Abstract:Practical gas sensing application requires sensors to quantify target analytes with high sensitivity and reproducibility. However, conventional surface enhanced Raman scattering (SERS) sensor lacks reproducibility and quantification arising from variations of "hot spot" distribution and measurement conditions. Here, a ratiodependent SERS sensor was developed for quantitative label-free gas sensing. Au@Ag-Au nanoparticles (NPs) were filtered onto anodic aluminum oxide (AAO) forming Au@Ag-Au@AAO SERS substrate. 4-MBA was encapsulated in the gap of Au@Ag-Au and served as the internal standard (IS) to calibrate SERS signal fluctuation for improved quantification ability. Combined with headspace sampling method, SO2 residue in traditional Chinese medicine (TCM) can be extracted and captured on the immediate vicinity of Au@Ag-Au surface. The intensity ratio I-613 (-1)(cm) /I-1078 (-1)(cm) showed excellent linearity within the range of 0.5 mg/kg-500 mg/kg, demonstrating superior quantification performance for SO2 detection. Signals for concentration as low as 0.05 mg/kg of SO2 could be effectively collected, much lower than the strictest limit 10 mg/kg in Chinese Pharmacopoeia. Combined with a handheld Raman spectrometer, handy and quantitative TCM quality evaluation in aspect of SO2 residue was realized. This ratiometric SERS sensor functioned well in rapid on-site SO2 quantification, exhibiting excellent sensitivity and simple operability.
A polylysine-based fluorescent probe for sulfite anion detection in aqueous media via analyte-induced charge generation and complexation

Abstract:Sulfite is widely used as a preservative in food and beverages; and human exposure to SO2 (which is easily transformed into sulfite) has become increasingly widespread. Overdose or inhalation of sulfite may induce many respiratory responses and a number of diseases. Hence, it is important to achieve rapid, sensitive and selective detection of sulfites. Herein, we report a robust fluorescent turn-on probe for sulfite anions via the analyte-induced charge generation and AIE effect (the aggregation induced emission effect). The two-component sensing system comprises a modified polylysine with benzoic aldehyde side groups and a positively charged tetraphenylethene bearing two quaternary ammonium groups. In the presence of sulfite anions, the benzoic aldehyde groups on polylysine side chains react with sulfite and turn into negatively charged groups, leading to the complexation between the polymer chains and the positively charged fluorophores (TPE-2N(+)) and eventually achieving the fluorescence enhancement of the system as a result of the AIE effect. The integration of the water-soluble biopolymer and AIE-active fluorophore ensures that the system is operable in totally aqueous media (100% water), which is beneficial for applications in food quality control, environmental pollution monitoring and biological assays. Furthermore the system exhibits highly selective and sensitive fluorescent turn-on detection for sulfite, with the detection limit of 3.6 mu M. Moreover, it can be used in real samples such as red wine, beer and rainwater, and there is no need for special sample pretreatment.
All-in-one: Accurate quantification, on-site detection, and bioimaging of sulfite using a colorimetric and ratiometric fluorescent probe in vitro and in vivo

Abstract:SO2 and its derivatives (SO32-/HSO3-) are used widely in food, beverages, and pharmaceutical production. However, they could induce multiple diseases in respiratory, nervous, and cardiovascular systems. Although several fluorescent probes have been developed for detecting SO32-/HSO3-, reports on rapid fluorescent probes for the on-site detection of SO2 derivatives are scarce. Herein, a colorimetric and ratiometric fluorescent probe 1 based on the intramolecular charge transfer (ICT) was reported. Probe 1 resulted in a 122 nm blue-shift in fluorescent emission and decrement of absorbance at 500 nm upon the addition of sulfite. Therefore, probe 1 could quantify SO32-/HSO3- using both UV-Vis and fluorescent methods (LOD: UV-Vis method 34 nM; fluorescent method 51 nM). Importantly, probe 1 was used for a rapid (60 s) and convenient (1 step, on-site) measurement of the SO2 derivatives in real samples (LOD: 0.47 µM) using smartphone based on the colorimetric method. The SO32-/HSO3--sensing mechanism was confirmed as the Michael addition reaction. Furthermore, the probe was used for the real-time monitoring of SO32-/HSO3- in A549 cells and zebrafish. In summary, an all-in-one fluorescent probe was successfully developed for the accurate quantification, on-site detection, and bioimaging of SO32-/HSO3-.

