Abstract:A facile and efficient transformation of arylboronic acids to their corresponding aryl thiocyanates has been successfully developed. Based on the CuCl-catalyzed oxidative cross-coupling reaction between arylboronic acids and trimethylsilylisothiocyanate (TMSNCS) under oxygen atmosphere, the transformation can be readily conducted at ambient temperature. The newly-developed protocol provided a competitive synthetic approach to aryl thiocyanates that can tolerate a broad range of reactive functional groups and/or strong electron-withdrawing groups.

Abstract:An operationally simple and inexpensive catalyst system was developed for the cross coupling of potassium thiocyanate with aryl iodides by using CuI as catalyst, 1, 10-phenanthroline as ligand, and tetraethylammonium iodide as activator. The procedure is applicable for the synthesis of diverse aryl thiocyanates without any exotic, poisonous reagents.

Abstract:Aryl thiocyanate compounds play a crucial role as building blocks in the synthesis of bioactive compounds and intermediates for various functional groups. Here, we developed a highly efficient and novel method for the direct ortho-Csp2–H thiocyanation of free anilines. This method employs copper acetate as a catalyst, stable and non-toxic ammonium thiocyanate as a thiocyanation source, and tert-butyl hydroperoxide as the oxidant, enabling the synthesis of ortho-thiocyanated anilines with good yields and broad substrate tolerance. This approach offers advantages such as short reaction time, and the use of inexpensive and readily available starting materials. Hitherto, no reports have been found in the literature for the ortho-thiocyanation of aromatic amines, making this reaction an important breakthrough in synthetic organic chemistry.

Abstract:We disclose a copper-photocatalyzed strategy that enables selective access to organic thiocyanates and isothiocyanates using benzylic thiocyanates as ATRA reagents. The electronic nature of the aromatic system is a crucial factor defining the outcome of the reaction for forging either the kinetic C–S or the thermodynamic C–N bonds. Mechanistic investigations support a radical pathway initiated by a reductive C–S bond cleavage of the substrates followed by a divergent inner-/outer-sphere interaction with copper depending on the electronic density of the formed intermediates. The developed protocol proceeds with high regio- and chemoselectivity and can also be applied for late-stage functionalization of bioactive molecules. The utility of the products is highlighted by their facile conversion to several building blocks that are relevant to organic synthesis.
