Abstract:A green and fully enzymatic procedure for the resolution of chiral alcohols through lipase/esterase-catalyzed acylation and subsequent lipase-catalyzed aminolysis using anhydrous ammonia was demonstrated. Both enantiomers can be obtained in high ee values (up to >99%) under ambient reaction conditions. The solvent and acyl donors can be recycled, and the enzyme can be reused for up to five times.
Green and Enantioselective Synthesis via Cascade Biotransformations: From Simple Racemic Substrates to High-Value Chiral Chemicals

Abstract:Asymmetric synthesis of chiral chemicals in high enantiomeric excess (ee) is pivotal to the pharmaceutical industry, but classic chemistry usually requires multi-step reactions, harsh conditions, and expensive chiral ligands, and sometimes suffers from unsatisfactory enantioselectivity. Enzymatic catalysis is a much greener and more enantioselective alternative, and cascade biotransformations with multi-step reactions can be performed in one pot to avoid costly intermediate isolation and minimise waste generation. One of the most attractive applications of enzymatic cascade transformations is to convert easily available simple racemic substrates into valuable functionalised chiral chemicals in high yields and ee. Here, we review the three general strategies to build up such cascade biotransformations, including enantioconvergent reaction, dynamic kinetic resolution, and destruction-and-reinstallation of chirality. Examples of cascade transformations using racemic substrates such as racemic epoxides, alcohols, hydroxy acids, etc. to produce the chiral amino alcohols, hydroxy acids, amines, and amino acids are given. The product concentration, ee, and yield, scalability, and substrate scope of these enzymatic cascades are critically reviewed. To further improve the efficiency and practical applicability of the cascades, enzyme engineering to enhance catalytic activities of the key enzymes using the latest microfluidics-based ultrahigh-throughput screening and artificial intelligence-guided directed evolution could be a useful approach.

Abstract:Chiral amines are key building blocks for the development of numerous bioactive compounds. In this study, we developed a concurrent chemoenzymatic cascade approach using ω-transaminase for the isomeric configuration inversion of a racemic amine mixture. One isomer was transaminated using ω-transaminase, generating coproduct ketones and an additional chiral substance. Then, the mixture underwent selective reductive amination of a ketone using a specially designed compatible nickel-based nanocatalyst, which transformed coproduct ketone to racemic amines while leaving the opposite enantiomer unchanged. The combination of the two steps in one reaction system functions as an overall isomeric configuration inversion system. Moreover, the desired chiral amines with an additional chiral substance were formed. The procedure consumed NH3 and generated H2O as the sole byproduct.

Abstract:Optically active β‐amino alcohols are very useful chiral intermediates frequently used in the preparation of pharmaceutically active substances. Herein, a novel cyclohexylamine oxidase (ArCHAO) was identified from the genome sequence of Arthrobacter sp. TYUT010‐15 with the ( R )‐stereoselective deamination activity of β‐amino alcohol. ArCHAO was cloned and successfully expressed in E. coli BL21, purified and characterized. Substrate specific analysis revealed that ArCHAO has high activity (4.15 to 6.34 U mg ‐1 protein) and excellent enantioselectivity toward the tested β‐amino alcohols. Using the purified ArCHAO, a wide range of racemic β‐amino alcohols were resolved, ( S )‐β‐amino alcohols were obtained in >99% ee. Deracemization of racemic β‐amino alcohols was conducted by ArCHAO catalyzed enantioselective deamination and transaminase catalyzed enantioselective amination, affording ( S )‐β‐amino alcohols in excellent conversion (78‐94%) and enantiomeric excess value (>99%). Preparative scale deracemization was carried out with 50 mM (6.859 g L ‒1 ) racemic 2‐amino‐2‐phenylethanol, ( S )‐2‐amino‐2‐phenylethanol was obtained in 75% isolated yield and >99% ee .

Abstract:Chiral amines are common drug building blocks and important active pharmaceutical ingredients. Preparing these functionalized compounds from simple materials, such as alkanes, is of great interest. We recently developed an artificial bioamination cascade for the C–H amination of cyclic alkanes by combining P450 monooxygenase, alcohol dehydrogenase, and amine dehydrogenase. Herein, this system has been extended to the synthesis of chiral aromatic amines. In the first hydroxylation step, process optimization increased the conversion to 77%. Two stereoselective ly complementary alcohol dehydrogenases and an amine dehydrogenase were selected for the bioconversion of aromatic hydrocarbons to amines. The amination reaction was optimized with respect to cofactor addition and enzyme dosage. Isopropanol was added to decrease ketone intermediate accumulation in the amination step, which further enhanced the overall conversion. This cascade system converted a panel of hydrocarbon substrates into the corresponding amines with excellent optical purity (>99% ee ) and moderate conversion ratios (13–53%).

