Abstract:Objective To study the association between serum total IgE level and β2-adrenoreceptor polymorphism (β2-AR) in asthmatic patients. Methods ELISA was used to determine serum total IgE and AS-PCR was used to determine β2-adrenoreceptor polymor- phism in 44 asthmatic patients. Results In β2-AR 16 locus genotype, the distribution frequencies of Arg/Arg, Gly/Gly showed increasing tendency, whereas Arg/Gly showed decreasing tendency, in normal serum total IgE group and increased serum total IgE group. But there was no significant difference between this two groups. In β2- AR 27 locus genotype, the distribution frequency of Gln/Gln genotype accounts for 76.2% and Gln/Glu genotype for 19.0% in increased serum total IgE group, while Gln/Gln genotype accounts for 9.0% and Gln/Glu geno- type for 73.9% in normal serum total IgE group. There was significant difference between two groups ( P<0.01 ). Conclusions Serum total IgE was correlated with β2-AR 27 locus genetic polymorphism, which may contribute to understand the mechanism of asthma in the peo- ple of the Han nationality in GuiZhou.

Abstract:Association and linkage studies of beta(2)-adrenergic receptor (beta(2)AR) polymorphisms in relation to the expression of asthmatic phenotypes and immune regulatory mechanisms have shown inconsistent results. This study was designed to analyze the relationship of particular combinations of single nucleotide polymorphisms (SNPs) or haplotypes of the beta(2)AR gene with bronchial asthma, bronchodilator response, and total IgE. By direct DNA sequencing, five SNPs (in positions -47, -20, 46, 79, and 252) of beta(2)AR gene were determined and combined with haplotypes in 201 asthmatic patients and 276 normal controls recruited from the Chinese Han population. Significantly higher bronchodilator response was observed in patients with homozygotic genotype 46A/A (13.40 +/- 3.48%), compared with those with homo-46G/G (7.25 +/- 3.11%) and heterozygotes 46A/G (7.39 +/- 3.14%), respectively (p < 0.0001). There was also a significant difference in bronchodilator response when beta(2)AR haplotypes were analyzed (p = 0.003). From two common SNPs at positions 46A/G and 79C/G, we had determined three haplotypes that constructed six haplotype pairs. Comparison of the mean delta forced expiratory volume in 1 second (FEV1) values for the six haplotype pairs showed significant difference. Subjects homozygous for 46A/79C (Arg16/Gln27) had the highest deltaFEV(1) (13.40 +/- 3.48%) and those with 46G/79C (Gly16/Gln27) homozygote had the lowest (6.43 +/- 0.55%). The two SNP haplotype pairs were significantly associated with delta FEV1 (p < 0.0001). Significantly higher total IgE levels were found in patients with homozygotic carriers of 79C genotypes (p = 0.022) and homozygotic haplotype -47 T/-20 T/46 A/79 C/252 G (p < 0.0001). These results indicate that the manifestation of asthma might be affected by either an individual beta(2)AR SNPs or beta(2)AR haplotype.
Polymorphism -509C/T in TGFB1 Promoter Is Associated With Increased Risk and Severity of Persistent Allergic Rhinitis in a Chinese Population.

Abstract:Background Polymorphism -509C/T in the promoter of transforming growth factor beta1 (TGFB1) gene is implicated in the pathogenesis of asthma. This polymorphism might also act to regulate the development of allergic rhinitis (AR). Objectives To investigate whether -509C/T is associated with AR susceptibility and severity in a Han Chinese population. Methods The study enrolled 263 patients with persistent AR and 249 healthy controls. AR patients were classified as mild or moderate/severe AR groups according to the Allergic Rhinitis and its Impact on Asthma classification. TGFB1 gene polymorphism -509C/T was genotyped with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Serum total Immunoglobulin E (IgE) and specific IgE levels were determined using an ImmunoCAP. Results Significant difference was found in the allele frequency of TGFB1 -509C/T between AR patients and healthy controls (P = .027) but not in the genotype frequency (P =.051). However, the genotype frequency of TGFB1 -509C/T showed significant difference between the mild AR group, the moderate/severe AR group, and the control group (P = .012); between the moderate/severe AR group and the control group (P =.036); between the mild AR group and the moderate/severe AR group (P = .038); but not between the mild AR group and the control group (P =.075). Conclusion TGFB1 promoter polymorphism -509C/T may be associated with the susceptibility and the severity of persistent AR of Han Chinese, but the functional relationship still needs clarification.
[Association Between Beta2-Adrenergic Receptor Haplotype/polymorphisms and Asthma Phenotype].

