Multiplex Allele-Specific Amplification From Whole Blood Directly

Abstract:Single nucleotide polymorphisms (SNPs) have been found to be related to the risk of many disorders and been used as important predictors of disease susceptibility. Here, we developed a multiplex allele-specific amplification method for detecting multiple SNPs from whole blood directly. To facilitate direct PCR amplification from whole blood, we increased the pH value of PCR solution and the concentration of magnesium ions, and added amplification facilitator glycerol to the PCR mixtures. Moreover, the effects of anticoagulants on PCR amplification and the effects of storage temperature and time on the stability of whole blood sample were also investigated. We established 3-plex, 5-plex, and 8-plex allele specific amplifications to determine thirteen SNPs and a deletion polymorphism in 141 Chinese individuals. The genotyping results were consistent with those obtained by sequencing method. The method developed in this study is time-saving, cost-effective, and easy-to-do, especially suitable for clinical prediction of disease susceptibility.
