Abstract:In this work, we propose a MgF2/Au/MgF2/Analyte based, angle-interrogation surface plasmon resonance sensor. This symmetrical structure results in higher surface electric field strengths, and longer surface propagation lengths and depths. A refractive index resolution of 8.1×10−8RIU in fluid protocol and 3.5×10−7RIU in atmosphere protocol is acquired by a simple SPR imaging system. In order to demonstrate the biomedical applications of this system, we detect Escherichia coli with different concentrations using dopamine film on the sensor surface to immobilize antibodies. A detection limit of 103cfu/mL of E. coli bacteria is acquired from this system.

Abstract:Gold nanoparticles (Au NPs) were almost chosen as the first option for biological and biosensor applications due to their enhancement and their outstanding properties. The combining of optical fiber with localized surface plasmon resonance (LSPR) for forming a biosensor is widely used in diagnosis. In this work, we report a fiber optical biosensor based on LSPR of Au NPs for the detection of bovine serum albumin (BSA) protein. BSA was functionalized on Au NPs immobilized fiber optic sensing head (length of 1 cm) via methanesulfonic acid (MSA) by carboxylic binding. It is the binding between the analytes with the surface-modified Au NPs that caused refractive index changes in the sensing medium led to changes in optical power at the output of the sensor. The detection limit of the LSPR fiber biosensor was found to be 0.18 ng/mL for the BSA detection with the low coefficient of variation (CV) at under 1%. We have demonstrated the effectiveness of combining multimode fiber with Au NPs to generate the biosensor as the label-free sensor that can be a feasible tool for highly sensitive, rapid response time, stable, and miniaturized point-of-care analytical systems.

Abstract:Nucleic acid (NA) is a widely-used biomarker for viruses. Accurate quantification of NA can provide a reliable basis for point-of-care diagnosis and treatment. Here, we propose a tilted fiber Bragg grating (TFBG)-based plasmonic fiber-optic spectral comb for fast response and ultralow limit NA detection. The TFBG is coated with a gold film which enables excitation of surface plasmon resonance (SPR), and single-stranded probe NAs with known base sequences are assembled on the gold film. To enhance sensitivity of refractive index (RI) for sensing a chosen combination of probe and target NAs around the TFBG surface, gold nanoparticles (AuNPs) are bonded to the target NA molecules as "RI-labels". The NA combination-induced aggregation of AuNPs induces significant spectral responses in the TFBG that would be below the detection threshold for the NAs in the absence of the AuNPs. The proposed TFBG-SPR NA sensor shows a fast response time of 30 seconds and an ultra-wide NA detection range from 1×10 −18 mol/L to 1×10 −7 mol/L. In the NA concentration range of 1×10 −12 mol/L (1 pM) to 10 5 pM, an ultra-high sensitivity of 1.534 dB/lg(pM) is obtained. The sensor achieves an ultra-low limit of detection down to 1.0×10 −18 mol/L (1 aM), which is more than an order of magnitude lower than the previous reports. The proposed sensor not only shows potentials in practical applications of NA detection, but also provides a new way for TFBG-SPR biochemical sensors to achieve higher RI sensitivity.

Abstract:A novel optical biosensor assembled with gold colloidal nanoparticles on gold plate surface has been constructed via 1,4-dithiothreitol and L-cysteic acid for detection of bio-molecules like antigen, antibody protein and DNA plasmid, which exhibited distinct optical properties of localized surface plasmon resonance (LSPR). The prepared nano gold sol were characterized by using UV-Vis absorption spectroscopy, transmission electron microscopy (TEM), and cyclic voltammetry (CV), indicating that the Au colloidals were sphere-shaped and narrow-dispersed size distribution with a diameter of 24 plusmn 0.2 nm. It caused 2.80 nm of red shift in the peak wavelength (lambdamax) of LSPR spectrum for the nanosensor exposed to c-Myc (3C7) mouse monoclonal IgG and 3.36 nm of red shift to the corresponding antibody by the immuno-reaction subsequently. The LSPR nanobiosensor has been successfully applied to recognition of a DNA plasmid of pCMV-Myc Mammalian Expression Vector.

