Abstract:beta-Amyloid (A beta) plaques in the blood vessels of the brain are associated with cerebral amyloid angiopathy (CAA), which is a common cause of stroke and vascular diseases. Imaging agents that can differentiate between A beta plaques in the brain and those on the walls of cerebrovascular vessels will provide non-invasive biomarkers to interrogate the pathogenesis of CAA and give insights into the mechanisms, significance and impact of A beta-CAA for developing effective therapies for CAA and stroke. A new series of Tc-99m-labeled benzothiazole and stilbene derivatives with positive charge were developed and evaluated for selectively targeting A beta plaques in the blood vessels of the brain. The rhenium complexes 6, 7, 13 and 14 displayed medium binding affinity to A beta(1-42) aggregates with K-i values of 162, 37, 366 and 78 nM, respectively. In vitro fluorescence staining of 7 and 14 demonstrated an intense labeling of A beta plaques associated with blood vessel walls on brain sections of a patient with Alzheimer's diseases. A relatively low initial brain uptake for [Tc-99m]7 and [Tc-99m]14, 0.18 and 0.24 ID% per gram, respectively, suggests that they may be useful SPECT imaging agents for selectively detecting A beta plaques associated with cerebral vessels in the living human brain.
99mTc(CO)3-Labeled Benzothiazole Derivatives Preferentially Bind Cerebrovascular Amyloid: Potential Use as Imaging Agents for Cerebral Amyloid Angiopathy

Abstract:Cerebral amyloid angiopathy (CAA) is a disorder affecting the elderly that is characterized by amyloid-beta (A beta) deposition in blood vessel walls of the brain. A series of Tc-99m(CO)3-labeled benzothiazole derivatives as potential SPECT imaging probes for cerebrovascular A beta deposition is reported. Rhenium surrogate displayed high affinities to A beta aggregates with Ki values ranging from 106 to 42 nM, and they strongly stained A beta deposits in transgenic mice (Tg) and Alzheimers disease (AD) patients. In vitro autoradiography on brain sections of Tg and AD patients confirmed that [Tc-99m]24 possessed sufficient affinity for A beta plaques, and [Tc-99m]24 could only label A beta deposition in blood vessels but not A beta plaques in the parenchyma of the brain of AD patients. Moreover, [Tc-99m]24 possessed favorable initial uptake (1.21% ID/g) and fast blood washout (blood2 min/blood60 min = 23) in normal mice. These preliminary results suggest that [Tc-99m]24 may be used as an A beta imaging probe for the detection of CAA.

Abstract:Timely diagnostic imaging plays a crucial role in managing cerebral amyloid angiopathy (CAA)-the condition in which amyloid β is deposited on blood vessels. To selectively map these amyloid plaques, we have designed amyloid-targeting ligands that can effectively complex with 68Ga3+ while maintaining good affinity for amyloid β. In this study, we introduced novel 1,4,7-triazacyclononane-based bifunctional chelators (BFCs) that incorporate a benzothiazole moiety as the Aβ-binding fragment and form charged and neutral species with 68Ga3+. In vitro autoradiography using 5xFAD and WT mouse brain sections (11-month-old) suggested strong and specific binding of the 68Ga complexes to amyloid β. Biodistribution studies in CD-1 mice revealed a low brain uptake of 0.10-0.33% ID/g, thus suggesting 68Ga-labeled novel BFCs as promising candidates for detecting CAA.

Abstract:In this study, we synthesized four novel (AlF)-F-18/19-labeled 2-phenylbenzothiazole derivatives conjugated to 1,4,7-triazacyclononane-1,4-diacetic acid via alkyl linkers and evaluated them as imaging agent targets to amyloid-beta (N beta) plaques deposited in the blood vessels of cerebral amyloid angiopathy (CAA) brain. The four ligands exhibited moderate-to-high binding ability to A beta(1-42) aggregates, of which complex 17 possessing the most potent affinity (K-i = 11.3 nM) was selected for further biological evaluations. In vitro fluorescent staining and in vitro autoradiography studies on brain sections from CAA patients proved that this ligand could label A beta deposits in blood vessels selectively. In biodistribution study, [F-18] 17 can hardly penetrate the blood-brain barrier (brain(2) (min) = 0.3% ID/g) and displayed a rapid blood washout rate (blood(2 min)/blood(60 min) = 25.2), which is favorable as CAA imaging agents. In conclusion, this (AlF)-F-18-labeled 2-phenyffienzothiazole complex was developed and proved to be a promising CAA positron emission tomography agent.

