Cold-Induced Lipid Dynamics and Gene Programs Highlight a Role of White-Adipose Browning in Health and Lifespan

Abstract:In mammals, cold exposure induces browning of white adipose tissue (WAT) and alters WAT gene expression and lipid metabolism to boost adaptive thermogenesis and maintain body temperature. In order to provide a comprehensive resource for describing the molecular signatures of cold-induced WAT browning, we applied RNA-sequencing (RNA-seq) and mass-spectrometry to study the adaptive changes of lipidomic and transcriptomic profiles, respectively, in inguinal WAT (iWAT) in response to cold. We showed that short-term (3-day) cold exposure induced browning of iWAT, increased energy expenditure and resulted in loss of body weight and fat mass. Lipidomics analysis showed that short-term cold exposure lead to dramatic changes of the overall composition of lipid classes in iWAT. Notably, cold exposure induced significant changes in the acyl chain composition of triacylglycerols (TAGs), as well as the levels of glycerophospholipids and sphingolipids in iWAT. RNA-seq and qPCR results showed that short-term cold exposure altered the expression of genes and pathways involved in fatty acid elongation, and the synthesis of TAGs, sphingolipids and glycerophospholipids.