Abstract:OBJECTIVE To analyze the association of single nucleotide polymorphisms (SNP)/ haplotypes of beta2-adrenergic receptor (beta2-AR) gene with bronchodilator response and total serum immunoglobulin E (IgE). METHODS Five SNP (in positions: -47, -20, 46, 79, 252) genotypes of beta2-AR gene in 201 asthmatic patients and 276 controls were determined by direct DNA sequencing, and the haplotypes were combined from 2006 February to 2007 February. Statistical analyses were performed with SPSS 11.5 software. The genotype frequencies for each polymorphism were tested for deviation from the Hardy-Weinberg equilibrium by chi2 goodness-of-fit analysis. The frequencies of the five polymorphic genotypes were compared with chi2 test, and the degree of linkage disequilibrium occurring between these polymorphic loci were analyzed by fisher' s exact. For the comparison of quantitative traits among different genotypes/haplotypes, ANOVA was used. Least significant difference (LSD) was used if there was statistical significant. RESULTS The bronchodilator response in patients with Argl6Argl6 was (13 +/- 4)L, significantly higher than those with Arg16Gly16 [(7 +/- 3) L and Gly16Gly16 (7 +/- 3)L, F = 81.55, P < 0.01]. When beta2-AR haplotypes were analyzed, bronchodilator response was highest in patients with haplotype Arg16Gln27/Arg16Gln27 [(13.4 +/- 3.5) L], compared to that with Gly16Gln27/Gly16Gln27 [(6.4 +/- 0.6) L], Gly16Glu27/Gly16Glu27 [(7.6 +/- 3.1)L], Gly16Gln27/Gly16Glu27 [(6.9 +/- 3.5)L], Gly16Gln27/Arg16Gln27 [(7.2 +/- 3.3) L, and Gly16Glu27/Arg16Gln27 (7.9 +/- 2.7) L, F = 32.55, P < 0.01]. The total IgE level was (2.51 +/- 0.33) IU/L in patients with genotype Gln27Gln27, significantly higher than that with Gln27Glu27 [(2.30 +/- 0.82) IU/L, F = 3.89, P < 0.05]. The total IgE level was significantly lowest in patients with haplotype Gly16Gln27/Arg16Gln27 (2.13 +/- 0.15) IU/L, compared to that with Arg16Gln27/Arg16Gln27 (2.56 +/- 0.14) IU/L, Gly16Glu27/Gly16Glu27 (2.40 +/- 0.16) IU/L, Gly16Gln27/Gly16Glu27 (2.54 +/- 1.26) IU/L, Gly16Gln27/Arg16Gln27 [(2.48 +/- 0.48) IU/L, F = 3.56, P < 0.01]. CONCLUSIONS These results indicate that depending on phenotypes studied, either an individual beta2-AR SNP or haplotype might affect disease manifestations.
Association Between Β2-Adrenergic Receptor Genetic Polymorphisms and Nocturnal Asthmatic Patients of Chinese Han Nationality

