Abstract:Chemical examination of the methanolic extract from the stem bark of Daphne aurantiaca led to the isolation of three new flavonoids (1-3), and 29 known flavonoids. All 32 compounds were isolated for the first time from Daphne aurantiaca. The isolates were tested for inhibitory activities against lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW 264.7 macrophages. Compounds 21 and 24 showed potent inhibitory activities against the production of NO with IC₅₀ values of 0.006 and 0.076 µM, respectively.

Abstract:Bioassay-directed fractionation led to the isolation of two new biflavonoids and 13 known biflavonoids from a sample of the dried stem barks of Daphne feddei. The structures of compounds 1 and 2 were elucidated as 2''-methoxy-daphnodorin C (1) and 2''-methoxy-2-epi-daphnodorin C (2) on the basis of detailed spectroscopic analysis and X-ray crystallography. All 15 biflavonoids were tested for inhibitory activities against lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW 264.7 macrophages. Compounds 1, 2, 8, 10, 14 and 15 showed varying degrees of inhibitory activities against the production of NO in tested concentration of 25, 50, 75 and 100 microg/ml.

Abstract:The stem and root barks of Daphne giraldii Nitsche, also known as "Zushima", is a traditional Chinese medicine with a long history of treating a variety of diseases such as rheumatism arthralgia, quadriplegia, headache, stomachache, waistache, and bruises. Chromatographic fractionation of the EtOH extracts of "Zushima" has led to the isolation of 26 flavonoids, including ten flavonols (1, 2, and 4−11), one flavone (12), two dihydroflavones (13 and 14), a dihydroflavonol (15), six flavans (3 and 16−20), three chalcones (21−23), and three 1,3-diarylpropanes (24−26). Daphgiflavones A−C (1−3) were new flavonoids. Their structures were elucidated by spectroscopic analysis. All compounds were evaluated for anti-inflammatory properties by testing their inhibitory activities against nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated murine RAW264.7 macrophages. Compounds 8, 11, 21, and 23 exhibited significant inhibition on NO production with IC50 values ranging from 2.26 to 6.88 μM, being more active than the positive control quercetin (IC50: 14.16 μM).

Abstract:In this study, two new ( 1, 13 ) and fourteen known ( 2 – 12, 14 – 16 ) compounds were isolated from the branches and leaves of Daphne retusa. On the basis of chemical evidence and spectral data analysis (UV, ECD NMR, and HR-ESI-MS), the structures of new compounds were elucidated. Furthermore, all compounds have been tested for their inhibitory effects on NO production in LPS-induced RAW 264.7 cells, and compound 3 showed obvious inhibitory effect. Through target screening and molecular docking technology, potential binding targets for compound 3 to exert anti-inflammatory effects have been predicted.

Abstract:Two new flavanes, named daphneflavan B (1) and daphneflavan C (2), along with two known biflavonoids, daphnodorin D1 (3) and daphnodorin D2 (4), were isolated from the roots of Daphne tangutica Maxim. Their structures were established on the basis of chemical, physicochemical, and spectroscopic evidences. Two compounds 3 and 4 were noted to have the most marked antitumor activity in vivo assay.

Abstract:Two new biflavonoids, daphnogirins A (1) and B (2), were obtained from the roots of Daphne giraldii. Their structures were established on the basis of the spectral data and X-ray diffraction data of the co-crystal of 1 and 2. Daphnogrins A and B have the same configuration at C-1 and opposite configurations at C-16 and C-17. Oxygen radical scavenging assay has indicated that they are of significant antioxidative activity.

Abstract:A new flavonoid glycoside, 5-methoxy-7-beta-D-glucopyranosyl-( - )-afzelechin (1), together with seven known flavonoids, 7-O-beta-D-glucopyranosyl-( - )-epiafzelechin (2), sakuranin (3), genkwanin (4), 4 ',5-dihydroxy- 3 ',7-dimethoxyflavone (5), luteolin-3 ',7-dimethyl ether-5-beta-D-glucoside (6), and luteolin-7-methyl ether-5-beta-D-glucoside (7), were isolated from the stem barks of Daphne giraldii. Their structures were identified on the basis of spectroscopic analysis. Compounds 2, 3, and 5-8 were first reported from the titled plant.

Abstract:Chemical examination of the methanolic extract from the stem bark of Daphne feddei led to the isolation of five new phenylpropanoids, 4,4'-dihydroxy-3,3'-dimethoxy-9-butoxy-9,9'-epoxylignan (1), 4,4'-dihydroxy-3,3'-dimethoxy-9-ethoxy-9,9'-epoxylignan (2), daphneresinol (3), armaosigenin (4), and isocubebin (5), together with 33 known phenylpropanoids. All 38 compounds were isolated for the first time from D. feddei. All compounds were tested for inhibitory activity against LPS-induced NO production in RAW 264.7 macrophages. Compounds 2, 8, 9, 12, 13, and 15 showed potent inhibitory activities against the production of NO with IC(50) values of 0.091, 0.047, 0.005, 0.088, 0.004, and 0.074 μM/mL, respectively.

