Abstract:The Special Issue of Micromachines entitled “Micro/Nanofluidic Devices for Single Cell Analysis” covers recent advancements regarding the analysis of single cells by different microfluidic approaches. To understand cell to cell behavior with their organelles and their intracellular biochemical effect, single cell analysis (SCA) can provide much more detailed information from small groups of cells or even single cells, compared to conventional approaches, which only provide ensemble-average information of millions of cells together. Earlier reviews provided single cell analysis using different approaches [1–3]. The author demonstrates invasive and noninvasive with time and non-time resolved SCA [1]; whereas some other literature provided destructive (with dyes, DNA, RNA, proteins and amino acids) and nondestructive (electroporation, impedance measurement and fluorescence based methods) cellular content analysis using microfluidic devices [3]. Further literature also suggest that single cell analysis is possible with capillary electrophoresis (CE) combined with a detection method such as electrochemical detection (ED), laser induced fluorescence (LIF) detection and mass spectrometry (MS) [4,5]. Abstract:Recent advances in cellular analysis revealed that the seemingly identical cells are heterogeneous in term of functionality, compositions, and genetic performance. These differences cause difficulty in the diagnostic for a specific model of disease. Detection of biomolecules such as DNA, RNA, and protein or analysis of cell(s), detection of cell surface molecules, and secreted protein, can help us to improve the understanding of a targeted disease and development of new diagnostic and therapeutic approaches. A single-cell includes the minute quantity of these target molecules. Microfluidic devices have the ability to capture a single-cell and its lysate into a pico or femtoliter volumes droplet, or micro-well thus preventing dilution and limiting diffusion. In this review we described the advancement and limitations in microfluidic techniques used toward single-cells analysis.

Abstract:Cells are the fundamental unit of life. Owing to cellular heterogeneity, traditional biochemical assays which analyze cells in bulk often overlook the rich information available when single cells are studied. Accurate and reliable determination of the chemical composition in individual cells would greatly improve the probability of discriminating infected cells from healthy ones and provide a solid foundation on study and development in various fields including biochemistry, medicine, and pathology clinic. Microfluidic chip system for single-cell analysis is now attracting broad interests. The ability of integrating the whole process on one microfluidic chip, including single cell injection, lysis, separation and detection of cellular constituents by chip-based CE, offers a new technical platform for single cell analysis. This article provides a review on recent advances in determining intracellular contents in individual cells using microfluidic devices. Focus areas include on-chip single cell manipulation by means of electroosmotic flow (EOF), pressure-driven flow combined with EOF and optical tweezer (OT) to perform sequentially single cell injection, lysis, separation and detection of cellular constituents by chip-based CE with highly sensitive detection. The approaches on on-chip derivatization are also addressed.

Abstract:Substantial evidence shows that the heterogeneity of individual cells within a genetically identical population can be critical to their chance of survival. Methods that use average responses from a population often mask the difference from individual cells. To fully understand cell-to-cell variability, a complete analysis of an individual cell, from its live state to cell lysates, is essential. Highly sensitive detection of multiple components and high throughput analysis of a large number of individual cells remain the key challenges to realise this aim. In this context, microfluidics and lab-on-a-chip technology have emerged as the most promising avenue to address these challenges. In this review, we will focus on the recent development in microfluidics that are aimed at total single cell analysis on chip, that is, from an individual live cell to its gene and proteins. We also discuss the opportunities that microfluidic based single cell analysis can bring into the drug discovery process.

Abstract:A core challenge in biomedical and life science research is to decipher cell-to-cell differences under pathological progression or external perturbation. Microfluidics-based integrated systems serve as powerful tools for accurately investigating cellular phenotypes and molecular responses in the state of physiological and pathological processes at the single-cell scale. Here, we review the advanced microfluidic strategies for single-cell research relevant to tumorigenesis and evolution, immune responses, and drug discovery The review will discuss the design of microfluidic devices for single-cell analysis, the current methods of microfluidic devices for single-cell isolation, and applications of microfluidics-based single-cell analysis in cell biology, disease diagnosis, and therapy. Specifically, we will focus on the microfluidics-based techniques for single-cell operation and recent strategies for single-cell measurements. Limitations and future perspectives of microfluidics-based single-cell analysis will also be discussed.

