Chemical Modifications in Eukaryotic Mrna Regulate Gene Expression

Abstract:Since the discovery of the first RNA modification in 1951, more than 160 chemical modifications have been identified in cellular RNA. Previously, most studies of RNA modifications have focused on abundant transfer RNA (tRNA), ribosomal RNA (rRNA), and small nuclear RNA (snRNA). In recent years, with the development of sequencing methods, dynamic modifications have also been identified in mRNA, which leads to the birth and development of “epitranscriptomics” field. In this review, we focus on chemical modifications to internal mRNA, such as N6-methyladenosine (m6A), N6,2′-O-dimethyladenosine (m6Am), N1-methyladenosine (m1A), pseudouridine (Ψ), 5-methylcytosine (m5C), and N7-methylguanosine (m7G). We provide an overview of their modification enzymes, high-throughput detection methods and biological functions, and discuss the current challenges and opportunities in the investigation of mRNA modifications.
