Abstract:Taxus is a genus of 12 species mainly distributed in the Northern Hemisphere, with four species in North America, one in Europe, and the remaining native to Southeast Asia. Taxus contains numerous compounds of taxane derivatives, polysaccharides, and flavanoids. The compound with the greatest significance for humans is an oxygen-rich diterpene called taxol, which can be used as an inhibitor of cancer cell growth. Many new biological activities of these chemical compounds have been discovered to fight against various cancers (e.g., ovarian, breast, esophagus, gastric, lung, and prostate cancers), anti-oxidation, cure inflammatory and HIV effects, and eliminate free radicals. This review revises the information on the morphological characteristics and geographical distribution, generalizes the relationship of special secondary metabolites among species, summarizes the main pharmacological activities of the compounds contained in plant, and also puts forward some effective strategies for the exploitation and protection of the Taxus species. These works may be helpful for the researcher and utilizer.

Abstract:To study on the 10-deacetyl baccatin III extract from the leaves and branches of Taxus mairei. A new method was proposed to extract and purify 10-DAB III, which was adopted as the parent nucleus of the high efficacious antitumour drug-taxol and docetaxel in the compositive synthesis. Optimization selection was also made in the extraction and purification conditions of taxol. The purity quotient of 10-DAB III obtained via this technics could reach 91%, with the yield above 70%. It has been testified that the technics was feasible in industrial practice.

Abstract:Objective:To isolate and purify 10-deacetyl baccatinⅢ(10-DABⅢ)from leaves and branches of Taxus mairei.Methods:10-DABⅢwas extracted and purified by liquid-liquid and back ex- traction,column chromatography and recrystallization,and elucidated by UV and HPLC.Results:The total yield of 10-DABⅢ,a precursor of anti-cancer drug taxol and docetaxel was 7.9% and the purity of 10-DASⅢwas 91%.Conclusion:The novel isolation process for 10-DABⅢwas warranted for the pilot manufacture.

Abstract:A new C - 14 oxygenated taxoid, 14β-hydroxy-baccatin Ⅵ (1), together with seven knowntaxoids, baccatin Ⅵ (2), baccatin Ⅳ (3), 1β-dehydroxybaccatin Ⅳ (4), taxayunnasin A (5), 2α-deacetyl-2α-benzoyl-13α-acetyltaxayuntin (6), 5α-hydroxy-2α, 7β, 9α, 10β, 13α-pentaacetoxy-4(20), 11-taxadiene (7) and taxacin (8) were isolated from the ethanolic extract of leaves and stems ofTaxus chinensis (Pilg) Rehd. The structures of the compounds were elucidated by spectroscopic tech-niques. The detailed 13 C and 1 H NMR data of the seven known compounds were measured in acetone- d6,and the NMR data of taxacin (8) were reported for the first time.

Abstract:Two new taxoids, 13,15-epoxy-13-epi-taxayunnasin A (1) and taxchinin N (2), were isolated from the leaves and stems of Taxus chinensis. Compound 2 is the first taxoid to be reported with an alpha,beta-unsaturated lactone at C-4, C-5, C-20, and C-2. The structure of 1 was elucidated by spectroscopic analysis and confirmed by semisynthesis from taxayunnasin A, while 2 was determined structurally using spectroscopic methods and by single-crystal X-ray diffraction.

Abstract:Taxchinin A, with a 11(15→1)-abeo-taxane skeleton, is a major, but inactive taxoid contained in leaves of Taxus chinensis . In our design of dual-functional antitumor abeo-taxane derivatives, two fragments from antitumor agents with different molecular targets (the N-acyl-3'-phenylisoserine side chain from the antimitotic agent paclitaxel and an α,β-unsaturated carbonyl system from NF-κB inhibitors) were incorporated into the scaffold of taxchinin A. The resulting compounds displayed broad inhibitory effects against proliferation of tumor cell lines, with notable selectivity toward colon cancer, melanoma, and renal cancer, when evaluated in the NCI-60 human tumor cell line screening panel. On the basis of the NCI-60 assay data, structure-activity relationship (SAR) correlations were elucidated. Mechanistic studies indicated that this new compound type can both destabilize microtubules and inhibit NF-κB activation, thereby inducing tumor cell apoptosis. This first report of the dual-functional taxoid-core compounds thus provides new opportunities for future drug development based on natural axoid scaffolds.

