A Label-Free Aptasensor Based on a Dual-Emission Fluorescent Strategy for Aflatoxin B1 Detection

Abstract:Aflatoxin B1 (AFB1) is a toxic mycotoxin that can contaminate many food products. For food safety monitoring, a rapid and convenient detection method is usually required. Herein, we reported a label-free dual-emission aptasensor based on fluorescence resonance energy transfer (FRET) for AFB1 detection. Tow fluorescent probes of thioflavin T (ThT) and trans-2-[4 '-(dimethylamino)styryl]-3-ethyl-1,3-benzothiazole (DMASEBT) were employed as dual-reporters in this system. And a chimeric aptamer B72 was utilized as the capturing probe. Due to the FRET effect between two fluorescent probes, fluorescent responses to the presence of AFB1 at two wavelengths can be observed. And the product of the changes of fluorescent intensities can be plotted with the concentrations to quantitively detect AFB1, with improved detection sensitivity. The detection limit was determined to be 0.01 ng/mL, which was lower than most of the currently reported method. And the selectivity of the detection in foodstuff was also confirmed. The proposed dual-emission aptasensor was applied in complex samples matrix (corn and peanut samples) with satisfactory results. This detection method is convenient and reliable, and the dual-emission fluorescent strategy could be widely used in the rapid detection of other mycotoxins.