Abstract:Asymmetric functionalization of alkenes allows the direct synthesis of a wide range of chiral compounds. Vicinal hydroxyazidation of alkenes provides a desirable path to 1,2-azidoalcohols; however, existing methods are limited by the control of stereoselectivity and regioselectivity. Herein, we describe a dual-enzyme cascade strategy for regiodivergent and stereoselective hydroxyazidation of alkenes, affording various enantiomerically pure 1,2-azidoalcohols. The biocatalytic cascade process is designed by combining styrene monooxygenase-catalyzed asymmetric epoxidation of alkenes and halohydrin dehalogenase-catalyzed regioselective ring opening of epoxides with azide. Additionally, a one-pot chemo-enzymatic route to chiral <i>β</i>-hydroxytriazoles from alkenes is developed via combining the biocatalytic cascades and Cu-catalyzed azide-alkyne cycloaddition.

Abstract:A palladium-catalyzed C-H functionalization/intramolecular asymmetric allylation cascade of N-sulfonyl benzamides with 1,3-dienes has been developed. In the presence of a chiral pyridine-oxazoline ligand, this protocol enables the synthesis of chiral 3,4-dihydroisoquinolones in yields of up to 83% with enantioselectivities of up to 96%, using environmentally friendly air as the terminal oxidant.

Abstract:An organocatalytic approach for direct conversion of racemic diarylmethanols to valuable chiral diarylmethylamines is described. Different from the previously reported elegant borrowing hydrogen approach, the present process employs a distinct complementary formal S(N)1 strategy. This approach enjoys excellent enantioselectivity, mild conditions, broad scope, and easy product derivatization. Mechanistically, control experiments also provided important insights into some notable features, such as substrate kinetic resolution and reversibility as well as the critical role of the ortho- hydroxy group in the substrate.

Abstract:Biocatalytic regio- and stereo-selective functionalization of renewable fatty acids for sustainable synthesis of value-added chiral chemicals is highly desirable but remains a great challenge. Herein, a three-step one-pot multienzyme cascade for the asymmetric synthesis of D-amino acids from renewable fatty acids was developed. Combination of P450 peroxygenase with two enantiocomplementary hydroxyacid oxidase(s) enabled the regioselective oxyfunctionalization of fatty acids into prochiral α-ketoacids with internal H2O2 recycling. An engineered D-amino acid dehydrogenase with formate dehydrogenase for self-recycling of the expensive cofactor NADPH was adopted for the reductive amination of α-ketoacids to D-amino acids. Various fatty acids (C6-C10) with different chain length can be efficiently converted into the corresponding D-amino acids with high yield (up to 99%) and excellent ee value (>99%). This study exploits the advantage of cascades and showcases the potential for synthesizing valuable chiral chemicals from inexpensive renewable feedstocks.

Abstract:Chiral alcohols are important intermediates for various fine chemicals and pharmaceuticals. Integrating chemical catalysis and efficient enzyme catalysis in sequential systems for the synthesis of chiral alcohols is considered an ecofriendly and promising approach. Herein, employing a highly selective carbonic anhydrase II and different chemical catalysts, we constructed three general chemoenzymatic sequential systems for chiral alcohol compound synthesis from alkanes, alkenes, and alkynes, respectively. Compared to classical approaches, the combination of chemical catalysis and promiscuous carbonic anhydrase catalysis is simple and efficient since it requires only mild reaction conditions and avoids expensive chiral ligands and cumbersome operation steps. In this integrated approach, a wide variety of readily available aryl alkanes, alkenes, and alkynes are transformed into valuable chiral alcohols with excellent enantioselectivity of up to 99% (nearly all above 90%). This unified strategy of combining enzymatic and chemical catalyses advances the general chemoenzymatic process for powerful and important chemical transformations.

Enhancing cofactor recycling in the bioconversion of racemic alcohols to chiral amines with alcohol dehydrogenase and amine dehydrogenase by coupling cells and cell-free system

Abstract:Alcohol dehydrogenase (ADH) and amine dehydrogenase (AmDH)-catalyzed one-pot cascade conversion of an alcohol to an amine provides a simple preparation of chiral amines. To enhance the cofactor recycling in this reaction, we report a new concept of coupling whole-cells with the cell-free system to enable separated intracellular and extracellular cofactor regeneration and recycling. This was demonstrated by the respective biotransformation of racemic 4-phenyl-2-butanol 1a and 1-phenyl-2-propanol 1b to (R)-4-phenylbutan-2-amine 3a and (R)-1-phenylpropan-2-amine 3b. Escherichia coli cells expressing S-enantioselective CpsADH, R-enantioselective PfODH, and NADH oxidase (NOX) was developed to oxidize racemic alcohols 1a-b to ketones 2a-b with full conversion via intracellular NAD+ recycling. AmDH and glucose dehydrogenase (GDH) were used to convert ketones 2a-b to amines (R)-3a-b with 89-94% conversion and 891-943 times recycling of NADH. Combining the cells and enzymes for the cascade transformation of racemic alcohols 1a-b gave 70% and 48% conversion to the amines (R)-3a and (R)-3b in 99% ee, with a total turnover number (TTN) of 350 and 240 for NADH recycling, respectively. Improved results were obtained by using the E. coli cells with immobilized AmDH and GDH: (R)-3a was produced in 99% ee with 71-84% conversion and a TTN of 1410-1260 for NADH recycling, the highest value so far for the ADH-AmDH-catalyzed cascade conversion of alcohols to amines. The concept might be generally applicable to this type of reactions.

