Abstract:The amorphous phase precipitates first during biomineralization and acts as the precursor for the subsequent mineralization of hard tissues including dentin. During this process, various biomineralization proteins with different, even opposite functions control the dimensions and phase states of the amorphous precursors that permeate the collagen matrix and then crystallize to form highly sophisticated organic-inorganic biological materials. In this study, a biomimetic strategy containing polyacylic acid and l-glutamic acid (l-Glu) was applied to promote the remineralization of Ca-depleted dentin collagen. Following the structural features of biomineralization-related non-collagenous proteins (NCPs), l-Glu was found to be capable of promoting the crystallization kinetics of the polyacylic acid-stabilized metastable amorphous to hydroxyapatite phase transformation. It is demonstrated that the dentin remineralization could be shortened within two days by using the cooperative effect of polyacrylic acid and l-glutamic acid. Furthermore, the resulting remineralized dentin layer has the same characteristics, including mechanical strength, as the natural ones. This biomimetic strategy highlights the combination of the two opposing factors of biomineralization components as the key to control the phase-transformation-based mineralization reactions with the organic matrix of dental tissues. In summary, a bio-inspired approach was followed to reconstruct collagen-mineralized tissues with biocompatible functions, morphologies, and characteristics.

Abstract:This study presents a biomimetic method for remineralizing dental hard tissues using an amyloid‐like protein matrix composite with amorphous calcium phosphate. This approach mimics natural mineralization processes, enhancing the regeneration and mechanical strength of demineralized enamel and dentine, providing a promising material for the treatment of early enamel lesions and dentine hypersensitivity. Numerous remineralizing coatings aim to prevent or treat early enamel lesions and occlude exposed dentinal tubules (DTs). Nevertheless, the pace of remineralization is inadequate, and the mechanical robustness of the newly established mineral layer fails to match the inherent strength. In this study, a biomimetic mineralization strategy aimed at replicating key events in biological mineralization, specifically focusing on the organic–inorganic composite matrix, is proposed. The material utilizes Tris(2‐carboxyethyl)phosphine (TCEP), which serves a dual role: stabilized amorphous calcium phosphate (ACP) (ACP@TCEP) nanoparticles as its inorganic component, and catalyzing the cleavage of intramolecular disulfide bonds in poly(ethylene glycol) (PEG) grafted lysozyme (lyso‐PEG) to facilitate the formation of an amyloid‐like protein matrix composite with ACP (ACP@lyso‐PEG nanocomplexes). ACP@lyso‐PEG nanocomplexes can rapidly and efficiently form an enamel‐like remineralization layer on the surface of damaged dental hard tissue, reaching ≈4.205 μm thickness after 3 days of acid‐etched enamel. Furthermore, achieving a depth of DTs occlusion exceeding 60 μm after 5 days, using a simple immersion process. The resulting mineralized layer exhibits mechanical strength comparable to natural teeth. This study introduces a conceptual biomimetic mineralization strategy for effective enamel repair or DTs occlusion in clinical practices, and offers potential insights into the mechanisms of biomineral formation.

Glutaraldehyde-induced Remineralization Improves the Mechanical Properties and Biostability of Dentin Collagen

Abstract:The purpose of this study was to induce a biomimetic remineralization process by using glutaraldehyde (GA) to reconstruct the mechanical properties and biostability of demineralized collagen. Demineralized dentin disks (35% phosphoric acid, 10s) were pretreated with a 5% GA solution for 3min and then cultivated in a calcium phosphate remineralization solution. The remineralization kinetics and superstructure of the remineralization layer were evaluated by Raman spectroscopy, transmission electron microscopy, scanning electron microscopy and nanoindentation tests. The biostability was examined by enzymatic degradation experiments. A significant difference was found in dentin remineralization process between dentin with and without GA pretreating. GA showed a specific affinity to dentin collagen resulting in the formation of a cross-linking superstructure. GA pretreating could remarkably shorten remineralization time from 7days to 2days. The GA-induced remineralized collagen fibrils were well encapsulated by newly formed hydroxyapatite mineral nanocrystals. With the nano-hydroxyapatite coating, both the mechanical properties (elastic modulus and hardness) and the biostability against enzymatic degradation of the collagen were significantly enhanced, matching those of natural dentin. The results indicated that GA cross-linking of dentin collagen could promote dentin biomimetic remineralization, resulting in an improved mechanical properties and biostability. It may provide a promising tissue-engineering technology for dentin repair.