Abstract:Sulfites play imperative roles in food crops and food products, serving as sulfur nutrients for food crops and as food additives in various foods. It is necessary to develop an effective method for the on-site quantification of sulfites in food samples. Here, 7-(diethylamino) quinoline is used as a fluorescent group and electron donor, alongside the pyridinium salt group as an electron acceptor and the C=C bond as the sulfite-specific recognition group. We present a novel fluorescent sensor based on a mechanism that modulates the efficiency of intramolecular charge transfer (ICT), CY, for on-site quantitative measurement of sulfite in food. The fluorescent sensor itself exhibited fluorescence in the near-infrared light (NIR) region, effectively minimizing the interference of background fluorescence in food samples. Upon exposure to sulfite, the sensor CY displayed a ratiometric fluorescence response (I447/I692) with a high sensitivity (LOD = 0.061 μM), enabling accurate quantitative measurements in complex food environments. Moreover, sensor CY also displayed a colorimetric response to sulfite, making sensor CY measure sulfite in both fluorescence and colorimetric dual-signal modes. Sensor CY has been utilized for quantitatively measuring sulfite in red wine and sugar with recoveries between 99.65% and 101.90%, and the RSD was below 4.0%. The sulfite concentrations in live cells and zebrafish were also monitored via fluorescence imaging. Moreover, the sulfite assimilated by lettuce leaves was monitored, and the results demonstrated that excessive sulfite in leaf tissue could lead to leaf tissue damage. In addition, the sulfate-transformed sulfite in lettuce stem tissue was tracked, providing valuable insights for evaluating sulfur nutrients in food crops. More importantly, to accomplish the on-site quantitative measurement of sulfite in food samples, a portable sensing system was prepared. Sensor CY and the portable sensing system were successfully used for the on-site quantitative measurement of sulfite in food.

Abstract:Sulfur dioxide (SO 2 ) is a typical additive that serves as an antioxidant, preservative, and decolorizer in industrial production. Yet, excessive SO 2 residues in the products pose a potential risk to human health. Although many fluorescent probes have been developed for the detection of SO 2 , effectively eliminating the interference from thiols on the selective recognition of SO 2 remains a challenge. Herein, we developed a coumarin-derived ratiometric fluorescent probe 1 for quantitatively sensing the SO 2 residues in actual samples. This probe can selectively recognize bisulfite (HSO 3 - ) based on the different p K a values between HSO 3 - and thiols. The working mechanism of this probe was further rationalized by the combination of experiments and quantum chemical calculations. Probe 1 was successfully used for the quantitative detection of SO 2 residues in medicine-food homology (MFH) samples with good sensitivity and accuracy. Furthermore, rapid visual and semi-quantitative detection of SO 2 can also be realized through a disposable test tube.

Abstract:A new sensitive fluorescent probe (CDs-AgNP/H 2 O 2 ) for detecting sulfite and bisulfite (SO 3 2− and HSO 3 − ) based on the inner-filter effect (IFE) between silver nanoparticles (AgNPs) and carbon dots (CDs) was developed. Because of the spectral overlap between the absorption of AgNPs and the excitation of CDs, the fluorescence of CDs can be quenched by AgNPs owing to the IFE. H 2 O 2 weakens the IFE and restores the fluorescence due to the oxidation of AgNPs by H 2 O 2 . However, the existence of SO 3 2− /HSO 3 − can quench the fluorescence again as a result of redox reaction between SO 3 2− /HSO 3 − and H 2 O 2 . The results showed a broad linear range of 20–200 μM with a low limit of detection (3.02 μM) toward SO 3 2− /HSO 3 − . The combination of IFE and redox reaction led to improvement of the sensitivity and selectivity. The probe was implemented to measure SO 3 2− /HSO 3 − in various agricultural products and foods with acceptable results (80.6 to 118.9% recovery).

Abstract:Sulfur dioxide (SO2) derivatives are typically employed as antioxidants in food and pharmaceutical processing. However, excessive sulfite intake could trigger serious health problems. Hence, it is urgent to establish a rapid and effective system for monitoring SO2. This study adopted a one-step hydrothermal method to synthesize dual-emitting nitrogen-doped carbon quantum dots (CECDs) and developed a ratiometric sensor for sulfite using CECDs-Cr (VI) composites. The emission intensity ratio (I440/I500) of the CECDs-Cr (VI) composites increased considerably with the addition of HSO3-. A method based on the ratiometric sensor was established for SO2 derivatives with advanced efficiency and excellent linearity over a broad concentration range of 0-500 μM (R2 = 0.9946). Four medicine-food homology materials (MFHMs) fumigated with sulfur have been accurately detected using this approach. Furthermore, a portable test tube was prepared to achieve rapid and semi-quantitative detection of SO2 residues and applied to real samples. This work presents an effective approach to develop a rapid on-site detection platform for sulfite residues in food and pharmaceuticals.

