Abstract:Optofluidic biolaser is an emerging technology for chemical and biomedical sensing. However, the sensitivity of optofluidic laser is still limited by the laser threshold condition. In this paper we report a DC-biased optofluidic biolaser for uric acid (UA) detection. Inside a Fabry-Perot laser cavity, a coupled enzyme reaction was employed to transform the detection of UA into the detection of analyte-converted hydrogen peroxide (H2O2). The fluorescent product of the enzyme-catalyzed reaction, resorufin, was employed as gain medium for optofluidic lasing. An optimized concentration of H2O2 was pre-added as a DC bias to counterbalance the requirement of high analyte concentration for lasing. UA detection with a limit of detection of 3.63 mu M was achieved with a low sample volume as small as 10 mu l. This work provides a sensitive technology using optofluidic biolaser for the detection of substance in human body fluids that can be converted into H2O2.

Abstract:The increased uric acid (UA) level is closely related to many diseases. However, the detection of UA remains great challenges due to the pain of blood testing and the complexity of common methods. Therefore, the detection of UA is of great significance for public health. In this paper, a new method for UA detection is proposed. The swellable hydrogel microneedles (hydrogel MNs) can be used for minimally invasive extraction of skin interstitial fluid (ISF). The colorimetric detection of trace UA in ISF can be successfully achieved by virtue of DNAzyme@MOFs with enhanced peroxidase activity. This study provides a powerful tool for minimally invasive, painless, rapid and convenient detection of UA in clinical applications. This study not only avoids the use of expensive and unstable natural enzymes but also expands the applications of DNAzyme@MOFs in biomedical detection.
Coiled Optical Nanofiber for Optofluidic Absorbance Detection.

Abstract:A challenge for optofluidic absorbance detection is the high concentration limit of detection due to the short optical path length. Herein, we introduce a concept of utilizing the coiled optical nanofiber for highly sensitive and robust optofluidic absorbance detection. Investigated by measuring the absorbance of FeCl3 solutions, the sensor shows a detection limit down to 10 μM with excellent reversibility in a concentration range of 0-5 mM. The sensitivity is 10-fold higher than that of standard absorbance measurement by using a 1 cm cuvette. Also, highly sensitive chloramphenicol sensing was demonstrated by using the enzyme-linked immunosorbent assay (ELISA) method, achieving a detection limit below 0.5 ng/L. The higher sensitivity and lower detection limit are caused by the large fractional power of evanescent field outside the nanofiber and the long detection length, which can effectively improve the absorption of the evanescent field, while the excellent reversibility is caused by the support of a polydimethylsiloxane (PDMS) pillar rather than by suspending the nanofiber in the microchannel. We envision that the present work may open up new opportunities for ultrasensitive chemical and biological sensing.
Enzyme Catalyzed Optofluidic Biolaser for Sensitive Ion Concentration Detection

Abstract:The enzyme horseradish peroxidase (HRP) has been extensively used in biochemistry for its ability to amplify a weak signal. By using HRP catalyzed substrate as the gain medium, we demonstrate sensitive ion concentration detection based on the optofluidic laser. The enzyme catalyzed reaction occurs in bulk solution inside a Fabry-Perot laser cavity, where the colorless, non-fluorescent 10-Acetyl-3,7-dihydroxyphenoxazine (ADHP) substrate is oxidized to produce highly fluorescent resorufin. Laser emission is achieved when pumped with the second harmonic wave of a Q-switched YAG laser. Further, we use sulfide anion (S2-) as an example to investigate the sensing performance of enzyme catalyzed optofluidic laser. The laser onset time difference between the sample to be tested and the reference is set to be the sensing output. Thanks to the amplification effects of both the enzymatic reaction and laser emission, we achieve a detection limit of 10 nM and a dynamic range of 3 orders of magnitude.