Abstract:Cellular heterogeneity of any tissue or organ makes it challenging to identify and study the impact and the treatment of any disease. In this context, analysis of cells at an individual level becomes highly relevant for throwing light on the heterogeneous nature of cells. Single cell analysis can be used to gain insights into an overall view of any disease, thereby holding great applications in health diagnosis, disease identification, drug screening, and targeted delivery. Various conventional methods, such as flow cytometry, are used to isolate and study single cells. Still, these methods are narrower in scope due to certain limitations, including the associated processing/run times, the economy of reagents, and sample preparation. Microfluidics, an emerging technology, overcomes such limitations and is now being widely applied to develop tools for the isolation, analysis, and parallel manipulation of single cells. This review systematically compiles various microfluidic tools and techniques involved in single cell investigation. The review begins by highlighting the applications of microfluidics in single cell sorting and manipulation, followed by emphasizing microfluidic platforms for single cell analysis, with a specific focus on optical sensing-based detection in a high-throughput fashion, and ends with applications in cancer cell studies.

Abstract:The use of nanoparticles (NPs) in nanomedicine holds great promise for the treatment of diseases for which conventional therapies present serious limitations. Additionally, NPs can drastically improve early diagnosis and follow-up of many disorders. However, to harness their full capabilities, they must be precisely designed, produced, and tested in relevant models. Microfluidic systems can simulate dynamic fluid flows, gradients, specific microenvironments, and multiorgan complexes, providing an efficient and cost-effective approach for both NPs synthesis and screening. Microfluidic technologies allow for the synthesis of NPs under controlled conditions, enhancing batch-to-batch reproducibility. Moreover, due to the versatility of microfluidic devices, it is possible to generate and customize endless platforms for rapid and efficient in vitro and in vivo screening of NPs' performance. Indeed, microfluidic devices show great potential as advanced systems for small organism manipulation and immobilization. In this review, first we summarize the major microfluidic platforms that allow for controlled NPs synthesis. Next, we will discuss the most innovative microfluidic platforms that enable mimicking in vitro environments as well as give insights into organism-on-a-chip and their promising application for NPs screening. We conclude this review with a critical assessment of the current challenges and possible future directions of microfluidic systems in NPs synthesis and screening to impact the field of nanomedicine.

Abstract:The investigation of human disease mechanisms is difficult due to the heterogeneity in gene expression and the physiological state of cells in a given population. In comparison to bulk cell measurements, single-cell measurement technologies can provide a better understanding of the interactions among molecules, organelles, cells, and the microenvironment, which can aid in the development of therapeutics and diagnostic tools. In recent years, single-cell technologies have become increasingly robust and accessible, although limitations exist. In this review, we describe the recent advances in single-cell technologies and their applications in single-cell manipulation, diagnosis, and therapeutics development.

Abstract:Biological analyses traditionally probe cell ensembles in the range of 103-106 cells, thereby completely averaging over relevant individual cell responses, such as differences in cell proliferation, responses to external stimuli or disease onset. In past years, this fact has been realized and increasing interest has evolved for single-cell analytical methods, which could give exciting new insights into genomics, proteomics, transcriptomics and systems biology. Microfluidic or lab-on-a-chip devices are the method of choice for single-cell analytical tools as they allow the integration of a variety of necessary process steps involved in single-cell analysis, such as selection, navigation, positioning or lysis of single cells as well as separation and detection of cellular analytes. Along with this advantageous integration, microfluidic devices confine single cells in compartments near their intrinsic volume, thus minimizing dilution effects and increasing detection sensitivity. This review overviews the developments and achievements of microfluidic single-cell analysis of intracellular compounds in the past few years, from proof-of-principle devices to applications demonstrating a high biological relevance.

Abstract:Understanding molecular, cellular, genetic and functional heterogeneity of tumors at the single-cell level has become a major challenge for cancer research. The microfluidic technique has emerged as an important tool that offers advantages in analyzing single-cells with the capability to integrate time-consuming and labor-intensive experimental procedures such as single-cell capture into a single microdevice at ease and in a high-throughput fashion. Single-cell manipulation and analysis can be implemented within a multi-functional microfluidic device for various applications in cancer research. Here, we present recent advances of microfluidic devices for single-cell analysis pertaining to cancer biology, diagnostics, and therapeutics. We first concisely introduce various microfluidic platforms used for single-cell analysis, followed with different microfluidic techniques for single-cell manipulation. Then, we highlight their various applications in cancer research, with an emphasis on cancer biology, diagnosis, and therapy. Current limitations and prospective trends of microfluidic single-cell analysis are discussed at the end.

