Physicochemical characterization and in vitro biological activities of water-extracted polysaccharides fractionated by stepwise ethanol precipitation from Rosa roxburghii Tratt fruit

Abstract:Three polysaccharide fractions RTFP-30, RTFP-50, and RTFP-80 were isolated from Rosa roxburghii Tratt fruit using 30%, 50%, and 80% (v/v) ethanol, respectively. The molecular weight, monosaccharide composition, functional groups, surface morphology, rheological properties, and in vitro biological activities of the fractions were investigated. RTFP-50 had the largest molecular weight and RTFP-80 the lowest. The three fractions were composed mainly of arabinose, galactose, glucose, and galacturonic acid in different molar ratios. They also exhibited different surface morphology, particle size, and rheological properties. The antioxidant, hypoglycemic, and hypolipidemic activities of these fractions were also investigated in vitro. RTFP-50 had the strongest ability to scavenge DPPH, ABTS and hydroxyl radicals, and the highest fat and cholesterol binding capacities. However, RTFP-80 exhibited the strongest α-glucosidase inhibition. This study broadens the sources of raw materials for functional foods and provides a scientific foundation for the use of R. roxburghii Tratt fruit.
