P1–115: L‐type Calcium Channel Blockers Rescue Beta‐amyloid‐42–induced Neurotoxicity by Inhibiting CREB and CDK5 Activation

Abstract:The patients of Alzheimer's disease show a continuous decline in memory and a progressive neurodegeneration in cerebral cortex, hippocampus and other kind of neurons. Alzheimer's disease characterized by two pathological hallmarks: amyloid plaques and neurofibrillary tangles. It has been proved that calcium homeostasis would be disrupted in human aging. Recently research show us that dense plaques have functional contact with the alteration of calcium signal in AD mice. Calcium channel blockers also produced positive effects in treating with Alzheimer's disease. Calcium channels basically can be divided into voltage-gated calcium channel and N-methyl-D-aspartate receptor channel, and voltage-gated calcium channel mainly belongs to type L, type Q, type P, type N and so on. Primary cell culture, Cell survival assay, Intracellular calcium assay, Quantitative real-time PCR(qRT-PCR), Cell harvesting, Protein quantification, Western blotting and statistical analysis. Here we demonstrate that elevation of [Ca 2+]i in newborn SD rats primary cells activates CDK5 and the transcription factor cAMP response element binding protein (CREB) by constitutive CaM kinase IV(CaMK IV) phosphorylation. Inhibition of increasing [Ca 2+]i by L-type Ca 2+ channel (LTCC) blockers nimodipine and verapamil each abolish CDK5 and CREB phosphorylation and transcriptional activity of CREB target gene including BDNF, c-fos and nNOS. The interference of LTCC-CaMKIV-CREB pathway caused by verapamil and nimodipine can significantly reduce cell death and render neuroprotective effects. This study shows that Aβ oligomers are associated with increased intracellular Ca 2+, while L-type calcium channel blockers can prevent such an influx and protect cells. Furthermore, our results suggest the possibility that L-type calcium channel clockers rescue Aβ42-induced neurotoxicity by inhibiting CREB and CDK5 activation.