Abstract:Two new bis-coumarin glycosides were isolated from the stem barks of Daphne giraldii NITSCHE. Their structures were elucidated as 6-O-alpha-L-rhamnnopyranosyl daphnogirin (1), and 6-O-beta-D-apiofuranosyl daphnogirin (2), on the basis of detailed spectroscopic analysis. Compounds 1 and 2 were tested inhibitory effects against production of nitric oxide (NO) in RAW264.7 cells, and cytotoxicity of human tumor cell lines A549, LOVO, QGY-7703, 6T-CEM. The results showed that compounds 1 and 2 neither reduced production of NO, nor inhibited human tumor cell lines.

Abstract:The mangrove ecosystem is a rich resource for the discovery of actinomycetes with potential applications in pharmaceutical science. Besides the genus Streptomyces, Micromonospora is also a source of new bioactive agents. We screened Micromonospora from the rhizosphere soil of mangrove plants in Fujian province, China, and 51 strains were obtained. Among them, the extracts of 12 isolates inhibited the growth of human lung carcinoma A549 cells. Strain 110B exhibited better cytotoxic activity, and its bioactive constituents were investigated. Consequently, three new isoflavonoid glycosides, daidzein-4-(2-deoxy--l-fucopyranoside) (1), daidzein-7-(2-deoxy--l-fucopyranoside) (2), and daidzein-4,7-di-(2-deoxy--l-fucopyranoside) (3) were isolated from the fermentation broth of strain 110B. The structures of the new compounds were determined by spectroscopic methods, including 1D and 2D nuclear magnetic resonance (NMR) and high-resolution electrospray ionization mass spectrometry (HR-ESIMS). The result of medium-changing experiments implicated that these new compounds were microbial biotransformation products of strain M. aurantiaca 110B. The three compounds displayed moderate cytotoxic activity to the human lung carcinoma cell line A549, hepatocellular liver carcinoma cell line HepG2, and the human colon tumor cell line HCT116, whereas none of them showed antifungal or antibacterial activities.

Abstract:A phytochemical investigation to search for new nitric oxide (NO) inhibitors resulted in the isolation of seven previously undescribed daphnane diterpenoids, thyrsoidpenes A-G, from the leaves of Trigonostemon thyrsoideus. Their structures including absolute configurations were elucidated on the basis of extensive NMR spectroscopic data analysis and the time-dependent density functional theory (TDDFT) electronic circular dichroism (ECD) calculations. Thyrsoidpenes B-G feature rare polycyclic caged structures of daphnane diterpenoid orthoester. The NO inhibitory effects were examined and all of the compounds showed inhibitory activities toward LPS-induced NO production in murine microglial BV-2 cells. The possible mechanism of NO inhibition of some bioactive compounds was also investigated using molecular docking, which revealed the interactions of bioactive compounds with the iNOS protein. All compounds were tested for inhibitory activity against LPS-induced NO production in RAW 264.7 macrophages. Compounds 7, 8, 9, 10, and 11 showed potent inhibitory activities against NO production with IC(50) values of 0.01, 0.01, 0.06, 0.07, and 0.03 microM, respectively.

Their structures were elucidated by extensive analysis of spectral data and comparison with the literature values. All compounds were tested for inhibitory activity against lipopolysaccharide-induced NO production in RAW 264.7 macrophages, and compounds 4 and 5 showed potent inhibitory activity with IC50 values of 0.161 and 0.127 μM, respectively.

Abstract:Cultivation of the actinobacteria strain Isoptericola chiayiensis, a mangrove-derived actinobacteria that was isolated from a mangrove soil collected in Chiayi County, resulted in the isolation of one new 2-furanone derivative, isopterfuranone (1), one new sesquiterpenoid, isopterchiayione (2), one new benzenoid derivative, isopterinoid (3), five new flavonoids, chiayiflavans A-E (4-8), and 4 metabolites isolated for the first time from nature source, methyl 3-(4-methyl-2,5-dioxopyrrolidin-3-yl)propanoate (9), 3-ethyl-4-methylpyrrolidine-2,5-dione (10), chiayiensol (11) and chiayiensic acid (12). Their structures were determined through in-depth spectroscopic and mass-spectrometric analyses. Most of the isolates showed potent inhibitory effects on NO production in LPS-stimulated RAW 264.7 murine macrophages cells with IC50 values ranging from 9.36 to 40.02 μM. Of these isolates, 4 and 5 showed NO inhibitory activity with IC50 values of 17.14 and 9.36 μM, stronger than the positive control quercetin (IC50 =36.95 μM). This is the first report on flavan metabolites from the genus Isoptericola.

Abstract:Twelve new abietane diterpenoids (1-12) and 31 known analogues (13-43) were isolated from a medicinal Chinese herb, Clerodendrum trichotomum Thunberg. The absolute configurations of 1-3 were established on the basis of ECD and X-ray crystallography data, whereas that of 4 was elucidated by comparison of experimental and calculated ECD data. Eight diterpenoids, 15,16-dehydroteuvincenone G (1), trichotomin A (4), 2α-hydrocaryopincaolide F (7), villosin C (20), 15-dehydro-17-hydroxycyrtophyllone A (22), demethylcryptojaponol (38), 6β-hydroxydemethylcryptojaponol (39), and trichotomone (43), exerted inhibitory effects against NO production with IC50 values of 5.6-16.1 μM. The structure-activity relationships of the isolated diterpenoids are also estimated.