Abstract:Biomineralization has intrigued researchers for decades. Although mineralization of type I collagen has been universally investigated, this process remains a great challenge due to the lack of mechanistic understanding of the roles of biomolecules. In our study, dentine was successfully repaired using the biomolecule polydopamine (PDA), and the remineralized dentine exhibited mechanical properties comparable to those of natural dentine. Detailed analyses of the collagen mineralization process facilitated by PDA showed that PDA can promote intrafibrillar mineralization with a decreased heterogeneous nucleation barrier for hydroxyapatite (HAP) by reducing the interfacial energy between collagen fibrils and amorphous calcium phosphate (ACP), resulting in the conversion of an increasing amount of nanoprecursors into collagen fibrils. The present work highlights the importance of interfacial control in dentine remineralization and provides profound insight into the regulatory effect of biomolecules in collagen mineralization as well as the clinical application of dentine restoration.
Reconstruction of a Demineralized Dentin Matrix Via Rapid Deposition of CaF2 Nanoparticles in Situ Promotes Dentin Bonding.

Abstract:Dentin bonding based on a wet-bonding technique is the fundamental technique used daily in clinics for tooth-restoration fixation and clinical treatment of tooth-related diseases. Limited bonding durability led by insufficient adhesive infiltration in the demineralized dentin (DD) matrix is the biggest concern in contemporary adhesive dentistry. This study proposes that the highly hydrated noncollagenous protein (NCP)-formed interfacial microenvironment of the DD matrix is the root cause of this problem. Meanwhile, the endogenous phosphate groups of the NCPs are used as pseudonuclei to rapidly induce the formation of amorphous CaF2 nanoparticles in situ in the interfacial microenvironment. The DD matrix is thus reconstructed into a novel porous structure. It markedly facilitates the infiltration of dentin adhesives in the DD matrix and also endows the DD matrix with anticollapsing capability when water evaporates. Whether using a wet-bonding or air-drying mode, the bonding effectiveness is greatly promoted, with the 12 month bonding strength being about twice that of the corresponding control groups. This suggests that the nanoreinforced DD matrix eliminates the dependence of bonding effectiveness on the moisture status of the DD surface controlled only by experiences of dentists. Consequently, this bonding strategy not only greatly improves bonding durability but also overcomes the technical sensitivity of bonding operations of the total-etched bonding pattern. This exhibits the potential to promote dentin bonding and is of great significance to dentistry.
Effect of Aspartic Acid on the Crystallization Kinetics of ACP and Dentin Remineralization.

Abstract:Type I collagen and non-collagen proteins are the main organic components of dentin. This study aimed to investigate the biomimetic remineralization of demineralized dentin by aspartic acid (Asp), which is abundant in non-collagenous proteins (NCPs). Asp was added to a mineralizing solution containing polyacrylic acid (PAA) to explore the mechanism of Asp regulating the pure amorphous calcium phosphate (ACP) phase transition process. The remineralization process and superstructure of the remineralized layer of demineralized dentin were evaluated and analyzed by transmission electron microscope (TEM) and scanning electron microscope (SEM), and the biological stability of the remineralized layer was investigated by collagenase degradation experiment. It demonstrated that Asp promoted the crystallization kinetics of PAA-stabilized amorphous calcium phosphate to hydroxyapatite (HAP), and shortened the remineralization time of demineralized dentin from 7 days to 2 days. The newly formed remineralized dentin had similar morphology and biological stability to the natural dentin layer. The presence of a large number of Asp residues in NCPs promoted the phase transformation of ACP, and further revealed the mechanism of action of NCPs in dentin biomineralization. This experiment also showed that Asp promoted the biomimetic remineralization of dentin; the morphology and hierarchical structure of remineralized layer was similar to that of natural teeth, and had good biological properties.