Abstract:A label-free immunosensor was designed and fabricated for sensitive detection of alpha-fetoprotein (AFP) based on the localized surface plasmon resonance (LSPR) of gold nanorods (GNRs). GNRs were fabricated through seedmediated growth and surface activation by alkanethiols for the attachment of antibodies that could directly capture of the target. A novel strategy using the absorbance ratio of transverse and longitudinal LSPR peak of GNRs as a function of AFP contraction was performed for AFP quantification. The limit of detection of the sensor is 0.25nM in the PBS, and the dynamic range spans 0.25nM to 14.3nM. It is expected that this simple and cost-effective LSPR biosensor can also be applicable to the detection of several other tumor markers that are present at low concentrations.

Abstract:A novel PDGF aptasensor is proposed in this study based on gold nanoparticles (NPs) triggered chemiluminescence (CL) upon target binding. The strategy chiefly depends on the formation of a sandwich-type immunocomplex among the capture antibody immobilized on the 96-well plate, target protein and aptamer-functionalized gold NPs. Introduction of target protein into the assay system leads to the attachment of gold NPs onto the surface of the plates and thus the assembled gold NPs could trigger the reaction between luminol and AgNO3 with a CL emission. Further signal amplification was achieved by a simple gold metal catalytic deposition onto the gold NPs. This new CL strategy might create a novel technology for developing simple aptasensors in the sensitive and selective detection of target protein in a variety of clinical, environmental and biodefense applications.
Optical Fiber-Based Localized Surface Plasmon Resonance Biosensor Using Gold Nanorods Encapsulated in Graphene Oxide Nanocomposite Film Applied for Immunoassay

Abstract:In this study, gold nanorods (GNRs) are immobilized on the surface of fiber probe using a self-assembly method for a localized surface plasmon resonance (LSPR)-based optical fiber biosensor. In order to promote the performances of GNRs films, graphene oxide (GO) is used to encapsulate GNRs, which forms the GNRs@GO structure. Furthermore, the GNRs and GNRs@GO fiber probes are used to detect different refractive indices of surrounding environments from 1.33 to 1.38. The results show that the GNRs fiber probes have the sensitivity of 48 nm/RIU with the linearity of = 0.988. By comparison, for GNRs@GO with 0.2 mg/mL of GO fiber probe, the sensitivity is increased to 218 nm/RIU, and the linearity is improved to = 0.991. After that, anti-mouse IgG is immobilized on the GNRs and GNRs@GO fiber probes for immunoassay. The results show that the limit of detection (LOD) of GNRs fiber probe is 147.9 ng/mL, and the sensitivity is 26.5 nm/ mL with the linearity of = 0.936 in the detecting range of 0.1– /mL. By comparison, for GNRs@GO with 0.2 mg/mL of GO fiber probe, the LOD is improved to 32.7 ng/mL, and the sensitivity as well as the linearity are increased to 61.1 nm/ mL and = 0.991, respectively. In addition, the optimal concentration of GO encapsulated with GNRs is 0.2 mg/mL, and the optimal response time is 20 min. The results also show that the GNRs@GO fiber probe has a good selectivity. Finally, the GNRs@GO structure shows the potential for applying in an optical fiber biosensor.

Abstract:Surface plasmon resonance (SPR) sensing is a real-time detection technique for measuring biomolecular interactions on gold surfaces. This study presents a novel approach using nano-diamonds (NDs) on a gold nano-slit array to obtain an extraordinary transmission (EOT) spectrum for SPR biosensing. We used anti-bovine serum albumin (anti-BSA) to bind NDs for chemical attachment to a gold nano-slit array. The covalently bound NDs shifted the EOT response depending on their concentration. The number of ND-labeled molecules attached to the gold nano-slit array was quantified from the change in the EOT spectrum. The concentration of anti-BSA in the 35 nm ND solution sample was much lower than that in the anti-BSA-only sample (approximately 1/100). With the help of 35 nm NDs, we were able to use a lower concentration of analyte in this system and obtained better signal responses. The responses of anti-BSA-linked NDs had approximately a 10-fold signal enhancement compared to anti-BSA alone. This approach has the advantage of a simple setup and microscale detection area, which makes it suitable for applications in biochip technology.

