We hypothesize that ERK is also involved in the development and differentiation of benign prostate epithelial and stromal cells. In the present study, we tested this hypothesis in the ERK2 knockout mice. METHODS ERK2 knockout mice Mapk1flox/flox in C57BL/6 background were provided by Dr. Landreth (Case Western University), FSP1-CRE mice in C57BL/6 background were by Dr. Bhowmick (Vanderbilt University) and AAR2PB-CRE mice in FVB background15 by Dr. Zhang (Northwestern University). We have crossed Mapk1flox/flox with FSP1-CRE mice to generate Mapk1fspKO mice with ERK2 knockout in the prostate stromal cells, and crossed Mapk1flox/flox with AAR2PB-CRE mice to generate Mapk1AAR2PbKO mice with homozygous deletion of the ERK2 gene in prostate epithelial cells expressing the CRE. The mice ventral prostate tissues were isolated in 6 weeks, the positive genotypes of knockout mice were confirm by PCR approach by using mice tail tissues. We studied 20 mice in each group, and the naïve C57BL/6 mice were used as the control. The immunohistochemistry staining for Ki67, CK18 and vimentin were performed to evaluate the proliferation and differentiation of prostate cells. RESULTS In comparison with controls, ERK knockout in epithelial cells (Mapk1AAR2PbKO) resulted in a significant atrophy of epithelial cells and much less secretion of prostate glands, and down-regulation of Ki67 and CK18 expression in epithelial cells. While ERK knockout in stromal cells (Mapk1fspKO) also induced down-regulation of Ki67 and vimentin, it did not change the phenotype of epithelial cells or the secretion of prostate grands. Furthermore, we found an up-regulation of both Ki67 and CK18 expression and epithelial cells when we knocked-out ERK in stromal cells, suggesting a close interaction between epithelial cells and stromal cells during the development. CONCLUSIONS Our findings suggest that ERK is important in the proliferation of both epithelial o and stromal cells and their functions including secretion. In addition, our findings suggest that ERK may mediate a crosstalk between epithelial and stromal cells during prostate development.