Abstract:Biomineralization is a well-regulated process mediated by many extracellular matrix proteins. Some collagenous proteins act as frameworks for depositing minerals in a preferred orientation while non-collagenous proteins (BSP, BPP, OC, etc.) function as regulators of the mineralization process, either as nuclei or inhibitors of hydroxyapatite. Biomimetic remineralization strategies should reproduce the dimension and structural hierarchy of apatite within a collagen matrix. Here we synthesized a peptide with sequence (EEEEEEEEDS(p)ES(p)S(p)EEDR) mimicking the function of BSP and DMP1 as calcium phosphate-and collagen-binding matrix protein analogues. More metastable calcium phosphate nanoprecursors were combined with the collagen matrix in acid-demineralized dentine pre-incubated with peptides during the process of remineralization in the phosphate-containing fluid. With the presence of our synthetic peptides, these nanoprecursors were transformed into polyelectrolyte-stabilized apatite nanocrystals that assembled at the "hole zones" (gap remineralization) and along the surface of the dentine collagen fibrils (surface remineralization). The transition from nanocrystals to larger apatite platelets was promoted by synthetic peptides via the formation of mesocrystal intermediates. It was concluded that deliberately designed peptide (EEEEEEEEDS(p)ES(p)S(p)EEDR) improved in situ remineralization of acid-etched dentine.

Dentin biomineralization is a process that simulates the mineralization of biological tissue in nature in which the remineralization of dentin collagen is induced and regulated by organic macromolecules. This review summarizes the process of remineralization of decalcified dentin regulated by non-collagenous protein analogues in vitro.
Remineralization of dentin induced by a compound of polyamide-amine and chlorhexidine in a resin dentin bonding microenvironment

Abstract:BACKGROUND: The purpose of this study was to investigate the effect of a complex of polyamide-amine dendrimer (PAMAM) and chlorhexidine gluconate (CG) on remineralization of dentin in an artificial simulated resin dentin bonding microenvironment.METHODS: The structure of this complex was characterized by FT-IR. Twelve standard dentin samples were randomly divided into four treatment fluid groups namely a PAMAM group, CG group, PAMAM + CG group, and deionized water group. A microenvironmental mineralization model was established <i>in vitro</i> with 50 µm gap width between resin and dentin. The dentin surface was observed by a scanning electron microscope (SEM), and the chemical structure of the surface was analyzed by X-ray energy spectrum (EDS), X-ray diffraction (XRD), and laser Raman spectroscopy.RESULTS: SEM showed the density of dentinal tubules exposed in the PAMAM group decreased after a 14-day immersion, with corn rod-shaped crystal structures gathered around the tubules. In addition, visible mineralization occurred in partial areas of the CG group, rod-shaped crystals and in comparison, dentinal tubules in the PAMAM + CG group were almost completely covered by flaky crystal structures. Raman spectrum analysis showed that crystals formed by PAMAM, CG, and PAMAM + CG solution all had strong phosphate characteristic peaks, indicating the presence of hydroxyapatite (HA), that of the PAMAM + CG group was the strongest. The EDS results showed that the Ca and P levels of the PAMAM group and the CG group were slightly higher than those of the deionized water group, while PAMAM + CG group significantly higher than the others, Ca/P value approaching 1.67. The results of XRD showed the characteristic peaks of hydroxyapatite detected by the PAMAM + CG group at 2θ=26.0 (002), 2θ=32.0 (211), and 2θ=33.0 (112) were high and sharp, with a few diffraction line burrs indicating it had high crystallinity and purity. The Scherrer equation results showed that the appearance and size of the grains formed by the PAMAM + CG group were basically consistent with those of healthy dentin.CONCLUSIONS: Altogether, the compound of polyamide-amine dendrimer and chlorhexidine could induce the remineralization of human dentin in a resin dentin bonding microenvironment with a gap of 50 µm to form a crystal structure similar to dentin hydroxyapatite.