Abstract:The American Journal of Chinese Medicine, Ahead of Print. Taxus, a rare and protected genus predominantly distributed across the Northern Hemisphere's temperate regions, has garnered global attention due to its significant potential in medical research and pharmaceutical development, bolstered by advancements in cultivation techniques and medical technology. This review primarily focuses on the chemical constituents and pharmacological activities of Taxus, underscoring the progress and potential of these components in clinical applications. Recent studies have revealed that Taxus contains not only taxane active components but also flavonoids and polysaccharides with distinct activities. These compounds from Taxus exhibit potent antitumor, anti-inflammatory, immunomodulatory, antibacterial, and antidiabetic properties with evident mechanisms of action. Notably, the representative compound, paclitaxel, has demonstrated significant efficacy in treating various cancers, such as ovarian, breast, and lung cancer. This paper also reviews the basic situation of Taxus drug formulations, with extracts primarily administered orally and monomeric taxanes typically via injection, reflecting a mature development stage with ongoing research into oral formulations. Finally, this review summarizes the pharmacokinetic characteristics of crucial compounds in Taxus, including their absorption, distribution, metabolism, and excretion patterns in the human body. These pharmacokinetic profiles provide crucial guidance for evaluating the overall dosing regimen of Taxus and its components. The paper concludes with a forward-looking analysis of the potential applications of these compounds in disease treatment, envisioning their role in the future of medical and pharmaceutical advancements.

Abstract:Paclitaxel is a well known anticancer compound. Its biosynthesis involves the formation of a highly functionalized diterpenoid core skeleton (baccatin III) and the subsequent assembly of a phenylisoserinoyl side chain. Despite intensive investigation for half a century, the complete biosynthetic pathway of baccatin III remains unknown. In this work, we identified a bifunctional cytochrome P450 enzyme [taxane oxetanase 1 (TOT1)] in Taxus mairei that catalyzes an oxidative rearrangement in paclitaxel oxetane formation, which represents a previously unknown enzyme mechanism for oxetane ring formation. We created a screening strategy based on the taxusin biosynthesis pathway and uncovered the enzyme responsible for the taxane oxidation of the C9 position (T9αH1). Finally, we artificially reconstituted a biosynthetic pathway for the production of baccatin III in tobacco.

Abstract:Abstract The ancient gymnosperm genus Taxus is the exclusive source of the anticancer drug paclitaxel, yet no reference genome sequences are available for comprehensively elucidating the paclitaxel biosynthesis pathway. We have completed a chromosome-level genome of Taxus chinensis var. mairei with a total length of 10.23 gigabases. Taxus shared an ancestral whole-genome duplication with the coniferophyte lineage and underwent distinct transposon evolution. We discovered a unique physical and functional grouping of CYP725A s (cytochrome P450) in the Taxus genome for paclitaxel biosynthesis. We also identified a gene cluster for taxadiene biosynthesis, which was formed mainly by gene duplications. This study will facilitate the elucidation of paclitaxel biosynthesis and unleash the biotechnological potential of Taxus .

Abstract:Taxol is one of the most famous natural diterpenoids and an important anticancer medicine. Taxol represents a formidable synthetic challenge and has prompted significant interest from the synthetic community. However, in all the previous syntheses of Taxol, there have been no reports of closing the desired eight-membered ring through C1–C2 bond formation. Furthermore, the existence of Taxol-resistant tumors and side effects of Taxol make the development of new approaches to synthesize Taxol and its derivatives highly desirable. Here, we report the asymmetric total synthesis of Taxol using a concise approach through 19 isolated intermediates. The synthetically challenging eight-membered ring was constructed efficiently by a diastereoselective intramolecular SmI2-mediated pinacol coupling reaction to form the C1–C2 bond. The unique biomimetic oxygen ene reaction and the newly developed facile tandem C2-benzoate formation and C13 side chain installation improved the efficiency of the synthesis. The mild oxygen ene reaction under light conditions would be an alternative reaction involved in Taxol biosynthesis. Abstract:The carbon skeleton of any organic molecule serves as the foundation for its three-dimensional structure, playing a pivotal role in determining its physical and biological properties. 1 As such, taxane diterpenes are one of the most well known natural product families, primarily owing to the success of their most prominent compound, paclitaxel, an effective anti-cancer therapeutic for more than 25 years. 2-6 In contrast to classical taxanes, the bioactivity of cyclotaxanes (also referred to as complex taxanes) remains significantly underexplored. The carbon skeletons of these two groups of taxanes differ significantly, and so would typically their own distinct synthetic approaches. Here, we report a versatile synthetic strategy based on the interconversion of complex molecular frameworks, providing general access to the wider taxane diterpene family. A range of classical and cyclotaxane frameworks was prepared including, among others, the first total syntheses of taxinine K ( 2 ), canataxapropellane ( 5 ) and dipropellane C from a single advanced intermediate. The synthetic approach deliberately eschews biomimicry, emphasizing instead the power of stereoelectronic control in orchestrating the interconversion of polycyclic frameworks.
