P3‐175: Presenilin‐1 Val97Leu Mutation Induces Hyperphosphorylation of Tau Through Activating Glycogen Synthase Kinase‐3

Abstract:Familial Alzheimer's disease (FAD) is mostly associated with presenilin 1 (PS1) gene mutations. In our previous studies, we found a novel mutation of PS1 (Val97Leu) in members of a Chinese family that had early onset AD. We also transfected human neuroblastoma SH-SY5Y cells with the Val97Leu mutant to generate a stable cell line and found the mutant showed pathological effects, including increased levels of extracellular and intracellular Aβ42. As it is not very clear whether PS1 mutation may regulate tau phosphorylation, in present study we study the effect of Val97Leu mutation on tau phosphorylation and analyze the role of glycogen synthase kinase-3 (GSK-3) in it. We transfected human neuroblastoma SH-SY5Y cells with wild-type PS1 and the Val97Leu mutant to generate a stable cell line. We detected the phosphorylation level of tau at many epitopes by Western blot. We observed the colocalization of phosphorylated tau and activated GSK-3 by immunofluorescence. The GSK-3 activity is measured by enzyme activity kit and antibodies by Western blot. We also use GSK-3 inhibitors (LiCl, SB415286 and AR-A014418) making the results convinced. 1. The Val97Leu mutant induced tau phosphorylation at Thr231, Ser396/404 and PHF-1 epitopes. 2. The elevated phosphorylation level of tau was due to activated GSK-3. PS1 Val97Leu mutant can involve in elevated tau phosphorylation level through activation of GSK-3.