Abstract:Background: As a result of the finding that the mutation of Arg into Gly at β2-adrenergic receptor (β2-AR)16 loci could promote the downregulation effect triggered by the β2-agonist, it was supposed that Gly16 might be associated with the downregulation of β2-AR in patients with nocturnal asthma. Objective: It was the aim of this study to analyze the association between β2-AR genetic polymorphisms and nocturnal asthmatic patients of Chinese Han nationality. Methods: A polymerase chain reaction allele-specific oligonucleotide hybridization assay was used to determine 16 and 27 loci alleles of β2-AR genetic polymorphisms in 25 nocturnal asthmatic patients (nocturnal asthma group), 22 non-nocturnal asthmatic patients (non-nocturnal asthma group), and 72 healthy people (control group). All people investigated were of Chinese Han nationality. Results: The distribution frequency of genotype Arg/Arg, Arg/Gly, and Gly/Gly at β2-AR 16 loci was 12, 16 and 72% in the nocturnal asthma group; and 27, 41 and 32% in the non-nocturnal asthma group. There was a significant increase in the frequency of genotype Gly/Gly and allele Gly in the nocturnal asthma group compared with the non-nocturnal asthma group (p < 0.01). However, there was no significant difference in the frequency of genotype Gly/Gly and allele Gly in the non-nocturnal asthma group, compared with the control group. There was no significance in the frequency of the genotypes and alleles of β2-AR 27 loci among the three groups (p > 0.05). Conclusion: The Gly16 polymorphism of β2-AR was overrepresented in nocturnal asthmatic patients, correlated with nocturnal asthma, and therefore appeared to be an important genetic factor in the expression of this asthmatic phenotype.
Association Between Polymorphisms/haplotypes of ADRB2 Gene and Asthma-Related Phenotypes in a South Chinese Han Population

Abstract:Objective The aim of this study was to evaluate the potential relationship between polymorphisms/haplotypes of β2-adrenergic receptor (ADRB2) gene and asthma-related phenotypes.Methods Six polymorphisms loci (T-2387C,5' LC Arg19 Cys,Arg16Gly,Glu27Gln,Thr164Ile and C523A) of the ADRB2 gene were genotyped by the TaqMan assay in 379 asthmatics and 435 healthy controls,the haplotypes also be analyzed.Total serum IgE level was measured by enzyme linked immunosorbent assay (ELISA).Results The mutant allele at 164 loci was not observed and only the homozygous wild-type was found in both asthmatic patients and control subjects,therefore it was not included in the statistical analysis.There were significant difference in both genotype frequencies and haplotype frequencies distribution of T-2387C,5'LC Arg19 Cys,Arg16Gly,Glu27Gln and C523A between asthmatic patients and control subjects (x2 were 2.45,4.12,0.43,5.81,0.44;2.39,0.80,1.42,0.20respectly,all P ＞0.05),and so were between the nocturnal asthma and non-nocturnal asthma (x2 were:0.77,1.51,1.68,1.51,1.03;1.05,0.45,1.01,0.40 respectly,all P ＞0.05).Total serum IgE levels in asthma group [(83.95±67.35) μg/L] was higher than normal subjects [(66.12±61.27) μtg/L] but there was no significant difference in total serum IgE levels between all genotypes in the five loci (all P ＞0.05).No association was found in relation to the serum levels of IgE between the four homozygous haplotypes accroding to five loci.But we found the individual who carring the Arg/Cys19 + Cys/Cys19,Gln/Glu27+Glu/Glu27 genotype in severe asthma had higher level of IgE values according to asthma severity.Conclusions There were no association between polymorphisms/haplotype of ADRB2 and asthma severity and nocturnal asthma,but we found that there might be asociation between-47,79 loci and the IgE values in severe asthma.