Abstract:Gold-nanoparticles can be used as a robust tool for bio-sensing based on localized surface plasmon resonance (LSPR). Both local refractive index change and self-assembly of the nanoparticles can lead to some spectral shifts of the LSPR extinction peak. Using different mechanisms of LSPR peak shifts, two types of transducers are set up to detect the streptavidin concentration through the specific recognition with biotin moleculars, which are conjugated to gold-nanoparticles. One is based on the self-assembly of gold-nanoparticles in solution, and the other is based on the increase of the local refractive index near the surface of the immobilized gold-nanoparticles after streptavidin–biotin binding. The sensitivity and efficiency of the two transducers are compared. Gold-nanoparticles are functionalized with polyelectrolyte, which can improve significantly the stability of the nanoparticles and reduce the unspecific binding of streptavidin. Polyelectrolyte functionalization also provides a simple way to conjugate gold-nanoparticles with charged molecules such as biotin and BSA–biotin.
Gold Nanoparticle-Enhanced Detection of DNA Hybridization by a Block Copolymer-Templating Fiber-Optic Localized Surface Plasmon Resonance Biosensor

Abstract:To overcome low surface coverage and aggregation of particles, which usually restricts the sensitivity and resolution of conventional localized surface plasmon resonance (LSPR) fiber-optic sensors, we propose a simple self-assembled templating technique that uses a nanometer thickness block copolymer (BCP) layer of poly(styrene-b-4-vinylpyridine) to form a 33 nm gold nanoparticle (AuNP) monolayer with high uniformity and density for LSPR sensing. The LSPR resonance wavelength for this PS-b-P4VP templated methodology is 592 nm and its refractive index sensitivity is up to 386.36 nm/RIU, both of which are significantly improved compared to those of conventional LSPR techniques. Calibrated by a layer-by-layer polyelectrolyte deposition procedure, the decay length of this LSPR sensor is calculated to be 78 nm, which is lower than other traditional self-assembled LSPR sensors. Furthermore, hybridization between target ssDNA, which is linked with capture ssDNA on the LSPR biosensor and DNA–AuNP conjugates, leads to a low detection limit of 67 pM. These enhanced performances are significant and valuable for high-sensitivity and cost-effective LSPR biosensing applications.

Abstract:Fiber-optic surface plasmon resonance (FOSPR) sensing technology has become an appealing candidate in biochemical sensing applications due to its distinguished capability of remote and point-of-care detection. However, FOSPR sensing devices with a flat plasmonic film on the optical fiber tip are seldom proposed with most reports concentrating on fiber sidewalls. In this paper, we propose and experimentally demonstrate the plasmonic coupled structure of a gold (Au) nanodisk array and a thin film integrated into the fiber facet, enabling the excitation of the plasmon mode on the planar gold film by strong coupling. This plasmonic fiber sensor is fabricated by the ultraviolet (UV) curing adhesive transferring technology from a planar substrate to a fiber facet. The experimental results demonstrate that the fabricated sensing probe has a bulk refractive index sensitivity of 137.28 nm/RIU and exhibits moderate surface sensitivity by measuring the spatial localization of its excited plasmon mode on Au film by layer-by-layer self-assembly technology. Furthermore, the fabricated plasmonic sensing probe enables the detection of bovine serum albumin (BSA) biomolecule with a detection limit of 19.35 μM. The demonstrated fiber probe here provides a potential strategy to integrate plasmonic nanostructure on the fiber facet with excellent sensing performance, which has a unique application prospect in the detection of remote, in situ, and in vivo invasion.

Abstract:Using the finite-difference time-domain (FDTD) method, optical properties are investigated for a sensor comprising a two-dimensional array of gold cubes placed on quartz pillars on a quartz substrate. Background condition is analyzed for an approximate ideal collective resonance. It is found that the resonance intensity is mainly determined by the quartz etch depth, and the spectra valley position depends mainly on the lattice period. Thickness of the gold film affects the wavelength of the transmission spectra slightly, which gives a low processing tolerance requirement for fabrication. Sensitivity of the biosensor is also discussed, spectral sensitivity as high as 596.7nm/refractive index unit (RIU) is obtained in the visible light region, making it an excellent candidate for liquid detection.

