Abstract:Minisci reaction is a convenient method for the functionalization of C—H in electron-defficient N-heterocycles.Different substituents could be introduced onto heterocycles through Minisci reaction,such as alkyl,acyl,silyl and alkylthio,etc.Water is eco-friendly and an ideal medium in organic synthesis,which has been applied in Minisci reaction.Recent progress in Minisci reaction in aqueous solution is reviewed herein.Minisci reaction is a convenient method for the functionalization of electron-defficient N-heterocycles.Different substituents could be introduced onto heterocycles through Minisci reaction,such as alkyl,acyl,silyl and alkylthio,boryl,etc.Thus,Minisci reaction is a powerful toolbox for the synthesis of substituted N-heterocylces.Water is eco-friendly and an ideal medium in organic synthesis,which has been applied in Minisci reaction.Although water was used from the discovery of Minisci reaction,Minisci reaction in aqueous solution is still underdeveloped.The recent progress in Minisci reaction in aqueous solution,including alkylation,arylation and acylation,is reviewed according to different radical precursors.

Abstract:Herein, we report a protocol for direct visible-light-mediated Minisci C-H alkylation of heteroarenes with unactivated alkyl halides using molecular oxygen as an oxidant at room temperature. This mild protocol is compatible with a wide array of sensitive functional groups and has a broad substrate scope. Notably, functionalization of (iso)quinolines, pyridines, phenanthrolines, quinazoline, and other heterocyclic compounds with unactivated primary, secondary, and tertiary alkyl halides proceeds smoothly under the standard conditions. The robustness of this protocol is further demonstrated by the late-stage functionalization of complex nitrogen-containing natural products and drugs.

Abstract:N-Heteroarenes are present in a wide variety of natural products, small-molecule drugs, organic materials, and ligands. Therefore, the methods for selective C-H functionalization of N-heteroarenes are highly sought-after for late-stage modification of pharmaceuticals. A useful tool for the synthesis of alkyl-substituted nitrogen-containing aromatic rings is the Minisci reaction, in which a protonated N-heteroarene is attacked by an alkyl radical under oxidative and acidic conditions. Classic Minisci reactions often require the use of excess oxidant, excess acid, and high temperature, which greatly limits the scope of the substrates. With the rapid development of photocatalysis in organic synthesis, in recent years, a variety of photocatalytic Minisci reactions have been reported, and successfully been applied to the synthesis of drugs. In this paper, the visible light mediated Minisci reactions in recent years are briefly reviewed.

Abstract:Minisci-type reaction is one of the important means to construct C(sp3)-H functionalization of heteroarenes. According to traditional methods, stoichiometric amounts of precious transition metal catalysts and chemical oxidants were required at high temperatures. Here, a green and gentle novel Minisci-type method was developed via visible-light-induced cross-dehydrogenative coupling of heteroarenes with aliphatic C(sp3)-H bonds under oxidant-free and transition-metal-catalyst-free conditions. Only the catalytic equivalent of CF3SO2Na and room temperature were required to maintain an efficient reaction.

Abstract:With the irradiation of visible light, alpha-heteroarylation of aliphatic alcohol was selectively achieved under gentle conditions in the present of iodobenzene dichloride. The Minisci-type cross-dehydrogenative coupling reaction was performed in the absence of metal and acid. Readily available primary alcohols and electron-withdrawing heteroarenes can be applied smoothly. Mechanism studies indicated that the PhICl radical and chlorine radical formed from iodobenzene dichloride were responsible for the selective hydrogen atom abstraction of alcohols.

Abstract:This study presents a green and practical visible-light-induced photosensitizer-free decarbonylative Minisci-type reaction using aldehydes as alkyl radical precursors. Mechanism experiments revealed that TFA could absorb light and activate N-heteroarenes, promoting the decarbonylation of aldehydes through auto-oxidation. The important role of TFA instead of traditional photosensitizers in acidic photocatalysis system has been proposed first. Furthermore, the photocatalytic system exhibits a broad substrate scope and synthetically useful yields. And the approach for late-stage functionalization of N-heteroarenes enabled synthesis of the bioactive molecule with potential applications in medicinal chemistry.

Abstract:A new strategy involving three-component functionalization of olefins with sulfoxyimidoylsulfonium salt and a nucleophilic reagent for the synthesis of β-amino alcohols and β-amino ethers by photoredox catalysis was developed. The sulfoxyimidoylsulfonium salt is important and serves as the N-centred radical precursor. β-Amino alcohols and β-amino ethers bearing various functional groups are synthesized in good yields under mild conditions. Furthermore, the advantages of this strategy are greater safety, no additives, and easy to obtain raw materials.

