Abstract:Several new 1,3,4-oxadiazole derivatives containing a pyrazole ring were designed and synthesized from ethyl acetoacetate and triethyl orthoformate as starting materials via multi-step reactions. The compound structures were confirmed by melting point, 1 H NMR and HRMS. They were evaluated for fungicidal and herbicidal activities. Four of the compounds exhibited moderate fungicidal activity against Colletotrichum species. Most of the compounds had moderate-to-good activity as a herbicide.

Abstract:The one-pot U-4CR and intramolecular O-alkylation sequence starting from 2-aminophenols in combination with α-bromoalkanoic acids, aldehydes, and isocyanides under controlled microwave heating has been established for a rapid access to highly functionalized 3,4-dihydro-3-oxo-2H-1,4-benzoxazines. With appropriate substitutions on the 1,4-benzoxazines, a microwave-assisted Cu-catalyzed intramolecular amidation was performed to furnish a novel class of heterocyclic conjugates of 1,4-benzoxazines with a 2-oxindole linked through a C–N single bond.

Abstract:In order to develop a novel herbicide containing the β-triketone motif, a series of 4-hydroxyl-3-(2-phenoxyacetyl)-pyran-2-one derivatives were designed and synthesized. The bioassay results showed that compound II15 had good pre-emergent herbicidal activity even at a dosage of 187.5 g ha-1. Moreover, compound II15 showed a broader spectrum of weed control when compared with a commercial herbicide 2,4-dichlorophenoxyacetic acid (2,4-D), and displayed good crop safety to Triticum aestivum L. and Zea mays Linn. when applied at 375 g ha-1 under pre-emergence conditions, which indicated its great potential as a herbicide. More importantly, studying the molecular mode of action of compound II15 revealed that the novel triketone structure is a proherbicide of its corresponding phenoxyacetic acid auxin herbicide, which has a herbicidal mechanism similar to that of 2,4-D. The present work indicates that the 4-hydroxyl-3-(2-phenoxyacetyl)-pyran-2-one motif may be a potential lead structure for further development of novel auxin-type herbicides.

Abstract:A series of novel 1,2,4-triazolo[4,3-a]pyridin-3(2H)-one derivatives were synthesized and identified by (1)H NMR, single crystal X-ray diffraction, elemental analysis or HRMS, and their herbicidal activities were determined at different concentrations. Abstract:To discover new acetyl-CoA carboxylase (ACCase) inhibiting-based herbicides, twenty-nine novel quinazolin-4(3H)-one derivatives were designed and synthesized based on the aryloxyphenoxypropionate motif. The bioassay results showed that most of the target compounds showed better pre-emergent herbicidal activity against monocotyledonous weeds in a greenhouse. Especially, when applied at 375 g ha−1 under pre-emergence conditions, compound QPP-7 displayed excellent herbicidal activity against monocotyledonous weeds (i.e., E. crusgalli, D. sanguinalis, P. alopecuroides, S. viridis, E. indica, A. fatua, E. dahuricu, S. alterniflora) with inhibition rate >90%, and displayed excellent crop safety to O. sativa, T. aestivum, G. spp, and A. hypogaea. The study of structure-activity relationship (SAR) revealed that the herbicidal activity of target compounds is strongly influenced by the spatial position of R group and the bulk of R1 group on quinazolin-4(3H)-one, and the (R = 6-F, R1 = Me) pattern is confirmed as the optimal orientation. Furthermore, the molecular docking study and the good inhibitory activity of QPP-7 against E. crusgalli ACCase enzyme (IC50 = 54.65 nM) indicated that it may be a ACCase inhibitor. Taken together, the present work demonstrated that compound QPP-7 could serve as a potential lead structure for further developing novel ACCase inhibiting-based herbicide.

