Abstract:Ruthenium-catalyzed sigma-bond activation-assisted meta-C-H functionalization has emerged as a useful tool to forge distal C-C bonds. However, given the limited number of mechanistic studies, a clear understanding of the origin of the site-selectivity and the complete reaction pattern is not available. Here, we present systematic computational studies on ruthenium-catalyzed C-H functionalization with primary, secondary, tertiary alkyl bromides and aryl bromides. The C-H scission and the C-C formation were carefully examined. Monocyclometalated ruthenium(II) complexes were identified as the active species, which then underwent inner-sphere single electron transfer (ISET) to activate the organic bromides. The site-selectivity results from the competition between the close-shell reductive elimination and the open-shell radical coupling. Based on this mechanistic understanding, a multilinear regression model was built to predict the site-selectivity, which was further validated by experiments.

Abstract:Radical‐mediated sp3 C–H functionalization has found broad applications in synthesis and late‐stage derivatization of complex molecules. The site‐selectivity of a given reaction is mainly dictated by the bond strength, polarity and steric accessibility of diverse C–H bonds as well as the characters of hydrogen atom acceptors. Unactivated sp3 C–H bonds usually follow the relative reactivity sequence of tertiary > secondary > primary, which is even recognized as the innate preferences of certain substrates. The development of innovative methods capable of overriding the "inherent" site‐selectivity is exceedingly attractive. Although it has been demonstrated that secondary C–H bonds could be selectively converted in the presence of weaker tertiary C–H bonds by tuning the steric and electronic properties of hydrogen atom acceptors, the selective activation of primary C–H bonds remains a daunting challenge. Experimental–Computational Synergy for Selective Pd(II)-Catalyzed C–H Activation of Aryl and Alkyl Groups

Abstract:C-H activation and functionalization are on the forefront of modern synthetic chemistry. Imagine if any C-H bond of a molecule could be converted to a C-X bond, where X is the target functionality-this would alter the synthetic blueprints for complex target molecules since it would provide novel disconnections in retrosynthetic analysis. Collaborations between many experimental and computational groups have led to rapid developments of new C-H functionalization methods. Our groups represent an example of this; we were brought together as part of the NSF-supported Center for Selective C-H Functionalization. Many examples of experimental-computational synergy for selective Pd(II)-catalyzed C-H activation of aryl and alkyl groups are described in this Account. We describe computations by the Houk group made in response to experimental stimuli by the Yu group. The first section discusses the experimental and computational investigations of oxazoline-directed stereoselective Pd(II)-catalyzed C(sp3)-H bond activation that occurs through the concerted metalation-deprotonation (CMD) pathway involving a monomeric Pd(II) complex. The second section involves two types of bidentate ligands, mono-N-protected amino acid (MPAA) and acetyl-protected aminoethyl quinoline (APAQ) ligands that promote the C-H activation reactions with the ligand as the internal base. In the MPAA-assisted remote C-H bond activation, the basic dianionic amidate ligand participates in the deprotonation of a specific C-H bond. This mechanism accounts for the improved reactivity and selectivity in C-H activation reactions with MPAA ligands. The chiral APAQ ligands enable asymmetric palladium insertion into prochiral C-H bonds on a single methylene carbon center. The dianionic amidate of the APAQ ligand acts as an intramolecular base to deprotonate the methylene C-H asymmetrically and facilitate chiral Pd-C bond formation. The origins of the dramatic differences of five-membered (relatively inactive) and six-membered (highly active) chelation in β-methylene C(sp3)-H activation reactions by a Pd(II) catalyst were explained with density functional theory (DFT) calculations. This is mainly due to the steric repulsions between the ArF group of the substrate and the quinoline group of the ligand. The steric repulsion between the ArF group of the substrate and the quinoline group of the APAQ ligand destabilizes the five-membered chelate transition structure, increasing the energy of the transition state. The third section discusses a mechanism involving a Pd-Ag heterodimeric complex intermediate in the template-directed, Pd(II)-catalyzed remote meta functionalization of toluene derivatives and benzoic acid derivatives. The nitrile directing group of the template coordinates with Ag while the Pd is placed adjacent to the meta-C-H bond in the transition state, leading to the observed high meta selectivity. The selective activation of remote meta-C-H bonds at various distances can be achieved by tuning the template. The dual role of AgOAc as both an oxidant and part of the heteronuclear active species in the mechanism involving PdAg(OAc)3 was determined by DFT calculations and is in accord with literature information about complexes. For the systems discussed in these three sections, the similarity is that they all proceed via the CMD mechanism. The differences lie in the proton acceptors and the active Pd species. Common CMD involves a monomeric Pd mechanism with acetate as the proton acceptor. Both MPAA and APAQ ligands react via monomeric Pd mechanisms with a ligand moiety (the amidate oxygen) as the proton acceptor. Nitrile-containing template-mediated meta-C-H activations proceed via a Pd-Ag heterodimeric mechanism, still with acetate as the proton acceptor. The interaction between our two groups, experts in experiment and computation, and the discoveries made possible by that interplay are highlighted in this Account.

