Abstract:The effects of acute and subchronic administration of dimethoate to rats on gamma-aminobutyric (GABA) concentration and receptor characteristics in cerebral cortex were investigated. After a single (acute exposure) and repeated treatment of dimethoate, cerebral cortex was found to inhibit ACh activity in a dose-dependent way and increase the affinities of GABA(A) receptor (P<0.05). Acute administration increased the concentration of GABA (P>0.05), without affecting the numbers of GABA(A) receptor. Whereas subchronic administration of dimethoate reduced the concentration of GABA (P>0.05), affecting the numbers of GABA(A) receptor. The doses in subchronic administration were much lower than those used in acute treatment. It is suggested that the GABAergic involvement may be a part of a compensatory inhibitory process to counteract the enhanced cholinergic activity induced by dimethoate, which probably is to counteract the seizures after acute intoxication and to protect learning and remember capacities loss after the repeated administration.