Abstract:Well-dispersed and dense layers of gold nanorods (AuNRs) on optical fibers are shown to regulate the longitudinal peak wavelength and enhance the sensing performances of localized surface plasmon resonance (LSPR) biosensors. A simple self-assembly method relying on a brush-like monolayer of poly(styrene)-b-poly(acrylic acid) (PS-b-PAA) diblock copolymer was used to immobilize AuNRs with various aspect ratios from 2.33 to 4.60 on optical fibers. Both the experimental and simulation results illustrated that the particle aspect ratio, deposition time (related to the coverage of AuNRs), and interparticle gap significantly affected the optical properties of the fiber-based LSPR biosensors. The highest refractive index (RI) sensitivity of the sensor was 753 nm/RIU, while the limit of detection for human IgG was as low as 0.8 nM. Compared with standard nanoparticle deposition methods of polyelectrolytes or alkoxysilanes, the RI sensitivity of the PS-b-PAA dip-coating method was approximately 3-fold better, a consequence of the higher particle coverage and fewer AuNR aggregates. The presented AuNR-based LSPR sensors could regulate the detection range by tuning the aspect ratios of AuNRs. Applicability is demonstrated via quantitative analysis of antigen-antibody interactions, DNA sensing, and surface-enhanced Raman scattering.

Abstract:The main challenge for all biosensors is how to measure the biological samples at very low concentrations. In this paper, a surface plasmon resonance (SPR) fiber biosensor using multi-layer gold nanoparticles (Au NPs)/Au film coupling to enhance sensitivity was proposed. The coupling effect between such longitudinally aligned multilayer nanoparticles increases the intensity of the local electromagnetic field and the depth of propagation to the outside. We used the finite element method to analyze the local enhancement effect of multi-layer Au nanospheres and Au nanorods, and the fabricated double-layer Au NPs (Au nanospheres and Au nanorods)/Au film coupling enhancement sensors. These two sensors' refractive index sensitivities are 15,747 nm/RIU and 25,642 nm/RIU, respectively. By changing biomolecules immobilized on the sensor's surface, the highly sensitive double-layer Au nanoparticle/Au film fiber sensor enables high-precision measurement of different biomarkers at low concentration. For detection of human IgG, the detection limit of gold nanosphere sensor is 10 ng/mL and gold nanorod sensor is 4.6 ng/mL.

Ultrasensitive Glucose Biosensor Using Micro-Nano Interface of Tilted Fiber Grating Coupled with Biofunctionalized Au Nanoparticles

Abstract:Monitoring glucose levels is one of the main diagnostic methods for preventing and treating diabetes. Conventionally monitoring of blood glucose through a finger-stick procedure brings pain. It is essential to develop biosensors for low concentration glucose detection in non-invasive samples. In this paper, a tilted fiber grating based localized surface plasmon resonance (LSPR) biosensor for ultrasensitive glucose detection has been demonstrated. The glucose biosensor was fabricated by immobilization of Au nanoparticles (AuNPs) onto an excessively tilted fiber grating (Ex-TFG) and subsequent biofunctionalization by coating polydopamine and concanavalin A onto the AuNPs/Ex-TFG surface. The biosensor has the response regions covering low, middle and high glucose concentrations. The resonance peak indicates a redshift of 0.24 nm, 0.06 nm and 0.48 nm in the concentration ranges of 1.0 nM $- 1.0\,\, \boldsymbol {\mu }\text{M}$ , $1.0\,\, \boldsymbol {\mu }\text{M}- 50.0\,\, \boldsymbol {\mu }\text{M}$ and $50.0\,\, \boldsymbol {\mu }\text{M}-5.0$ mM, respectively. The limit of detection (LOD) of the biosensor in pure phosphate buffered saline is 2.5 nM. The high sensitivity and specific selectivity for D-glucose detection are attributed to strong coupling between the evanescent fields from transverse magnetic (TM) cladding modes of the Ex-TFG and the localized electric fields from LSPR of the AuNPs and their biofunctionalization, and resulting in remarkable enhancement of localized effective refractive index with slightly increased D-glucose concentration. Especially, the glucose biosensor based on the Ex-TFG coupled with the biofunctionalized AuNPs, has been demonstrated to be of practical utility to detect the glucose in artificial urine, which has potential applications in non-invasive glucose monitoring for early diagnosis and better management of diabetes.