Abstract:ABSTRACTBackground and objective: Adrenergic β2 receptors (ADRB2) play an important role in regulating pulmonary function. Many previous studies have investigated possible associations between polymorphisms in the ADRB2 gene and asthma, but have yielded conflicting results. Furthermore, little is known regarding the possible role of the Arg19Cys polymorphism in susceptibility to asthma among Chinese.Methods: This case–control association study involved 238 patients with asthma and 265 healthy subjects from a Han population in southwest China. For all subjects, the 5′ leader cistron Arg19Cys, Arg16Gly and Gln27Glu polymorphisms in the ADRB2 gene were characterized by direct sequencing. Genotype, allele and haplotype frequencies were determined. In addition, to evaluate the association between the ADRB2 polymorphisms and lung function, bronchodilator response to inhaled β2 agonists (400 µg of albuterol) was assessed in the asthmatic patients.Results: There were no significant differences in genotype or allele frequencies for the three ADRB2 polymorphisms between the two cohorts. The Arg19/Arg16/Gln27 haplotype was more frequent among asthmatic patients than control subjects (odds ratio 2.24, 95% confidence interval (CI): 1.05–4.73, P = 0.04). Moreover, the Arg19/Cys19 genotype was associated with a lower FEV1% (mean difference −4.5, 95% CI: −12.5 to 3.6, P = 0.02) and FEV1/FVC (mean difference 8.9, 95% CI: 8.5–9.4, P = 0.01). The bronchodilator response to albuterol was also marginally lower in individuals who were homozygous for the Arg19 genotype (mean difference 4.2, 95% CI: 3.7–4.8, P = 0.03).Conclusions: The Arg19/Cys19 genotype was an independent risk factor for lower FEV1% and FEV1/FVC. Asthmatic patients with the Arg19/Arg19 genotype showed decreased responsiveness to albuterol. Furthermore, the Arg19/Arg16/Gln27 haplotype may contribute to increased susceptibility to asthma in the Chinese population.
Relationship Between Genetic Polymorphisms of Β_2-Adrenergic Receptor and Childhood Asthma

Abstract:Objectives To explore the correlation between asthmatic bronchitis and asthma at molecular level. Methods The polymorphisms of β2-adrenergic receptors (β2-AR) in 217 children with asthma,119 children with asthmatic bronchitis and 50 healthy children were detected with PCR restriction fragment analysis,and their plasma concentration of total IgE were determined by enzyme linked immunosorbent assay. Results (1) There was no significant difference in the frequencies of genotypes and alleles of β2-AR 16 loci among the three groups(P 0.05). (2) The frequency of Glu/Glu genotype of β2-AR 27 loci in the asthmatic group was significantly lower than that in the control group(P 0.05). There was no significant differences between the asthmatic bronchitis group and the control group or the asthmatic group in Glu/Glu frequency(P 0.05). The frequency of Glu27 allele in the asthmatic group was significantly lower than that in the control group,while the frequency of Gln27 allele in the two groups was on the contrary(P 0.05). (3) There were no significant differences in total plasma IgE among the three genotypes of β2-AR 16 loci. The total IgE level in Gln/Gln genotype was significantly higher than that in Glu/Glu and Gln/Glu of the asthma group and the asthmatic bronchitis group(P 0.05),however no differences were observed between Glu/Glu and Gln/Glu. Conclusions Asthmatic bronchitis is correlated with asthma in genetic polymorphism of β2-AR 27 locus. Strong correlation with earlier asthma can be considered when the asthmatic bronchitis with both Gln/Gln genotype of β2-AR 27 loci and high total plasma IgE. The polymorphisms of the β2-adrenergic receptors can help to make earlier diagnosis for childhood asthma,which also provide evidences at molecular level for the correlation study between asthmatic bronchitis and asthma.

Abstract:Objective To analyze the relationship between β2-drenergic receptor(β2-AR) genetic polymorphism and asthma in the people of the Miao nationality of Guizhou.Methods Allele Specific-PCR techniqu was used to determine 16,27 loci of β2-AR genetic polymorphisms in 31 unrelated patients with asthma,and 37 healthy controls.Results The frequency of genotype β2-AR 16 loci:Arg/Arg genotype accounted for 16.1%,Arg/Gly 61.3% and Gly/Gly 22.6% in asthma group;Arg/Arg for 13.5%,Arg/Gly 70.3% and Gly/Gly 16.2% in control group.The frequency of genotype β2-AR 27 loci:Gln/Gln genotype accounted for 32.3%,Gln/Glu 54.8% and Glu/Glu 12.9% in asthma group;Gln/Gln 37.8%,Gln/Glu 51.4% and Glu/Glu 10.8% in control group.There was no significant diference in the frequencies of genotypes and alleles β2-AR 16,27 loci in both groups.Conclusion β2-AR 16,27 loci genetic polymorphism is not correlated with asthma in the people of the Miao nationality of Guizhou.