Abstract:Four Tc-99m-labeled dibenzylideneacetone derivatives and corresponding rhenium complexes were successfully synthesized and biologically evaluated as potential imaging probes for A beta plaques using SPECT. All rhenium complexes (5a-d) showed affinity for A beta((1-42)) aggregates (K-i = 13.6-120.9 nM), and selectively stained the A beta plaques on brain sections of transgenic mice. Biodistribution in normal mice revealed that [Tc-99m]5a-d exhibited moderate initial uptake (0.31%-0.49% ID/g at 2 min) and reasonable brain washout at 60 min post-injection. Although additional optimizations are still needed to facilitate it's penetration through BBB, the present results indicate that [Tc-99m]5a may be a potential SPECT probe for imaging A beta plaques in Alzheimer's brains. Abstract:An array of complexes, Tc-99m/Re-labeled cyclopentadienyl tricarbonyl and 2-phenyl/pyridylbenzothiazoles, conjugated through an ester linkage were tested as potential beta-amyloid (A beta) probes for SPECT imaging. The [CpRe/Tc-99m(CO)(3)] complexes were prepared by double ligand transfer reactions from ferrocene precursors, and the X-ray structure of one rhenium surrogate revealed a classical "piano stool" like geometry. Several of the rhenium complexes displayed high affinity for A beta(1-2) aggregates in in vitro inhibition assays, and they could intensely stain Ap deposits on brain sections from transgenic mice and Alzheimer's disease (AD) patients. Complex [Tc-99m]4h strongly binds to Ap deposits in blood vessels of the brain section of AD patients in in vitro autoradiography. Biodistribution experiments in normal mice revealed that Tc-99m-labeled tracers exhibited moderate degree of initial brain uptake (0.54% -1.06% ID/g at 2 min). These tracers targeting A beta plaques of AD patients warrant further structure optimization to improve their ability to penetrate blood-brain barrier, moreover, they may be suitable for developing imaging probes for targeting amyloid aggregates outside of the brain. Abstract:Two uncharged 99mTc-labeled phenylbenzoxazole derivatives were biologically evaluated as potential imaging probes for β-amyloid plaques. The 99mTc and corresponding rhenium complexes were synthesized by coupling monoamine–monoamide dithiol (MAMA) and bis(aminoethanethiol) (BAT) chelating ligand via a pentyloxy spacer to phenylbenzoxazole. The fluorescent rhenium complexes 6 and 9 selectively stainined the β-amyloid plaques on the sections of transgenic mouse, and showed high affinity for Aβ(1–42) aggregates (Ki=11.1nM and 14.3nM, respectively). Autoradiography in vitro indicated that [99mTc]6 clearly labeled β-amyloid plaques on the sections of transgenic mouse. Biodistribution experiments in normal mice revealed that [99mTc]6 displayed moderate initial brain uptake (0.81% ID/g at 2min), and quickly washed out from the brain (0.25% ID/g at 60min). The preliminary results indicate that the properties of [99mTc]6 are promising, although additional refinements are needed to improve the ability to cross the blood–brain barrier.

Abstract:Based on the conjugate strategy, two neutral 99mTc labeled 2-(1-(6-(dialkylamino)naphthalen-2-yl)ethylidene)malononitrile (DDNP) and 1-(6-(dialkylamino)naphthalen-2-yl)ethanone (ENE) derivatives, and their corresponding rhenium complexes were synthesized. In vitro fluorescent staining indicated that the corresponding rhenium derivatives selectively stained the β-amyloid (Aβ) plaques in the brain sections of AD model mice with low background. Compared with FDDNP and FENE, the affinities of the corresponding rhenium derivatives to Aβ aggregates decreased about 10–14-fold. In vivo biodistribution experiments in normal mice showed that 99mTc-MAMA-ENE displayed medium initial brain uptake (0.65%ID/g at 2min) with a reasonable washout from the brain (0.19%ID/g at 2h) while 99mTc-MAMA-DDNP showed a low brain uptake (0.28%ID/g at 2min). Further optimize these 99mTc-labeled tracers in order to improve their binding affinities to Aβ plaques and diffusion through the blood brain barrier may generate useful imaging agents for SPECT.

