Abstract:A general synthesis of 4-hydroxylcarbazoles by domino vinylogous conjugate addition/cyclization/elimination/aromatization of easily prepared 3-nitroindoles with alkylidene azlactones under mild and transition-metal-free conditions has been developed. This method was also applicable to other nitrosubstituted benzofused heterocycles such as 3-nitrobenzothiophene, 2-nitrobenzothiophene, and 2-nitrobenzofuran. The valuable tetracyclic carbazole derivatives, such as 6H-oxazolo[4,5-c]carbazole and 3,6-dihydro-2H-oxazolo[4,5-c]carbazol-2-one, were readily prepared from the product, demonstrating synthetic utility of this method.

Abstract:Photo‐induced cascade difluoromethylation‐cyclization of N‐alkene tethered indoles was developed using tetrazole sulfone reagent as a CF2H radical precursor. A variety of indoles with ester, cyano, ketone, halide, ether, aldehyde group occurred smoothly to give the desired products in moderate yields. Polycyclic indoles are prevailed in natural products, pharmaceuticals and biologically active compounds. Herein is described photo‐induced difluoromethylation‐cyclization of N‐alkene tethered indoles to synthesis of tetrahydropyrido[1,2‐a]indole using 5‐((difluoromethyl)sulfonyl)‐1‐phenyl‐1H‐tetrazole as a CF2H radical source. This photochemical reaction tolerates an array of functional groups like ester, cyano, ketone, halide, ether, aldehyde and heteocycles. Mechanistic experiments indicated that the difluoromethyl radical and the single electron transfer were involved.

Abstract:A green and efficient protocol for the preparation of benzo[a]carbazoles via visible-light-promoted and electron donor-acceptor (EDA) complex-driven intermolecular cyclization of 2-arylindoles with Z-α-bromocinnamaldehydes in the absence of external photocatalysts, transition metals, and oxidants was reported. This new approach demonstrates an intermolecular cyclization model using indole derivatives as electron donors under visible light. Mechanistic investigations have showed that 2-arylindoles with Z-α-bromocinnamaldehydes form EDA complexes, which undergo sequential single-electron transfer, radical coupling, 6π-electrocyclization, and dehydroaromatization to generate benzo[a]carbazoles under visible light irradiation. The current photochemical method features readily accessible starting materials, mild conditions, simple operation, and a broad substrate scope.

Abstract:A catalyst‐, acid‐ and base‐free, environmental‐friendly method for the synthesis of 2H‐benzo[g]furo[2,3‐e]indazoles, 2H‐benzo[g] thieno[2,3‐e]indazoles and 2H‐benzo[g]pyrrolo[2,3‐e]indazoles via a UV light irradiation of 3‐phenyl‐4‐(2‐heteroaryl)pyrazoles (aryl=furanyl, thiophenyl and N‐methylpyrrolyl) in EtOH/H2O at room temperature under argon atmosphere was described. Irradiation of 3‐(2‐hydroxyphenyl)‐4‐(2‐heteroaryl)pyrazoles showed a high chemo‐selectivity to obtain dehydrogenation product 2H‐benzo[g]furo/thieno/pyrrolo[2,3‐e]indazols‐10‐ol. The mechanism of photocyclization was expound that through the process of 6π‐electroncyclization, [1,5]‐hydrogen shift, pyrazole tautomerism, 1,3‐eneamine tautomerism and evolution H2.This article is protected by copyright. Abstract:Vinyl radicals were generated from 3-iodoflavones under a mercury lamp and tandem cyclization reactions occurred with five-membered heteroarenes entailing two consecutive C-C bond formations to synthesize benzo[e] chromeno[2,3-g] indol-13(1H)-one derivatives. The tandem cyclization reactions worked in acetonitrile without any additives such as transition metals, ligands and oxidants, giving rise to a broad variety of novel polycyclic xanthone frameworks in good yield under mild and environmentally friendly reaction conditions.

Abstract:Three novel indolo[3,2-b]carbazoles derivatives were successfully synthesized by condensation reaction and structurally characterized by elemental analysis, mass spectrometry and proton nuclear magnetic resonance spectroscopy methods, which belong to donor-π-acceptor systems comprising an indolo[3,2-b]carbazole group as an electron donor and two benzothiazole rings as electron acceptors. The thermal, electrochemical and photophysical properties of the compounds were characterized by thermogravimetric analysis combined with electrochemistry, UV–vis absorption spectroscopy and fluorescence spectroscopy. On the other hand, the geometries, molecular orbitals, charge-injection and transport properties were determined by quantum chemical calculations. The results show that the compounds synthesized exhibit good thermal stability and high fluorescence quantum yields, indicating the potential application as optoelectronic materials.

Abstract:Indolocarbazole alkaloids and their derivatives were discovered to have potent protein kinase and topoisomerase I inhibitory activities. Disclosed herein is the photochemical synthesis of the indolocarbazole ring system from N-allenyl-2-iodoanilines. The tandem protocol included visible-light-mediated 5-exo-trig radical cyclization and subsequent radical dimerization, followed by acid-promoted deprotection and intramolecular Mannich cyclization. This strategy showed exceptional functional group tolerance and was successfully applied in the concise synthesis of natural products tjipanazoles B and D.

Abstract:An efficient methodology for the synthesis of dibenzo[a,c]carbazoles via annulation of 2,3-diphenyl-1H-indoles in EtOH under UV light irradiation (lambda = 365 nm) along with hydrogen evolution is described. This method exhibits the advantages of mild reaction conditions, no requirement of any oxidants and catalysts, and release of hydrogen as the only byproduct. Notably, the mechanism investigation confirms that the trans-4b,8a-dihydro-9H-dibenzo[a,c]carbazole intermediate could convert into cis-4b,8a-dihydro-9H-dibenzo[a,c]carbazole, which relies on the nitrogen atom of the indole ring. This is followed by intramolecular dehydrogenation which yields the dibenzo[a,c]carbazoles.

Abstract:An approach to hydrocarbazoles bearing an all‐carbon quaternary center at C4a position was developed via a Brønsted acid‐initiated Diels‐Alder cycloaddition/retro‐aza‐Michael addition cascade process from azepino[4,5‐b]indoles and commercially available dienophiles. The method provided a range of hydrocarbazoles in 63–99% yields. The practicality of this transformation was demonstrated by a scale‐up experiment and various transformations to several hydrocarbazole derivatives and the tetracyclic indoline scaffold. Moreover, hetero‐Diels‐Alder cycloadditions of azepino[4,5‐b]indoles were explored with 4‐phenyl‐3H‐1,2,4‐triazole‐3,5(4H)‐dione (PTAD) and 82–99% yields were obtained.

Abstract:A series of novel quinone-fused cyclopenta[2,1-b]indoles are designed and conveniently constructed by an iron(II)-catalyzed annulation reaction from indolynaphthoquinone and 1,3-dicarbonyl compounds. This process involves Michael addition /radical cyclization/aromatization, medium to excellent yields are achieved to give cyclization product. These compounds have near-infrared absorption and can produce O2−•. Electrochemical tests have also confirmed that compound 3af had excellent ability to transfer electron. In addition, 3af demonstrated excellent Type I PDT effects in tumor cells.
