Effects Of Low Levels Of Mono-Phthalate On Inter-Sertoli Tight Junction In Two-Compartment Sertoli Cell Culture

Abstract:Di-n-butyl phthalate (DBP) and Di-2-ethylhexyl phthalate (DEHP) are the most commonly used plasticizers in China. In epidemiological surveys, these two phthalates could be found in biological samples of almost all people. Although reproductive and developmental toxicity of phthalates had already been confirmed in wildlife and experimental animals, little is known about whether the low concentration of phthalates could affect the development of reproductive organs in animals or humans. In this study, the in vitro two-compartment primary Sertoli cell culture model was used to explore the potential toxicity of low concentration of phthalates on Sertoli cells of rat testis. Mono-n-butyl phthalate (MBP) and mono-2-ethylhexyl phthalate (MEHP), the metabolites of DBP and DEHP, were chosen as target chemicals because they were considered as the main toxicants of DBP and DEHP in most literatures. By the methods of transmission electron microscope, transepithelial electrical resistance assay (TEER), immunofluorescence localization and RT-PCR, the effects of MBP and MEHP on the tight junction of rat Sertoli cells were studied and compared. Results showed that low levels of mono-phthalates (>= 150 mu mol/L in MBP and >= 30 mu mol/L in MEHP) could destroy Sertoli cell monolayer (decline the TEER value), induce ridge line of Sertoli cells losing, and affect the tight junction structure and expression of tight junction related protein such as ZO-1, F-actin and Occludin. The toxicity of MEHP on Sertoli cells was higher than that of the same dose of MBP. Comparing with other parameters (TEER, ultrastructure, ZO-1 and F-actin), Occludin can be considered as a sensitive biomarker of phthalate effects on rat testis.