Abstract:A series of dibenzylideneacetones were synthesized and evaluated as imaging probes for β-amyloid plaques. They displayed high binding affinity to Aβ(1-42) aggregates (K(i) = 6.4 for 8, K(i) = 3.0 for 9), and the high binding were confirmed by in vitro autoradiography with AD human and transgenic mouse brain sections. Two of them were selected for (18)F-labeling directly on the benzene ring. In biodistribution experiments, [(18)F]8 and [(18)F]9 displayed high initial uptakes (9.29 ± 0.41 and 5.38 ± 0.68% ID/g) and rapid washouts from the normal brain (brain(2 min)/brain(60 min) ratios of 21.6 and 13.4). These preliminary results suggest that [(18)F]8 and [(18)F]9 may be used as potential PET imaging agents for the detection of Aβ plaques in the brain.
Synthesis and Biodistribution of 125I-Labelled Benzothiazole Derivatives As Β-Amyloid Imaging Agents

Abstract:In order to develop ~(123)I labelled probe for SPET imaging of A β plaques in Alzheimer's (disease), Four ~(125)I labelled benzothiazole derivatives are synthesized and their radiochemical purity are more than 95%. The biodistribution results in normal mice indicate that the radiolabelled probes have higher initial brain uptake:10.28, 3.62, 3.33 and 3.71%/g at 2 min post i.v. injection for 3′-~(125)I-BTA,3′-~(125)I-CBTA,3′-~(125)I-BTA-Ac and 3′-~(125)I-CBTA-Ac, respectively. Brain uptake ratios at 2 min to 60 min of 3′-~(125)I-BTA,3′-~(125)I-BTA-Ac and 3′-~(125)I-CBTA-Ac are 7.3, 6.2 and 5.7 respectively. The results reveal that 3′-~(123)I-BTA is a very potential agent for SPECT imaging of A β plaques in Alzheimer's disease.
99Mtc-Labeled 2-Arylbenzothiazoles: Aβ Imaging Probes with Favorable Brain Pharmacokinetics for Single-Photon Emission Computed Tomography.

Abstract:A series of 2-arylbenzothiazole derivatives conjugated with bis(aminoethanethiol) (BAT) chelating groups were designed and synthesized. A competitive binding assay-based screening was used to select seven rhenium complexes with potent binding affinity toward A beta(1-42) aggregates (K-i < 50 nM) for Tc-99m labeling and further evaluation. The Tc-99m-labeled probes showed good affinity and specificity to A beta plaques in Tg mouse brain tissue in in vitro autoradiography studies. Moreover, [Tc-99m]14b exhibited favorable brain pharmacokinetics in normal mice (2.11% ID/g at 2 min and 0.62% ID/g at 60 min). Ex vivo autoradiography revealed extensive labeling of A beta plaques by [Tc-99m]14b in the brain of Tg mice. Furthermore, we performed the first single-photon emission computed tomography (SPECT) imaging study in nonhuman primates with Tc-99m-labeled A beta probes. The semiquantitative data showed that [Tc-99m]14b penetrated the brains of rhesus monkeys. These results indicate that [Tc-99m]14b could be utilized as a SPECT imaging probe for A beta plaques.

Abstract:Four neutral Tc-99m/Re-labeled 2-arylbenzoxazole derivatives conjugated to bis (aminoethanethiol) (BAT) chelating ligand via a short propoxy spacer were synthesized and evaluated. In vitro binding assay showed that they displayed binding affinities to A beta(1-42) aggregates (K-i = 15.86-393.18 nM). In vitro autoradiography studies further confirmed the high and specific binding of [Tc-99m]20 to beta-amyloid plaques on brain sections of transgenic mice. Biodistribution study of [Tc-99m]17-20 in normal mice displayed moderate initial brain uptake (0.96-1.55%ID/g at 2 min), and fast washed out from the brain (0.14-0.40%ID/g at 60 min), especially for [Tc-99m]20 with a brain(2min)/brain(60min) ratio of 8.86. 18F-Labeled Benzyldiamine Derivatives As Novel Flexible Probes for Positron Emission Tomography of Cerebral Β-Amyloid Plaques.

Abstract:Early noninvasive visualization of cerebral β-amyloid (Aβ) plaques with positron emission tomography (PET) is the most feasible way to diagnose Alzheimer's disease (AD). In this study, a series of flexible benzyldiamine derivatives (BDA) were proposed for binding to aggregated β-amyloid 1-42 (Aβ1-42) with high adaptability, high binding affinity (6.8 ± 0.6 nM), and rapid body excretion. The methylthio (12) and ethoxyl (10) derivatives were further labeled with 18F directly on their benzene ring and examined as PET probes for Aβ plaque imaging. [18F]12 displayed 4.87 ± 0.52% ID/g initial uptake and prompt washout from normal brain in biodistribution studies. MicroPET-CT imaging indicated sufficient retention of [18F]12 but lower white matter uptake in the brain of an AD transgenic mouse model compared with that of commercial [18F]AV-45. Our experimental results provide new insights for developing targeting ligands possessing a flexible framework for use as efficient Aβ probes for PET imaging of AD brain.
QSAR Study of 125I-Labeled 2-(4-aminophenyl)benzothiazole Derivatives As Imaging Agents for Β-Amyloid in the Brain with Alzheimer’s Disease

