A Class of Protein Coding RNAs Bind to PRC2 and Regulate Histone Methylation

Abstract:Background: Polycomb repressive complex 2 (PRC2), mediating the modification of H3K27me3, plays an important role in transcriptional repression. However, the mechanism of PRC2 recruitment remains unclear. Methods: The Bayesian gene regulatory network analysis, GSEA (gene-set enrichment analysis) and WGCNA (weighted gene co-expression network analysis) were performed to study the association between PRC2 binding RNAs and genes downstream of PRC2. The interaction between RNA and EZH2 was determined by RNA-pulldown assay. The gene expression was explored by qRT-PCR. Findings: Like non-coding RNAs, a class of protein coding RNAs bind to PRC2 and regulate gene expression through histone epigenetic modifications rather than their protein function. The alteration of TSLP expression by the typical protein-coding gene TMEM117 is EZH2 dependent. Nucleus enrichment and RNA recognition motif of PRC2 containing maybe the reasons that these protein coding RNAs bind to PRC2 and regulate histone methylation. The same regulatory pattern is found in mice as well.
