Abstract:Alpine meadows and alpine steppes are two major grassland types distributed on the Tibetan Plateau. Due in large part to the differences in hydrothermal and nutrient conditions following the thawing of lakeshore permafrost, alpine meadows and alpine steppes which are characterized by disparate above- and below-ground biomass, could emerge together in the grassland transition zone between meadows and steppes of the Tibetan Plateau. Bacterial communities are essential components of alpine grassland ecosystems and respond rapidly to environmental changes. Despite their ecological significance, it remains poorly elucidated whether and how the assembly patterns of bacterial communities differed between alpine meadows and alpine steppes. Here, to disentangle the assembly mechanisms of bacterial communities from alpine meadows and alpine steppes, we collected samples from three diverse habitats (i.e., sediments, rhizosphere soils and bulk soils) in both alpine meadow and steppe ecosystems on the Tibetan Plateau. Our results indicated that in both meadows and steppes, rhizosphere bacterial communities exhibited higher alpha-diversity but lower beta-diversity compared to the bacterial communities in sediments and bulk soils. However, the close relationships of bacterial communities between different habitats weakened from meadows to steppes. Null model analysis indicated that the importance of environmental selection shaping bacterial community assemblages in all habitats decreased from meadows to steppes, whereas the role of dispersal limitation showed an opposite pattern. Moreover, pH was the primary driver of phylogenetic turnover of bacterial communities in the steppes across all habitats, whereas the dominant drivers of phylogenetic turnover of bacterial communities in meadows varied with habitat types. Overall, our findings provide novel insights into understanding the differences in microbial communities between meadows and steppes in the grassland transition zone on the Tibetan Plateau.

Abstract:Alpine ecosystem stability and biodiversity of the Tibetan plateau are facing threat from dry valley vegetation uplift expansion, a process which is highly connected to variations in the soil microbial community and soil nutrients. However, the variation of microbial community properties and their relationship to soil nutrients have scarcely been explored in Tibetan dry valleys, which is a gap that hampers understanding the dry valley ecosystem’s response to vegetation change. In this study, we sampled grasslands (G), a grass-shrub transition area (T), and shrublands (S) along an uplift expansion gradient and investigated the link between microbial community properties and soil nutrients. The results showed that shrub degradation by grass expansion in Tibetan dry valley was accompanied by increasing relative phosphorus (P) limitation, which was the main driver for bacterial and fungal composition variation as it offered highest total effect on PC1 (0.38 and 0.63, respectively). Total phosphorus (TP) was in the center module of bacterial and fungal network under shrub soil and even acted as key nodes in fungal networks. During the replacement by grass, TP was gradually marginalized from both bacterial and fungal center network module and finally disappeared in networks, with ammonia and nitrate gradually appearing in the bacterial network. However, TC and total nitrogen (TN) were always present in the center modules of both fungal and bacterial network. These support that a TP variation-induced compositional and network functional shift in the microbial community was a potential reason for vegetation uplift expansion in Tibetan dry valley. This study highlighted the effect of TP on microbial community properties during dry valley vegetation uplift expansion and offered basic information on Tibetan alpine dry valley ecosystem’s response to climate change.

Abstract:To understand effects of soil microbes on soil biochemistry in alpine grassland ecosystems under environmental changes, we explored relationships between soil microbial diversity and soil total nitrogen, organic carbon, available nitrogen and phosphorus, soil microbial biomass and soil enzyme activities in alpine meadow, alpine steppe and cultivated grassland on the Qinghai-Tibetan plateau under three-year warming, enhanced precipitation and yak overgrazing. Soil total nitrogen, organic carbon and NH4-N were little affected by overgrazing, warming or enhanced precipitation in three types of alpine grasslands. Soil microbial biomass carbon and phosphorus along with the sucrase and phosphatase activities were generally stable under different treatments. Soil NO3-N, available phosphorus, urease activity and microbial biomass nitrogen were increased by overgrazing in the cultivated grassland. Soil bacterial diversity was positively correlated with, while soil fungal diversity negatively with soil microbial biomass and enzyme activities. Soil bacterial diversity was negatively correlated with, while soil fungal diversity positively with soil available nutrients. Our findings indicated soil bacteria and fungi played different roles in affecting soil nutrients and microbiological activities that might provide an important implication to understand why soil biochemistry was generally stable under environmental changes in alpine grassland ecosystems.

