Abstract:Recently, we have developed highly regio- and stereoselective carbometalation of 2-alkynols and 2,3-allenols. The organometallacyclic intermediates may be trapped with I(2) to afford 3-iodoalk-2-en-1-ols. These 3-iodoalk-2-en-1-ols may readily undergo the Sonogashira coupling with terminal propargyl alcohols to form 4-alkyn-2-ene-1,6-diols. Subsequent cycloisomerization in DMA or CH(2)Cl(2) with Au(PPh(3))Cl and AgOTf as the catalyst would afford polysubstituted 2-(1-alkenyl)furans; with PdCl(2) as the catalyst and the reaction in DMA in the presence of allylic bromides, the same substrates afforded polysubstituted 2-(1,4-alkadienyl)furans. In both types of catalyzed cyclization reactions, the elimination of H(2)O promoted the aromatization to form the furan ring. Different alkyl or aryl groups could be introduced into different positions of furans due to the substituent-loading capability of 3-iodoalkenols and diversity of the terminal propargyl alcohols and allylic bromides.

Abstract:The copper-catalyzed decarboxylative [3+2] annulation of ethynyl benzoxazinanones with 3-oxo-3-arylpropanenitriles has been developed, producing tetrasubstituted furan derivatives in moderate to high yield. This reaction was generally compatible with a wide range of substrates. Notably, the intermediate copper-allenylidenes worked as a C2 synthon in the cycloaddition reaction.

Abstract:Synthesis of new polysubstituted furans incorporating reactive phosphorus ylides or N ‐acyliminophosphoranes was achieved by the heterocyclization of flexible α‐acyl ketene dithioacetals. This one‐pot tandem reaction was initiated by acyl‐directed desulfurative Sonogashira coupling, followed by 1,6‐addition of phosphine to the in situ‐generated enynone and subsequent 5‐ exo ‐ dig cyclization, as well as phosphoranation. The selectivity toward the two products, i.e. α‐phosphorus ylides vs. α‐ N ‐acyliminophosphoranes‐substituted furans, was determined by both electron‐deficient acetylenes and copper salts.

Abstract:Tri- or tetrasubstituted furans have been prepared from terminal activated olefins and acyl chlorides or anhydrides by a multicomponental convergent synthesis mode. Instead of stoichiometric nBu3P, only catalytic nBu3P or nBu3P=O is needed to furnish the furans in modest to excellent yields with a good functional group tolerance under the aid of reducing agent silane. This synthetic method features a silane-driven catalytic intramolecular Wittig reaction as a key annulation step and represents the first successful application of catalytic Wittig reaction in multicomponent cascade reaction.

Abstract:Comprehensive Summary Furans bearing alkynyl substituents are highly useful in organic synthesis. However, the methodologies to access these important furan derivatives are rather limited. We herein report an efficient synthesis of alkynylated furan derivatives based on Pd‐catalyzed oxidative cross‐coupling reaction between allenyl ketones and terminal alkynes. This novel synthesis of alkynylated furans with wide substrate scope is operationally simple and tolerates various functional groups. Mechanistically, the formation of the palladium carbene through cycloisomerization and the subsequent alkynyl migratory insertion are proposed as the key steps in the transformation. The reaction reported in this manuscript further demonstrates the generality of the carbene‐based cross coupling. This article is protected by copyright. All rights reserved.

Abstract:A novel palladium-catalyzed intermolecular dearomatization of furans with alkynes via a three-component formal [3 + 2] spiroannulation/allylic substitution cascade reaction has been successfully developed for the stereoselective assembly of spiro 2,5-dihydrofuran frameworks. High step economy and efficacy as well as excellent stereoselectivity were achieved for a broad substrate scope. Two new C-C bonds and one new C-O bond were generated sequentially in a one-pot manipulation. The yielded spiro 2,5-dihydrofuran skeleton bearing a tetrasubstituted carbon center constitutes the core structure for plenty of useful natural products or corresponding analogues. This work represents a significant advancement in the dearomatization strategy for furan heterocycles and provides a practical methodology for expedited access to complex spiro dihydrofuran scaffolds.
