Abstract:The chiral phosphorus derivatizing agent (CDA) 1 was prepared from optically pure (S)-1,1-bis-2-naphthol. It was first used in the determination of the enantiomeric excess of chiral alcohols and amines by means of P-31 NMR spectroscopy. It showed that, for the chiral aromatic alcohols, no apparent kinetic resolution was noted and good base-line separation was observed. Furthermore, the chemical shift difference (Deltadelta) of P-31 NMR spectroscopy was much larger than those determined by the use of other chiral phosphorus derivatizing agents reported previously. However, for aliphatic alcohols, it showed not only obvious kinetic resolutions but incomplete base-line separation. Moreover, we also found that the use of CDA 1 was suitable for the determination of enantiomeric excess of chiral primary amines. (C) 2002 John Wiley Sons, Inc.

Abstract:A three-component protocol involving enantiopure Δ-[Ir(ppy)2(MeCN)2](PF6) (ppy is 2-phenylpyridine) and salicylaldehyde as chiral auxiliaries was successfully applied to discriminate the absolute configuration and determine the enantiopurity of primary amines and amine alcohols via 1H NMR spectroscopy. The assembly reaction is rapid and quantitative, generating a pair of diastereomers that can be determined directly without physical separation. Single crystal structural analyses indicate that the shielding effects on the ligands imposed by a pair of diastereomers are different and generate sufficient resolution NMR signals for identification. The enhancement of stability via chelating coordination to Ir(III) ion and more than one pair of diastereotopic resonances in different detection regions of the three-component protocol ensure a high degree of accuracy in quantifying the ee value of chiral amines. The absolute errors in the ee determinations by 1H NMR spectroscopy in different detection windows are within 2.0%. The linear relationship between the experimentally measured ee values and the gravimetrically prepared samples is found to be excellent. This finding would provide a complementary method for the discrimination and determination of the enantiopurity of chiral primary amines and amine alcohols in the screening of asymmetric reactions.

Abstract:P-31 NMR has become a widely applied spectroscopic probe of the structure of phosphorus-containing compounds. Meanwhile: the application of P-31 NMR has been rapidly expanded to biochemistry and medicinal chemistry of phosphorus-containing compounds because the growing importance of the phosphorus compounds is now widely realized. We report here the results of P-31 NMR study on some phosphorus-containing compounds, namely, O-alkyl O-4-nitrophenyl methyl phosphonates with different alkyl chain-length (MePO-n), 4-nitrophenyl alkylphenylphosphinates with different alkyl chain-length (PhP-n), diethyl phosphono- acetonitrile anion and diethyl phosphite anion Our results indicate that P-31 NMR can not only be applied to not only the study of the hydrolytic reactions of MePO-8 and PhP-8 bur also be applied to the study of thr presence of the: anions of diethylphosphonoaceronitrile and diethyl phosphite in nucleophilic reactions.

Abstract:Here, we have documented a new protocol to determine d/l-amino acids by derivatizing amino acids via a chiral phosphinate. (RP)-l-Menthyl phenylphosphinate was able to bond both primary and secondary amines, as well as improve the sensitivity of analytes in MS. Eighteen pairs of amino acids were successfully labeled except for Cys which has a thiol group on the side chain, and the chirality of amino acids can be discriminated by 31P NMR. Seventeen pairs of amino acids were separated by a C18 column within 45 min of elution, and resolution values ranged from 2.01 to 10.76. The lowest limit of detection was 10 pM acquired at parallel reaction monitoring, in which two factors collectively contributed that the ability of protonation of phosphine oxide and the sensitivity of parallel reaction monitoring. Chiral phosphine oxides might be a promising tool in future chiral metabolomics.

Abstract:This paper using (S)-diethyl (1-diazo-2-(1,3-dioxoisoindolin-2-yl)propyl)phosphonate as starting material which catalysted by 5%Pd-C, prepared(S)-diethyl(2-(1,3-dioxoisoindolin-2-yl)propyl)phosphonate in the presence of glacial acetic acid and i-PrOH. The effect of catalyst amount, the glacial acetic acid amount and the reaction time were discussed. The results indicate that the optimum conditions were the ratio of compound a, 5%Pd-C and glacial acetic acid was 1∶0.2∶0.6 and reacted 2 h. The obtalned compound b was identified by 1HNMR,13CNMR,31PNMR and MS and the enantiomeric excess was 94.5%.

Abstract:The phosphorus stereocentre, present in a diversity of compounds ranging from phosphonates to phosphines, plays a crucial role in various fields, in particular catalysis, materials science, agrochemistry and drug development. The unique structural and stereochemical features of intrinsically homochiral phosphorus units form the incentive for major current efforts towards developing stereoselective synthesis methodology of P-chirogenic compounds. Despite advances in metal-catalysed C–P bond formation, generating homochiral phosphorus stereocentres remains challenging. Here we present an efficient and flexible method for the synthesis of various classes of P-chirogenic compounds through the functionalization of phosphonamidates with excellent stereoselectivity. Key to this is an asymmetric C–P cross-coupling reaction between phosphoramidites and aryl halides with axial-to-central transfer of chirality and the use of 1,1′-bi-2-naphthol as an inexpensive chiral auxiliary. Preliminary mechanistic studies revealed the formation of an unexplored chiral amino–phosphonium intermediate. This transformation sets the stage for ready access to P-chirogenic phosphinates, phosphine oxides and phosphines.

