Abstract:A general and efficient copper-catalyzed benzylation reaction of triazolopyridine with N-tosylhydrazones was developed. This reaction forms a C(sp(2))-C(sp(3)) bond through cross-coupling, and represents an exceedingly practical method to afford 3-benzylated triazolopyridines in moderate to good yields. A proposed mechanistic pathway underlying this reaction was outlined. This catalytic transformation should enable broad synthetic applications in functionalization chemistry, allowing the synthesis of new pharmaceutically relevant triazolopyridine derivatives.

Abstract:A Cu-catalyzed tandem synthesis of azole-fused pyrimido[1,2-c]quinazolines and imidazo[1,2-c]quinazolines has been developed. The reaction is based on a C-N cross-coupling/C-H functionalization reaction of 2-(2-bromophenyl)-1,4,5,6-tetrahydropyrimidines with azoles. Abstract:The copper-catalyzed azide-alkyne cycloaddition (CuAAC) reaction is regarded as a prime example of "click chemistry", but the asymmetric click cycloaddition of internal alkynes still remains challenging. A new asymmetric Rh-catalyzed click cycloaddition of N-alkynylindoles with azides was developed, providing atroposelective access to C-N axially chiral triazolyl indoles, a new type of heterobiaryl, with excellent yields and enantioselectivity. This asymmetric approach is efficient, mild, robust and atom-economic, and features very broad substrate scope with easily available Tol-BINAP ligands.

Abstract:A CuI-catalyzed three-component coupling of trifluoromethyl ketone N-tosylhydrazones, alkynes and azides has been developed. The reactionrepresents a straightforward method to access difluoromethylene substituted 1,2,3-triazoles. Mechanistically, it has been proposed that the reaction follows a pathway involving the formation of Cu(I)triazolide intermediate, Cu(I) carbene formation, migratory insertion, and -fluoride elimination. Thetransformation is featured by mild conditions, wide substrate scope and high efficiency.

Abstract:A novel metal-free, efficient cascade reaction has been developed to construct 1,2,3-triazole-fused 1,4-diazepinone skeletons. Mechanism investigation indicated that sodium azide has not only served as a 1,3-dipoles synthon in [3 + 2] cycloaddition but also prompted C-N bond formation. Furthermore, the potential utility of this protocol was demonstrated by scale-up synthesis of 1,2,3-triazole-fused diazepinone derivatives and the derivatization of them.

Abstract:The first metal-catalyzed [6 + 3] cycloaddition of tropone with azomethine ylides has been developed. With the use of a chiral ferrocenylphosphine-copper(I) complex as the catalyst, the asymmetric variant of the [6 + 3] cycloaddition has also been successfully achieved. The reactions proceeded smoothly under mild conditions, affording piperidine-fused bicyclic heterocycles in moderate to high yields with good to excellent diastereo- and enantioselectivies. The procedures are operationally simple and the catalysts are cheap and readily accessible, thus providing a practical approach to piperidine-fused bicyclic heterocycles.

Abstract:A Cu/Pd‐catalyzed tandem radical cyclization and C‐H activation/carbene insertion of α‐bromocarbonyls with α‐diazoesters has been developed for the construction of fused quinolin‐2(1H)‐one scaffolds. The reaction proceeded well to afford a series of 3,5‐dihydrocyclopenta[de]quinolin‐2(1H)‐one derivatives. Additionally, bioactive molecules such as sesamol and estrone could be modified by using this method.

Abstract:A Rh(II)-catalyzed dearomative intramolecular [3 + 2] dipolar cycloaddition involving the indolic C2-C3 carbon-carbon double bond has been developed. The reaction was launched from the triazole moiety within the substrate and proceeded efficiently under mild conditions. A wide range of functional groups could be tolerated. These features render the current reaction a highly useful tool for the synthesis of polycyclic indole alkaloids, as showcased by a rapid assembly of the core structure of Aspidosperma and the related alkaloids.

Abstract:An unprecedented copper-catalyzed tandem cross-coupling/[2 + 2] cycloaddition of 1,6-allenynes with diazo compounds was reported, chemo- and regioselectively providing 3-azabicyclo[5.2.0] frameworks in moderate to excellent yields under mild reaction conditions. Moreover, the products readily convert to highly functionalized quinolines via oxidative radical rearrangement.

Abstract:Herein, a copper‐catalyzed photoinduced [3+3] cycloaddition reaction of α‐acidic isocyanides with 2,5‐diaryltetrazoles is described. The reaction proceeded smoothly with high regioselectivity, affording a range of important 1,2,4‐triazines under mild conditions. The obtained 1,2,4‐triazines were converted into triazine alcohol, Weinreb amide and triazinone easily, which further illustrate the utility of this [3+3] cycloaddition process.

Abstract:An iodine‐promoted one‐pot cascade oxidative annulation reaction has been developed for the synthesis of [1,2,3]triazolo[1,5‐a]quinolines from methyl azaarenes and N‐tosylhydrazines. The reaction has a broad substrate scope and can be easily scaled up to gram‐scale. 1,2,3‐Triazoles are an important skeletal structure for the construction of C‐C and C‐P bonds, 2‐cyclopropylquinolines and imidazo[1,5‐a]quinolines, for which different synthetic applications were explored.