Abstract:Biomineralization of collagenous mineralized tissues (CMT), in vivo, stands as a precisely cell-controlled process. An organic insoluble collagenous matrix forms, then inorganic mineral is deposited on the matrix. The exact mechanism of action remains elusive and most researchers agree the amorphous mineral phase is crucial in CMT's development. Artificial demineralized dentin can be remineralized via amorphous calcium phosphate (ACP) precursor stage stabilized by 500 mu g ml(-1) polyacrylic acid (PAA). This remineralization is a step-by-step progression. ACP initially forms in the collagen matrix at its base, transforms into hydroxyapatite, and migrates towards the surface. The state of ACP, which is labile in liquid environments, can be controlled by PAA in a dose-dependent manner. Thus, the concentration of PAA is a key parameter influencing dentin remineralization. Low concentration of PAA (100 mu g ml(-1)) fails to maintain liquidity of ACP, resulting in superficial remineralization. Conversely, high concentration of PAA (1000 mu g ml(-1)) retards phase transformation of ACP and, thereby, inhibits remineralization. The purpose of this study is to demonstrate the balance between meta-stability and crystallization of ACP precursor phase. Our findings also provide evidence for the role of ACP during biomineralization and reveal a possible approach to repair CMT.

Abstract:OBJECTIVE: The objective of this study was to develop a rapid and effective method to remineralize human carious-like enamel using chimaeric peptide-mediated nanocomplexes of carboxymethyl chitosan/amorphous calcium phosphate (CMC/ACP), mimicking the mineralizing pattern of the oriented assembly of ACP guided by amelogenin in the biomineralization of enamel.METHODS: CMC/ACP nanocomplex solution was first synthesized through the successive addition of carboxymethyl chitosan, calcium chloride, and dipotassium phosphate into distilled water. ACP nanoparticles were degraded by 1% NaClO from CMC/ACP nanocomplexes. The morphology of the particles at different periods was tested by transmission electron microscopy (TEM). The chimaeric peptides were added to guide the arrangement of ACP nanoparticles and to bind ACP nanoparticles to the demineralized enamel surface specifically. X-ray diffraction (XRD)/scanning electron microscope (SEM)/confocal laser scanning microscopy (CLSM)/nano-indentation tests were applied to check the remineralization effects.RESULTS: CMC/ACP nanocomplexes were obtained and could be kept without precipitation for a long time. After the degradation of NaClO and guidance of chimaeric peptides, ACP nanoparticles were arranged into oriented arrays before transforming into crystals, and the enamel-like crystals were tightly bound onto the demineralized surface. The newly formed enamel-like crystals were nearly well-organized and equipped with strong mechanical properties.

Abstract:Achieving oriented and ordered remineralization on the surface of demineralized dental enamel, thereby restoring the satisfactory mechanical properties approaching those of sound enamel, is still a challenge for dentists. To mimic the natural biomineralization approach for enamel remineralization, the biological process of enamel development proteins, such as amelogenin, was simulated in this study. In this work, carboxymethyl chitosan (CMC) conjugated with alendronate (ALN) was applied to stabilize amorphous calcium phosphate (ACP) to form CMC/ACP nanoparticles. Sodium hypochlorite (NaClO) functioned as the protease which decompose amelogenin in vivo to degrade the CMC-ALN matrix and generate HAP@ACP core-shell nanoparticles. Finally, when guided by 10 mM glycine (Gly), HAP@ACP nanoparticles can arrange orderly and subsequently transform from an amorphous phase to well-ordered rod-like apatite crystals to achieve oriented and ordered biomimetic remineralization on acid-etched enamel surfaces. This biomimetic remineralization process is achieved through the oriented attachment (OA) of nanoparticles based on non-classical crystallization theory. These results indicate that finding and developing analogues of natural proteins such as amelogenin involved in the biomineralization by natural macromolecular polymers and imitating the process of biomineralization would be an effective strategy for enamel remineralization. Furthermore, this method represents a promising method for the management of early caries in minimal invasive dentistry (MID).

