Expression and clinical significance of microRNA-143 in serum of bladder cancer patients

Abstract:Objective:To explore the expression of microRNA-143 in the serum of bladder cancer patients and its correlation with clinical features and prognosis.Methods:A retrospective study was performed on 68 patients (an observation group) diagnosed with bladder cancer and treated in the Qilu Hospital of Shandong University and Tengzhou Central People's Hospital from June 2013 to June 2016 and 40 healthy individuals (a control group) in the same period. There were 51 males and 17 females in the observation group, with an age of (54.5±6.7). There were 32 males and 8 females in the control group, with an age of (54.8±8.6). Real-time polymerase chain reaction (RT-PCR) was used to detect the expression levels of microRNA-143 in the serum of the bladder cancer patients and the healthy subjects. The expression of microRNA-143 in the serum of the bladder cancer patients and its correlation with clinical pathological indicators (pathological grade, metastasis, and clinical stage) and prognosis were analyzed.Results:There were no statistical differences in the general data between these two groups (all P>0.05). The expression level of microRNA-143 in the serum of the bladder cancer patients was significantly lower than that of the healthy people [(0.68±0.32) vs. (0.87±0.26), P<0.05]. The expression level of microRNA-143 correlated with pathological grade, lymph node metastasis, distant metastasis, and clinical stage (all P<0.05). The median expression level of microRNA-143 in the serum of the bladder cancer patients was 0.652. Among the patients, the 34 patients with an expression of microRNA-143 lower than the median were set as a low expression group, and the 34 patients with an expression of microRNA-143 higher than the median were set as a high expression group. The survival time in the low expression group was significantly lower than that in the high expression group [(37.53±1.85) months vs. (44.97±2.00) months, P<0.05]. Conclusions:MicroRNA-143 is lowly expressed in the serum of bladder cancer patients. Its expression level is correlated with clinical stage, pathological grade, lymph node metastasis, distant metastasis, and prognosis.