Abstract:Introduction The functions of terrestrial ecosystems are mainly maintained by bacteria, as a key component of microorganisms, which actively participate in the nutrient cycling of ecosystems. Currently, there are few studies have been carried out on the bacteria contributing to the soil multi-nutrient cycling in responding to climate warming, which hampers our obtainment of a comprehensive understanding of the ecological function of ecosystems as a whole. Methods In this study, the main bacteria taxa contributing to the soil multi-nutrient cycling under the long-term warming in an alpine meadow was determined based onphysichemical properties measurement and high-throughput sequencing, and the potential reasons that warming altered the main bacteria contributing to the soil multi-nutrient cycling were further analyzed. Results The results confirmed that the bacterial β-diversity was crucial to the soil multi-nutrient cycling. Furthermore, Gemmatimonadetes, Actinobacteria, and Proteobacteria were the main contributors to the soil multi-nutrient cycling, and played pivotal roles as keystone nodes and biomarkers throughout the entire soil profile. This suggested that warming altered and shifted the main bacteria contributing to the soil multi-nutrient cycling toward keystone taxa. Discussion Meanwhile, their relative abundance was higher, which could make them have the advantage of seizing resources in the face of environmental pressures. In summary, the results demonstrated the crucial role of keystone bacteria in the multi-nutrient cycling under the climate warming in the alpine meadow. This has important implications for understanding and exploring the multi-nutrient cycling of alpine ecosystems under the global climate warming.

Abstract:Currently, the impact of changes in precipitation and increased nitrogen(N) deposition on ecosystems has become a global problem. In this study, we conducted a 8-year field experiment to evaluate the effects of interaction between N deposition and precipitation change on soil bacterial communities in a desert steppe using high-throughput sequencing technology. The results revealed that soil bacterial communities were sensitive to precipitation addition but were highly tolerant to precipitation reduction. Reduced precipitation enhanced the competitive interactions of soil bacteria and made the ecological network more stable. Nitrogen addition weakened the effect of water addition in terms of soil bacterial diversity and community stability, and did not have an interactive influence. Moreover, decreased precipitation and increased N deposition did not have a superimposed effect on soil bacterial communities in the desert steppe. Soil pH, moisture content, and NH 4 + -N and total carbon were significantly related to the structure of bacterial communities in the desert steppe. Based on network analysis and relative abundance, we identified Actinobacteria, Proteobacteria, Acidobacteria and Cyanobacteria members as the most important keystone bacteria that responded to precipitation changes and N deposition in the soil of the desert steppe. In summary, we comprehensively analyzed the responses of the soil bacterial community to precipitation changes and N deposition in a desert steppe, which provides a model for studying the effects of ecological factors on bacterial communities worldwide.

Abstract:Warming and precipitation anomalies affect terrestrial carbon balance partly through altering microbial eco-physiological processes (e.g., growth and death) in soil. However, little is known about how such processes responds to simultaneous regime shifts in temperature and precipitation. We used the O-water quantitative stable isotope probing approach to estimate bacterial growth in alpine meadow soils of the Tibetan Plateau after a decade of warming and altered precipitation manipulation. Our results showed that the growth of major taxa was suppressed by the single and combined effects of temperature and precipitation, eliciting 40-90% of growth reduction of whole community. The antagonistic interactions of warming and altered precipitation on population growth were common (~70% taxa), represented by the weak antagonistic interactions of warming and drought, and the neutralizing effects of warming and wet. The members in and genera had high growth rates under changed climate regimes. These results are important to understand and predict the soil microbial dynamics in alpine meadow ecosystems suffering from multiple climate change factors.