Abstract:Visible light promoted difunctionalization of alkynes is reviewed. The difunctionalization reaction is achieved by different reagents. Radicals such as carbon (sp3), carbon (sp2), and other heteroatom (P, S, N, Se, O, and halide) radicals initiated by visible light can undergo radical addition to a carbon-carbon triple bond. Upon further transformation, the desired difunctionalized products are obtained. Some organometallic complexes can be activated by visible light; the difunctionalization of alkynes is catalyzed by these species. Other reagents like 1,3-dipole precursors could also react with alkynes to give difunctionalization products; here, the 1,3-dipole derivatives are obtained by visible light photocatalysis. So far, the strategy has been succeeded in the formation of C–C bonds and C–X bonds. Several valuable chemical skeletons have been constructed under mild conditions. However, high regio- and stereoselectivities in some direct difunctionalization methodologies are yet to be achieved.

Abstract:Herein we disclose a metal-free, N/O-centered radical-promoted Minisci reaction, in which the coupling of various heteroarenes with simple alkanes proceeds under mild conditions. The reaction conditions are neutral; no extra acid is added to preactivate N-heteroarenes in the Minisci reaction. The N-/O-centered radicals are generated directly from amide (TsNHMe) or alcohol (CF3CH2OH) under visible-light irradiation. This green and eco-friendly synthetic process may find potential use in medicinal chemistry.

Abstract:A Minisci-type C-H alkylation reaction of N-heteroarenes with aliphatic alcohols via beta-scission of alkoxy radical intermediates under photoredox-catalyzed conditions was developed. The use of the benziodoxole acetate (BI-OAc) oxidant is critical to achieving high efficiency under mild conditions using a slight excess of an alcohol reactant. Primary, secondary, and tertiary alcohols all are compatible. Reactions of various complex N-heteroarenes including drug molecules and steroid natural products have been demonstrated.

Abstract:We disclosed a visible-light-induced alkylarylation reaction of unactivated alkenes via a metal-free radical addition/aryl translocation cascade sequence. Distal olefinic sulfonate was designed as a unique molecular scaffold allowing for a dominoprocess to synthesize valuable alkylarylated alcohols in good yields with excellent diastereoselectivity, featuring mild reactionconditions, broad substrate scope, and excellent functional group tolerance. The mechanism investigation suggests that a visible-light-induced radical chain process dominates the cascade transformation.

Abstract:Herein, we report a mild protocol for metal-, photocatalyst-, and light-free Minisci C-H alkylation reactions of N-heteroarenes with alkyl oxalates derived from primary, secondary, and tertiary alcohols. The protocol uses environmentally benign persulfate as a stoichiometric oxidant and does not require high temperatures or large excesses of either of the substrates, making the procedure suitable for late-stage C-H alkylation of complex molecules. Notably, several pharmaceuticals and natural products could be functionalized or prepared with this protocol, thus demonstrating its utility.

Abstract:Visible-light-induced three-component 1,2-alkylarylation of alkenes and alkyl radical addition/cyclization of acrylamides have been realized via a photocatalytic halogen-atom transfer (XAT) process. This metal-free protocol utilizes readily available tertiary alkylamine as both an electron donor and XAT reagent for the activation of alkyl halides using naphthalimide (NI)-based organic photocatalysts. This process features broad substrate scope and good functional group tolerance under mild conditions, and could be effectively applied to a variety of medicinally relevant substrates.

Abstract:Herein, a photocatalytic strategy for the synthesis of secondary alcohols by nucleophilic addition of an alkene with an aldehyde is described. This operationally simple methodology opens an approach for the synthesis of alcohols using commercially available reagents in moderate to excellent yields. Mechanistic studies indicate that the formation of the radical anion from alkene via single-electron transfer is the key step in this reaction.

Abstract:One-pot, multicomponent reactions are known for their green and efficient nature. We report a novel three-component reaction of alkyl amines, alkyl glyoxylates, and unactivated alkyl bromides under visible-light-induced palladium catalysis, yielding N-alkyl unnatural α-amino acid derivatives. This method offers mild conditions, broad substrate scope, and excellent functional group tolerance without requiring stoichiometric organometallic reagents. The approach has promising applications in protein engineering and drug discovery.