Abstract:Objective To analyze the association betweenβ 2 AR genetic polymorphism and cAMP in patients with asthma.Methods The 16,27 locus alleles of β 2 AR genetic polymorphisms were determined by using AS PCR techniques and the cAMP was determined by using RIA techniques in 44 patients with asthma.Results The relativity between cAMP and genotype β 2 AR 16 loci:the level of cAMP was (18.03±4.37)pmol/ml in Arg/Arg,(18.54±14.22)pmol/ml in Arg/Gly and(13.00±4.07)pmol/ml in Gly/Gly.There were significant difference cAMP compared with three groups;(15.99±4.89)pmol/ml in Gln/Gln genotype,(16.14±4.34)pmol/ml in Gln/Glu and(13.69±7.88)pmol/ml in Glu/Glu,There were no significant difference in cAMP compared with three groups in β 2 AR 27 loci.Conclusion β 2 AR 16 locus genetic polymorphism is correlated with cAMP in asthma.
The IL6R Gene Polymorphisms Are Associated with Sil-6R, IgE and Lung Function in Chinese Patients with Asthma.

Abstract:Background: sIL-6R is involved in a variety of inflammatory diseases. The present study analyzed the potential associations between two IL6R gene polymorphisms (rs2228145 and rs12083537) and asthma in a Han Chinese population. Methods: A cohort of 394 patients and 395 healthy controls were genotyped to detect the two polymorphisms using SNaPshot. In 66 asthma patients and 49 controls, peripheral eosinophil and serum immunoglobulin E (IgE) levels were determined using a routine blood test, and sIL-6R levels were measured by ELISA. Results: No statistically significant differences were detected between the patients and controls in the distribution of the two independent IL6R polymorphisms (p>0.05). However, rs2228145 C and rs12083537 G were significantly associated with decreased lung function in patients with asthma; the rs2228145 A–C variant was also associated with increased sIL-6R and IgE levels. In addition, sIL-6R levels were positively associated with IgE and inversely associated with lung function in patients with asthma. Conclusions: Our results provide evidence that rs2228145 C and rs12083537 G are associated with poor lung function in patients with asthma. Furthermore, the rs2228145 A–C variant is associated with levels of sIL-6R and IgE.
Beta-2 adrenergic receptor (ADRB2) gene polymorphisms and the risk of asthma: a meta-analysis of case-control studies.

Abstract:BACKGROUND AND OBJECTIVE:A number of studies have assessed the relationship between beta-2 adrenergic receptor (ADRB2) gene polymorphisms and asthma risk. However, the results are inconsistent. A meta-analysis that focused on the association between asthma and all ADRB2 polymorphisms with at least three case-control studies was thus performed. METHODS:A literature search of the PubMed, Embase, Web of Science, CNKI, and Wangfang databases was conducted. Odds ratios with 95% confidence intervals were used to assess the strength of associations. RESULTS:Arg16Gly, Gln27Glu, Thr164Ile, and Arg19Cys single nucleotide polymorphisms (SNPs) were identified in 46 case-control studies. The results showed that not all of the SNPs were associated with asthma in the overall population. Significant associations were found for the Arg16Gly polymorphism in the South American population via dominant model comparison (OR = 1.754, 95% CI = 1.179-2.609, I2 = 16.9%, studies = 2, case = 314, control = 237) in an analysis stratified by ethnicity. For the Gln27Glu polymorphism, a protective association was found in children via recessive model comparison (OR = 0.566, 95% CI = 0.417-0.769, I2 = 0.0%, studies = 11, case = 1693, control = 502) and homozygote genotype comparison (OR = 0.610, 95% CI = 0.434-0.856, I2 = 0.0%, studies = 11, case = 1693, control = 1502), and in adults via dominant model comparison (OR = 0.864, 95% CI = 0.768-0.971, I2 = 46.9%, n = 18, case = 3160, control = 3433). CONCLUSIONS:None of the ADRB2 gene polymorphisms were reproducibly associated with a risk of asthma across ethnic groups in the general population.

