Abstract:Ultrasound (US)-assisted electrochemical oxidation (EO) has previously demonstrated its ability to degrade perfluorooctanoic acid (PFOA). However, the mechanisms of direct electron transfer and indirect oxidation by reactive oxygen species (ROS), as well as the involvement of these ROS, are unclear, particularly with the peroxydisulfate (PDS) electrolyte and the added effect of US. In this study, an US/EO/PDS system was first investigated, achieving nearly 100 % and 80.01 % defluorination for PFOA and perfluorooctane sulfonate, respectively (within 3 h). This US/EO/PDS system exhibited remarkable stability, pH tolerance, and practicality. Further experimental and theoretical studies revealed that the predominant mechanism of PFOA defluorination was indirect oxidation via ROS, rather than direct oxidation at the anode. Although none of these ROS could initiate the degradation of PFOA without additional energy, both ROS (SO center dot(4) > center dot OH > O-1(2) > O center dot(2)) and US/EO contributed to the initial step of PFOA defluorination. Furthermore, US enhanced the generation of ROS and the direct oxidation (electron transfer) ability of EO. The electrons were transferred from PFOA to PDS via the electrode assembly. This study clarified, for the first time, the synergistic effect of US/EO/PDS and the involvement of direct and indirect ROS oxidation in defluorination of PFOA, which shows potential for developing US and EO technology for complete degradation of polyfluoroalkyl substances.

Abstract:Perfluorooctanoic acid (PFOA) is an artificially synthesized perfluorinated chemical widely used in industries. It is often released into the environment without treatment, which causes pollution in groundwater. Recently, we have reported a rapid and efficient removal of PFOA in aqueous solution by using a fountain-strip dielectric barrier discharge reactor (SF-DBD). This design allows for the gaseous-liquid interaction to happen in a large space at atmospheric pressure, so it is a promising method to efficiently remove PFOA from water. Recently, we reported the effects of the process parameters, including power mode, pulse time, sinusoidal wave discharge, the discharge gas, initial concentration, pH, conductivity, and positive and negative discharges, on the efficiency of this method for PFOA degradation. Understanding the reaction mechanism is key to further improve the efficiency of the system. In this work, we reported the decomposition mechanism of the SF-DBD for PFOA degradation. The mass spectrum (MS) showed that PFOA was degraded to perfluoroheptanoic acid, perfluorohexanoic acid, perfluoropentanoic acid, perfluorobutanoic acid, perfluoropropionic acid, and trifluoroacetic acid after the plasma treatment. The optical emission spectroscope (OES) and the radical scavenger experiments indicated that the excited argon atoms and hydroxyl radicals played a major role in PFOA degradation, while the contributions from the solvated electrons (e(aq)(-)), superoxide anion radical (center dot O2-), and singlet oxygen (1O(2)) were negligible in initiating the cleavage reaction.

Abstract:The perfluoro. carboxylic acid fluorides prepared by electrochemical perfluorination (ECPF) method were analyzed by gas chromatography/mass spectrometry. Methanol was used to convert the perfluoro. carboxylic acid fluorides into the methyl esters at 60 degrees C. The effects of column length, polarity and film thickness on the separation efficiency were investigated. The KB-1MS capillary column (90 m x 0. 25 mm x 1. 0 mu m) was employed in the GC/MS analysis. The fragmentation patterns and mass spectrometric characteristics of perfluorinated alicyclic acid methyl esters, aliphatic carboxylic acid methyl esters and mono. unsaturated perfluorinated aliphatic carboxylic acid methyl esters were analyzed and deduced from known patterns of perfluoro-organic mass spectrometry. In this work, five perfluoro. carboxylic acid fluorides including two isomers were identified and the perfluorocyclohexylcarboxylic acid fluoride accounted for about 65% of the total perfluoro-carboxylic acid fluorides in the product.

Abstract:Regarding the emerging concerns of the widely occurred and environmentally persistent per- and polyfluoroalkyl substances (PFASs), one intriguing and unsolved scientific question for environmental microbiologists, chemists, and engineers is that whether microbial reductive defluorination of perfluorinated compounds exist in nature. Due to the strong dissociation energy of carbon‒fluorine (C‒F) bonds in PFASs, no convincing evidence for biological cleavage of C‒F bonds from > C 2 perfluorinated structures has ever been reported. We for the first time show C‒F bond cleavage via reductive defluorination by an organohalide-respiring microbial community for two PFASs, perfluoro(4-methylpent-2-enoic acid) and 4,5,5,5-tetrafluoro-4-(trifluoromethyl)-2-pentenoic acid. Comprehensive biotransformation pathways are further elucidated. This study brings valuable fundamental knowledge of microbial dehalorespiration, which opens avenues for the future exploration of PFAS environmental fate and bioremediation strategies.

