Abstract:A novel [3 + 2] annulation between para-quinone methides and vinylcyclopropanes for the synthesis of spiro[4.5]deca-6,9-diene-8-ones has been described. The palladium and phosphine-thiourea cooperative catalysis system played an important role in high yields and diastereoselectivities. The reaction exhibited good functional group tolerance and scalability.

Abstract:A novel tandem 1,6-addition/cyclopropanation/rearrangement reaction of vinylogous para -quinone methides with 3-chlorooxindoles has been developed, providing dispirooxindole–cyclopentane–cyclohexadienones with vicinal quaternary carbon centers. A novel tandem 1,6-addition/cyclopropanation/rearrangement reaction of vinylogous para -quinone methides with 3-chlorooxindoles is reported. This method provides an efficient approach to various functionalized dispirooxindole–cyclopentane–cyclohexadienones with vicinal quaternary carbon centers. Computational studies have been conducted to provide insight into the origin of diastereoselectivity and predict a stepwise diradical mechanism. The anti-tumor activities of these structurally unique dispirooxindole derivatives are also reported.

Abstract:para‐TsOH‐promoted cascade reaction of easily prepared ortho‐propynol phenyl azides for the synthesis of 4‐methoxy quinolines has been developed. This reaction proceeded in a new way to generate 4‐methoxy quinolines through a concerted nucleophilic mechanism of propargyl methyl ether. It is worth noting that propargyl methyl ether can be successfully transformed into 4‐methoxy quinolines only when substrates contain a strong electron‐donating group (OR).

Abstract:Here, we present an unprecedented formal [3 + 2] cycloaddition of para-quinone methides with vinyl epoxides/cyclopropanes to deliver a wide range of spiro[4.5]decanes in high efficiency and stereoselectivity. The commercial availability of the catalysts and reagents, together with the convenient procedure, makes it an attractive method in asymmetric synthesis.

Abstract:A wide range of isoquinoline ring-modified Quinap oxides with different steric and electronic variations have been constructed through a palladium-catalyzed imidoylative cyclization of arylethenyl isocyanides with 2-diphenylphosphinyl-1-naphthyl bromides. The Pd-catalysis plays dual roles in the formation of both axial C–C bond and isoquinoline ring. This de novo synthetic strategy features good functional group tolerance, high yields and easy scale-up, providing Quinap derivatives with substitution patterns that could not be obtained using coupling reactions. Chiral ligands 7 and 12 can be readily prepared by transformation of the resulting Quinap oxide to their BINOL esters, and have been proven to be superb chiral ligands for the copper-catalyzed enantioselective A3-coupling and alkynylation of quinoline reactions. In general, the enantioselectivies obtained using ligands 7 and 12 are excellent, and the ee values are higher than those using Quinap as ligand, even three times higher in some cases. Mechanism studies revealed that a monomeric copper(I) complex bearing a single chiral ligand was formed and served as the catalytically active species.

Abstract:A chiral squaramide catalyzed approach constructing spiro-3,4-dihydrocoumarin motif by the enantioselective 1,6-addition/acetalization reactions of 1-oxotetralin-2-carbaldehydes and ortho-hydroxyphenyl-substituted para-quinone methides followed by an oxidation was developed. The reactions proceeded smoothly with a wide range of p-QMs and 1-oxotetralin-2-carbaldehydes to generate corresponding products in high yields with excellent diastereoselectivities (>19:1 dr) and enantioselectivities (up to 99% ee).

Abstract:A palladium-catalyzed divergent synthesis of spiro-cyclohexadienone with a five- or a six-membered ring by a cycloaddition reaction of vinylidenecyclopropane-diesters (VDCP-diesters) with p-quinone methides (p-QMs) was disclosed. This protocol features a switchable process between [3+2] and [4+2] cycloadditions tuned by subtle choice of the phosphine ligand. The substrate scopes have been investigated and the reaction mechanism is clarified by mechanistic studies; and DFT calculations also reveal that the coordination modes of ligands with substrates and the bite angle of ligands play critical roles for the product regioselectivity.

Abstract:The para-quinone methides (p-QMs) are activated by Lewis acid and then attacked by diazo compounds. The following rearrangement leads to nitrogen gas extrusion and C-C double bond formation to constitute a metathesis reaction process. Therefore, the diazoester is transformed into tetrasubstituted alkenes, whereas the diazo-oxindole delivers the quinolinone products. Furthermore, the 13C-labeling experiments were also conducted to elucidate a possible mechanism.