Abstract:Eight new 5,7-disubstituted-2-(5-aryl-2-furoylimino)-[1,2,4]-thiadiazolo[2,3-alpha] pyrimidine derivatives were synthesized under ultrasonic irradiation. All of them are new compounds and their structures were exactly confirmed by IR, H-1 NMR spectra and elemental analysis. Our method using the ultrasonic irradiation under the condition of phase transfer catalyst (PTC) has the advantages of short reaction time and good yield. The preliminary biological tests show that some of the target compounds have some inhibitory activities against weeds.

Abstract:In order to develop novel herbicides, a series of novel acylthiourea derivatives containing methylsulfone were synthesized and characterized by melting point, elemental analysis, infrared spectroscopy, mass spectrometry and 1H NMR spectroscopy. Furthermore, the preliminary herbicidal activity of title compounds was determined against Echinochloa crusgal, Polypogon fugax, Digitaria adscendens, Brassica napus, Amaranthus retroflexus and Abutilon theophrasti. The results of the petri dish experiments showed that a small part of title compounds had a good inhibitory effect on Digitaria adscendens and Brassica napus. The results of the greenhouse herbicidal activity experiments indicated that the compounds 4p, 4w and 4x exhibited more high herbicidal activity against Brassica napus and Digitaria adscendens than the commercial herbicide bensulfuron-methyl. Moreover, except for compound 4i, all test compounds performed well in crop safety evaluation. Next, acetohydroxyacid synthase (AHAS) enzyme activity experiments were carried out, and the results showed that compound 4x had the highest inhibition rate reaching 89.12%. Finally, in order to screen AHAS inhibitor, molecular docking was performed by using AHAS protein and target ligand molecules 4a-4x. The results of molecular docking were similar to bioassay experiments. Compound 4p, 4w and 4x were considered potential AHAS inhibitors.

Abstract:A series of novel 1,2,4‐triazolo[4,3‐a]pyridines were synthesized, and their structures were characterized by 1H NMR, MS, elemental analysis, and single‐crystal X‐ray diffraction analysis. The antifungal activities were evaluated. The antifungal activity results indicated that the compound 2b, 2g, 2p, and 2i exhibited good activities. The activity of compound 2b, 2g, 2p, and 2i can compare with the commercial pesticide. The 3D‐QSAR model was developed using CoMFA method. Both the steric and electronic field distributions of CoMFA are in good agreement in this work and will be very helpful in designing a new set of analogues.

Abstract:A novel series of novel 1,2,4-triazolo[4,3-a]pyridines were designed and synthesized under microwave irradiation condition. The structures were identified by H-1 NMR, MS and elemental analysis, and the antifungal activities of them were evaluated at 100 ppm. It was found that some of the 1,2,4-triazolo[4,3-a] pyridines displayed good antifungal activity. Among them, compound 2e exhibited good antifungal activity against Stemphylium lycopersici (Enjoji) Yamamoto and Fusarium oxysporum. Sp. Cucumebrium.

Abstract:A practical method of four-step synthesis towards novel (E)-5-(methoxyimino)-3,5-dihydrobenzo[e][1,2]oxazepin-4(1H)-one antifungals is presented, where a commercially available pesticide and pharmacology intermediate, (E)-methyl 2-(2-(bromomethyl)phenyl)-2-(methoxyimino)acetate (1), was used as starting material. These compounds were confirmed by 1H NMR, 13C NMR, high-resolution mass spectroscopy and X-ray crystal structure. Via in vitro fungicidal evaluation, the moderate to high activities of several compounds against eight phytopathogenic fungi were demonstrated. Especially, the fungicidal activities of compounds 5-03 and 5-09 were comparable to those of the controls azoxystrobin and trifloxystrobin in precise virulence measurements for four fungi. These results suggested that dihydrobenzo[e][1,2]oxazepin-4(1H)-one analogues could be considered as potential fungicidal candidates for crop protection.