Abstract:Optofluidic laser (OFL) is an emerging technology that integrates microfluidics, optical microcavity, and the liquid gain medium. Thanks to the strong light-matter interaction in the laser cavity, OFL could perform bio-chemical sensing with high performance. Here, we review the recent advances in optofluidic lasers and highlight their applications in biochemical analysis with unique properties. We introduce the physical mechanisms as well as the structure and material employed in the optofluidic laser to achieve high sensitivity, disposability, fast response and high throughput. In addition, we discuss the cross properties between these performance indicators to show how to achieve overall high performance. The effects of new materials including both gain materials and enhancer materials in laser-based sensors are briefly discussed. Furthermore, the comparison with other optical biochemical sensors and the prospects of OFLs are also given.

Due to the optical feedback provided by the laser, the small difference in ion concentrations can be amplified. Consequently, a detection limit of 10 nM is achieved with a dynamic range as large as three orders of magnitude, representing significant improvement over the traditional fluorescence and colorimetric methods. This work will open a door to a new catalytic-laser-based chemical sensing platform for detecting a wide range of species that could inhibit the catalytic reaction.

Abstract:A novel fiber optofluidic laser biosensor based on nanoparticle-functionalized thin-walled hollow optical fiber is developed for sensitive detection of enzyme horseradish peroxidase (HRP). High sensitivity is achieved thanks to the high surface-to-volume ratio of the nanomaterials. Two kinds of attractive nanomaterials, i.e., Au nanorods and SiO2 nanoparticles, are selected as demonstrative models to be coated onto the inner surface of thin-walled hollow fiber through electrostatic adsorption and biochemical cross-linking, respectively, while the streptavidin-biotin conjugation mechanism acts as a bridge for the specific recognition of enzyme. The nanoparticle-functionalized fiber optofluidic lasers have lower lasing threshold, compared to the control group without nanomaterial. We experimentally demonstrate that the nanomaterials enhanced fiber optofluidic laser biosensors can detect HRP in the range of 7515000 pM with a good linearity in semi-logarithmic scale, and with a sensitivity enhancement of 57.4 by using the SiO2 nanoparticles. We believe that this nanoparticle-enhanced fiber optofluidic laser biosensor could provide an efficient strategy for biomolecule detection such as enzyme-linked immunosorbent assay (ELISA).

Abstract:Microstructures can improve both sensitivity and assay time in heterogeneous assays (such as ELISA) for biochemical analysis; however, it remains a challenge to perform the essential wash process in those microstructure-based heterogeneous assays. Here, we propose a sequential bioconjugation protocol to solve this problem and demonstrate a new type of fiber optofluidic laser for biosensing. Except for acting as an optical microresonator and a microstructured substrate, the miniaturized hollow optical fiber (HOF) is used as a microfluidic channel for storing and transferring reagents thanks to its capability in length extension. Through the capillary action, different reagents were sequentially withdrawn into the fiber for specific binding and washing purposes. By using the sequentially bioconjugated FOFL, avidin molecules are detected based on competitive binding with a limit of detection of 9.5 pM, ranging from 10 pM to 100 nM. It is demonstrated that a short incubation time of 10 min is good enough to allow the biomolecules to conjugate on the inner surface of the HOF. Owing to its miniaturized size, only 589 nL of liquid is required for incubation, which reduces the sample consumption and cost for each test. This work provides a tool to exploit the potential of microstructured optical fibers in high-performance biosensing.

Abstract:Optofluidics organically integrates microfluidics and photonics and is an emerging technology in biological and chemical analysis. In this paper, we overview the recent studies in bio-chemical sensing applications of optofluidics. Particularly, we report the research progress in our lab in developing diverse optofluidic devices using two unique configurations: thin-walled capillary based optofluidic ring resonator (OFRR) and multi-hole capillary based optofluidic platforms. The first one has been developed to be OFRR-based label-free biosensor, microfluidic laser based intra-cavity sensors, and on-column optical detectors for micro-gas chromatography (mu GC), while the second one has been developed to be optofluidic Fabry-Perot based label-free biosensor and optofluidic Surface-Enhanced Raman Spectroscopy (SERS) biosensor. All of these devices take advantage of superior fluidic handling capability and high sensitivity, and have been used in detecting various biological and chemical analytes in either liquid or vapor phase.