Abstract:The grassland and shrubland are two major landscapes of the Tibetan alpine meadow, a region very sensitive to the impact of global warming and anthropogenic perturbation. Herein, we report a study showing that a majority of differences in soil microbial community functional structures, measured by a functional gene array named GeoChip 4.0, in two adjacent shrubland and grassland areas, were explainable by environmental properties, suggesting that the harsh environments in the alpine grassland rendered niche adaptation important. Furthermore, genes involved in labile carbon degradation were more abundant in the shrubland than those of the grassland but genes involved in recalcitrant carbon degradation were less abundant, which was conducive to long-term carbon storage and sequestration in the shrubland despite low soil organic carbon content. In addition, genes of anerobic nitrogen cycling processes such as denitrification and dissimilatory nitrogen reduction were more abundant, shifting soil nitrogen cycling toward ammonium biosynthesis and consequently leading to higher soil ammonium contents. We also noted higher abundances of stress genes responsive to nitrogen limitation and oxygen limitation, which might be attributed to low total nitrogen and higher water contents in the shrubland. Together, these results provide mechanistic knowledge about microbial linkages to soil carbon and nitrogen storage and potential consequences of vegetation shifts in the Tibetan alpine meadow.

Abstract:I calculate the screening length and its relation to elastic constants from mean-field models of superionic conductors. The Debye formula for a continuous fluid's screening length is replaced by one that accounts for the discrete nature of the lattice. Interactions alter the screening length and in some cases give oscillations in the charge structure. The screening lengths derived here are exact not only at low temperature where there are few defects, but also at high temperature where the effective interaction strength is small. The mean-field treatment correctly gives the decrease in an elastic constant observed at the superionic transition for a variety of crystals. This decrease is best explained by a change in the defect creation entropy with density. The effect of doping on the elastic constant also agrees well with experiment. The extension to the mean-field models given here provides a consistent picture of the response of a superionic crystal to electrical, mechanical and thermal stresses, as well as to doping.

Abstract:<p>Soil microorganisms play an important role in regulating the feedback of Alpine steppe ecosystems to future climate change. However, the interaction effect of warming and increasing precipitation on soil microorganisms remains unclear, in the face of an ongoing warmer and wetter climate on the Tibetan Plateau. In this study, we investigate the multi-factorial effects on soil microbial diversity, community structure, and microbial interactions in a three-year climate change experiment established in an Alpine steppe on the Tibetan Plateau, involving warming (+2 °C), +15% increasing precipitation and +30% increasing precipitation. Compared to warming, warming plus increasing precipitation alleviated the decrease in microbial diversity, and increased the dissimilarities in microbial community structures, largely influenced by water and substrate availability. We further observed differences in moisture increased the differences in microbial diversity and dissimilarities in microbial community structures across different precipitation levels under ambient temperature. Interestingly, warming plus increasing precipitation could create more ecological niches for microbial species to coexist but may lessen the strength of microbial interactions in contrast to increasing precipitation alone. Collectively, our findings indicate that microbial responses to future climate change in Alpine steppe soils will be more complex than those under single-climate-factor conditions.</p>

Abstract:Composition and traits of soil microbial communities that closely related to their ecological functions received extensive attention in the context of climate changes. We investigated the responses of soil bacterial community structure, traits, and functional genes to the individual warming, precipitation increases, and the combination of warming and precipitation increases in an alpine grassland in the Qinghai-Tibet Plateau that is experiencing warming and wetting climate change. Soil properties, plant diversity and biomass were measured, and the ecological processes and environmental factors driving bacterial community changes were further explored. Results indicated that the Shannon diversity of soil bacterial communities decreased significantly only under the combination treatment, which might due to the decreased plant diversity. Soil bacterial community composition was significantly correlated with soil pH, and was affected obviously by the combination treatment. At the taxonomic classification, the relative abundance of Xanthobacteraceae and Beijerinckiaceae increased 127.67 and 107.62%, while the relative abundance of Rubrobacteriaceae and Micromonosporaceae decreased 78.29 and 54.72% under the combination treatment. Functional genes related to nitrogen and phosphorus transformation were enhanced in the combination treatment. Furthermore, weighted mean ribosomal operon copy numbers that positively correlated with plant aboveground biomass increased remarkably in the combination treatment, indicating a trend of life-history strategies shift from oligotrophic to copiotrophic. Stochastic processes dominated soil bacterial community, and the proportion of stochasticity increased under the combination treatment. Our study highlights the significant effects of simultaneous warming and precipitation increase on soil bacterial community.

