Long Non-Coding RNA NEAT1 Inhibits Hepatocyte Proliferation in Fulminant Hepatic Failure Through H3K27me3 Methylation-Dependent Promotion of LATS2

Abstract:Background: Apoptosis is an important cellular pathological process in D-galactosamine (D-GalN)/lipopolysaccharide (LPS)-stimulated liver injury. Long noncoding RNAs (lncRNA) are involved in liver injury, but their mechanisms of action in fulminant hepatic failure (FHF) remain unclear. The aim of this study is to investigate the effect of NEAT1, an lncRNA, on apoptosis and proliferation of hepatocytes.Methods: Initially, we collected FHF clinical samples and established an in vitro cell model and an in vivo rat model of FHF by using (D-GalN)/LPS. The expression pattern of NEAT1 and LATS2 were then determined by RT-PCR and ELISA assay. Following the verification of the binding relationship between NEAT1, EZH2 and LATS2, gain-of- and loss-of-function experiments were applied to analyze the effects of NEAT1, EZH2 and LATS2 on hepatocyte proliferation and apoptosis, determined by EdU and Tunnel assay respectively. In addition, involvement of Hippo-YAP pathway was explored.Results: NEAT1 was highly expressed in FHF tissues, FHF cells and FHF rat model, in contrast LATS2 was downregulated. LATS2 was negatively correlated with NEAT1. After D-GalN/LPS treatment, NEAT1 inhibited the expression of LATS2 and cell proliferation, but promoted apoptosis. Furthermore, mechanistic investigation indicated that NEAT1 recruited EZH2 to LAST2 promoter region and mediate the methylation of H3K27me3, ultimately activate the Hippo-YAP axis. Besides, down-regulation of NEAT1 or overexpression of LAST2 inhibits liver injury caused by FHF in vivo.Conclusion: The NEAT1 modulate hepatocyte apoptosis in fulminant hepatic failure by recruiting EZH2 to LATS2 promoter region and promotes H3K27me3 methylation, highlighting new therapeutic directions in FHF.Funding: This study was supported by Hunan Provincial Natural Science Foundation Youth Fund Project (No. 2017JJ3427).Declaration of Interest: The authors declare that they have no competing interests.Ethical Approval: The study was approved by the ethics committee of the Third Xiangya Hospital of Central South University and the informed consents were singed by all participants or their guardians.