Abstract:Enantiomerically pure amines are crucial intermediates for a wide range of pharmaceuticals, natural products, and bioactive compounds. Asymmetric intermolecular hydroamination of alkenes is one of the most atom-economical methods for the synthesis of chiral amines. However, the direct stereoselective hydroamination of alkenes remains a significant challenge. Herein, we developed a novel fully biocatalytic system to enable the direct stereoselective hydroamination of aryl alkenes to produce chiral amines. The process involves sequential alkene asymmetric hydration catalyzed by a hydratase (HD), followed by in situ oxidation of alcohol into ketone by two enantiocomplemetary alcohol dehydrogenases (ADHs), and finally, enantioselective transformation of ketone into enantiomerically pure amine by a ω-transaminase (TA). The addition of lactate dehydrogenase (LDH) facilitated the connection between alcohol oxidation and pyruvate reduction, making the cascade reaction redox self-sufficient and driving the process towards the desired target product. In vitro cascade biocatalysis for asymmetric hydroamination of 4 hydroxystyrenes 1a-f was first investigated by using the combined enzymes (HD/ADHs/LDH/TA) with trace amount of NAD+ (0.05 mM) and pyridoxal-5'-phosphate (PLP) (0.1 mM), affording both enantiomers of amines 4a-f in 30-90% conversions and >99% ee. Furthermore, whole-cell-based cascade biocatalysis was achieved by using the constructed recombinant Escherichia coli cells co-expressing the five enzymes without additional NAD+ cofactor, (R)- and (S)-amines 4a−f could be obtained in 35−93% conversions and >99% ee. Finally, the preparative experiments were demonstrated by using the whole-cell biocatalyst E. coli (RFM-ERR-CB) and E. coli (RFC-ERR-CB) with substrate 1a, affording (S)-4a and (R)-4a in 54.8% and 59.2% yields and >99% ee. This research achievement provides a high selective, high atom-economical, green and sustainable method for synthesizing chiral amines from alkenes via formal alkene asymmetric hydroamination.

Abstract:The integration of organocatalysis and enzyme catalysis in one-pot cascade processes allows for the efficient construction of complex molecular architectures with high levels of stereocontrol. However, challenges related to reaction compatibility between both processes are often a limitation for the development of efficient synthetic routes. In this study, we describe the combination of an enzymatic aerobic oxidation followed by the squaramide-mediated asymmetric formation of C-P and C-C bonds to access important building blocks such as chiral α-hydroxy phosphonates and β-nitro alcohols in good yields and enantiomeric ratios. This sequential process is conducted in a one-pot fashion within a biphasic system and represents a pioneering example of a chemoenzymatic cascade involving aerobic biooxidation and an organocatalytic step operating under hydrogen-bond activation mode.

Abstract:A general and highly efficient method for asymmetric sequential hydrogenation of alpha,beta-unsaturated ketones has been developed by using an iridium/f-Ampha complex as the catalyst, furnishing corresponding chiral alcohols with two contiguous stereocenters in high yields with excellent diastereo- and enantioselectivities (up to 99% yield, >20 : 1 dr and >99% ee). Control experiments indicated that the C=C and C=O bonds of the enones were hydrogenated sequentially, and the final stereoselectivities were determined by the dynamic kinetic resolution of ketones. Moreover, DFT calculations revealed that an outer sphere pathway was involved in both reduction of C=C and C=O bonds of enones. The synthetic utility of this method was demonstrated by a gram-scale reaction with very low catalyst loading (S/C = 20 000) and a concise synthetic route to key chiral intermediates of the antiasthmatic drug CP-199,330.

Abstract:Enantiopure 1,2-diols are widely used in the production of pharmaceuticals, cosmetics, and functional materials as essential building blocks or bioactive compounds. Nevertheless, developing a mild, efficient and environmentally friendly biocatalytic route for manufacturing enantiopure 1,2-diols from simple substrate remains a challenge. Here, we designed and realized a step-wise biocatalytic cascade to access chiral 1,2-diols starting from aromatic aldehyde and formaldehyde enabled by a newly mined benzaldehyde lyase from Sphingobium sp. combined with a pair of tailored-made short-chain dehydrogenase/reductase from Pseudomonas monteilii (PmSDR-MuR and PmSDR-MuS) capable of producing (R)- and (S)-1-phenylethane-1,2-diol with 99% ee. The planned biocatalytic cascade could synthesize a series of enantiopure 1,2-diols with a broad scope (16 samples), excellent conversions (94%-99%), and outstanding enantioselectivity (up to 99% ee), making it an effective technique for producing chiral 1,2-diols in a more environmentally friendly and sustainable manner.