Breaking Conventional Site Selectivity in C–H Bond Activation: Selective Sp3 Versus Sp2 Silylation by a Pincer-Based Pocket

Abstract:A deeply ingrained assumption in the conventional understanding and practice of organometallic chemistry is that an unactivated aliphatic C(sp3)-H bond is less reactive than an aromatic C(sp2)-H bond within the same molecule given that they are at positions unbiasedly accessible for activation. Herein, we demonstrate that a pincer-ligated iridium complex catalyzes intramolecular dehydrogenative silylation of the unactivated δ-C(sp3)-H (δ to the Si atom) with exclusive site selectivity over typically more reactive ortho δ-C(sp2)-H bonds. A variety of tertiary hydrosilanes undergo δ-C(sp3)-H silylation to form 5-membered silolanes, including chiral silolanes, which can undergo further oxidation to produce enantiopure β-aryl-substituted 1,4-diols. Combined computational and experimental studies reveal that the silylation occurs via the Si-H addition to a 14-electron Ir(I) fragment to give an Ir(III) silyl hydride complex, which then activates the C(sp3)-H bond to form a 7-coordinate, 18-electron Ir(V) dihydride silyl intermediate, followed by sequential reductive elimination of H2 and silolane. The unprecedented site selectivity is governed by the distortion energy difference between the rate-determining δ-C(sp3)-H and δ-C(sp2)-H activation, although the activation at sp2 sites is much more favorable than sp3 sites by the interaction energy.

Abstract:We discovered a cooperative gold/silver catalysis mechanism in the oxidative cross-coupling reaction between 1,2,4,5-tetrafluorobenzene and N-TIPS-indole, using DFT calculations. A silver(I)-catalyzed CMD mechanism is responsible for the C-H activation of 1,2,4,5-tetrafluorobenzene, and C-H acidity determines the chemoselectivity. A gold(III)-catalyzed S(E)2Ar mechanism is responsible for the C3-H activation of N-TIPS-indole, and arene nucleophilicity determines the chemo- and regioselectivity. The orthogonal chemoselectivity control provides a mechanistic guide for dual C-H activation reactions.
Designing Electrocatalytic Mediators and Understanding Site-selectivity for C(sp3)-H Activation Using First-Principles Calculations

Abstract:Selective C-H bond activation is one of the most critical molecular transformations in synthesizing chemicals, pharmaceuticals, and natural product intermediates with broad applications. Recent efforts have focused on developing electrocatalytic mediators that rapidly and selectively activate specific C-H bonds. These mediated activations offer multiple benefits over direct electrochemical oxidation as they can occur at lower overpotentials, leading to higher faradaic efficiency and selectivity with reduced solvent oxidation. Our previous work described the development of N-alkyl ammonium ylides as a new class of electro-oxidative mediators. Despite its importance, the underlying principles of designing efficient mediators and understanding their site-selectivity are yet to be fully elucidated. The work discussed herein scrutinized mediator design using density functional theory calculations to highlight the critical features of mediators that govern C(sp3)-H activation. The design of newer mediators is guided by scaling relationships between the thermodynamic descriptors associated with the elementary steps involved in C(sp3)-H activation. We subsequently examine the results from detailed transition state calculations to elucidate the site-selectivity for C(sp3)-H activation for various substrates with quinuclidine and ylide mediators. The results show the critical interplay of thermodynamic, steric, and electronic features of the substrate and mediator that govern the corresponding site-selectivity. Finally, we present unifying trends across multiple substrates and mediators to understand the site-selectivity for mediated electrocatalytic C(sp3)-H activations and push our efforts toward predicting regio-selectivity in the future.