Abstract:Objective To investigate the influence of genetic polymorphisms within β2-adrenoceptor(β2-AR)gene on the development of childhood asthma and the clinical outcomes of the individual response to long-acting β2-agonist(LABA)therapy in Chinese Han children.Methods Arg16Gly,Glu27Gln and Thr164Ile polymorphisms were detected within β2-adrenoceptor(β2-AR)in 212 children with asthma as well as in 52 normal subjects by DNA direct sequencing.And the relationship between polymorphisms and asthma was analyzed.A prospective pharmacogenetic study was carried out on selected 89 children with moderate persistent asthma aged between 5 and 11 years(mean age 7.57 ± 2.26 years).These children were given Salmeterol(Seretide)by inhalation for 12 weeks and lung function was tested before and after treatment.The influence of β2-AR polymorphisms on children's lung function before and after inhalation of LABA was analyzed.Results The frequencies of both of Arg16Gly G and Glu27Gln G allele in asthma patients were significantly higher than those in the control group(38.7% vs.31.7%,P 0.01 and 7.1% vs.0,P 0.01 respectively).There was no significant difference of 164 allele polymorphisms between asthmatics and normal subjects(P 0.05).After treatment 69.66% patients'conditions were within control and the lung function was significantly improved in FEV1(forced expiratory volume in 1 second),FVC(forced vital capacity),PEF(peak expiratory flow)and FEF25,50,75(Forced expiratory flow between 25% and 75% of vital capacity).There was no difference among the three Arg16Gly groups and also that three Glu27Gln groups in clinical symptom control(P 0.05).Conclusions β2-AR Arg16Gly G and Glu27Gln G but Thr164Ile are statistically related with the development of childhood asthma.It shows that no pharmacogenetic effect of β2-AR variation on therapeutic response to LABA in childhood asthma.
Association of CD14 Gene Polymorphism with Atopic Diseases in Chinese Han Ethnic Group Children

Abstract:OBJECTIVE:To investigate the distribution characteristics of the single nucleotide polymorphisms (SNPs) of the human CD14 gene in Chinese Han ethnic group children in Wenzhou, and their association with atopic diseases.METHODS:Totally 113 cases were recruited in atopic disease group who met the following criteria: 2 - 12 years old, clinically diagnosed as asthma or allergic rhinitis or atopic dermatitis, elevation of serum total IgE levels and serum specific IgE. Sixty-seven healthy children were enrolled in control group. The related regions of CD14 gene were sequenced to identify and characterize the SNPs, and plasma TIgE and SIgE were detected by immunoassay system and uniCAP system, respectively. The frequency of genotypes and alleles between two groups, as well as the levels of IgE in different genotypes, were compared.RESULTS:CD14/-159 SNP was present in Han ethnic group population of Wenzhou. The frequency of each genotype was 57.0% (TT), 28.0% (TC), 15.0% (CC) in normal children, and 46.9% (TT), 35.4% (TC), 17.7% (CC) in atopic children. No significant difference was found in the distribution of CD14/-159 polymorphism between atopic children and healthy control (chi(2) = 1.918, P > 0.05) according to Hardy-Weinberg principle statistics. There were no significant difference in frequency of each genotype between boys and girls. No significant difference was found in the total plasma IgE levels among groups of TT genotypes [(2520 +/- 460) IU/L], TC genotypes [(2400 +/- 460) IU/L] and CC genotype [(2500 +/- 460) IU/L] (F = 0.807, P > 0.05).CONCLUSION:CD14/-159 SNP is present in Han ethnic group children in Wenzhou, and other SNP in CD14 gene was not found. TT genotype was the primary genotype in CD14/-159 SNP in the children studied. No relationship between CD14/-159 SNP and atopic disease or serum total IgE level was found.