Abstract:In order to develop small molecular probes for imaging of A beta plaques in the brain with Alzheimer's disease, four benzothiazole derivatives as AP probes, 3'-I-125-BTA, 3'-I-121-CBTA, 3'-I-121-BTA-Ac, and 3'-I-125-CBTA-Ac were successfully prepared. The four probes displayed the following initial brain uptake: 10.28 %ID/g, 3.62 %ID/g, 3.33 %ID/g and 3.71 %ID/g at 2-minute post iv injection, respectively. The study of quantitative structure-activity relationship (QSAR) indicated that the initial brain uptake was correlated with molecule volume (V-m) and dipole moment (D-p), butthe D-p was the main factor. 3'-I-125-BTA has the highest initial brain uptake among the four A beta probes because of the smallest D-p.

Abstract:Five iodinated 2-phenyl-1H-benzo[d]imidazole derivatives were synthesized and evaluated as potential probes for β-amyloid (Aβ) plaques. One of the compounds, 4-(6-iodo-1H-benzo[d]imidazol-2-yl)-N,N-dimethylaniline (12), showed excellent affinity for Aβ1-42 aggregates (Ki=9.8 nM). Autoradiography with sections of postmortem Alzheimer's disease (AD) brain revealed that a radioiodinated probe [125I]12, labeled Aβ plaques selectively with low nonspecific binding. Biodistribution experiments with normal mice injected intravenously with [125I]12 showed high uptake [4.14 percent injected dose per gram (% ID/g) at 2 min] into and rapid clearance (0.15% ID/g at 60 min) from the brain, which may bring about a good signal-to-noise ratio and therefore achieve highly sensitive detection of Aβ plaques. In addition, [125I]12 labeled amyloid plaques in vivo in an AD transgenic model. The preliminary results strongly suggest that [125I]12 bears characteristics suitable for detecting amyloid plaques in vivo. When labeled with 123I, it may be a useful SPECT imaging agent for Aβ plaques in the brain of living AD patients.
Preparation and Biological Evaluation of Re/99 Tcm (CO) 3 +-diphenyldiazene Derivative as Candidates for Imaging Aβ Plaques in the Brain

Abstract:To develop the non-invasive and diagnostic agents for Alzheimer's disease(AD),99Tcm(CO)3+-L(L:(E)-2-(4-((4-(dimethylamino)phenyl)diazenyl)phenylamino)acetic acid,a diphenyldiazene derivative) was prepared,radiochemical purity was above 95%.Initial autoradiography results suggested that 99Tcm(CO)3+-L showed selective binding to the Aβ plaque-like structures in the brain section from the AD transgenic mice(Tg C57,APP,PS1 12-month-old),further,99Tcm(CO)3+-L showed the same binding sites with fluorescence-stained by Re(CO)3+-L.Biodistribution of 99Tcm(CO)3+-L in normal mice demonstrated low uptake in brain(5 min:(0.52 ± 0.11)% ID/g) whilst slow clearance from the brain tissues at 120 min post-injection(0.28 ± 0.04)%ID/g.The corresponding Re analogue was also synthesized,and the nature of its lowest electronically excited state was determined by studies of the UV-vis absorption and fluorescence emission properties at room temperature.In addition,the fluorescent staining of the Re-complex was performed in comparison to Thioflavin-T and autoradiography results of 99Tcm(CO)3+-L.In conclusion,these results are encouraging for further exploration of 99Tcm(CO)3+-L as a SPECT imaging agent for Aβ plaques in the AD brain.

