Abstract:As part of our continuing effort for development of novel anti-inflammatory agents, the highly potential agent CCY1a, which we reported recently, was selected as lead compound to synthesize a series of its derivatives for evaluation. Most of the newly synthesized compounds exhibited superior inhibitory activity than both the lead compound and the positive control (trifluoperazine) toward fMLP-stimulated neutrophil superoxide formation. (2E)-3-[2-(Benzyloxy)-5-methoxyphenyl]-acrylaldehyde (31) was among the most potent with action mechanism different from CCY1a in that it does not act as cAMP-elevating agent but inhibits the increase in cellular Ca2+ with greater potency.

Abstract:A versatile and economical reaction of diketene (1), aryl amines 2, cyclic 1,3-diketones 3, primary amines 4, and aryl aldehydes 5 was explored to synthesize 3,4-dihydropyran-3-carboxamide derivatives under mild conditions. Three stereogenic centers are generated in the products, and the structure of the major diastereomer of 6{1,1,3,1} was identified by X-ray diffraction and 2D NMR spectroscopy. The scope and limitation investigation provided two series of (2S,3R,4S)-chromene-3-carboxamides in good to excellent yields with high diastereoselectivity. Two products, 6{5,3,1,1} and 6{7,3,1,1}, exhibited in vitro anti-inflammatory activity with significant inhibition of pro-inflammatory cytokine IL-6 and TNF-α expression in lipopolysaccharide (LPS)-treated Raw 264.7 cells.

Abstract:As part of a continuing effort for the development of novel anti-inflammatory and antiallergic agents, the highly potential agent, 2-chloro-3-methoxycarbonylethylcarboxamido-1, 4-naphthoquinone (2), was selected as the lead com pound to synthesize a series of derivatives. The structure-activity relationships in this series were examined. Some of the compounds showed significant anti-inflammatory and antiallergic activities. Upon further study of the mechanism of action, it was found that 2-methoxycarbonypropylcarboamido-3-chloro-1, 4-naphthoquinone (3) and 2acetamido-3-chloro-1, 4-naphthoquinone (4) had a direct inhibitory effect on the activator protein 1 (AP-1) and Nuclear factor-κB (NF-κB).

Abstract:A series of new substituted derivatives of 2-cyano-3-methvlthio-3'-aminoacrylates(acrylamides) have been designed and synthesized. Their structures were confirmed by H-1 NMR and elemental analysis. The bioassay test indicates that most of these new compounds show good Hill reaction inhibitory activity.

Abstract:Ethoxycarbonylmethyl ursolate obtained by a reaction of ursolic acid with ClCH2COOC2H5 reacted with hydrazine hydrate to give hydrazinocarbonylmethyl ursolate (3). Reaction of the hydrazide 3 with keto compounds give the corresponding hydrazones 4a similar to 4g, which could be readily convered to 3-acetyl-2-[(un)substituted phenyl]-2,3-dihydro-1,3,4-oxadiazol-5-methyl 3-O-acetylursolate (5a similar to 5g) in good yields by treatment with Ac2O. The structures of new products were characterized by H-1 NMR, IR and MS spectra. By the preliminary bioassay test some of such Compounds show good anti-inflammatory activity. For example, compounds 5a (40 mg.kg(-1)), 5d (40 mg.kg(-1)) and 5g (40 mg.kg(-1)) were better than the parent ursolic acid.

Abstract:In order to search for new compounds with stronger anti-inflammatory activity and less side effects, phenyl 5-chloro-2-oxindole-1-carboxylate, obtained from 5-chloro-2-oxindole via esterification with phenyl chloroformate and subsequent hydrolyzation, reacted with 4-(methylsulfonyl) benzoyl chloride and 4-dimethylaminopyridine, followed by acidification with hydrochloric acid to yield phenyl 5-chloro-3-(hydroxy-p-methylsulfonylphenylmethylene)-2-oxo-2,3-dihydroindole-1-carboxylate (II1), which was condensed with corresponding amines, followed by acidification with hydrochloride acid to afford the target compounds II2 therefore similar to II15, respectively. Fifteen novel title compounds were synthesized, and their structures were confirmed by IR, H-1 NMF, MS spectra and elemental analyses. II10, II11, and II15 exhibited marked anti-inflammatory activity in xylene-induced mice ear swelling model. Moreover, II10 and II11, showed significant anti-inflammatory activity comparable to diclofenac sodium and tenidap sodium (P > 0.05) in carrageenan-induced rat paw edema model. II11, had less gastrointestinal side effect than diclofenac sodium (P < 0.05) and tenidap sodium (P < 0.01).
