The route could be expanded to the synthesis of α,γ-disubstituted butenolide units of naturally occurring annonaceous acetogenins. Abstract:An insecticidal compound 6β-propoxycelangulin V was synthesized from celangulin V and 1-propanol through anhydrous iodination and Williamson reactions.The purpose of the study was to develop the microsynthetic method of 6β-propoxycelangulin V.The powdered root bark of Chinese bittersweet(Celastrus angulatus Max.)was extracted by benzene under reflux,followed by purification by macroporous-resin and silica-gel chromatography.The desired product was purified by RP-HPLC and identified by HRMS,1H NMR and 13C NMR.The total yield was about 17%,and the purity was higher than 98%.This method could be applied in radiosythesis of 6β-([1-14C]propoxy)celangulin V.

Abstract:A highly stereocontrolled synthesis of the C18–C28 ketone fragment of the 16-membered plecomacrolide micromonospolide B has been accomplished. The C21–C23 syn–anti stereotriad is secured by the anti-selective aldol condensation of the ephedrine-derived chiral propionate with (E,E)-hexa-2,4-dienal and Sharpless asymmetric allylic epoxidation–regioselective reductive epoxide ring opening, respectively. The overall yield of this 14-step sequence is 18.4% and the target C18–C28 ketone was obtained in enantiomerically pure form.

Abstract:New insecticides are needed for control of disease-vectoring mosquitoes and this research evaluates the activity of new carbamate acetylcholinesterase (AChE) inhibitors. Biochemical and toxicological characterization of carbamates based on the parent structure of terbam, 3-tert-butylphenyl methylcarbamate, was performed. In vitro enzyme inhibition selectivity (Anopheles gambiae versus human) was assessed by the Ellman assay, as well as the lethality to whole insects by the World Health Organization (WHO) paper contact assay. Bromination at the phenyl C6 position increased inhibitory potency to both AChEs, whereas a 6-iodo substituent led to loss of potency, and both halogenations caused a significant reduction of mosquitocidal activity. Similarly, installation of a hexyl substituent at C6 drastically reduced inhibition of AgAChE, but showed a smaller reduction in the inhibition of hAChE. A series of 4-carboxamido analogs of the parent compound gave reduced activity against AgAChE and generally showed more activity against hAChE than AgAChE. Replacement of the 3-t-buyl group with CF3 resulted in poor anticholinesterase activity, but this compound did have measurable mosquitocidal activity. A series of methyl- and fluoro- analogs of 3-trialkylsilyl compounds were also synthesized, but unfortunately resulted in disappointing activity. Finally, a series of sulfenylated proinsecticides showed poor paper contact toxicity, but one of them had topical activity against adult female Anopheles gambiae. Overall, the analogs prepared here contributed to a better understanding of carbamate structure–activity relationships (SAR), but no new significant leads were generated.

Abstract:A class of structurally simplified analogues of the naturally occurring annonaceous acetogenins were developed, amongst which some non-THF analogues showed remarkable cytotoxicities against tumor cell lines, as well as good selectivity between human tumor cells and normal cells. The synthetic routes were significantly shortened because of the removal of the chiral centers bearing the THF rings on the natural templates. This simplification also provides access to the parallel synthesis of these mimics by a combinatorial strategy. The remaining stereogenic centers at the positions a to the ethereal links were introduced by the Chiron approach from the easily accessible chiral building blocks 6a and/or 6b, made in turn from L-ascorbic acid or D-mannitol, while the one in the butenolide segment was taken from L-lactate. All four diastereomeric non-THF analogues 2a-2d showed remarkable activity against the HCT-8 cell line, and better differentiation was found when testing against the HT-29 cell line. It was also discovered that both the butenolide and ethylene glycol subunits play essential roles in the cytotoxicities against tumor cell lines, while the 10-substituted hydroxy group and the absolute configuration of methyl group at the butenolide moiety are less important for their activity.

Abstract:We describe here the highly stereoselective total synthesis of the Laurencia C15 acetogenins (3Z)- and (3E)-elatenynes having a 7,12-dibromo-6,9-cis-10,13-cis adjacent bis-tetrahydrofuran (THF) core. The present synthesis features a highly stereoselective, protecting group-dependent, chelate-controlled intramolecular amide enolate alkylation (IAEA) for the synthesis of key intermediate 7-hydroxy-6,7-cis-6,9-cis-THF intermediate 10, deployment of the sequential ate complex (n-BuLi/DIBAL-H) reduction/Keck allylation/cross metathesis (CM) protocol for the stereoselective introduction of the C(10)-C(15) unit, a sequential Sharpless asymmetric dihydroxylation (SAD)/intramolecular Williamson etherification for the construction of the 10,13-cis-THF ring, and a modified Nakata chloromethanesulfonate-mediated SN2 displacement for the 7,12-dibromo functionality. Furthermore, our strategy based on chelate-controlled IAEA methodology would provide access to any member of the C15 adjacent bis-THF acetogenin class.

Abstract:An enantioselective synthesis of the acyl side-chain 3 of polyoxypeptins 1 and 2 was achieved by the Sharpless AE, with subsequent regioselective opening of the epoxyalcohol using a Grignard reagent in the presence of CuI, and an aldol condensation using Seebach's ester. The method reported has the advantage of a concise route and excellent enantiomeric purity, as well as starting from readily available chemical substrates and the use of inexpensive reagents.