Abstract:Photoinduced reactions of bicyclopropylidene (BCP) with para-quinones (p-quinones) including benzoquinones, naphthoquinones, and anthraquinones were found to proceed via different cycloaddition pathways and lead to diverse polycyclic products bearing spiropropyl moiety. Photocycloaddition of BCP with benzoquinones gave spirooxetanes as the primary products, which upon irradiation were able to rearrange into the spiro[4.5]deca-6,9-diene-2,8-diones as secondary photoproducts. Chemoselectivity of the photocycloaddition of BCP with naphthoquinones relies largely on the substitution groups linked to the C═C in between the two carbonyl groups to give different types of products. Photoreaction of BCP with 9,10-anthraquinone gave not only the spirooxetane product, but also a novel spiro[indan-1,1'-phthalan]-3'-one product whose formation might be initiated by a transannular attack of the C4 cyclopropyl radical to the para-carbonyl group. Mechanisms involved in the formation of diverse primary or secondary products in the photoreactions of BCP with p-quinones were proposed. Some of the photoreactions also hold potentials as useful synthetic protocols for important spiropolycyclic compounds such as sesquiterpenes.

Abstract:A DBU‐promoted [3+2] cyclization/retro‐Mannich cascade reaction of N‐aminoisoquinolinium and N‐aminoquinolinium derivatives with para‐quinone methides has been established, employing a C=C double bond cleavage. A broad range of N‐aminoquinolinium salts, N‐aminoisoquinolinium salts, and para‐quinone methides are well tolerated, providing the corresponding rearrangement products. Moreover, scaled‐up reactions and diverse derivatizations of the products were also investigated and discussed.

Abstract:An asymmetric 1,6-conjugate addition reaction of dialkyl diazomethylphosphonates to para-quinone methides promoted by phase-transfer catalysis has been developed. A series of chiral diarylmethylated diazomethylphosphonates were accessed with up to 85% yields and 99% ee enantioselectivities. The resulting products were further transformed into bioactive compounds, namely, a chiral dihydrocinnoline phosphonate and a chiral α-aminophosphonate, bearing diarylmethine stereogenic centers.

Abstract:A three-component reaction of cyclobutanone oxime esters, DABCO·(SO2)2 and N-alkyl-N-methacryloyl benzamides is described. This reaction proceeds without the addition of any oxidant or transition metal, affording sulfonyl-containing isoquinoline-1,3-(2H,4H)-diones in moderate to good yields. Various functional groups are tolerated well in this transformation. Mechanistic studies suggest that a radical pathway is involved, including β-scission, sulfur dioxide insertion, and intramolecular cyclization processes.

Abstract:Reported in this work is a water triggered chemo-divergent enantioselective spiro-annulation and cascade reaction of ortho-quinone methides (o-QMs) with alpha-thiocyanato indanones catalyzed by a chiral organic base. In the case of spiro-annulation, the use of trace amount of water as additive is critical to achieve high enantioselectivity (up to 96 % ee). We found that a cascade reaction was enabled by just tuning the ratio of water in solvent. Accordingly, two new highly efficient asymmetric reactions for the divergent synthesis of spiro- and fused-indanone scaffolds with excellent enantioselectivities (up to 99 % ee) were developed. Mechanistic investigations suggest that interfacial hydrogen bonding may play an important role in achieving the switchable reaction pathways.

Abstract:An asymmetric (4 + 1) annulation of 3‐diazooxindoles/4‐diazooxisoquinolines with para‐quinone methides, catalyzed by a chiral phosphoric acid, has been described. A wide range of spiro[dihydrobenzofuran‐2,3'‐oxindoles/2,4'‐oxisoquinoline] derivatives were afforded with excellent diastereo‐ and enantioselectivities. In this study, the possible reaction pathway was proposed and the synthetic applications were shown by a tenfold scale‐up conversion as well as the further transformations into other structurally more complex spirocyclic compounds. The significance of this protocol is highlighted by its metal‐free participation with heterocyclic diazo compounds as the direct nucleophile and extremely high efficiency in a straightforward and mild reaction process to access the structurally‐diverse spiro‐heterocyclic 2,3‐dihydrobenzofuran derivatives with good to excellent stereocontrol.