Abstract:Sulfur dioxide and its derivative sulfite widely existed in air, water as the environment pollutant. Sulfite is also commonly used as preservative and additive in fresh fruits, vegetables, wines and pharmaceutical materials. Due to sulfite closely related with human diseases, it is very urgent for the sensitive and rapid quantification of sulfite in various samples. In our study, a turn-on near infrared (NIR) fluorescent probe (MDQ) was developed for sulfite detection based on a Michael addition reaction, with high sensitivity (LOD 4.16 nmol/L), selectivity and fast response time (400 s). Using MDQ, a quantify method for sulfite in traditional Chinese medicines (TCMs) was developed with advantages of high precision, accuracy and convenient operation. Furthermore, according to the photophysical property of MDQ, a portable fluorescence detector is designed to quantify sulfite for TCMs and surface water in Dalian city of China. Therefore, the developed fluorescent probe MDQ and portable fluorescent detector as a rapid inspection instrument were successfully used to real-time monitor the sulfite in various complex samples.

A highly sensitive ratiometric fluorescence probe for sensing and imaging sulfite in food samples and living cells

Abstract:In this work, a ratiometric and chromogenic fluorescent probe 1 was synthesized for the detection of SO32-. The probe 1 at PBS (10 mM, pH = 7.4) presented a marked emission band at 661 nm. Upon addition of SO32- ions, a highly emissive adduct with a marked fluorescence at 471 nm were obtained through a Michael addition. The probe 1 displayed a noticeable fluorescence ratiometric response with a large shift (190 nm) in emission wavelength. The probe can quantitatively detect SO32- with high specificity, fast response (within 130 s) as well as low detection limit (13 nM), and a large Stokes shift (139 nm). Fluorescence imaging of HeLa cells indicated that 1 could be used for monitoring the intrinsically generated intracellular SO32- in living cells by ratiometric fluorescence imaging. Furthermore, 1 could be application in real water and sugar samples with high sensitivity and good recoveries.

Abstract:A new fluorescent probe 7-(diethylamino)-3-((1e,3e)-5-oxo-5-phenylpenta-1,3-dien-1-yl)-2H-chromen-2-one (SPH), based on Michael addition mechanism, was designed and synthesized for selective detection of sulfite. The probe was constructed by incorporating an α,β-unsaturated ketone conjugated with a C˭C bond into the coumarin fluorophore as a specifical reaction site for sulfite utilizing its nucleophilic property. The extra conjugated C˭C bond induced obvious red-shifts in both absorption and emission maxima, and remarkably promoted the nucleophilic addition rate. Once treated with sulfite, the solution's color changed from orange to yellow companied with a strong blue-green fluorescence. The ratio of fluorescence intensity at 488nm and 630nm (I488/I630) was linear with sulfite concentration over the range of 0-80μM with a detection limit of 0.23μM. High selectivity and good competition of the probe toward sulfite over other anions and thiols enable us to monitor sulfite levels in realistic samples as well as in living HeLa cells.

Abstract:A simple 4-hydroxynaphthalimide-derived colorimetric and ratiometric fluorescent probe ( 1 ) containing a receptor of levulinate moiety was designed and synthesized to monitor sulfite. Probe 1 could quantificationally detect sulfite by a ratiometric fluorescence spectroscopy method with high selectivity and sensitivity. Specially, probe 1 exhibited a 100 nm red-shifted absorption spectrum along with the color changes from colorless to yellow, and 103 nm red-shifted emission spectra upon the addition of sulfite. Thus, 1 can serve as a “naked-eye” probe for sulfite. Further, the recognition mechanism of probe 1 for sulfite was confirmed using nuclear magnetic resonance and electrospray ionization mass spectrometry. Also, the preliminary practical application demonstrated that our proposed probe provided a promising method for the determination of sulfite.
Simple methods for rapid determination of sulfite in food products

Abstract:Two simple methods for sulfite determination in food products were proposed. The modified para-rosaniline (PRA)-formaldehyde method showed a much broader linear range (0.05–5.0mgl−1 as SO2) than the commonly used procedure for the sulfur dioxide detection in the atmosphere (0.05–1.0mgl−1 as SO2). By using a standard reference color card, this method only needed 5min to complete a test. The 5,5′-dithiobis(2-nitrobenzoic acid) (DTNB) method was another good alternative for rapid determination of sulfite since it only needed one main reagent and the method was robust and easy to operate with the linear range of 0.10–4.3mgl−1 as SO2. When applying to real food samples, the DTNB method had good recoveries for all the tested samples and the results agreed well with those obtained by the official iodometric titration. The modified PRA-formaldehyde method worked well with all the tested dried food products, though matrix effect of sulfite binding was observed for the beer samples. Thus, the modified PRA-formaldehyde method has advantages of sensitivity and rapidity, but the DTNB method offers a wider range of applications. Both methods provided practical ways for in situ determination of sulfite by non-professional operators with the modified PRA-formaldehyde method suited for food samples with less sulfite binding problems.

