Abstract:Xanthene dyes are a series of brilliant fluorescent dyes exhibiting superior photophysical properties, which facilitate their applications in various field. Traditionally, they are prepared via acid-catalyzed condensations between electron-rich resorcinol ( m-aminophenols ) and phthalic anhydride. Recently, structurally complicated xanthenes are often in need for bioimaging applications and not readily accessed with the aforementioned methods.This has promoted the development of novel pathways for their synthesis, which are summarized in this review.The scope and limitations of each methods are briefly discussed.

Abstract:Novel xanthenoid dyes by replacing the central oxygen atom of the xanthene dyes with less electron-rich bridging groups have been intensively sought after primarily for their long spectral wavelengths. However, the new scaffolds are likely prone to nucleophilic attack at their central methane carbon, as the result of the reduced electron density of the fluorochromic scaffolds. We envisage that the bridging group may be harnessed to sterically shield the central methane carbon from incoming nucleophiles and render high stability and synthesized xantheno-xanthene dyes. Additionally, the xantheno-bridging group can be modified via electrophilic aromatic substitution to introduce functionalities, e.g., sulfonate groups.

Abstract:The historical dye Indian yellow, derived from euxanthic acid formed from 1,7-dihydroxyxanthone (euxanthone) and methyl (tri- O -acetyl-α- D -glucopyranosyl bromide) uronate, has significantly influenced the art world due to its vibrant color and unique production process. Studying Indian yellow is important for its historical relevance and impact on various art forms, as well as the challenges in its synthetic production. Herein, this work investigates the synthesis of the two main components, a novel method for obtaining euxanthone, and attempts to produce euxanthic acid and Indian yellow. All key intermediates and desired compounds have successfully been synthesized with good to high isolated yields, and characterized using different analytical and spectroscopic techniques. A proposed mechanism for euxanthone synthesis via 2,6,2′,5′-tetramethoxybenzophenone formation is also offered. During this process, 2,7-dihydroxyxanthone has also been synthesized, revealing an equilibration reaction that produced three isomeric tetramethoxybenzophenones, confirmed by both GC/MS and NMR. Following the synthesis of euxanthone and clarification of the equilibration, the production of Indian yellow via euxanthic acid formation has further been explored.

Abstract:Fluorescent dyes are an indispensable part of the scientific enterprise. Xanthene-based fluorophores, such as fluorescein and rhodamine, have been in continual use across numerous fields since their invention in the late 19th century. Modern methods to synthesize and expand the scope of xanthene dye chemistry have enabled new colors, enhanced stability, and improved brightness. Modifications to the 3-position of xanthene dyes have been, until recently, less well-explored. Here, we discuss how small changes to the identity of the substituent at the 3-position of fluoresceins and rhodamines can profoundly alter the properties of xanthene dyes, with the potential to unlock new applications at the interface of chemistry and biology.

Abstract:Disperse reactive dyes with appropriate chemical structure are key for the coloration of natural fibers in the water-free environmentally friendly medium of supercritical carbon dioxide with various advantages. The objective of this work is to design and synthesize a novel anthraquinonoid disperse reactive dye involving a versatile bridge group to improve the coloration properties of the dye in supercritical carbon dioxide. Cross-coupling condensation based on an Ullmann reaction between N-phenylethylenediamine and 1-chloroanthraquinone in a ligand-free system is investigated by optimizing the synthesis parameters. Notable influences are observed from the dosages of N,N-dimethyl formamide and potassium hydroxide, as well as the system temperature and reaction duration, on the isolated yield of the dye precursor. An optimized process is also recommended for synthesizing the designed novel dye. Then, the chemical structure, color characteristics, and coloration properties of the obtained dye are further investigated and successfully characterized by utilizing Fourier-transform infrared analysis, 1H and 13C nuclear magnetic resonance spectroscopy, UV-vis absorption spectroscopy, elemental analysis, and liquid chromatography-mass spectrometry. Additionally, practical coloration experiments are performed with cotton, silk, and wool in a supercritical carbon dioxide medium.

Abstract:Near-infrared dyes are sought after for their potential in biomedical applications. Benzoxanthene dyes with a non-oxygen bridging atom are expected to exhibit longer absorption and emission wavelengths than their oxygen-containing counterparts, yet their synthesis remains unaddressed. Herein, we report the first syntheses of non-oxygen-bridged benzoxanthenes starting from a 7-bromo-2-aminonaphthalene derivative, and discuss their photophysical properties as well as their potential for in vivo imaging.

Abstract:Series of coumarins containing heterocyclic substituent in 3-position were prepared. Sulfonyl chloride, a reactive group, was drawn into the heterocycle, and further condensed with amines to give series of new compounds that contain N-alkyl sulfamide. The spectral properties and dyeing character of these new dyes are discussed. A bathochromic shift (10 nm), enhanced fastness in light and sublimation (1–2 grade) and larger molar extinction coefficient (increase 10 4 L.mol −1 .cm −1 ) are apparent with these new compounds. In the meantime, larger solubility appears with these new compounds.