Abstract:Enzymatic cleavage of C─F bonds in per- and polyfluoroalkyl substances (PFAS) is largely unknown but avidly sought to promote systems biology for PFAS bioremediation. Here, we report the reductive defluorination of α, β-unsaturated per- and polyfluorocarboxylic acids by Acetobacterium spp. The microbial defluorination products were structurally confirmed and showed regiospecificity and stereospecificity, consistent with their formation by enzymatic reactions. A comparison of defluorination activities among several Acetobacterium species indicated that a functional fluoride exporter was required for the detoxification of the released fluoride. Results from both in vivo inhibition tests and in silico enzyme modeling suggested the involvement of enzymes of the flavin-based electron-bifurcating caffeate reduction pathway [caffeoyl-CoA reductase (CarABCDE)] in the reductive defluorination. This is a report on specific microorganisms carrying out enzymatic reductive defluorination of PFAS, which could be linked to electron-bifurcating reductases that are environmentally widespread.

Abstract:Decomposition of perfluorocarboxylic acids (PFCAs) is of great significance due to their global distribution, persistence and toxicity to organisms. In this study, the photodegradation of a series of PFCAs (∼C2C12) in water by a medium-pressure mercury lamp was experimentally and theoretically examined. We found that photolysis of PFCAs all follow pseudo-first-order kinetics with the rate constant (kapp) increasing with carbon chain lengths, except for trifluoroacetic acid (TFA) which cannot be degraded by the polychromatic irradiation. Product analysis showed that the PFCAs were mainly decomposed into shorter carbon chain length PFCAs in a stepwise manner, with the accumulation of TFA and fluoride ions as the end products. Moreover, a small amount of perfluoroolefins (CnF2n) was determined as gas-phase products. Wiberg bond order calculations confirmed the cleavage of the CC bond between carboxylic carbon and the adjacent carbon as the first reaction step, and density functional theory-based calculations revealed that kapp value is correlated with some molecular structural parameters. In the case of mixture irradiation, the evolution profiles of individual PFCAs were different from that in single-component systems, due to the dynamic balance between production and degradation. This work reveals the main molecular descriptors controlling the degradation rate of different PFCAs species, and improves the general understanding on the photodegradation mechanisms, which will provide useful information for future researches.

Abstract:An improved extraction (ion pairing) and cleanup (ENVI-carb and solid phase extraction) method was developed for analysis of perfluorinated compounds (PFCs) in human whole blood samples from China. Ten PFCs including PFOS, PFHxS, PFOSA, PFDoDA, PFUnDA, PFDA, PFNA, PFOA, PFHpA, and PFHxA were detected in the blood samples (n = 30) from five cities (Jintan, Nanjing, Guiyang, Beijing, and Shenyang). PFOS was found to be the dominant PFC ranging from 0.446-83.1 ng/mL. Total fluorine (TF) and extractable organic fluorine (EOF) also were measured in the blood samples using combustion ion chromatography for fluorine. Analysis of known PFCs and extractable organic fluorine showed that known PFCs could account for >70% of EOF in samples from Beijing, Shenyang, and Guiyang, whereas known PFCs could only account for similar to 30% of EOF in samples from Jintan. Results of the present study indicated the presence of substantial amounts of unidentified organic fluorine in human blood samples from Jintan. Characterization and identification of these unidentified fluorinated compounds will be instructive.
On the Chemical Reduction and Electro- Reduction Behavior of Perfluorodecalin

Abstract:The chemical reduction and electro reduction behavior of perfluorodecalin have been investigated under the illumination of UV light. The IR spectra of the resulting products showed that the same functional group, acyl fluoride, could be formed in both reduction processes. The results of 19 F NMR and UV vis spectra revealed that there were other different functional groups formed in the two reduction processes. In the electro chemical reduction , conjunctive double bonds could be formed in the same and different carbon ring. In the chemical reduction, using Zn and Mg as reducing agents, aromatic structure was formed in the products.

Abstract:Per- and polyfluoroalkyl substances (PFAS) are found in a variety of places including cosmetics, rain jackets, dust, and water. PFAS have also been applied to occupational gear to protect against water and oils. However, PFAS have been identified as immunosuppressants and perfluorooctanoic acid (PFOA), a specific PFAS, has been identified as carcinogenic. Since there is a risk for dermal exposure to these compounds, there is a need to characterize their dermal absorption. Using in vitro flow-through diffusion, skin permeabilities were determined for 14C-labeled perfluorooctanoic acid (PFOA), perfluorohexanoic acid (PFHxA), and perfluorobutanoic acid (PFBA) using porcine skin. Tests were conducted over 8 h with either acetone or artificial perspirant as the vehicle. PFBA was found to have greater permeability than PFHxA, likely due to having a smaller molecular weight. The dosing vehicle did not appear to impact permeability rates but impacted the disposition through the skin model. While these PFAS compounds showed a low permeability rate through the skin membranes, they can stay in the skin, acting as a reservoir.

Abstract:Many clearing methods achieve high transparency by removing lipid components from tissues, which damages microstructure and limits their application in lipid research. As for methods which preserve lipid, it is difficult to balance transparency, fluorescence preservation and clearing speed. In this study, we propose a rapid water-based clearing method that is fluorescence-friendly and preserves lipid components. FLUID allows for preservation of endogenous fluorescence over 60 days. It shows negligible tissue distortion and is compatible with various types of fluorescent labeling and tissue staining methods. High quality imaging of human brain tissue and compatibility with pathological staining demonstrated the potential of our method for three-dimensional (3D) biopsy and clinical pathological diagnosis.