Abstract:Recent years have witnessed the growing interest in the remote functionalization of alkenes for it offers a strategy to activate the challenging C-H bonds distant from the initiation point via alkene isomerization/functionalization. However, the catalytic enantioselective isomerization/functionalization with one single transition metal catalyst remains rare. Here we report a highly regio- and enantioselective cobalt-catalyzed remote C-H bond borylation of internal alkenes via sequential alkene isomerization/hydroboration. A chiral ligand featured twisted pincer, anionic, and non-rigid characters is designed and used for this transformation. This methodology, which is operationally simple using low catalyst loading without additional activator, shows excellent enantioselectivity and can be used to convert various internal alkenes with regio- and stereoisomers to valuable chiral secondary organoboronates with good functional group tolerance.

Abstract:Most chelation-assisted aliphatic C-H activation proceeds through a kinetically favored five-membered cyclometalated intermediate. Here, we report the first site-selective alkenylation of δ-C(sp(3))-H in the presence of more accessible γ-C(sp(3))-H bonds via a kinetically less favored six-membered palladacycle. A wide range of functional groups are tolerated, and the unique protocol can be applied to the synthesis of chiral piperidines. Moreover, mechanistic insights have been conducted to elucidate the origin of the unusual site-selectivity.

Abstract:The mechanism and origins of selectivities in [Ni(NHC)]-catalyzed intramolecular (5 + 2) cycloadditions and homo-ene reactions of vinylcyclopropanes (VCPs) and alkynes have been studied using density functional theory. The preferred mechanism involves oxidative alkyne-alkene cyclization to form a metallacyclopentene intermediate, in contrast to a cyclopropane cleavage pathway in the reaction with Rh(I) catalysts. The selectivity between the (5 + 2) and homo-ene products is determined in the subsequent competing reductive elimination and β-hydride elimination steps. Two similar-sized N-heterocyclic carbene (NHC) ligands, SIPr and ItBu, yielded reversed product selectivity, favoring the (5 + 2) and homo-ene products respectively. This is attributed to the anisotropic steric environment of these NHC ligands, which positions the bulky substituents on the ligand toward different directions and leads to distinct steric control in the reductive elimination and β-hydride elimination transition states.
Study of Pd-catalyzed Selective Mono- and Di-C(sp3)-H Bond Activation: A Bi-ligand Model

Abstract:Controlling the number of C-H bond activation is a long-standing challenge in organic synthesis. Recently, Yu's group demonstrated that in Pd-catalyzed alanine's arylation, pyridine-type ligands favor a mono-C-H bond activation, while quinoline-type ligands favor a di-C-H bond activation. To disclose the underlying principles, a theoretical study (density functional theory (DFT)) has been carried out. Our study indicates that a mono-ligand model, which is generally adopted in the community, does not reproduce the experimentally observed mono-/di-selectivity, while a bi-ligand model can rationalize the experimental observations well, including the observed diastereoselectivity in diarylation. The electron-rich pyridine-type ligands with less steric congestion can promote the C-H bond activation reaction of alanine derivatives. The quinoline-type ligands have a better π back-donation interaction with the metal, which makes a more active C-H bond activation than the pyridine-type ligands for this reaction. This bi-ligand model, which is a necessity, allows the understanding and future design of a dual ligand effect in C-H bond activation.
Site-Selective C(sp3)–H Oxidation of Aliphatic Substrates Devoid of Functional Groups

Abstract:Although tremendous progress in the field of C(sp3)–H oxidation has been achieved over the last decades, the selective oxidation of non-activated positions on hydrocarbon skeletons is still highly challenging. It usually requires the presence of a suitable functional group in proximity to the desired oxidation site. Here we present a novel approach for catalyst-directed C–H oxidation that relies on substrate binding via the solvophobic effect in fluorinated alcohols, and thus is independent of functional groups on the substrate. Enabled by the novel supramolecular catalyst Mn(mcp)-RS2, the preferential oxidation at the fifth position on aliphatic substrates was observed.