Abstract:Developing new reactive disperse dyes and applying them to dye natural fiber substrates in the supercritical fluid of carbon dioxide (SCF–CO2) is of great importance for ecological protection and the sustainable development of the textile industry. In this work, a reactive disperse dye with an anthraquinonoid chromogen in blue hue and a dichlorotriazine reactive group was successfully designed and synthesized. The precursor of the anthraquinonoid derivative reactive disperse dye was synthesized with a one-pot amination reaction method by employing the reactions between 1, 4-dihydroxyanthraquinone or its cryptochrome 1, 4-diaminoanthraquinone and aniline, ethylenediamine. Then, the precursor was subjected to continuously react with a reactive group of cyanuric chloride to obtain a dichlorotriazine type reactive disperse dye. Moreover, the chemical structures and color characteristics of the final dye, as well as the precursor, were characterized and confirmed by Fourier transform infrared spectroscopy (FT-IR), 1H nuclear magnetic resonance (1H NMR), and ultraviolet–visible absorption spectroscopy (UV–Vis). Additionally, the anthraquinonoid derivative reactive disperse dye was preliminarily applied to the coloration of cotton and wool substrates in SCF-CO2. The coloration results show that the staining fastness and the dry, wet rubbing fastness of the reactive disperse dye on both cotton and wool substrates were satisfactory.

Abstract:A synthesis of the carbopyronine dye Carboxy ATTO 647N from simple materials is reported. This route proceeds in 11 forward steps from 3-bromoaniline with the key xanthone intermediate formed using a new oxidation methodology. The step utilizes an oxidation cycle with base, water, iodine, and more than doubles the yield of the standard permanganate oxidation methodology, accessing gram-scale quantities of this late-stage product. From this, Carboxy ATTO 647N was prepared in only four additional steps. This facile route to a complex fluorophore is expected to enable further studies in fluorescence imaging.

Abstract:A novel N-coumarin derivative, namely 9,10-[di-p-(7-diethylamino-coumarin-3-yl) thiopheneyl]anthracene ((CTh)2A), containing anthracene as the core and 3-thiophene N-coumarin as the substituent was synthesized, and its structure was confirmed by 1H NMR and IR spectroscopies. The optical, electrochemical and thermal properties were investigated. Thermogravimetric analysis and PL spectra reveal the high thermal and good photoluminescence characteristics. The coumarin derivative exhibits blue photoluminescence with high fluorescence quantum yield in solution (up to 40%). The results show that the derivative would serve as promising organic light-emitting diode luminescent material.

Abstract:In the present paper, a series of xanthene-2-carboxylic acids and xanthone-2-carboxylic acids with electron withdrawing groups on the 7-position had been synthesized.Xanthone-2-carboxylic acid(Ⅷ) was converted to(Ⅸ) by Huang-Minlon reduction. A by-product, 2, 2'-dihydroxy-diphenylmethane(Ⅹ), was also obtained in this reaction.Elemental analysis and MS of Compound(Ⅻ_d) was consistent with the assigned structure, but its ~1HNMR spectrum in CF_3COOH solution was identical with that of(Ⅻ_e) obtained by refluxing compound(Ⅻ_d) in acetic anhydride. It follows that compound(Ⅻ_e) was formed through cyclization and dehydration of compound(Ⅻ_d) in CF_3COOH.The results of passive cutaneous anaphylaxis(PCA) assay in rats showed that compound(Ⅷ), (Ⅻ_b) and(Ⅻ_e) have 50～60% protective effect(po, 200mg/kg) and(Ⅻ_d) 54%(ⅳ, 10 mg/kg).

Abstract:Xanthates are widely used in organic synthesis for the construction of various molecules and have numerous applications as bioactive compounds. Here, we present their preparation via a straightforward photocatalytic decarboxylative xanthylation process involving readily available oxime esters and xanthate dimers. This approach provides access to a wide variety of xanthate compounds in moderate to good yields, including xanthates derived from relevant pharmaceutical and bioactive molecules.

Abstract:Two polymeric dyes containing the anthraquinone structure were prepared via solution polycondensation. Their monomers were prepared through reactions between 1,5-dichloroanthraquinone and nucleophiles. The polymeric dyes were characterized with NMR and thermogravimetry. Their ultraviolet spectra, intrinsic viscosity, and solubility were also measured. [Syntheses of Xanthone Derivatives].

Abstract:In the present paper, fourteen derivatives (Ⅱ～Ⅴ, Ⅷ, Ⅸ) of 1,3-dihydroxyxanthone have been synthesized. Compound Ⅱ was obtained by condensation of 1,3-dihydroxyxanthone with the corresponding halides in the presence of base Compounds Ⅲ and Ⅳ were prepared by treating 1,3-dihydroxyxanthone with epichlorohydrin in the presence of piperidine. Amination of Ⅳ gave compound Ⅴ. Passive cutaneous anaphylaxis (PCA) assay in rats showed that compound Ⅱ_c gave 63% protective effect (5 mg/kg iv). In order to facilitate the introduction of electron-withdrawing substituents, unsuccessful attempts have been made to remove the ketonic group of Ⅰ andⅧ_b by zinc amalgam-HCl reduction or of Ⅸ. by Huang Min lon reaction, the latter suffering demethylation under drastic alkaline conditions.