Crosslinking Improve Demineralized Dentin Performance and Synergistically Promote Biomimetic Mineralization by CaP_PILP

Abstract:OBJECTIVE: to explore the effects of optimal crosslinking (chemical treatment) on demineralized dentin matrix and the possible synergism with calcium phosphate polymer-induced liquid precursor (CaP-PILP) bionic remineralization (physical treatment), and offer benefit to the clinic of resin-dentin bonding and dentin hypersensitivity.METHODS: demineralized dentin was treated with glutaraldehyde (GA), carbodiimide (EDC), and procyanidin (PA) for crosslinking, followed by CaP-PILP biomimetic remineralization. The morphology, surface mechanical and physio-chemical properties, and enzymatic resistance were evaluated regardless of the modification.RESULTS: the collagen fibers appeared morphologically complete with higher surface microhardness and characteristic peaks of amide I-III bands were visible after GA, PA, and EDC crosslinking. Collagen collapse and dissolution were seen in untreated demineralized dentin with enzyme attack, while the collagen fiber structure remained intact in GA- and PA-treated specimens. The lamellar mineral phase was visible at 2 days and the dentin tubules were almost completely enclosed at 4-6 days after PA crosslinking and mineralization. However, demineralized collagen fibers and open tubules were still visible between the dentinal tubules on day 8 in the control group.CONCLUSION: the structure integrity, enzyme resistance, and mechanical properties of the collagen fiber network could be significantly preserved by GA and PA crosslinking than EDC and no treatment. While, strongest synergistic effects were observed in PA on bionic remineralization by CaP-PILP, and further significantly improve the quality and shorten the duration of mineralization. These findings would be beneficial for dental clinical practice of resin-dentin bonding and dentin hypersensitivity.

Abstract:Objective: This review elucidates the mechanisms underpinning intrafibrillar mineralization, examines various amorphous calcium phosphate (ACP) stabilizers employed in dentin's intrafibrillar mineralization, and addresses the challenges encountered in clinical applications of ACP-based bioactive materials. Methods: The literature search for this review was conducted using three electronic databases: PubMed, Web of Science, and Google Scholar, with specific keywords. Articles were selected based on inclusion and exclusion criteria, allowing for a detailed examination and summary of current research on dentin remineralization facilitated by ACP under the influence of various types of stabilizers. Results: This review underscores the latest advancements in the role of ACP in promoting dentin remineralization, particularly intrafibrillar mineralization, under the regulation of various stabilizers. These stabilizers predominantly comprise non-collagenous proteins, their analogs, and polymers. Despite the diversity of stabilizers, the mechanisms they employ to enhance intrafibrillar remineralization are found to be interrelated, indicating multiple driving forces behind this process. However, challenges remain in effectively designing clinically viable products using stabilized ACP and maximizing intrafibrillar mineralization with limited materials in practical applications. Significance: The role of ACP in remineralization has gained significant attention in dental research, with substantial progress made in the study of dentin biomimetic mineralization. Given ACP's instability without additives, the presence of ACP stabilizers is crucial for achieving in vitro intrafibrillar mineralization. However, there is a lack of comprehensive and exhaustive reviews on ACP bioactive materials under the regulation of stabilizers. A detailed summary of these stabilizers is also instrumental in better understanding the complex process of intrafibrillar mineralization. Compared to traditional remineralization methods, bioactive materials capable of regulating ACP stability and controlling release demonstrate immense potential in enhancing clinical treatment standards.
Remineralization of Artificial Dentin Caries Using Dentin and Enamel Matrix Proteins

Materials

Abstract:To assess the remineralizing potential of dentin matrix proteins and enamel matrix derivatives (DMPs and EMDs) after application on artificially induced dentin lesions, given the hypothesis that these materials increase the mineral uptake, binding, and mineralization. Forty-eight caries-free human premolars were used. Teeth were cut, polished, and embedded, leaving an open window on the root surface, of which one-third was covered with a flowable composite to preserve the healthy untreated dentin. Then, samples were demineralized in Buskes solution for 33 days. A micro-CT scan prior to treatment was performed. Next, the samples were randomly allocated into four groups: (A) An untreated negative control (CON), (B) application of porcine dentin matrix proteins (DMP), (C) treatment with enamel matrix derivatives (EMD, Emdogain, Straumann), and (D) amine fluoride application (AMF, Elmex fluid, GABA). All samples were placed in artificial saliva for 21 days. A second micro-CT scan was performed, after which the change in gray scaling within a defined region of interest (0.25 mm3) was analyzed. ANCOVA was applied to discover statistical differences between the different treatments. Both, treatment with AMF; (P = 0.011 versus CON) as well as with DMP (P = 0.043 versus CON) yielded a statistically significant difference compared to the control treatment. EMD treatment was not found to differ (P > 0.05). Mainly the top layer of the defects showed clear signs of remineralization, which was also evident in CON. This study was able to visually confirm the remineralization potential of demineralized dentin especially after DMP application, which, however, did not outperform AMF. Based on this, additional studies combining proteins and fluorides are now warranted and ongoing.

