Abstract:A practical Pd-catalyzed reaction was developed to achieve C-H activation/C-B cross-coupling of acetanilides under acidic conditions. The new reaction shows a good functional group tolerance and an exclusive mono-selectivity. This C-H borylation method may provide a generally applicable route for the conversion of C-H moieties into many other types of bonds.

Abstract:Room-temperature ortho-arylation and acetoxylation of anilides have been achieved using cationic palladium (Pd[TFA](+)) as catalyst and (NH4)(2)S2O8 as oxidant. Preliminary investigation of the mechanism suggests that palladium may have different oxidation states in the catalytic cycles of these two transformations.

Abstract:Decarbonylative borylation of carboxylic acids is reported. Carbon electrophiles are generated directly after reagent-enabled decarbonylation of the in situ accessible sterically-hindered acyl derivative of a carboxylic acid under catalyst controlled conditions. The scope and the potential impact of this method are demonstrated in the selective borylation of a variety of aromatics (>50 examples). This strategy was used in the late-stage derivatization of pharmaceuticals and natural products. Computations reveal the mechanistic details of the unprecedented C-O bond activation of carboxylic acids. By circumventing the challenging decarboxylation, this strategy provides a general synthetic platform to access arylpalladium species for a wide array of bond formations from abundant carboxylic acids. The study shows a powerful combination of experiment and computation to predict decarbonylation selectivity.

Abstract:A convenient and efficient palladium-catalyzed synthesis of aromatic ketones and isoindolobenzimidazoles has been developed via selective aromatic CH bond acylation. The protocol uses palladium acetate as the catalyst, readily available carboxylic acids as the acylating reagents, trifluoroacetic anhydride as the activated agent of the acids, and the corresponding aromatic ketones and isoindolobenzimidazoles were obtained in good to excellent yields. This finding should provide a new and useful strategy for synthesis of aromatic ketones and isoindolobenzimidazoles.

Abstract:The functionalization of remote unactivated C(sp3)-H and the reaction selectivity are among the core pursuits for transition-metal catalytic system development. Herein, we report Pd-catalyzed γ-C(sp3)-H-selective alkylation and alkenylation with removable 7-azaindole as a directing group. Acid and base were found to be the decisive regulators for the selective alkylation and alkenylation, respectively, on the same single substrate under otherwise the same reaction conditions. Various acrylates were compatible for the formation of C(sp3)-C(sp3) and C(sp3)-C(sp2) bonds. The alkenylation protocol could be further extended to acrylates with natural product units and α,β-unsaturated ketones. The preliminary synthetic manipulation of the alkylation and alkenylation products demonstrates the potential of this strategy for structurally diverse aliphatic chain extension and functionalization. Mechanistic experimental studies showed that the acidic and basic catalytic transformations shared the same six-membered dimer palladacycle.

Abstract:Carbazole alkaloids have great potential for applications in biomedicine and materials science. Direct C-H functionalization construction of carbazole derivatives, however, has been a great challenge. Herein, we theoretically investigated a palladium-catalyzed site-selective direct C-H functionalization (alkylation and acylation) strategy for free NH-carbazole by density functional theory (DFT) methods. Calculations show that C-H activation is the rate determining step for the conversion, with a potential barrier of 23.4 kcal/mol. Meanwhile, the six-membered ring C-H activation intermediate formed at the C1-position is the most stable and kinetically favorable under the synergistic effect of norbornene (NBE) and palladium(II) catalysts. In contrast, the large tension prevents the formation of activation intermediates at the C2, C3, or C4-positions, leading to the excellent site-selectivity for the transformation. We also investigated the mechanism of key acylation step, oxidative addition, under the same conditions. In agreement with experiments, benzoic anhydride was theoretically rationalized to act as an acylating agent in this conversion with an oxidative addition potential barrier of 13.3 kcal/mol. Further calculations also indicate the possible great potential of acyl halides as acylating agents following the protocol, with an oxidative addition barrier of only 2.6 kcal/mol.

Abstract:Pd(OAc)2-catalyzed oxidative acetoxylation of benzylic C-H bonds utilizing a bidentate system has been explored. A variety of picolinoyl- or quinoline-2-carbonyl-protected toluidine derivatives react with PhI(OAc)2 in the presence of Pd(OAc)2 to afford the acetoxylated products in synthetically useful yields. A broad of functionalities, such as CH3, F, Cl, Br, I, COCH3, CO2Et, SO2CH3, and NO2, were tolerated. This transformation provides easy access to 2-hydroxymethylaniline derivatives.

Abstract:Reported herein is a Pd-catalyzed regioselective C-H activation method that is used for C-H deuteration, carbonylation, halogenation, and oxidation of arene substrates substituted by two N-heterocycles. When conducted in acetic acid (AcOH), these reactions occur at the five-membered palladacycle sites, whereas they switch to the six-membered palladacycle sites in trifluoroacetic acid (TFA). This controllable regioselective C-H activation is applied for late-stage functionalization of bioactive molecules. A mechanism study indicated that the regioselectivity is achieved by Brönsted acid-Lewis base interactions and electronic effects (in TFA) and the different kinetic stabilities of palladacycle intermediates (in AcOH).

Abstract:A palladium/norbornene-catalyzed ortho acylation for the efficient synthesis of functionalized alkyl aryl ketones is reported. Studies on the electronic and steric properties of mixed aryl anhydrides indicated that the cross-coupling favored with the electron-enriched aryl acyl group. DFT calculation on the oxidative addition of Pd(II) with 2,4,6-(Cl)3C6H2CO2C(O)Ph suggested that 2,4,6-(Cl)3C6H2C(O)–O bond cleavage was more kinetically disfavored than that of the PhC(O)–O bond by 11.7 kJ/mol.

Abstract:Efficient copper-catalyzed aerobic oxidative C-H and C-C functionalization of 1-[2-(arylamino)aryl]ethanones leading to acridones has been developed. The procedure involves cleavage of aromatic C-H and acetyl C-C bonds with intramolecular formation of a diarylketone bond. The protocol uses inexpensive Cu(O2CCF3)2 as catalyst, pyridine as additive, and economical and environmentally friendly oxygen as the oxidant, and the corresponding acridones with various functional groups were obtained in moderate to good yields.