Abstract:A novel chiral monodentate spiro phenylphospholane ligand 4 was prepared from a readily accessible, enantiomerically pure 1,1 '-spirobiindane-7,7 '-diol in high yield. This ligand has proven to be efficient for Pd-catalyzed enantioselective allylation of aldehydes with allylic alcohols. Aromatic, heteroaromatic, and aliphatic aldehydes gave homoallylic alcohols in good enantioselectivities (up to 83% ee) and excellent anti diastereoselectivities (up to 99:1 dr).

Abstract:Aminophosphine 2 was obtained by deprotection of amidophosphine 1, which was derived from L-proline. Deprotonation of aminophosphine 2 with n-BuLi, and then being captured by 2-MeO-pyridine gave chiral pyridyl N,P-ligands 4. Reaction of aminoalcohol and diethyl carbonate afforded oxazolidione 6, which was converted to compounds 7 by the reaction with triethyloxonium tetrafluoroborate. Chiral N,P-ligands 8 were obtained by the reaction of compounds 7 with aminophosphine 2. Two kinds of chiral N,P-ligands shown excellent catalytic activity in Ag(I) catalytic asymmetric 1,3-dipolar cycloaddition, and up to 93% yield and 15% ee were obtained.

Abstract:Within pharmaceutical research, ensuring the enantiomeric purity of chiral compounds is critical. Specifically, chiral amines are a crucial category of compounds, due to their extensive therapeutic uses. However, the enantiomeric analysis of these compounds, particularly those with significant steric hindrance, remains a challenge. To address this issue, our research introduces a novel chiral 19F-tagged NNO palladium pincer probe, strategically engineered with an open binding site to accommodate bulky amines. This probe facilitates the enantiodifferentiation of such amines, as evidenced by the distinct 19F NMR signals generated by the enantiomers. Moreover, our findings highlight the probe's applicability in the chiral discrimination of various psychoactive substances, underscoring its potential for the identification of illegal stimulant use and contributing to forensic investigations.

Abstract:A novel biscinchona alkaloid-catalyzed highly enantioselective desymmetrization reaction of bisphenol compounds with achiral Morita-Baylis-Hillman carbonate agents was developed. Through the asymmetric allylic alkylation strategy, a broad range of optically active P-stereogenic phosphine oxides were generated with excellent to good yields (up to 99%) and high enantioselectivities (up to 98.5:1.5 e.r.). The reaction was further investigated by the linear free energy relationship (LFER) analysis. A possible transition state was proposed and furthered verified by theoretical calculations.

Abstract:P-Stereogenic phosphinamides represent important structural elements in chiral organocatalysts and bioactive compounds. Herein, we report Pd(II)-catalyzed enantioselective C-H alkynylation using cheap commercially available l-pyroglutamic acid as a chiral ligand. A range of structurally diverse P-stereogenic phosphinamides was prepared in good yields with high enantioselectivities via desymmetrization and kinetic resolution. A tailor-made congested directing group, N-ethyl-N-(3-methylpyridin-2-yl)amino, was crucial for the reactivity.

Abstract:Palladium complexes of chiral aminophosphine phosphinites were investigated for the asymmetric allylic alkylation of 1,3-diphenyl-2-propenyl acetate with dimethyl malonate. The chirality of the carbon bonded to O-PPh2 in the ligands was found to be very important to the enantioselectivity of the catalysts. Introduction of this stereogenic center has led to significantly improved enantioselectivities.

Abstract:A new BINOL-derived chiral bifunctional phosphine has been designed and successfully prepared, which features a cyclic substructure of 1,3,5-diazaphosphinane. This chiral phosphine possesses good air stability in solid state, and it can be conveniently used as a relatively more nucleophilic phosphine organocatalyst. Preliminary investigations showed that it could generally afford fair to excellent yields but only modest enantioselectivity in the (aza)-Morita-Baylis-Hillman reactions of activated olefins such as ethyl acrylate and methyl vinyl ketone with aldehydes or imines. Abstract:A new chiral sensor assembly with enantiomeric Ir(III) complex Delta-[Ir(ppy)(2)(MeCN)(2)](PF6) (ppy is 2-phenylpyridine) and 3-hydroxypyridine-2-carboxaldehyde as anxiliaries for chiral amines and amino derivatives, including alpha-amines, alpha-amino alcohols, alpha-amino esters, beta-amines and beta-amino alcohols as well as amino ether, was developed on the basis of H-1 NMR spectroscopy. The assembly reaction is rapid and quantitative in equivalent under mild conditions, generating the corresponding diastereomers that are applied to discriminate the absolute configuration and quantitatively determine the enantiomeric excess (ee) directly without physical separation. The chelating coordination to Ir(III) ion much enhances the stability of the formed Schiff base ligand. Furthermore, more than one pair of diastereotopic resonances in wide detection regions ensures a convenient and high degree of accuracy in quantifying the ee value of chiral amines and amino derivatives. The absolute errors of the ee determinations by H-1 NMR spectroscopy in different detection windows are within 1.9%. The linear relationship between the experimentally measured ee values and the gravimetrically prepared samples is found to be excellent.