Abstract:A series of 3-aminocarbonyl-2-oxazolidinethione derivatives containing a substituted pyridine ring were designed and synthesized. The structures of all of the title compounds were characterized by 1H NMR, 13C NMR, IR, and HRMS. Their agricultural bioactivities were evaluated, and some of these compounds exhibited good herbicidal activities against Echinochloa crusgalli, Sorghum vulgare, Digitaria sanguinalis, Eclipta prostrasta, Cucumis sativus, and Brassica campestris, which were associated mainly with their steric properties and lipophilicities based on the structure-activity relationship discussion.

Abstract:A series of novel thiochroman-4-one derivatives incorporating carboxamide and 1, 3, 4-thiadiazole thioether moieties were synthesized. Bioassay results indicated that the EC50 values of compound 6-chloro-N-(5-(methylthio)-1, 3, 4-thiadiazol-2-yl)-4-oxothiochromane-2-carboxamide (5a) against Xanthomonas oryzae pv. Oryzae (Xoo) and Xanthomonas axonopodis pv. Citri (Xac) were 24 and 30 μg/mL, respectively, which were even better than those of bismerthiazol and thiadiazole copper. Meanwhile, compound 6-methyl-4-oxo-N-(5-(propylthio)-1, 3, 4-thiadiazol-2-yl)thiochromane-2-carboxamide (5m) showed a better antifungal activity against Botrytis cinerea (B. cinerea), with an inhibition rate of 69%, than carbendazim. As far as we know, this is the first report on the antibacterial and antifungal activities of this series of novel thiochroman-4-one derivatives incorporating carboxamide and 1, 3, 4-thiadiazole thioether moieties.

Abstract:In the face of increasing resistance to the currently used commercial herbicides and the lack of success in identifying new herbicide targets, alternative herbicides need to be developed to control unwanted monocotyledon grasses in food crops. Here, a panel of 29 novel sulfonylurea-based compounds with ortho-fluoroalkoxy substitutions at the phenyl ring were designed and synthesized. Pot assays demonstrated that two of these compounds, 6d and 6u, have strong herbicidal activities against Echinochloa crus-galli, Eleusine indica, Alopecurus aequalis, and Alopecurus japonicus Steudel at a dosage of 15 g ha-1. Furthermore, these two compounds exhibited <5% inhibition against wheat at a dosage of 30 g ha-1 under post-emergence conditions. 6u also exhibited <5% inhibition against rice at a dosage of 30 g ha-1 under both post-emergence and pre-emergence conditions. A kinetics study demonstrated that 6d and 6u are potent inhibitors of Arabidopsis thaliana acetohydroxyacid synthase (AHAS; EC 2.2.1.6) with potent Ki values of 18 ± 1.1 and 11.9 ± 4.0 nM, respectively. The crystal structure of 6u in complex with A. thaliana (At)AHAS has also been determined at 2.7 Å resolution. These new compounds represent new alternative herbicide choices to protect wheat or rice from invading grasses.

Abstract:Herein, a series of novel 4-chromanone-derived compounds incorporating an acylhydrazone moiety was synthesized and their structures were confirmed using 1 H NMR, 13 C NMR, and HRMS technologies. Bioassay results demonstrated that some of the target compounds exhibited moderate to good antibacterial activities against Xanthomonas oryzae pv. oryzae ( Xoo ) and Xanthomonas oryzae pv. oryzicolaby ( Xoc ), as well as antifungal activities against Lasiodiplodia theobromae ( L. theobromae ) and Fusarium proliferatum ( F. proliferatum ).

Abstract:Nine novel types of 2-(1,2-benzisothiazol-3-yloxy)-N-(1-aryl-3-cyanopyrazol-5-yl)-acetamides were synthesized, and their inhibition effects on selected bacteria (heterotrophic bacteria) and algae (marine chlorella) were evaluated. Results showed that 2-(1,2-benzisothiazol-3-yloxy)-N-(3-cyano-1-(2,4-dimethylphenyl)pyrazol-5-yl) acetamide achieved the highest yield of 81% with good bioactivity against heterotrophic bacteria and marine chlorella.
