Dha reduces insulin resistance and inflammatory response in the liver of diabetic rats by regulating tnf-a/tak1/jnk signaling pathway

Abstract:Objective: To analyze how DHA reduces insulin resistance and inflammation in the liver of diabetic rats by regulating the TNF-a/TAK1/JNK signaling pathway. Methods: A total of 30 clean SD rats were recruited and given a high-fat diet + streptozotocin injection to establish a rat T2DM model, which was divided into control, T2DM, and T2DM + DHA groups, with 10 rats per group. Fasting glucose and insulin levels were determined for each group, and the insulin resistance index (HOMA-IR) was calculated. The expressions of G6Pase, PEPCK mRNA, and the inflammatory factors TNF-alpha, IL-1 beta, and IL-6 in rat liver tissue were detected using real-time quantitative PCR. TAK1 and JNK protein expressions in each group of rats were detected using a western blot method. Result: Fasting blood glucose, insulin, and HOMA-IR in the T2DM group were significantly higher compared to the control group (P<0.05), and the HOMA-IR level in the T2DM + DHA group was significantly lower than that in the T2DM group (P<0.05). The expression of G6Pase and PEPCK mRNA in the T2DM group was significantly higher than that in the control group (P<0.01). The expression of G6Pase and PEPCK mRNA in the T2DM + DHA group was significantly lower than that in the T2DM group, and the difference was statistically significant (P<0.01). The expression of TNF-alpha, IL-1 beta, and IL-6 mRNA in liver tissue of the T2DM group was significantly higher compared to the control group (P<0.01) but significantly lower than those in the T2DM group; the difference was statistically significant (P<0.01). The TAK1 and JNK protein phosphorylation levels in the liver tissue of the T2DM group were significantly higher than those in the control and T2DM+DHA groups (P<0.01), and the difference was statistically significant (P<0.01). Conclusions: DHA can significantly reduce insulin resistance and inflammatory response in the liver of T2DM rats. The mechanism involved with this reduction may be related to TNF-alpha-mediated TNF-alpha/TAK1/JNK signaling pathway downregulation.