Abstract:Regulating the reactivity of nucleophiles is important in synthetic chemistry. As a weaker nucleophile is also a stronger Brønsted acid, it can be selectively deprotonated by a Brønsted base to give an anionic species that shows higher reactivity. Following this idea, the reactivity of nucleophiles can be controlled by using an appropriate Lewis acid/Brønsted base pair catalyst. Cuprous/aminolithium catalyzed hydrofunctionalization of acrylonitrile with an alcohol and an amine were chosen as model reactions. As the acidity of the alcohol is significantly higher than that of amine, the former is more easily deprotonated. Therefore, an alcohol shows higher reactivity than an amine in the presence of a Lewis acid/Brønsted base pair catalyst, reversing the selectivity trend.

Abstract:The functionalization of C–H bonds in organic molecules is one of the most direct approaches for chemical synthesis. Recent advances in catalysis have allowed native chemical groups such as carboxylic acids, ketones and amines to control and direct C( sp 3 )–H activation 1,2,3,4 . However, alcohols, among the most common functionalities in organic chemistry 5 , have remained intractable because of their low affinity for late transition-metal catalysts 6,7 . Here we describe ligands that enable alcohol-directed arylation of δ -C( sp 3 )–H bonds. We use charge balance and a secondary-coordination-sphere hydrogen-bonding interaction—evidenced by structure–activity relationship studies, computational modelling and crystallographic data—to stabilize L-type hydroxyl coordination to palladium, thereby facilitating the assembly of the key C–H cleavage transition state. In contrast to previous studies in C–H activation, in which secondary interactions were used to control selectivity in the context of established reactivity 8,9,10,11,12,13 , this report demonstrates the feasibility of using secondary interactions to enable challenging, previously unknown reactivity by enhancing substrate–catalyst affinity.

The synergy between computation and experiment elucidated the mechanistic model and controlling factors of selectivities in these transformations, leading to advances in innovative C-O bond activation and functionalization methods.Toward the rational design of C-O bond activation, our collaborations with the Jarvo group bridged the mechanistic models of C(sp2)-O and C(sp3)-O bond activations. We found that the nickel catalyst cleaves the benzylic and allylic C(sp3)-O bonds via two general mechanisms: the stereoinvertive SN2 back-side attack model and the stereoretentive chelation-assisted model. These two models control the stereochemistry in a wide array of stereospecific Ni-catalyzed cross-coupling reactions with benzylic or allylic alcohol derivatives. Because of the catalyst distortion, the ligands can differentiate the competing stereospecific C(sp3)-O bond activations. The PCy3 ligand interacts with nickel mainly through σ-donation, and the Ni(PCy3) catalyst can undergo facile bending of the substrate-nickel-ligand angle, which favors the stereoretentive benzylic C-O bond activation. The N-heterocyclic carbene SIMes ligand has additional d(metal)-p(ligand) back-donation with nickel, which leads to an extra energy penalty for the same angle bending. This results in the preference of stereoinvertive benzylic C-O bond activation under Ni/SIMes catalysis. In addition to ligand control, a Lewis acid can increase the selectivity for stereoinvertive C(sp3)-O activation by stabilizing the SN2 back-side attack transition state. The oxygen leaving group complexes with the MgI2 Lewis acid in the stereoinvertive activation, leading to the exclusive stereoinvertive Kumada coupling of benzylic ethers. We also identified that the competing C(sp3)-O bond activation models have noticeable differences in charge separation. This leads to the solvent polarity control of the stereospecificity in C(sp3)-O activations. Low-polarity solvents favor the neutral stereoretentive C-O bond activation, while high-polarity solvents favor the zwitterionic stereoinvertive cleavage.In sharp contrast to the nickel catalysts, the C(sp2)-O bond activation under palladium catalysis mainly proceeds via the classic three-membered ring oxidative addition mechanism instead of the chelation-assisted mechanism. This is due to the lower oxophilicity of palladium, which disfavors the oxygen coordination in the chelation-assisted-type activation. The three-membered ring activation model selectively cleaves the weak C-O bond, resulting in the exclusive chemoselectivity of acyl C-O bond activation in Pd-catalyzed cross-coupling reactions with aryl carboxylic acid derivatives. This explains the overall acylation in the Pd-catalyzed Suzuki-Miyaura coupling with aryl esters. In collaboration with the Szostak group, we revealed that the three-membered ring model applies in the Pd-catalyzed C-O bond activation of carboxylic acid anhydride, which stimulated the development of a series of Pd-catalyzed decarbonylative functionalizations of aryl carboxylic acids.
