Abstract:Nine new tirucallane-type triterpenoids, ficutirucins A-I (1-9), were isolated from the fruit of Ficus carica. Their structures were established on the basis of spectroscopic data and chemical methods. All isolates were evaluated for their cytotoxic activities against three human cancer cell lines, MCF-7, HepG-2, and U2OS. Compounds 1-3, 6, 7, and 9 exhibited moderate cytotoxic activities with IC50 values of 11.67-45.61, mu M against one or more of the three cancer cell lines.

Abstract:Eight new cycloartane triterpenoids (1–8), along with eight known analogues (9–16), were isolated from the roots of Cimicifuga foetida L. Their structures were elucidated by spectroscopic analysis and acidic hydrolysis. All of the new compounds were evaluated for their in vitro cytotoxicity against five human tumor cell lines (HL-60, SMMC-7721, A-549, MCF-7 and SW480), and compound 2 showed inhibitory activities with IC50 values raging from 2.61 to 3.32μM.

Abstract:Three new triterpenoids (1-3), including two new 19 (10 -> 9 beta) abeo-tirucallane derivatives (1-2) and a new tirucallane-type triterpenoid (3), together with two known tirucallane-type triterpenoids (4-5) were isolated from the ethanol extract of the fruits of Chukrasia tabularis. Their structures were elucidated on the basis of 1D and 2D NMR spectroscopic analyses and MS data. The isolated triterpenoids 1 and 2 exhibited anti-inflammatory activities with IC50 values of 75.98 +/- 2.12 and 24.86 +/- 1.16 mu M, respectively. Compound 5 showed cytotoxic activities against the selected tumor cell lines.

Abstract:Eleven triterpenoids were isolated from the fruits of Phellodendron chinense Schneid, and their structures were determined by spectroscopic analysis. The results show that four new tirucallane-type triterpenoids 1, 2, 5, and 6 and seven known compounds 3, 4, 7, 8, 9, 10, and 11 were isolated. Structurally, compound 6 was uncommon; it has a chlorine atom instead of a methyl group at the C-20 position. The cytotoxicities of the compounds was evaluated against the in vitro proliferation of four human tumor cell lines HEL, K562, MDA, and PC3 using adriamycin as the positive control. Compound 1 showed a similar cytotoxicity as the positive control; compounds 3 and 10 showed moderate cytotoxicities compared to the control (P<0.05). This indicates that these compounds have great potential for the development of new antitumor drugs.

Abstract:Six new 9,19-cycloartane triterpenes (1–6) were isolated from the aerial parts of Cimicifuga yunnanensis. The new chemical structures were determined by extensive analyses of 1D and 2D NMR spectroscopy. Compounds 1 and 2 are the first 9,19-cycloartane triterpenes characterized by CH3-18 shifting from C-13 to C-12 in the Cimicifuga spp. The evaluation of inhibition activity against human HL-60, SMMC-7721, A-549, MCF-7, and SW480 cell lines indicated that compounds 1–6 showed different levels of cytotoxic activities with IC50 values ranging from 1.2 to 27.8μm.

Abstract:Four undescribed coumarin derivatives, ficusalt A (1) and ficusalt B (2), a pair of racemic coumarins, (±) ficudimer A (3a/3b), along with ten known amides, were isolated from the roots of Ficus hirta. Their structures were elucidated by several spectroscopic data analyses, including HRESIMS, NMR, and X-ray single-crystal diffraction. The cytotoxic activities of all compounds against HeLa, HepG2, MCF-7, and H460 cell lines were detected using the MTT assay. Among these, 5 showed the highest activity against HeLa cells. Subsequently, the apoptotic, anti-invasive, and anti-migration effects of 5 on HeLa cells were determined by flow cytometer, transwell invasion assay, and wound-healing assay, respectively. The result suggested that 5 distinctly induced the apoptosis in HeLa cells and inhibited their invasion and migration. Further studies on anticancer mechanisms were conducted using Western blotting. As a result, 5 increased the cleavage of PARP and the expression of pro-apoptotic protein Bax. Moreover, 5 notably upregulated the phosphorylation of p38 and JNK, whereas inhibited the expression of p-ERK and p-AKT. Our results demonstrated that 5 could be a potential leading compound for further application in the treatment of cervical cancer.

