Orthodontic force regulates the osteogenic differentiation of periodontal ligament stem cells in animal study

Abstract:Objective:To study the effects of different orthodontic forces on the proliferation and differentiation of PDLSCs.Methods:The tooth movement model of rats was established with 50 g and 150 g force, respectively. PDLSCs from control, 50 g and 150 g force groups were isolated and cultured after 7 days of force applied. The biological characteristics and molecular signals of PDLSCs were studied.Results:Compared with the control group, the first molar of the rats moved mesially after 50 g force was applied for 7 days. The movement distance of the 150 g force group was greater than that of the 50 g force group. For PDLSC differentiation, the osteogenic and adipogenic differentiation ability of PDLSCs in 50 g force group was higher than that of control group, while the osteogenic differentiation ability of PDLSCs from 150 g force group was significantly lower. The adipogenic differentiation potential of PDLSCs from 150 g force group was higher than that of control group. Mechanically, the expression of active-β-catenin in PDLSCs was increased when 50 g was applied, while its expression was decreased when 150 g was applied, both are compared to control group.Conclusions:Different orthodontic forces have different effects on PDLSCs. 50 g force promotes the osteogenic and adipogenic differentiation of PDLSCs, while 150 g force inhibits the osteogenic differentiation of PDLSCs.
