Abstract:A phosgene- and metal-free synthesis of unsymmetrical arylurea derivatives utilizing 3-substituted dioxazolones and commercially available amines has been developed. The 3-substituted dioxazolones serve as the precursors of thein situgenerated isocyanate intermediates in the presence of non-toxic sodium acetate as a base and methanol as a solvent under mild heating conditions. A series of unsymmetrical phenylureas includingN, N'- mono-, di-, and trisubstituted derivatives are chemoselectively obtained in moderate to excellent yields from a broad scope of both 3-substituted dioxazolones and amines without a significant amount of the symmetrical byproducts. In many cases, the resulting unsymmetrical phenylureas are easily separated by filtration allowing no chromatographic purification. These investigations provide an opportunity for facile, practical, and eco-friendly synthesis of unsymmetrical diarylureas. Application of this method is demonstrated toward the gram-scale preparation of anti-cancer drugs, sorafenib and the 3-cycle continuous synthesis in conjugation with the recycle of sodium acetate and methanol process of known herbicide, daimuron.

Abstract:3-[(E)-5-(2,3,4,5-Tetr~ethoxy-6-methylphenyl)-3-methylpent-3-enyl]-2,2-dimethyloxirane (8) is a key intermediate for total synthesis of coenzyme QlO (1) and has previously been synthesized from p-cresol via bromination, methylation, Grignard reaction and epoxidation.'S2 However, this procedure is not attractive for the large-scale preparation of 8 due to drawbacks such as low yield (ca. 30% overall yield), low reaction temperature (-78°C). chromatographic separation and the use of several expensive and hazardous reagents. Therefore, a practical method for synthesis of 8 is desirable. Herein, we report a new efficient and convenient method for the preparation of 8 from commercially available starting material 2.

The structure of related compounds was identified by IR, H-1 NMR and C-13 NMR. The method could be carried out economically on a huge scale.

Abstract:A parallel process for the new synthesis of 2,7-dimethyl-2,4,6-octatriene-1,8-dial was investigated.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-butenal was obtained by a process involving aldehyde protection,ester exchange and catalytic oxidation from 4-acetoxy-2-methyl-2-butenal.A new catalytic oxidation process was proposed to get 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal by oxidating 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol with copper(Ⅰ) chloride、2,2,6,6-tetramethyl-1-piperidinyloxy and oxygen.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol was chlorizated with SOCl_2 in DMF to get 2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane.2,7-Dimethyl-2,4,6-octatriene-1,8-dial was acquired in an overall yield of 48% by one-pot synthesis method from 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal,2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane and triethylphosphite.

Abstract:A procedure for synthesis of 2-methoxyphenothiazine (1) has been developed, starting from resorcinol and aniline by condensation, following methylation and cyclization. p-Toluenesulfonic acid and polysubstituted aromatics were employed as the catalyst of condensation and the solvent of cyclization, respectively, to improve the yield. The use of parallel experiments, statistical experimental design, and multivariate modeling made the total yield of the procedure as high as 74.2%. J. Heterocyclic Chem., (2012).

Abstract:A simple and efficient strategy for the chemoselective synthesis of (2 Z ,5 Z )-3-benzyl/alkyl-5-(2-oxo-5-arylfuran-3(2 H )-ylidene)-2-(phenylimino)thiazolidin-4-one derivatives is developed via a sequential three-component reaction of readily available phenacyl bromides, pyridine and methyl 2-(3-benzyl/alkyl-4-oxo-2-(phenylimino)thiazolidin-5-ylidene)acetates in acetonitrile at room temperature. The advantages of this one-pot sequential Michael addition/1,2-H shift/elimination of pyridine/intramolecular cyclization are highlighted by its low energy requirements (short reaction times at room temperature), excellent yields, the absence of a metal catalyst, and mild conditions. The product structures are characterized by spectroscopic techniques and single-crystal X-ray analysis. Abstract:Aromatic abietane diterpenoids, known for their wide range of biological and pharmaceutical activities, have attracted considerable interest from synthetic and medicinal chemists. Although some aromatic abietane diterpenoids are naturally abundant, many are only available in limited quantities, constraining their use in detailed biological investigations. We initiated a project to synthesize these compounds, and as our first approach, we describe the formal synthesis of ferruginol methyl ether. Starting from tiglic aldehyde ( 6 ) and 2-bromo-5-methoxybenzoic acid ( 11 ), we synthesized the triene ( 3 ) in nine steps. The key transformation involved an acid-catalysed intramolecular Diels-Alder (IMDA) cyclisation of triene ( 3 ), leading to the formation of 12-methoxy-19-norpodocarpa-4(18),8,11,13-tetraene ( 14 ) as the primary product, with an overall yield of 17.3 % across ten steps. While the yield is modest, this approach provides a novel pathway for synthesizing abietane-type compounds. We are actively exploring strategies to improve the overall yield and developing an asymmetric synthesis variant.

Abstract:Purpose To prepare 1-methyl-3-ethyl-3-(3-hydroxyphenyl)-hexahydro-1H-azepin hydrochloride (Ⅰ) from m-methoxyacetophenone. Methods 1-methyl-3-ethyl-3-(3-hydroxyphenyl)-hexahydro-1H-azepin hydrochloride(Ⅰ) was synthesized by eight steps with m-methoxyacetophenone as starting material.The eight steps were Willgerodt rearrangement,formation of acyl chloride,acylation,cyclization,C-ethylation,Wolff-Kishner-Huang reduction and reduction by Lithium aluminium hydride and salt formation. Results 1-methyl-3-ethyl-3-(3-hydroxyphenyl)-hexahydro-1H-azepin Hydrochloride(Ⅰ) was prepared in eight steps from cheap material and reagent on normal condition. Conclusions This method is applicable to industrial production.

Abstract:Aim To synthesize 5'(S)-1,1-dioxolane-5-oxo-(5'-ethyl-5'-hydroxyl-2'H,5'H,6'H-6-oxopyrano)-[3',4'-f]-Δ6(8)-tetrahydroindolizine,a key intermediate for the total synthesis of anticancer drugs 20(S)-camptothecin and its deriveatives.Methods Target compound was synthesized from 1,1-dioxolane-5-oxo-(5'-ethyl-2'H,5'H,6'H-6-oxopyrano)-[3',4'-f]-Δ6(8)-tetrahydroindolizine,via a three-step process including iodine catalyzed oxidation,kinetic resolution and base-hydroxylation.Results and conclusion The synthetic procedure was simple to manipulate and the target compound is obtained with ee 96% optical purity in 15.7% overall yield.

Abstract:A synthetic method for 2-formylcyclopent-2-enecarboxylic acid is described. This procedure comprises two steps. The first step is a [2 + 2] cyclo-addition reaction, and the second step is a hydrolysis and ring-opening reaction. A plausible mechanism of the hydrolysis and ring-opening reaction was supposed. Then, the effects of different solvents, base mol ratio, concentration of base, and reaction temperature on the yield of the second step were studied. Finally, under the obtained optimized conditions, the product was achieved and isolated in 59.4 % yield, which was more than double the ones reported previously. An improved and efficient procedure for the synthesis of 2-formylcyclopent-2-enecarboxylic acid has been developed.