Abstract:The disequilibrium between demineralization and remineralization of teeth, especially of dentin, may lead to serious consequences like dental caries that are considered to affect people's quality of life. The employment of biomimetic analogs of proteins to duplicate biomineralization, which is a well-regulated process mediated by extracellular matrix proteins, may provide new insights to solve these problems. Here we report the use of a modified multifunctional dendrimer, synthesized by the introduction of phosphate groups via a Mannich-type reaction onto poly(amidoamine) (PAMAM) dendrimers, to biomimetically remineralize dentin. The phosphorylated PAMAM dendrimers were demonstrated to act, along with an amorphous calcium phosphate stabilizing agent, polyacrylic acid (PAA), as biomimetic analogs of noncollagenous proteins to induce the remineralization of demineralized dentin. Phosphorylated PAMAM dendrimers treated demineralized dentin discs were immersed in a remineralizing solution containing PAA for up to 7 days. The success of this remineralization was examined using attenuated total reflection Fourier-transform infrared spectroscopy (ATR-FTIR), X-ray diffraction (XRD), energy-dispersive X-ray spectroscopy (EDS) and electron microscopy. These results showed that demineralized dentinal collagen fibrils were successfully phosphorylated by the treatment of phosphorylated PAMAM dendrimers and embedded with calcium-deficient hydroxyapatite after remineralization. The surfaces of demineralized dentin discs were covered with newly induced crystals and the patent dentinal tubules were occluded. A good biocompatibility was also determined. Thus, phosphorylated PAMAM dendrimers could be applied as a minimally invasive method of management of dentin caries, employed to improve the resin-dentin bonding stability and also be used in the treatment of dentin hypersensitivity.

Abstract:Biomineralization of collagen fibers is regulated by non-collagenous proteins and small biomolecules, which are essential in bone and teeth formation. In particular, small biomolecules such as succinic acid (SA) exist at a high level in hard tissues, but their role is yet unclear. Here, our work demonstrated that SA could significantly promote intrafibrillar mineralization in two- and three-dimensional collagen models, where the relative mineralization rate was 16 times faster than the control group. Furthermore, the FTIR spectra and isothermal experimental results showed that collagen molecules could interact with SA via a hydrogen bond and that the interaction energy was about 4.35 kJ mol-1. As expected, the SA-pretreated demineralized dentin obtained full remineralization within two days, whereas it took more than four days in the control group, and their mechanical properties were considerably enhanced compared with those of the demineralized one. The possible mechanism of the promotion effect of SA was ultimately illustrated, with SA modification strengthening the capacity of the collagen matrix to attract more calcium ions, which might create a higher local concentration that could accelerate the mineralization of collagen fibers. These findings not only advance the understanding of the vital role of small biomolecules in collagen biomineralization but also facilitate the development of an effective strategy to repair hard tissues.