Abstract:Soil microbes play a fundamental role in maintaining nutrient biogeochemical cycles. To understand the distribution of soil bacterial communities on grassland plateaus, high-throughput sequencing was used to compare bacterial communities in soils from swamp meadows (SM), alpine meadows (AM), alpine steppes (AS), and desert steppes (DS) at the eastern edge of the Qinghai–Tibetan Plateau (QTP) in China. We then compared response to nitrogen addition between SM and DS soils in microcosms. Bacterial α-diversity decreased from SM > AM > AS > DS. Variations in soil properties across grassland types was associated with different soil bacterial communities corresponding to bacterial species associated with nutrient cycles to those associated with degradation. Soil moisture, pH, and total phosphorus were the main drivers of these differences. Nitrogen addition decreased bacterial diversity but had inconsistent effects on soil bacterial communities in SM and DS, which may also indicate that different alpine grassland soil types have unique bacterial communities. Alpine grassland degradation significantly affects bacterial communities, and the response to nitrogen addition depends on the alpine grassland type. These results allow for better predictions of soil bacteria community-level responses to geochemical and environmental change in alpine areas.

Abstract:Global change may be a severe threat to natural and agricultural systems, partly through its effects in altering soil biota and processes, due to changes in water balance. We studied the potential influence of changing soil water balance on soil biota by comparing existing sites along a natural water balance gradient in the Qinghai-Tibetan Plateau. In this study, the community structure of bacteria, archaea and eukaryotes differed between the different soil water conditions. Soil moisture was the strongest predictor of bacterial and eukaryotic community structure, whereas C/N ratio was the key factor predicting variation in the archaeal community. Bacterial and eukaryotic diversity was quite stable among different soil water availability, but archaeal diversity was dramatically different between the habitats. The auxotype of methanogens also varied significantly among different habitats. The co-varying soil properties among habitats shaped the community structure of soil microbes, with archaea being particularly sensitive in terms of community composition, diversity and functional groups. Bacterial and archaeal phylogenetic community turnover was mainly driven by deterministic processes while stochastic processes had stronger effects on eukaryotic phylogenetic community turnover. Our work provides insight into microbial community, functional group and phylogenetic turnover under different soil conditions in low-latitude alpine ecosystem.

Compositional Shifts and Co-occurrence Patterns of Topsoil Bacteria and Micro-Eukaryotes Across a Permafrost Thaw Gradient in Alpine Meadows of the Qilian Mountains, China

Abstract:Soil microorganisms are pivotal in the biogeochemical cycles of alpine meadow ecosystems affected by permafrost thaw. However, the mechanisms driving microbial community responses to environmental changes, such as variations in permafrost active layer thickness (ALT), are poorly understood. This study utilizes next-generation sequencing to explore the composition and co-occurrence patterns of bacterial and micro-eukaryotic communities in soils along a permafrost thaw gradient. Our findings revealed a decrease in alpha diversity within bacterial communities along the permafrost thaw gradient, while micro-eukaryotic community exhibited an increase. Although shifts were observed in the composition of these communities in both permafrost and seasonally frozen soils, these variations were not statistically significant. Bacterial communities differed more significantly between frozen soil types than within them, a pattern not observed in eukaryotic communities. LEfSe analysis identified more potential biomarkers in bacterial communities than in micro-eukaryotic ones. Furthermore, bacterial co-occurrence networks showed more nodes, edges, and positive linkages compared to those of micro-eukaryotes. Soil texture, ALT, and bulk density significantly influenced bacterial community structures, particularly affecting the abundance of Acidobacteria, Proteobacteria, and Actinobacteria phyla. Conversely, fungal communities (including Nucletmycea, Rhizaria, Chloroplastida, and Discosea groups) were more influenced by electrical conductivity, vegetation coverage, and ALT. This study underscores the differential responses of soil bacteria and micro-eukaryotes to permafrost thaw, highlighting implications for microbial community stability under global climate change.