Abstract:Three new cucurbitane-type triterpenoids, including (23E)-cucurbita-3β-hydroxy-25-methoxy-5,23-diene (1), (23E)-cucurbita-3β-hydroxy-5,23,25-triene (2), and cucurbiata-3β-hydroxy-5,25-dien-24-one (3), have been isolated with four known triterpenoids (4-7) from the dried whole plants of Ainsliaea henryi. The chemical structures of compounds 1–3 were elucidated by 1D NMR (1H, 13C and DEPT), 2D NMR (COSY, HMBC, HSQC and NOESY) and HREIMS analyses. Among these compounds, compound 3 exhibited strong cytotoxicity against human cancer cell lines MGC 803 (IC50=8.33±0.94μM) and CCRF-CEM (IC50=4.34± 0.41μM), while compound 6 significantly inhibited the proliferation of the A549 and HCT 116 cell lines with IC50 values of 2.14±0.25 and 6.84±0.39μM, respectively.

Abstract:Four new 9,19-cycloartane triterpenoids, cimilactone E (1), cimilactone F (2), 2′-O-(E)-butenoyl-23-epi-26-deoxyactein (3), and 2′,12β-O-diacetylcimiracemonol-3-O-β-d-xylopyranoside (4), together with four known constituents (5–8) were isolated from the roots of Cimicifuga foetida. The new structures were elucidated by extensive spectroscopic analysis. In addition, compounds 7 and 8 showed significant Wnt signaling pathway inhibitory activity, with IC50 values of 3.33 and 13.34 μM, respectively, using the luciferase reporter gene assay.

Abstract:A new guaiane-type sesquiterpene glycoside, torilolone 8-O-beta-D-glucopyranoside (1), together with a known analogue compound, torilolone 11-O-beta-D-glucopyranoside (2), was isolated from the fruits of Daucus carota L. Their chemical structures were elucidated on the basis of MS, NMR spectroscopic analyses coupled with chemical degradation. The cytotoxic activities of two isolated compounds were evaluated against two human gastric cancer cell lines BGC-823 and AGS using MTT assay.

Abstract:A phytochemical investigation of the fruits of Citrullus colocynthis resulted in the isolation of 21 structurally diverse cucurbitane triterpenoids, including 9 previously undescribed ones, colocynins A-I ( 1–9 ). Their absolute configurations were elucidated by means of quantum chemical electronic circular dichroism (ECD) calculations, CD exciton chirality method, and single-crystal X-ray crystallography. Colocynins A-C ( 1–3 ) represent the first examples of nonanorcucurbitane-type triterpenoids. An anti-acetylcholinesterase activity assay showed that 6 , 10 , 13 , 18 , and 20 exhibited inhibitory activities, with IC 50 values ranging from 5.0 to 21.7 μM. In addition, 18 and 21 showed significant cytotoxicity against PACA, A431, and HepG2 cells, with IC 50 values ranging from 0.042 to 0.60 and 3.6–14.4 μM, respectively.

Abstract:Three new tigliane-type diterpenoids were isolated from the methanolic extract of the twigs and leaves of Croton caudatus, trivially named crotusins A-C (1-3). The structures of compounds 1-3 were elucidated on the basis of extensive spectral methods. These new compounds were highly oxygenated and heavily substituted. Cytotoxic activity against five human tumor cell lines was assessed for compounds 1-3 of which compound 3 showed significant inhibitory activity with IC50 values ranging from 0.49 to 4.19 mu M against these cells, while crotusins A and B exhibited moderate activity.

