O2‐12‐04: Extracellular Domain of P75ntr Attenuates Beta‐amyloid‐induced Neurotoxicity and Tau Hyperphosphorylation

Abstract:Amyloid-beta peptide (Aβ) plays critical roles in the development of Alzheimer's disease (AD), causing neuronal death, neuritic degeneration and dysfunction in synaptic transmission via the neurotrophin receptor p75NTR pathway. Aβ also induces tau hyperphosphorylation, another critical event in AD pathogenesis, but the pathway for this event remains unclear. The present study was to validate the use of p75NTR extracellular domain to block Aβ neurotoxicity via blocking binding of Aβ to p75NTR. The gene for extracellular domain of human p75NTR (p75NTR-ECD) was cloned and fused with Fc domain of human IgG. Recombinant p75NTR-ECD was expressed in and purified from E.Coli. Human SH-SY5Y neuroblastoma cells were cultured in medium containing various concentration of Aβ, with or without p75NTR-ECD. The cell viability was measured with MTT assay, neurite growth was examined by measuring the length of neurites, and tau phosphorylation was investigated with Western blot. p75NTR expression was detected in SH-SY5Y cells with immunohistochemistry and Western blotting. Aβ oligomers induced neuron death, neurite degeneration and tau hyperphosphorylation of SH-SY5Y cells at different doses. Addition of p75NTR-ECD to the culture medium rescued the Aβ induced cell death, attenuated neurite growth inhibition and tau hyperhposphorylation in a dose dependent manner. p75NTR might mediate Aβ-induced tau hyperphosphorylation. p75NTR-ECD can block Aβ-induced neurotoxicity and tau hyperphosphorylation. Our previous study found that p75NTR-ECD can inhibit Aβ aggregation and disaggregate Aβ fibrils. Given that p75NTRE-ECD is physiologically shedded from membrane, p75NTR-ECD is endogenous protective molecule against Aβ, and is a promising therapeutic molecule with multiple functions targeting Aβ neurotoxicity and deposition in the brain of AD.