Abstract:Carbon cycling and associated microbial responses in both subsoil and topsoil was studied in a Tibetan alpine grassland over 4 years of warming and altered precipitation. Compared to the unchanged topsoil carbon, subsoil carbon exhibited a stronger response to the interactive effect of warming and altered precipitation. More than twice as many populations whose MAG abundances correlated significantly with the variations of carbon content, components and fluxes were observed in the subsoil than topsoil, suggesting their potential contribution in mediating subsoil carbon cycling. Subsoil is a large organic carbon reservoir, storing more than half of the total soil organic carbon (SOC) globally. Conventionally, subsoil is assumed to not be susceptible to climate change, but recent studies document that climate change could significantly alter subsoil carbon cycling. However, little is known about subsoil microbial responses to the interactive effects of climate warming and altered precipitation. Here, we investigated carbon cycling and associated microbial responses in both subsoil (30–40 cm) and topsoil (0–10 cm) in a Tibetan alpine grassland over 4 years of warming and altered precipitation. Compared to the unchanged topsoil carbon (β = .55, p = .587), subsoil carbon exhibited a stronger response to the interactive effect of warming and altered precipitation (β = 2.04, p = .021), that is, warming decreased subsoil carbon content by 28.20% under decreased precipitation while warming increased subsoil carbon content by 18.02% under increased precipitation.Furthermore, 512 metagenome‐assembled genomes (MAGs) were recovered, including representatives of phyla with poor genomic representation. Compared to only one changed topsoil MAG, 16 subsoil MAGs were significantly affected by altered precipitation, and 5 subsoil MAGs were significantly affected by the interactive effect of warming and precipitation. More than twice as many populations whose MAG abundances correlated significantly with the variations of carbon content, components and fluxes were observed in the subsoil than topsoil, suggesting their potential contribution in mediating subsoil carbon cycling. Collectively, our findings highlight the more sensitive responses of specific microbial lineages to the interactive effects of warming and altered precipitation in the subsoil than topsoil, and provide key information for predicting subsoil carbon cycling under future climate change scenarios.

Effects of warming and isolation from precipitation on the soil carbon, nitrogen, and phosphorus, and their stoichiometries in an alpine meadow in the Qinghai–Tibet Plateau: A greenhouse warming study

Abstract:Introduction: In the Qinghai–Tibet Plateau (QTP), alpine meadows are among the most noticeable reflection of global climate change. However, effects of global warming on soils hosting alpine meadows in the QTP, such as reduced moisture because of low precipitation, remain unclear. Methods: Here, the soil moisture content (SMC), pH, dissolved organic carbon (DOC), ammonium nitrogen (NH 4 + –N), nitrate nitrogen (NO 3 − –N) and available phosphorus (AP) contents in the QTP were analyzed. The changes in and stoichiometries of total carbon, nitrogen, and phosphorus (TC, TN, and TP), microbial biomass carbon, nitrogen, and phosphorus (MBC, MBN, and MBP), β-1,4-glucosidase (BG), β-1,4-N-acetylglucoaminosidase (NAG), leucine aminopeptidase (LAP), and acid phosphatase (ACP) in the 0–30 cm layer of soils associated with warming in a greenhouse in the QTP from 2015 to 2020 were characterized. Results: We found that warming in the greenhouse significantly decreased the SMC, NO 3 − –N, MBC, MBN, MBP, BG, LAP, ACP, and enzymatic C:N ratio. The warming increased the DOC, NH 4 + –N, AP, MBC:MBN, and enzymatic N:P ratios noticeably. The pH, TC, TN, TP, C:N, C:P, N:P, MBC:MBP, MBN:MBP, and enzymatic C:P ratios were minimally affected. Conclusion: The results showed that warming and isolation from precipitation promoted mineralization of N and P in the soil but did not significantly alter the cycling of elements in soils in an alpine meadow.