Abstract:OBJECTIVE: The aim of this study was to investigate the association between CD14 gene promoter SNPs with serum total-IgE and eosinophil levels in atopic asthma and non-atopic asthma in Chinese Han.METHODS: A total of 152 patients with asthma were divided into atopic asthma (n = 100) and non-atopic asthma (n = 52) groups for this study. Six CD14 gene SNPs were analyzed using PCR and gene sequencing. Serum total-IgE and eosinophil levels were measured. The association between genotype frequencies of the CD14 gene loci with total-IgE and eosinophil levels in atopic asthma and non-atopic asthma was evaluated by the ANOVA test method. Hundred and sixteen healthy subjects constitute the control group.RESULTS: We found that serum total-IgE and eosinophil levels were significantly higher in individuals with atopic asthma when compared to individuals with non-atopic asthma (p < 0.01). For non-atopic asthma, the total-IgE levels of the heterozygous genotypes were significantly higher than the corresponding levels for the homozygous genotypes in CD14-260C > T, CD-651C > T, CD-911A > C and CD-1247A > G (p < 0.01). In atopic asthma, there was no statistical significance for either serum total-IgE or eosinophil levels among the genotypes of the CD14 gene SNPs. In addition, allele A frequency of CD14-1247A > G was significantly different between the atopic asthma and non-atopic asthma groups (p = 0.025).CONCLUSIONS: There was a statistical association between the serum total-IgE level and the CD14 gene promoter SNPs in the non-atopic asthma group. The eosinophil level was not found to be statistically associated with the CD14 gene promoter SNPs in either the atopic asthma or non-atopic asthma groups.

Abstract:■ Abstract Background: Research increasingly suggests that asthma is a familial and hereditary disorder in the pathogenesis of which genetic and environmental factors play an important role. Objective: To investigate the single and combined associations between 8 single-nucleotide polymorphism (SNP) loci in 5 genes and the development of asthma in children of Chinese Han nationality. Methods: The study population comprised 192 children with asthma and an equal number of healthy controls. Asthma was diagnosed in accordance with American Thoracic Society criteria. Polymerase chain reaction-restriction fragment length polymorphism was used to detect the genotypes of the SNP loci. Results: No statistically signifi cant differences (P>.05) were found between the experimental and control group in genotype distribution among 6 loci (IL-13 C1112T, IL-13 C1923T, IL-4 C590T, IL-4RA I75V, FcεR1ß E237G, and ß2-ADR Q27E). However, signifi cant diversity was observed among FcεR1ß C109T (P=.002) and ß2-ADR R16G (P=.000). Furthermore, the frequency of FcεR1ß C109T T/T and ß2- ADR R16G A/A in the asthma group was signifi cantly higher than in the control group (odds ratio (OR)=1.96, P=.001; OR=2.58, P=.000, respectively). Carriers of both FcεR1ß C109T T/T and ß2-ADR R16G A/A had a more signifi cant risk of developing asthma than those with only a single polymorphism. Conclusion: The 6 loci (IL-13 C-1112T, IL-13 C1923T, IL-4 C-590T, IL-4RA I75V, FcεR1ß E237G and ß2-ADR Q27E) make little contribution to the development of asthma in children of Chinese Han nationality. FcεR1ß C-109T and ß2-ADR R16G are signifi cantly associated with childhood asthma. FcεR1ß C-109T T/T and ß2-ADR R16G A/A have a signifi cant and combined effect on the development of asthma.