Abstract:The interplay between light photons and nanomaterials can give rise to many striking phenomena. Surface plasmon resonance (SPR), which is one such plasmonic sensing mechanism, is gaining much attention from the biomedical fraternity. Two-dimensional (2D) nanomaterials like transition-metal dichalcogenides (TMDCs) and graphenes are recently discovered nanomaterials that have favorable properties like high optical absorption. These materials have been applied extensively in the fields of drug delivery and cancer therapy. However, their applications in the area of plasmonic biosensing need to be explored more. Hence, using commercial SPR sensors, we demonstrate the effectiveness of heterostructured monolayer nanosheets of graphene and WS2 for a plasmonic-based detection system of a specific ligand–analyte interaction between bovine serum albumin (BSA) and anti-BSA. Our detection technique does not require any complex functionalization scheme as the graphene–WS2 nanosheet-coated gold chip can attach protein molecules through hydrophobic interactions. Double-Antibody Sandwich Immunoassay and Plasmonic Coupling Synergistically Improved Long-Range SPR Biosensor with Low Detection Limit

Abstract:A long-range surface plasmonic resonance (LR-SPR) biosensor modified with double-antibody sandwich immunoassay and plasmonic coupling is demonstrated for human-immunoglobulin G detection with a low limit of detection (LOD). The double-antibody sandwich immunoassay dramatically changes the average refractive index of the medium layer on the sensor surface. The near-field electron coupling between the localized surface plasmon and the long-range surface plasmon leads to a significant perturbation of the evanescent field. The large penetration depth and the long propagation distance of the long-range surface plasmonic waves facilitate the LR-SPR sensor in the detection of biological macromolecules. The unique light absorption characteristic of the nanocomposite material in the sensor provides the in situ self-compensation for the disturbance. Therefore, besides the inherent advantages of optical fiber sensors, the developed biosensor can realize the detection of biomolecules with high sensitivity, low LOD and high accuracy and reliability. Experimental results demonstrate that the LOD of the biosensor is as low as 0.11 μg/mL in the detection of the phosphate-buffered saline sample, and the spike-and-repetition rate is 105.56% in the detection of the real serum sample, which partly shows the practicability of the biosensor. This indicates that the LR-SPR biosensor provides better response compared with existing similar sensors and can be regarded as a valuable method for biochemical analysis and disease detection.

Abstract:Tilted fiber Bragg grating, which has the advantages of both fiber Bragg grating and long-period fiber grating, has been widely studied for sensing in many fields, especially in the field of biochemistry. Calmodulin, which has a wide distribution in eukaryotes, can regulate several enzymes such as adenylate cyclase and guanylate cyclase and mediates several cellular processes such as cell proliferation and cyclic nucleotide metabolism. The abnormal levels of calmodulin in the body will result in serious effects from metabolism to nerve growth and memory. Therefore, it is important to measure the calmodulin concentration in the body. In this work, we propose and experimentally demonstrate a plasmonic tilted fiber Bragg grating-based biosensor for calmodulin detection. The biosensor was made using an 18° tilted fiber Bragg grating with a 50 nm-thick gold nanofilm coating the surface of the fiber, and transient receptor potential channels were bonded onto the surface of the gold nanofilm to serve as bio-detectors for calmodulin detection. Experimental results showed that the limit of detection using our biosensor was 0.44 nM. Furthermore, we also demonstrated that the interaction between calmodulin and transient receptor potential channels was quite weak without calcium in the solution, which agrees with the biology. Our proposed biosensor has a simple structure, is easy to manufacture, and is of small size, making it a good choice for real-time, label-free, and microliter-volume biomolecule detection.