Abstract:Understanding how microbial processes develop and change in alpine meadow soils is key to global initiatives toward environmental sustainability and local land management. Yet, how microbial interactions mediate soil multifunctionality in disturbed and managed alpine meadows remains understudied. Here, we investigated multiple community metrics, particularly microbial network properties and assembly processes, of soil bacterial and fungal communities and their links to certain soil functions along a degradation-restoration sequence of alpine meadows in the Qinghai-Tibetan Plateau. Meadow degradation caused significant declines in soil hydraulic conductivity (e.g., higher bulk density, reduced soil porosity and water content) and nitrogen availability, leading to lowered soil multifunctionality. Meadow degradation only caused weak changes in microbial abundance, alpha diversity, and community composition, but remarkably reduced bacterial network complexity, to a less extent for fungal network properties. Short-term artificial restoration with productive grass monocultures did not restore soil multifunctionality, in turn even destabilized bacterial network and favored pathogenic over mutualistic fungi. Soil fungi community are more stable than bacteria in disturbed alpine meadows, and they evolved with distinct assembly strategies (stochastic-dominant versus deterministic-driven processes, respectively). Further, microbial network complexity, positively and better predicts soil multifunctionality than alpha diversity. Our work shows how microbial interaction complexity may enhance soil multifunctionality in degraded alpine meadow ecosystems, noting that meadow restoration with low plant species diversity may failed in restoring multiple ecosystem functions. These findings would help predict the outcomes of global environmental changes and inform management strategies in regional grassland conservation and restoration.

Abstract:The alpine ecosystem as one of the most representative terrestrial ecosystems has been highly concerned due to its susceptibility to anthropogenic impacts and climatic changes. However, the distribution pattern of alpine soil bacterial communities and related deterministic factors still remain to be explored. In this study, soils were collected from different altitudes and slope aspects of the Mount (Mt.) Shergyla, Tibetan Plateau, and were analyzed using 16S rRNA gene-based bioinformatics approaches. Acidobacteriota and Proteobacteria were identified consistently as the two predominant phyla in all soil samples, accounting for approximately 74% of the bacterial community. The alpha diversity of the soil bacterial community generally increased as the vegetation changed with the elevated altitude, but no significant differences in alpha diversity were observed between the two slopes. Beta diversity analysis of bacterial community showed that soil samples from the north slope were always differentiated obviously from the paired samples at the south slope with the same altitude. The whole network constituted by soil bacterial genera at the Mt. Shergyla was parsed into eight modules, and Elev-16S-573, Sericytochromatia, KD4-96, TK10, Pedomicrobium, and IMCC26256 genera were identified as the "hubs" in the largest module. The distance-based redundancy analysis (db-RDA) demonstrated that variations in soil bacterial community thereof with the altitude and slope aspects at the Mt. Shergyla were closely associated with environmental variables such as soil pH, soil water content, metal concentrations, etc. Our results suggest that environmental variables could serve as the deterministic factors for shaping the spatial pattern of soil bacterial community in the alpine ecosystems.

Abstract:Biological soil crusts (BSCs), a vital component of ecosystems, are pivotal in carbon sequestration, nutrient enrichment, and microbial diversity conservation. However, their impact on soil microbiomes in alpine regions remains largely unexplored. Therefore, this study aimed to determine the influence of BSCs on alpine grassland soil microbiomes, by collecting 24 pairs of soils covered by biological and physical crusts along a transect on the Qinghai-Tibetan Plateau. We found that BSCs significantly increased the contents of soil moisture, organic carbon, total nitrogen, and many available nutrients. They also substantially altered the soil microbiomes. Specifically, BSCs significantly increased the relative abundance of Cyanobacteria, Verrucomicrobiota, and Ascomycota, while decreasing the proportions of Gemmatimonadota, Firmicutes, Nitrospirae, Mortierellomycota, and Glomeromycota. By contrast, microbial abundance and α-diversity demonstrated low sensitivity to BSCs across most study sites. Under the BSCs, the assembly of prokaryotic communities was more affected by homogeneous selection and drift, but less affected by dispersal limitation. Conversely, soil fungal community assembly mechanisms showed an inverse trend. Overall, this study provides a comprehensive understanding of the effects of BSCs on soil properties and microbial communities, offering vital insights into the ecological roles of BSCs.