Abstract:We experimentally demonstrated a chemical sensing system for Cu2+ detection based on an optofluidic laser. A Fabry–Perot cavity within a square quartz capillary fluidic channel was built to enable optofluidic lasing. Rh6G-HS (Rh6G, hydrazine hydrate, salicylaldehyde), a rhodamine derivative was used as the laser gain medium, and the switching mechanism of spirolactam (fluorescence: “off”) to ring-opened amide (fluorescence: “on”) was used to provide strong fluorescence emission with the addition of Cu2+ ions. As the Cu2+ ion concentration increased, the optofluidic laser slope efficiency was enhanced and the lasing threshold was reduced. Under optimized conditions, the chemosensor exhibited a detection limit of $300~\mu \text{M}$ and satisfactory selectivity toward Cu2+ ions over other metal ions. This method may provide a new way to detect metal ions and could be applied to new applications in environmental protection and biochemical research.

Abstract:A highly sensitive integrated optical biosensor based on the cascade of a Fabry-Perot cavity laser and a microring resonator is investigated theoretically and experimentally. The free spectral ranges of the microring and the laser cavity are designed to be about the same in order to apply intensity interrogation. The variation of the refractive index of the fluidic sample results in a large shift in the envelope function of the total transmission spectrum of the sensor, and consequently, a large change in the transmitted power. The simple light-source-integrated sensing scheme achieved a high sensitivity of 1000 dB/RIU in the preliminary experiment, demonstrating its principle and potential for low-cost practical applications.

Abstract:Optofluidic bio-lasers are currently of high interest for sensitive, intra-cavity, biochemical analysis. In comparison with conventional methods such as fluorescence and colorimetric detection, lasers provide us with a method for amplifying small concentration differences in the gain medium, thus achieving high sensitivity. Our previous research has demonstrated that sandwich IL-6 ELISA performed in capillary-based optofluidic laser cavity was able to achieve ultrahigh detection sensitivity (LOD between 1-10 fg/ml) with a small sample volume (~20 μL). However, such approach has several limitations such as low repeatability and long assay time (~8 hours in total, 7 hours for laser measurements). Here, we developed a novel on-chip ELISA laser platform by directly fabricating micro-wells on dielectric mirrors for immunosorbent reactions. Polystyrene microbeads of 30 μm in diameter were placed in the wells to optically enhance the resonance cavity during laser measurement, thus significantly improving reliability, shortening assay time (~1.5 hours, 30 minutes for laser measurements) while maintaining the attractive features such as small sample volume and very high sensitivity (LOD ~0.1 pg/mL for IL-6). This work pushes the ELISA laser one step closer to solving problems in realworld biochemical analysis.

Abstract:Optofluidic laser is an emerging method for biochemical sensing. The topic on, which kind of structure is the best to integrate the laser resonator and microfluidic channel, is worth to be explored. Here, we report a hollow optical fiber optofluidic laser based on a dye-doped polymer film. This fiber microstructure is a promising candidate that serves both as a laser microresonator and a compact microfluidic channel. The refractive index of the polymer film is higher than silica so that the whispering gallery mode can be supported. The coating process is explored in detail. The laser spectra and threshold properties of the HOF microlaser are investigated. The dye-doped polymer film can be controlled to be as thin as 1.6 μm and a threshold of below 1 μJ/mm2 can be reached. Thanks to the strong light-liquid interaction enabled by the polymer coating on the inner interface of HOF, sensitive refractive index detection is demonstrated by monitoring the laser wavelength shift, which has the potential for wavelength division multiplexing. This laser sensor is promising for label-free biochemical detection.
Dual optofluidic distributed feedback dye lasers for multiplexed biosensing applications

