CONSPECTUS: All living organisms and soft matter are intrinsically responsive and adaptive to external stimuli. Inspired by this fact, tremendous effort aiming to emulate subtle responsive features exhibited by nature has spurred the invention of a diverse range of responsive polymeric materials. Conventional stimuli-responsive polymers are constructed via covalent bonds and can undergo reversible or irreversible changes in chemical structures, physicochemical properties, or both in response to a variety of external stimuli. They have been imparted with a variety of emerging applications including drug and gene delivery, optical sensing and imaging, diagnostics and therapies, smart coatings and textiles, and tissue engineering. On the other hand, in comparison with molecular chemistry held by covalent bonds, supramolecular chemistry built on weak and reversible noncovalent interactions has emerged as a powerful and versatile strategy for materials fabrication due to its facile accessibility, extraordinary reversibility and adaptivity, and potent applications in diverse fields. Typically involving more than one type of noncovalent interactions (e.g., hydrogen bonding, metal coordination, hydrophobic association, electrostatic interactions, van der Waals forces, and π-π stacking), host-guest recognition refers to the formation of supramolecular inclusion complexes between two or more entities connected together in a highly controlled and cooperative manner. The inherently reversible and adaptive nature of host-guest molecular recognition chemistry, stemming from multiple noncovalent interactions, has opened up a new platform to construct novel types of stimuli-responsive materials. The introduction of host-guest chemistry not only enriches the realm of responsive materials but also confers them with promising new applications. Most intriguingly, the integration of responsive polymer building blocks with host-guest recognition motifs will endow the former with further broadened responsiveness to external stimuli and accordingly more sophisticated functions. In this Account, we summarize recent progress in the field of responsive polymeric materials containing host-guest recognition motifs with selected examples and highlight their versatile functional applications, whereas small molecule-oriented host-guest supramolecular systems are excluded. We demonstrate how the introduction of host-guest chemistry into conventional polymer systems can modulate their responsive modes to external stimuli. Moreover, the responsive specificity and selectivity of polymeric systems can also be inherited from the host-guest recognition motifs, and these features provide extra advantages in terms of function integration. The following discussions are categorized in terms of design and functions, namely, host-guest chemistry toward the fabrication of responsive polymers and assemblies, optical sensing and imaging, drug and gene delivery, and self-healing materials. A concluding remark on future developments is also presented. We wish this prosperous field would incur more original and evolutionary ideas and benefit fundamental research and our daily life in a more convenient way.

Abstract:Covalent polymers connected by non-covalent interactions constitute a fascinating set of materials known as supramolecular polymer networks (SPNs). A key feature of SPNs is that the underlying covalent polymers endow the resulting self-assembled materials with features, such as structural and mechanical integrity, good processability, recyclability, stimuli-responsiveness, self-healing, and shape memory, that are not recapitulated in the case of classic covalent polymer systems. The unique nature of SPNs derives from the controlled marriage of traditional covalent polymers and macrocycle-based host-guest interactions. As a consequence, supramolecular polymeric networks have played important roles in a number of diverse fields, including polymer science, supramolecular chemistry, materials science, biomedical materials, and information storage technology. In this Review, we summarize advances made in the area of functional SPNs, with a focus on original literature reports appearing in the past five years. The treatment is organized according to the key macrocycle-based host-guest interactions used to produce various SPNs. The role of the underlying polymer backbones is also discussed.

Abstract:Supramolecular polymers based on host-guest molecular recognition have emerged as promising platforms for the development of smart materials. However, the studies on them are primarily conducted in solution and/or in the gel state. In contrast, little is known about dynamic properties and applications of supramolecular polymers in bulk. Herein, we present a self-cross-linking supramolecular polymer network (SPN) as a model system to understand the bulk properties controlled by noncovalent interactions. Specifically, the SPN monomer is composed of two benzo-21-crown-7 (B21C7) host units and two dialkylammonium salt guest moieties on a four-arm core, wherein complementary host-guest complexation drives the formation of the SPN with [2]pseudorotaxane linkages between B21C7 and ammonium motifs. The dynamic and reversible behaviors of the linkages are evaluated by measurement of viscoelasticity. The results indicate that the host-guest molecular recognition becomes highly dynamic at elevated temperature. Moreover, the relatively high activation energy of the SPN manifests itself as a new type of thermoplastic material with network topology freezing glass transition. Finally, we demonstrate how these findings provide insights into the malleability and processability of the SPN by simple demos. The fundamental understanding gained from the research on this SPN in bulk will facilitate the advancement and application of supramolecular materials.