Abstract:Alzheimer's disease (AD) is a debilitating neurodegenerative dementia that involves substantial neuronal loss. Extracellular deposition of neurotoxic beta-amyloid (A beta) plaques in the brain has been recognized as the central histological characteristic of AD. In the past decade, precise detection of the A beta plaques at preclinical AD with positron emission tomography (PET) or single photon emission computed tomography (SPECT) has achieved continued development. A big category of A beta imaging agents was benzoheterocycles which derived from Thioflavin-T (ThT), a traditional amyloid binding dye. This review summarizes the past and current status of radioactive benzoheterocycles designed to selectively bind to A beta plaques. Separate sections discuss the chemical synthesis, in vitro and in vivo investigations of radiolabeled benzothiazole, benzoxazole, benzofuran, benzothiophene, indole, imidazopyridine and quinoxaline analogs to act as PET/SPECT candidates for imaging A beta plaques. Synthesis and evaluation of novel benzothiazole derivatives based on the bithiophene structure as potential radiotracers for β-amyloid plaques in Alzheimer’s disease

Abstract:In this study, six novel benzothiazole derivatives based on the bithiophene structure were developed as potential β-amyloid probes. In vitro binding studies using Aβ aggregates showed that all of them demonstrated high binding affinities with Ki values ranged from 0.11 to 4.64nM. In vitro fluorescent staining results showed that these compounds can intensely stained Aβ plaques within brain sections of APP/PS1 transgenic mice, animal model for AD. Two radioiodinated compounds [125I]-2-(5′-iodo-2,2′-bithiophen-5-yl)-6-methoxybenzo[d]thiazole [125I]10 and [125I]-2-(2,2′-bithiophen-5-yl)-6-iodobenzo[d]thiazole [125I]13 were successfully prepared through an iododestannylation reaction. Furthermore, in vitro autoradiography of the AD model mice brain sections showed that both [125I]10 and [125I]13 labeled the Aβ plaques specifically with low background. In vivo biodistribution studies in normal mice indicated that [125I]13 exhibited high brain uptake (3.42% ID/g at 2min) and rapid clearance from the brain (0.53% ID/g at 60min), while [125I]10 showed lower brain uptake (0.87% ID/g at 2min). In conclusion, these preliminary results of this study suggest that the novel radioiodinated benzothiazole derivative [125I]13 may be a candidate as an in vivo imaging agent for detecting β-amyloid plaques in the brain of AD patients.

Abstract:Technetium-99m-labeled cyclopentadienyl tricarbonyl complexes conjugated with the 2-phenylbenzothiazole binding motif were synthesized. The rhenium surrogates 20, 21, 22 and 23 were demonstrated to have moderate to high affinities for A beta(1-42) aggregates with K-i values of 142, 76, 64 and 24 nM, respectively. During the fluorescence staining of brain sections of transgenic mice and patients with Alzheimer's disease, these rhenium complexes demonstrated perfect and intense labeling of A beta plaques. Moreover, in in vitro autoradiography, Tc-99m-labeled complexes clearly detected beta-amyloid plaques on sections of brain tissue from transgenic mice, which confirmed the sufficient affinity of these tracers for A beta plaques. However, these compounds did not show desirable properties in vivo, especially showing poor brain uptake (below 0.5% ID g(-1)), which will hinder the further development of these tracers as brain imaging agents. Nonetheless, it is encouraging that these Tc-99m-labeled complexes designed by a conjugate approach displayed sufficient affinities for A beta plaques.
Preliminary Characterization and In Vivo Studies of Structurally Identical 18 F- and 125 I-Labeled Benzyloxybenzenes for PET/SPECT Imaging of β-Amyloid Plaques

Abstract:With the assistance of molecular docking and 3D-QSAR models established previously, structurally identical 18 F- and 125 I-labeled benzyloxybenzene derivatives were designed to achieve the early detection of Aβ plaques by PET/SPECT imaging. In competition binding assay, ligands 7a and 12a displayed high binding affinities to Aβ 42 aggregates with K i values of 19.5 nM and 23.9 nM, respectively. Specific plaque labeling was observed on the in vitro autoradiography of brain sections from AD patients and Tg mice. In biodistribution, [ 125 I]7a, [ 18 F]7a, [ 125 I]12a and [ 18 F]12a all exhibited high initial brain uptakes (>5% ID/g at 2 min). [ 125 I]7a and [ 125 I]12a cleared fast from the normal brain regions, while corresponding [ 18 F]7a and [ 18 F]12a showed slow washout rates. Dynamic microPET/CT and microSPECT/CT imaging data in normal ICR mice were in accordance with in vivo biodistribution results. In vivo metabolism results indicated that the different clearance profiles between the structurally identical 18 F- and 125 I-labeled tracers could be attributed to different biochemical characteristics of the radiometabolites. Radioiodinated benzyloxybenzene derivatives exhibited good in vivo biostability in brain. Ex vivo autoradiography further confirmed the strong in vivo Aβ labeling ability of [ 125 I]7a. These new fluorinated and iodinated benzyloxybenzenes can develop into PET/SPECT dual imaging agents targeting Aβ plaques.