Abstract:A phytochemical study on the rhizomes of Cimicifuga foetida resulted in the isolation of two new cycloartane triterpenoids (1 and 2), eight new cycloartane glycosides (3-10), and six known cycloartane glycoside analogues (11-16). The structures of 1-10 were determined by application of spectroscopic methods, with the absolute configuration of 1 determined by X-ray crystallography. Compounds 1-6, as three pairs of epimers at C-10 and C-24, belong to a seven-membered-ring variant of 9,10-seco-9,19-cycloartane triterpenoids, and glycosides 3-10 were found to be 3-O-β-D-xylopyranosides. The cytotoxicity of the isolates was evaluated against five selected human tumor cell lines, and the known compounds 15 and 16 showed cytotoxicity against the hepatocellular carcinoma SMMC-7721 cell line with IC50 values of 5.5 and 6.3 μM, respectively.

Abstract:Five new cucurbitane triterpenoids (hemslepenside B–E (1–3 and 5)), one new cucurbitane (16,25-O-diacetyl-cucurbitane F (4)), and six analogues (6–11) were isolated from the roots of Hemsleya jinfushanensis. The structures of 1–5 were elucidated using infrared absorption spectroscopy (IR), high resolution electrospray ionization mass spectrometry (HR-ESI-MS), and nuclear magnetic resonance spectroscopy (NMR). These five new compounds exhibited cytotoxic effects against lung adenocarcinoma (H460), colon cancer (SW620) and human prostate cancer cells (DU145), and compound 4 showed significant cytotoxic activity, with IC50 values of 0.046, 0.18 and 0.87 μg/mL. This finding suggests that cucurbitane triterpenoids isolated from H. jinfushanensis should be studied further as potential anti-cancer drugs.

Abstract:Two new cucurbitane triterpenoids, 7β-hydroxycucurbitacin F-25-O-acetate (1) and 2β,3β,20(S),26,27-pentahydroxy-16α,23(S)-epoxycucurbita-5,24-dien-11-one (2) along with eleven known cucurbitane triterpenoids (3–13, resp.) were isolated from the rhizomes of Hemsleya amabilis Diels. The chemical structures of the new isolated compounds were elucidated unambiguously by spectroscopic data analysis. The cytotoxic activities of the isolated cucurbitane triterpenoids were evaluated against the HeLa human cancer cell lines. Hemslecin A (5), the main ingredient of H. amabilis, exhibited the significant cytotoxicity with IC50 value of 0.389μM.

Abstract:Two new triterpenoids, 30-O-beta-D-glucopyranosyloxy-2alpha,3alpha,24-trihydroxyurs-12,18-diene-28-oic acid O-beta-D-glucopyranosyl ester (1) and 2alpha,3beta,3,30-tetrahydroxyurs-12,18-diene-28-oic acid O-beta-D-glucopyranosyl ester (2) were isolated from roots of Actinidia valvata Dunn. Their structures were elucidated by means of extensive spectroscopic studies. Both these two new compounds showed moderate cytotoxic activity in vitro against BEL-7402 and SMMC-7721 tumor cell line.

Abstract:Two new cucurbitane-type triterpenoid saponins, 2,20 β ,22 β -trihydroxy-16 α ,23( R )-epoxycucurbita-1,5,24-triene-3,11-dione 2- O - β -D-glucopyranoside ( 1 ), 2,20 β ,22 α -trihydroxy-16 α ,23( S )-epoxycucurbita-1,5,11,24-tetraene-3-one 2- O - β -D-glucopyranoside ( 2 ) were isolated from the fruit of Citrullus colocynthis (L.) Schrad. Their structures were elucidated by mass spectrometry, IR, 1D, and 2D NMR spectroscopy, etc. Besides, both of the compounds showed significant hepatoprotective activities at 10 μ M against paracetamol-induced HepG2 cell damage.