Abstract:OBJECTIVE To explore the correlation of gene polymorphism of interleukin 4 receptor alpha chain (IL-4Rα) and total serum IgE levels in children of bronchial asthma in Guiyang area. METHODS A total of 160 asthmatic children at Affiliated Hospital, Guiyang Medical College from August 2010 to February 2011 were selected as asthma group. There were 96 males and 64 females with an average age of (6.9 ± 3.3) years. During the same period, 143 healthy children from outpatient physical examination center of this hospital were chosen as control group. There were 75 males and 68 females with an average age of (4.9 ± 3.9) years. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and allele-specific polymerase chain reaction (AS-PCR) were used to determine the polymorphisms of Q576R on exon 12 and I75V on exon 5 of IL-4Rα in both groups. The total serum IgE levels were determined by enzyme-linked immune sorbent assay (ELISA). Comparisons of two groups were made for genotype and allele frequency, serum total IgE and different genotypes, different groups of total serum IgE levels. RESULTS Three genotypes of AA, AG and GG existed on Q576R and I75V loci between two groups. The genotypic frequencies of two loci had significant differences in two groups (χ(2) = 30.972, 16.776, both P < 0.001). The subjects with variant allele on Q576R and I75V loci had higher risks for asthma than those without variant allele (χ(2) = 33.236, 13.845, both P < 0.001). The total serum levels of IgE in asthma group were higher than those in control group ((312 ± 297) vs (61 ± 48) U/ml, t = 9.959, P < 0.001). However, these three genotypes of two loci showed no significant inter-group difference (all P > 0.05). In mutation allele carriers and non-carriers of Q576R locus, total IgE levels showed no significant difference (t = -0.028, 1.279; P = 0.978, 0.203). Differences in total IgE levels between I75V polymorphic variant allele carriers and non-carriers were also not statistically significant in control group (t = -0.763, P = 0.447). However, in asthma group, total IgE levels of I75V polymorphic variant allele carriers were significantly higher than those of non-carriers (t = -2.112, P = 0.036). CONCLUSIONS The 576 and 75 loci of IL-4Rα are important candidate genes of asthmatic children in Guiyang area. And the serum total IgE levels of variant allele G at 75 locus in IL-4Rα are higher than those without it.

Abstract:Previous studies of association of beta 2 adrenergic receptor gene (β2-AR) with bronchial asthma was focused in the coding region polymorphisms,these polymorphisms may be associated with the asthmatic susceptivity,phenotype and response to treatment.But there are many inconsistent results,and these differences could not be fully explained by β2-AR gene coding region and the 5'untranslated region polymorphism.In recent years a large number of studies have found not only the coding region,5'and 3' untranslated region polymorphisms can also change the β2-AR function and affect treatment response in asthma.All variants that complete haplotypes on the β2-AR gene should be identified and analyzed.This article reviews the recent development of β2 AR gene coding region,5'and 3' untranslated region polymorphisms,especially the 3'untranslated region of poly-C repeat length variation on patients with asthma plays an important role on pulmonary function and β2-AR agonist treatment response.

Abstract:Asthma is a common respiratory disease associated with airway hyperactivity and high total serum immunoglobulin E (IgE) levels. The asthmatic phenotypes are widely considered to be caused by environmental effects on genetically predisposed individuals. Interleukin (IL)-4 and IL-13 act on B cells and regulate the IgE production, and the single nucleotide polymorphisms (SNPs) in the IL-4 and IL-13 genes are implicated in the pathogenesis of asthma. To study the association of IL-4 and IL-13 gene polymorphisms with asthma, we sequenced the promoter regions and exons of IL-4 and IL-13 genes in two groups: one (spouses group) consisted of 13 pairs of asthmatic patients (cases) and their unaffected spouses (controls); the other (parents group) consisted of 11 pairs of asthmatic children (cases) and their unaffected father/mother (controls). In total, seven polymorphisms were identified, one novel and six previously reported. However, according to the results of the statistical analysis we performed, no statistically significant association of these SNPs and asthma was observed (p > 0.05). Asthma is largely determined by the accumulation of several certain genetic variations other than one or two SNPs. Future function studies may be able to help us reveal the significance of genetic variants we identified in this study.