Abstract:N'-Nitro-2-hydrocarbylidenehydrazinecarboximidamides and methoxyacrylates are two types of important agrochemicals. By combining their key fragments into one framework, a series of the title compounds was designed and synthesized. Some of these compounds have shown excellent insecticidal activities for aphides, for example, the LC50 values of compounds 6-06, 6-11 and 6-19 against M. perslcae and H. amygdale were found to be 1.9/14.4, 13.4/1.5 and 0.3/38.4 mg L-1 respectively. They were also effective against the mycelium growth of B. cinerea in vitro. Compound 6-04 could control (similar to 100%) cucumber anthrax and rice blast at 100 mg L-1 in vivo, and inhibit the spore germination (similar to 100%) of vegetable gray mold at 6.25 mg L-1. These compounds could be considered as potential insecticidal or fungicidal candidates for crop protection. The results of structure-activity relationship (SAR) studies are discussed.

Abstract:(R)‐β‐piperonyl‐γ‐butyrolactones are key building blocks for the synthesis of podophyllotoxin, which have demonstrated remarkable potential in cancer treatment. Baeyer–Villiger monooxygenases (BVMOs)‐mediated asymmetric oxidation is a green approach to produce chiral lactones. While several BVMOs were able to oxidize the corresponding cyclobutanone, most BVMOs gave the (S) enantiomer while Cyclohexanone monooxygenase (CHMO) from Brevibacterium sp. HCU1 gave (R) enantiomer, but with a low enantioselectivity (75% ee). In this study, we use a strategy called "focused rational iterative site‐specific mutagenesis" (FRISM) at residues ranging of 6 Å from substrate. The mutations by using a restricted set of rationally chosen amino acids allows the formation of a small mutant library. By generating and screening less than 60 variants, we achieved high ee of 96.8%. Coupled with the cofactor regeneration system, 9.3 mM substrate was converted completely in a 100‐mL scale reaction. Therefore, our work reveals a promising synthetic method for (R)‐β‐piperonyl‐γ‐butyrolactone with the highest enantioselectivity, and provides a new opportunity for the chem‐enzymatic synthesis of podophyllotoxin.

Abstract:The development of novel and safe insecticides remains an important need for a growing world population to protect crops and animal and human health. New chemotypes modulating the insect nicotinic acetylcholine receptors have been recently brought to the agricultural market, yet with limited understanding of their molecular interactions at their target receptor. Herein, we disclose the first crystal structures of these insecticides, namely, sulfoxaflor, flupyradifurone, triflumezopyrim, flupyrimin, and the experimental compound, dicloromezotiaz, in a double-mutated acetylcholine-binding protein which mimics the insect-ion-channel orthosteric site. Enabled by these findings, we discovered novel pharmacophores with a related mode of action, and we describe herein their design, synthesis, and biological evaluation.

Abstract:A pair of chemical isomeric structures of novel N-tert-butylphenyl thenoylhydrazide compounds I and II were designed and synthesized. Their structures were characterized by MS, IR, (1)H NMR, elemental analysis and X-ray single crystal diffraction. The regioselectivity of the Meerwein arylation reaction and the electrophilic substitution reaction of N-tert-butyl hydrazine were studied by density functional theory (DFT) quantum chemical method. The larvicidal tests revealed that some compounds I had excellent larvicidal activity against Culex pipiens pallens. As the candidates of insect growth regulators (IGRs), the larval growth inhibition and regulation against Culex pipiens pallens were examined for some compounds, especially I1 and I7. Compounds I1 and I7 were further indicated as an ecdysteroid agonist by reporter gene assay on the Spodoptera frugiperda cell line (Sf9 cells). Finally, a molecular docking study of compound I7 was conducted, which was not only beneficial to understand the structure-activity relationship, but also useful for development of new IGRs for the control of mosquitos.

Abstract:Linear annonaceous acetogenin mimetic AA005 has been found to exhibit potent antitumor activity and significant selectivity between human normal and cancerous cells. Utilizing the concept of Click chemistry, a convergent modular fragment-assembly approach has been newly developed and successfully applied to the synthesis of three representative AA005-like molecules via formation of small nitrogen-containing heterocycles or amide bond. These nitrogen-containing analogues were found to exhibit low micromolar inhibitory activities against the growth of several cancer cell lines.

Abstract:The regioselective oxidation of three lathyrane diterpenoids (lathyrol, 1; 7β-hydroxylathyrol, 2; and Euphorbia factor L3, 3) catalyzed by the fungi Mortierella ramanniana CGMCC 3.03413, Mucor circinelloides CICC 40242, and the actinomycete Nocardia iowensis sp. nov. NRRL 5646 was investigated. Ten new metabolites (4–13) including two unprecedented cyclopropane-rearranged products (4 and 5) have been obtained. Metabolites (6, 10 and 11) were further converted chemically to eight acylated derivatives (6a, 10a–10f and 11a). The structures of all compounds were elucidated on the basis of extensive NMR and MS data analyses. All the metabolites and enzyme-chemical products were evaluated for their cytotoxicities against three human cancer cell lines as well as their multidrug resistance (MDR) reversing effects in adriamycin (ADM)-resistant human MCF-7 breast cancer cells (MCF-7/ADM).

Abstract:To explore candidate pesticides from non-food bioactive products, twenty-five new matrine-type alkaloid analogs, 15-chloro-18-(acyl)oximinylmatrines (6a–6y), were semi-synthesized from matrine isolated from the roots of Sophora flavescens Aiton (Fabaceae). The spatial configurations of six derivatives were determined by X-ray crystallography. Analogs 6f (R = (CH2)9CH3) and 6g (R = (CH2)10CH3) all showing the pronounced insecticidal and acaricidal activities against two threatening pests Mythimna separata Walker and Tetranychus cinnabarinus Boisduval, could be used as the lead compounds for further structural optimization. It suggested that R as the aliphatic alkane with long straight chain was necessary for the insecticidal and acaricidal activities of matrine oximino esters.