Abstract:Integrated optofluidic devices have become subjects of high interest for rapid biosensor devices due to their unique ability to combine the fluidic processing of small volumes of microfluidics with the analysis capabilities of photonic structures. By integrating dynamically reconfigurable optofluidic lasers on-chip, complex coupling can be eliminated while further increasing the capabilities of sensors to detect an increasing number of target biomarkers. Here, we report a polydimethylsiloxane-based device with two on-chip fluidic distributed feedback (DFB) laser cavities that are integrated with an orthogonal analyte channel for multiplexed fluorescence excitation. One DFB grating is filled with 4-(dicyanomethylene)-2-methyl-6-(4-dimethylaminostyryl)-4H-pyran dissolved in dimethyl sulfoxide. The second grating is filled with rhodamine 6G dissolved in a diluted ethylene glycol solution. We present characterization of both lasers through analysis of the lasing spectra for spectral narrowing along with a power series to observe threshold behavior. We then demonstrate simultaneous detection of two different fluorescent microbeads as a proof of concept for scalable, single biomarker analysis using on-chip optofluidic lasers.

Abstract:Optofluidic lasers (OFLs) are an emerging technological platform for biochemical sensing, and their good performance especially high sensitivity has been demonstrated. However, high-throughput detection with an OFL remains a major challenge due to the lack of reproducible optical microcavities. Here, we introduce the concept of a distributed fibre optofluidic laser (DFOFL) and demonstrate its potential for highthroughput sensing applications. Due to the precise fibre geometry control via fibre drawing, a series of identical optical microcavities uniformly distributed along a hollow optical fibre (HOF) can be achieved to obtain a one-dimensional (1D) DFOFL. An enzymatic reaction catalysed by horseradish peroxidase (HRP) can be monitored over time, and the HRP concentration is detected by DFOFL-based arrayed colorimetric detection. Experimentally, five-channel detection in parallel with imaging has been demonstrated. Theoretically, spatial multiplexing of hundreds of channels is achievable with DFOFL-based detection. The DFOFL wavelength is tuned over hundreds of nanometers by optimizing the dye concentration or reconfiguring the liquid gain materials. Extending this concept to a two-dimensional (2D) chip through wavelength multiplexing can further enhance its multi-functionality, including multi-sample detection and spectral analysis. This work opens the door to high-throughput biochemical sensing.
Simple enzyme-free detection of uric acid by an in situ fluorescence and colorimetric method based on Co-PBA with high oxidase activity

Abstract:In this work, we prepared a simple and low-cost cobalt-doped Prussian blue analog (Co-PBA), which can directly oxidize 10-acetyl-3,7-dihydroxyphenoxazine and 3,3',5,5'-tetramethylbenzidine (TMB) to produce resorufin (ox-AR) with high fluorescent quantum yield and ox-TMB with blue color, respectively, without the need for unstable H2O2. Using the Michaelis-Menten curve and Lineweaver-Burk equation, the Michaelis-Menten constant of Co-PBA and the substrate TMB was found to be 0.033 mM, which was much lower than horseradish peroxidase and other reported nanozymes, showing satisfactory substrate affinity. Uric acid (UA) can cause erosion of the Co-PBA structure, and it significantly reduces the catalytic activity of Co-PBA, resulting in the decrease of the fluorescence emission signal of ox-AR and the absorption signal of ox-TMB. Based on this, a simple, sensitive, and fast fluorescence/colorimetric dual-mode uric acid detection platform was established. The detection range for UA by fluorescence method is 0.625-40 μM, and the detection limit (LOD, S/N = 3) is as low as 0.389 μM. The detection system was applied to serum samples with good recovery and can be used for field detection of UA in biological samples under different environments to meet different needs.

Abstract:A coupled optofluidic ring laser sensor is fabricated, and its ultrasensitive bulk refractive index and surface mass sensing properties are demonstrated. The sensor consists of a cylindrical ring laser and a thin-walled optofluidic capillary ring resonator. Coupling of the whispering gallery mode in the two resonators generates a largely magnified sensitivity due to the Vernier effect. A sensitivity of 2510 nm/RIU and magnification factor of 355 are achieved experimentally in aqueous solution, which corresponds to a noise equivalent detection limit of 1.6 x 10(-5) RIU for bulk RI detection. The new sensor configuration provides a convenient way to achieve ultrasensitive biological and chemical sensing. (C) 2012 Optical Society of America