Abstract:Recent advances in single-cell and multicellular microfluidics technology have provided powerful tools for studying cancer biology and immunology. The ability to create controlled microenvironments, perform high-throughput screenings, and monitor cellular interactions at the single-cell level has significantly advanced our understanding of tumor biology and immune responses. We discuss cutting-edge multicellular and single-cell microfluidic technologies and methodologies utilized to investigate cancer–immune cell interactions and assess the effectiveness of immunotherapies. We explore the advantages and limitations in the wide range of 3D spheroid and single-cell microfluidic models recently developed, highlighting the various approaches in device generation and applications in immunotherapy screening for potential opportunities for point-of-care approaches. Abstract:In a forest of a hundred thousand trees, no two leaves are alike. Similarly, no two cells in a genetically identical group are the same. This heterogeneity at the single-cell level has been recognized to be vital for the correct interpretation of diagnostic and therapeutic results of diseases, but has been masked for a long time by studying average responses from a population. To comprehensively understand cell heterogeneity, diverse manipulation and comprehensive analysis of cells at the single-cell level are demanded. However, using traditional biological tools, such as petri-dishes and well-plates, is technically challengeable for manipulating and analyzing single-cells with small size and low concentration of target biomolecules. With the development of microfluidics, which is a technology of manipulating and controlling fluids in the range of micro- to pico-liters in networks of channels with dimensions from tens to hundreds of microns, single-cell study has been blooming for almost two decades. Comparing to conventional petri-dish or well-plate experiments, microfluidic single-cell analysis offers advantages of higher throughput, smaller sample volume, automatic sample processing, and lower contamination risk, etc., which made microfluidics an ideal technology for conducting statically meaningful single-cell research. In this review, we will summarize the advances of microfluidics for single-cell manipulation and analysis from the aspects of methods and applications. First, various methods, such as hydrodynamic and electrical approaches, for microfluidic single-cell manipulation will be summarized. Second, single-cell analysis ranging from cellular to genetic level by using microfluidic technology is summarized. Last, we will also discuss the advantages and disadvantages of various microfluidic methods for single-cell manipulation, and then outlook the trend of microfluidic single-cell analysis.

Abstract:Cell-to-cell differences exist ubiquitously in microbial communities. This individual heterogeneity, often manifested in cell phenotypic function (e.g., antibiotic resistance), can be critical in determining the fate of a microbial community. However, studying such heterogeneity in microbial communities remains a significant challenge due to their enormous diversity and complex cell-cell interactions. Here we review recent advances in microfluidic technologies for detecting, manipulating, and sorting microbial populations at the single-cell level, which significantly advanced our understanding of microbial behaviour and their roles in the microbial ecosystem. We will highlight microfluidic systems with label-free detection methods, including optical imaging and Raman spectroscopy, due to their advantages in investigating real-world microbial communities. We will showcase these technologies in emerging applications, including rapid diagnosis of pathogens and antibiotic resistance, chemotaxis, and Raman-activated cell sorting to search for natural microbial cells of desirable phenotypic function.

Abstract:Microdevices, since their inception in the last decade of 20th century, have changed our outlook toward science and technology due to their potential applications in fields ranging from optics, semiconductors, and microelectronics industry to drug discovery and development, point-of-care clinical diagnostics, sensitive bioanalytical systems, and other areas of the biological sphere. Microfluidics comprise constraining liquids on a micrometer or submicrometer scale. The fast growth and widespread research pertaining to microfluidic systems have been sustained by their appositeness for abovementioned biological applications that can be carried out using smaller sample volumes and at a fraction of the cost and time as compared to conventional macroscale systems. In this chapter, we review the potential applications of microfluidic platforms for drug discovery applications comprising high throughput screening in target selection, lead identification/optimization, and preclinical testing. We also discuss the potential applications of microfluidics in chemical analysis as well as analysis of metabolites in blood for studying pathology, which can potentially replace conventional benchtop equipment by miniaturized chips that are portable and can be easily handled.

Abstract:The commonly existing cellular heterogeneity plays a critical role in biological processes such as embryonic development, cell differentiation, and disease progress. Single-cell omics-based heterogeneous studies have great significance for identifying different cell populations, discovering new cell types, revealing informative cell features, and uncovering significant interrelationships between cells. Recently, microfluidics has evolved to be a powerful technology for single-cell omics analysis due to its merits of throughput, sensitivity, and accuracy. Herein, the recent advances of microfluidic single-cell omics analysis, including different microfluidic platform designs, lysis strategies, and omics analysis techniques, are reviewed. Representative applications of microfluidic single-cell omics analysis in complex biological studies are then summarized. Finally, a few perspectives on the future challenges and development trends of microfluidic-assisted single-cell omics analysis are discussed.