Abstract:Reversible detection of bisulfite (HSO 3 - ) and hydrogen peroxide (H 2 O 2 ) is highly desirable in biology and food safety owing to their redox pair function in biological systems and their utilization in food industry. In this work, a ratiometric fluorescent probe ( TPA-Y ) with aggregation-induced emission (AIE) properties based on triphenylamine fluorophore was designed and synthesized for reversibly monitoring of HSO 3 - /H 2 O 2 . Reaction of HSO 3 - with TPA-Y can result in ratiometric fluorescence changes ( I 444 / I 629 ), and the emission spectrum blue shift reaches 185 nm, which can effectively avoid spectral overlap. Meanwhile, the solution color changed obviously from orange to colorless, enabling naked eye detection of HSO 3 - . TPA-Y was capable of quantitatively detect HSO 3 - in sugar and wine samples with the recovery rates ranging from 94.52 to 102.18%. In addition, TPA-Y is suitable to prepare paper strips and anti-counterfeiting ink for HSO 3 - detection. In addition, the original TPA-Y can be released after the addition product ( TPA-Y-HSO 3 - ) is oxidized by H2O2, and the HSO 3 - /H 2 O 2 redox cycle can repeated for at least six times. The probe TPA-Y can be applied to fluorescence imaging of reversible and dynamic interconversion between HSO 3 - and H 2 O 2 in living MCF-7 cells.

Determination of sulfite in food by flow injection analysis.

Abstract:A method is described for the determination of sulfite levels in food products by flow injection analysis (FIA). The method is based on the decolorization of malachite green by SO2, which is isolated from the flowing sample stream by means of a gas diffusion cell. The FIA method has a detection limit in food sample extracts of 0.1 ppm SO2 (3 times peak height of blank), which corresponds to 1-10 ppm SO2 in a food product, depending on the extraction procedure used. At the 5 ppm SO2 level in a food extract, the precision of replicate injections is +/- 1-2%. The method was tested on a variety of both sulfite-treated and untreated food products and the results compared favorably with those obtained by the Monier-Williams, colorimetric (pararosaniline), and enzymatic (sulfite oxidase) methods. The average differences from the FIA results were 19, 11, and 12%, respectively, for those samples (n = 12) above 50 ppm SO2. At lower levels the results were somewhat more erratic due to inaccuracies of the various methods at low concentrations.

Abstract:The emission of venenous sulfur dioxide (SO2) and its derivatives from industrial applications such as coking, transportation and food processing has caused great concern about public health and environmental quality. Probes that enable sensitivity and specificity to detect SO2 derivatives play a crucial role in its regulations and finally mitigating its environmental and health impacts, but fluorescent probes that can accurately, rapidly and on-site detect SO2 derivatives in foodstuffs and environmental systems rarely reported. Herein, a near-infrared (NIR) fluorescent probe (ZTX) for the ratiometric response of bisulfite (HSO3-) was designed and synthesized by regulating the structure of high-performance HSO3- fluorescent probe SL previously reported by us based on structural analyses, theoretical calculations and related literature reports. The Michael addition reaction between the electronic-deficient C=C bond and HSO3- destroys ZTX's π-conjugation system and blocks its intramolecular charge transfer (ICT) process, resulting in a significant fading of the fuchsia solution and the bluish-purple fluorescence turned light blue fluorescence. Fluorescent imaging of HSO3- in live animals utilizing ZTX has been demonstrated. The quantitative analysis of HSO3- in food samples using ZTXvia a smartphone has been also successfully implemented. Simultaneously, the ZTX-based test strips were utilized to quantificationally determine HSO3- in environmental water samples by a smartphone. Consequently, probe ZTX could provide a new method to understand the physiopathological roles of HSO3-, evaluate food safety and monitor environment, and is promising for broad applications.