Abstract:Proanthocyanidin (PA) has demonstrated promise as a dental biomodifier for maintaining dentin collagen integrity, yet there is limited evidence regarding its efficacy in dentin repair. The aim of this study was to investigate the effect of PA on dentin remineralization through the polymer induced liquid precursor (PILP) process, as well as to assess the mechanical properties of the restored dentin. Demineralized dentin was treated with a PA-contained remineralization medium, resulting in the formation of PA-amorphous calcium phosphate (ACP) nanoparticles via the PILP process. The kinetics and microstructure of remineralized dentin were examined through the use of Fourier transform infrared spectroscopy(FTIR), attenuated total reflectance-FTIR, scanning electron microscopy, transmission electron microscopy. The results showed that the application of PA facilitated the process of dentin remineralization, achieving completion within 48 h, demonstrating a notable reduction in time required. Following remineralization, the mechanical properties of the dentin exhibited an elastic modulus of 15.89 ± 1.70 GPa and a hardness of 0.47 ± 0.08 GPa, which were similar to those of natural dentin. These findings suggest that combining PA with the PILP process can promote dentin remineralization and improve its mechanical properties, offering a promising new approach for dentin repair in clinical practice.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Objective</h3><p>To assess the ultrastructural change of demineralized dentin collagen during calcium phosphate polymer-induced liquid precursor (Ca/P-PILP) mediated remineralization process and to evaluate the biomimetic remineralization potential of high concentration Ca/P-PILP at demineralized artificial caries dentin lesion, additionally to investigate the bond interfacial integrity as well as the bonding strength of the biomimetic remineralized artificial caries dentin lesion.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Methods</h3><p>Demineralized dentin collagen of 5 μm thick was biomimetically remineralized with low, medium concentration Ca/P-PILP for 10 days and high concentration Ca/P-PILP for 10, 15, 20 days. Artificial caries dentin lesion at a thickness of 150 ± 50 μm was biomimetically remineralized with high concentration Ca/P-PILP for 20 days. The biomimetic remineralization of demineralized dentin collagen was observed by scanning electron microscopy (SEM). The biomimetic remineralization intensity and depth of artificial caries dentin lesion was assessed by Electron Probe Micro Analyzer (EPMA). The bonding interfacial integrity between remineralized artificial caries dentin and composite resin was observed by Swept-source optical coherence tomography (SS-OCT) and the bonding strength of remineralized artificial caries dentin was evaluated by micro-tensile bond strength analysis (μTBS).</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>Solely PAA-PASP solution and solely saturated Ca/P solution can't achieve dentin collagen remineralization. Increased concentration of Ca/P-PILP and prolonged remineralization time can enhance the biomimetic remineralization intensity of demineralized dentin collagen. After treating with high concentration Ca/P-PILP, a 150 ± 50 μm thick layer of demineralized artificial caries dentin lesion was not fully remineralized, and the biomimetic remineralization intensity reached up to 88.0%. Furthermore, a better bonding interfacial integrity with less microgap and increased bond strength at both baseline level and aging level were observed when artificial caries dentin lesion was biomimetically remineralized with high concentration Ca/P-PILP.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Significance</h3><p>Biomimetic remineralization of demineralized caries dentin lesion promotes its clinical properties for resin composited adhesive restoration.</p>

Abstract:Remineralization of enamel plays a crucial role in the progression of carious process and the management of early caries lesion. Based on the influence of phosphorylated proteins in biomineralization, the objective of this study was to synthesize nano-complexes of phosphorylated chitosan and amorphous calcium phosphate (Pchi–ACP), and evaluate their ability to remineralize enamel subsurface lesions in vitro. Pchi was synthesized using a previously established chemical method. The biomimetic remineralizing solution containing nano-complexes of Pchi–ACP was prepared by adding CaCl 2 and K 2 HPO 4 into Pchi–ACP solution (0.5 % w/v) in sequence. The final concentrations of calcium and phosphate ions were 10 and 6 mM, respectively. The nano-complexes of Pchi–ACP were characterized by Fourier-transform infrared (FTIR), X-ray diffraction (XRD), transmission electron microscopy (TEM), and selected area electron diffraction (SAED). During testing the enamel lesions were treated with Pchi–ACP and fluoridated remineralizing solutions, respectively. The remineralizing of enamel lesions was examined with field emission electron microscope (FE-SEM) and Micro-CT. ACP was stabilized by Pchi to form nano-complexes that were soluble in water. The size of Pchi–ACP nano-complexes particles was determined to be less than 50 nm. XRD and SAED results confirmed their amorphous phases. FE-SEM and Micro-CT results showed that the remineralizing effect of Pchi–ACP on enamel lesions was similar to that of fluoride. However, the remineralizing rate of Pchi–ACP treatment was significantly higher than that of fluoride treatment ( P < 0.05). This study highlighted the potential of nanoparticles functionalized with a natural analogue involved in biomineralization, to remineralize early enamel caries.