Abstract:Supramolecular polymers constructed by orthogonal self-assembly based on multiple hydrogen bonding and macrocyclic host-guest interactions have received increasing attention due to their elegant structures, outstanding properties, and potential applications. Hydrogen bonding endows these supramolecular polymers with good adaptability and reversibility, while macrocyclic host-guest interactions give them good selectivity and versatile stimuli-responsiveness. Abstract:Nature uses the power of non-covalent interactions as the basis for many kinds of adhesion phenomena. Inspired by nature, scientists have prepared various synthetic adhesive materials that rely on a number of non-covalent interactions at the interfaces. Commonly used non-covalent interactions include hydrogen bonding, - stacking, charge transfer interactions, electrostatic interactions, hydrophobic interactions, macrocycle-based host-guest interactions, among others. Within this context, macrocycle-based host-guest interactions are of particular interest. Often they give rise to distinct properties, such as multiple combined noncovalent interactions and a diversity of stimuli-based responsiveness. In this tutorial review, we will summarise recent advances in adhesive supramolecular polymeric materials that rely primarily on macrocycle-based host-guest interactions. An overview of future challenges and a perspective of this sub-field are also provided.

Abstract:Supramolecular chemistry involving macrocyclic hosts is a highly interdisciplinary and fast-growing research field in chemistry, biochemistry, and materials science. Host-guest based supramolecular assemblies, as constructed through non-covalent interactions, are highly dynamic in nature, and can be tuned easily using their responses to various external stimuli, providing a convenient approach to achieve excellent functional materials. Macrocyclic hosts, particularly cyclodextrins, cucurbit[n]urils, and calix[n]arenes, which have unique features like possessing hydrophobic cavities of different sizes, along with hydrophilic external surfaces, which are also amenable towards easy derivatizations, are versatile cavitands or host molecules to encapsulate diverse guest molecules to form stable host-guest complexes with many unique structures and properties. Interestingly, host-guest complexes possessing amphiphilic properties can easily lead to the formation of various advanced supramolecular assemblies, like pseudorotaxanes, rotaxanes, polyrotaxanes, supramolecular polymers, micelles, vesicles, supramolecular nanostructures, and so on. Moreover, these supramolecular assemblies, with varied morphologies and responsiveness towards external stimuli, have immense potential for applications in nanotechnology, materials science, biosensors, drug delivery, analytical chemistry and biomedical sciences. In this perspective, we present a stimulating overview, discussing simple host-guest systems to complex supramolecular assemblies in a systematic manner, aiming to encourage future researchers in this fascinating area of supramolecular chemistry to develop advanced supramolecular materials with superior functionalities, for their deployment in diverse applied areas.

Extending host–guest chemistry from solution to the solid state results in complexes that are generally more robust, enabling easier processing and broadened applications. Macrocyclic compounds in the solid state can encapsulate guests with larger affinities than their soluble counterparts. This is crucial for use in applications such as separation science and devices. In this Review, we summarize recent progress in macrocycle-based solid-state host–guest chemistry and discuss the basic physical chemistry of these complexes. Representative macrocycles and their solid-state complexes are explored, as well as potential applications. Finally, perspectives and challenges are discussed.

Abstract:Supramolecular nanostructures assembled by polymeric amphiphiles have been intensively studied during the last two decades. Such nanocarriers may be engineered to possess on-demand bio-responsitivity for the prevention, diagnosis, and treatment of human diseases. The successful development of several nanoassembly-based polymer therapeutics further encouraged scientists to develop nano-vehicles to achieve controlled release, enhanced efficacy, improved specificity and reduced toxicity. Different from the abundant existing literatures on the hydrophobically or electrostatically driven self-assemblies and their therapeutic applications, this article reviews host-guest interaction mediated nanoassemblies, especially those constructed using cyclodextrins as the host entities. The excellent biocompatibility, complexation capacity, and chemical-sensitivity of cyclodextrin make cyclodextrin-containing polymers attractive to construct host-guest nanoassemblies. Such nanocarriers may be advantageous also because of the broad availability of cyclodextrins, their flexibility for structure/property modulation and their chemical-responsive characteristics.