Abstract:Sulfur dioxide (SO2), widely used as an antioxidant and preservative in food production, has been associated with detrimental cardiovascular and neurological effects when consumed excessively. This highlights the pressing need to develop a fast and sensitive probe capable of high-throughput screening for the quantitative determination of SO2 in food. Herein, we synthesized a new fluorescent probe, namely B3, specifically designed for high-throughput detection of SO2 in food. The vinyl chloride aldehyde within the B3 structure engages in a nucleophilic addition reaction with SO2, contributing to B3 ' s exceptional selectivity for SO2, and fast response time within 9 s. Furthermore, by integrating B3 with a microplate reader, we effectively achieved high-throughput detection of SO2 concentration up to 45 mu M within a pH range of 5.5 to 8.0 in real food samples. This accomplishment serves as a significant contribution to ensuring consumer safety and facilitating health assessments.

Abstract:Sulfite, widely used in food and pharmaceutical industries, poses significant health and safety concerns, necessitating efficient detection methods. This study introduces three novel red fluorescent probes (MCC-BT, MCC-TI, and MCC-DM), synthesized by integrating chromene derivatives with various electron-withdrawing groups. These probes effectively detect sulfite in both food samples and living cells, leveraging a mechanism transition from intramolecular charge transfer (ICT) to photo-induced electron transfer (PET). They demonstrate low detection limits (2.0 μM, 0.42 μM, and 0.34 μM, respectively) and offer advantages including large Stokes shifts, minimal interference, and colorimetric capabilities. We also developed smartphone-compatible test paper strips for convenient sulfite detection in real-world samples. The probes' effectiveness is further evidenced through sulfite quantification in Traditional Chinese Medicine ingredients. Additionally, their robust fluorescent imaging potential for cellular sulfite sensing highlights their significant contribution to analytical chemistry and biomedical research.

Abstract:A method for the determination of sulfite in sulfuring ginger by anion-exchange chromatography with suppressed conductivity detection was developed.The chromatographic separation was carried out on a NJ-SA-4A anion exchange column,using 1.92 mmol/L Na2CO3 + 1.80 mmol/L NaHCO3 +2%(V/V) acetone as mobile phase.Triethanolamine(TEA) was added in the samples to prevent oxidation of the sulfite and to eliminate the interference of metal ions.The linear range of the calibration curve was 0.02-2.00 mg/L with a correlation coefficient of 0.9998.The RSD for the peak area of sulfite anion was 1.9%,and the detection limit of sulfite anion was 0.015 mg/L.The spiked recoveries were between 84.0% and 93.5%.This method was easy to be operated and had a better accuracy.It provides a reliable basis for the detection of sulfuring ginger.

Abstract:Sulfite, a widely used food additive, is subject to regulated labeling. The extraction of sulfite as the stable hydroxymethylsulfonate (HMS) form and its quantitative analysis by liquid chromatography-tandem mass spectrometry (LC-MS/MS) has been recognized for their good sensitivity, selectivity, and versatility across various food materials. This study aimed to develop a cost-effective and simpler method for sulfite quantitation, while maintaining the superior sensitivity and selectivity of mass spectrometry (MS). To achieve this, we introduced paper spray ionization (PSI), an ambient desorption ionization technique that could achieve the direct measurement of analytes without employing separation. We also employed a novel internal standard (IS) structurally similar to the analyte, replacing the more expensive isotopically labeled IS. Although the PSI-MS/MS method developed in this study exhibited slightly lower analytical performance compared to the conventional LC-MS/MS, it remained effective for sulfite analysis in dried fruits.

Abstract:Maintaining redox homeostasis in organisms plays a crucial role in human life and health, and changes in the levels of SO2 and H2O2, as typical representative substances of reductants and oxidants, directly affects intracellular redox homeostasis. Hence, it is necessary to manufacture fluorescent probes that can detect HSO3 − and H2O2. In this work, a benzopyrylium salt containing benzothiazole derived fluorescent probe HBTO-H with dual properties of aggregation-induced emission (AIE) and excited-state intramolecular proton transfer (ESIPT) has been developed. The probe displays rapid and reversible test of HSO3 − and H2O2 in DMF/Tris (v:v = 2:8, pH = 7.3, Tris 10 mM) mixed solvent with high selectivity, good anti-interference ability and a wide applicable pH range (6–11). The detection limits of HBTO-H for HSO3 − and H2O2 are 0.187 μM and 0.152 μM, respectively. The response mechanisms of HBTO-H toward HSO3 − and H2O2 have been demonstrated to be Michael addition reaction and its retro-reaction based on 1H NMR and HRMS analyses. In addition, HBTO-H has been successfully applied to check HSO3 − in sugar and wine specimens. Moreover, HBTO-H was proved to be mitochondria-targetable and can alternately detect exogenous HSO3 − and H2O2 in MCF-7 cells, which is desired to be applied to trace HSO3 − in organisms in the future.
