Abstract:Light and temperature in plants are perceived by a common receptor, phytochrome B (phyB). How phyB distinguishes these signals remains elusive. Here, we report that phyB spontaneously undergoes phase separation to assemble liquid-like droplets. This capacity is driven by its C terminus through self-association, whereas the intrinsically disordered N-terminal extension (NTE) functions as a biophysical modulator of phase separation. Light exposure triggers a conformational change to subsequently alter phyB condensate assembly, while temperature sensation is directly mediated by the NTE to modulate the phase behavior of phyB droplets. Multiple signaling components are selectively incorporated into phyB droplets to form concentrated microreactors, allowing switch-like control of phyB signaling activity through phase transitions. Therefore, light and temperature cues are separately read out by phyB via allosteric changes and spontaneous phase separation, respectively. We provide a conceptual framework showing how the distinct but highly correlated physical signals are interpreted and sorted by one receptor.

Abstract:We demonstrate an extension of a UV–Vis spectroscopy method to determine the phase boundaries for thermosensitive colloids as an alternative to the time-consuming sedimentation method. The Bragg attenuation peak from colloidal crystallites was monitored during the quasi-equilibrium colloidal crystal melting. The melting and freezing boundaries of the coexistence region were determined via a blue shift of Bragg's peak and the disappearance of peak area. We confirm this method using poly(N-isopropylacrylamide) (PNIPAM) particles at different charge densities and temperatures far below the lower critical solution temperature. At low pH, the particles behave as thermosensitive hard spheres.

Abstract:Light and temperature are two distinct but closely linked environmental factors that profoundly affect plant growth and development. Biomolecular condensates are membraneless micron-scale compartments formed through liquid–liquid phase separation, which have been shown to be involved in a wide range of biological processes. In the last few years, biomolecular condensates are emerged to serve as phase separation-based sensors for plant sensing and/or responding to external environmental cues. This review summarizes the recently reported plant biomolecular condensates in sensing light and temperature signals. The current understanding of the biophysical properties and the action modes of phase separation-based environmental sensors are highlighted. Unresolved questions and possible challenges for future studies of phase-separation sensors are also discussed.

Abstract:Heat shock factor 1 (HSF1) is the critical orchestrator for a cell responding to heat shock, and its dysfunction is linked to cancers and neurodegenerative diseases. HSF1 undergoes phase separation upon heat shock, and its activity is regulated by various post-translational modifications (PTMs). Despite its importance, the molecular details underlying HSF1 phase separation, temperature sensing, and post-translational modifications (PTMs) regulation remain poorly understood. Here, we discovered that HSF1 exhibits temperature-dependent phase separation with a lower critical solution temperature (LCST) behavior due to entropy contribution from solvent molecules, providing a new conceptual mechanism accounting for HSF1 activation. We employed a synergistic approach combining coarse-grain simulation and nuclear magnetic resonance spectroscopy to reveal the residue-level molecular details of the interactions driving the phase separation of wild-type HSF1 and its distinct PTM patterns at various temperatures. The identified interaction sites were further validated with biochemistry assays and mapped interface accounts for HSF1 functions reported. Importantly, the amino acid substitution experiment reveals the molecular grammar for temperature-dependent HSF1 phase separation is species-specific and physiologically relevant. These findings delineate chemical code that integrates protein PTM patterns with accurate phase separation for body physiological temperature control in animals.

Abstract:Detecting temperature is crucial for the survival of living organisms. Though the thermo transient receptor potential (thermoTRP) channels, such as TRPV1 or TRPM8, have been identified as prototypic heat or cold sensors, respectively, how they detect temperature remains elusive. Here we first identified groups of clustered residues in these channels that undergo burial/exposure conformational rearrangements during temperature activation by analyzing available protein structures or hydroxyl radical footprinting-mass spectroscopy (HRF-MS). By systematically perturbing water-protein interactions at these residues, we found that the temperature sensitivity in these channels were modulated in accordance with the sidechain hydrophobicity. The changes in energy associated with changes in water-protein interactions were sufficient for thermo activation. Abstract:Energy transfer is usually applied in ratiometric thermometry, but it often decreases sensitivities due to much reduced distinguishment in the thermal responses of two different-colored emitters. Herein, a feasible strategy to restrain energy transfer is utilized for achieving sensitive high-temperature detection, simply by increasing the dopant concentration to induce microphase separation. Atomic force microscopy phase images reveal that this phase separation becomes dominant when the doping ratio reaches above 40%. This results in suppression of energy transfer, which is evidenced by systematic photophysical investigations. On this basis, by using heat-resistant emitters, a series of inexpensive and easily prepared solid-film organic high-temperature ratiometric thermometers are developed. They exhibit a broad eye-detective sensing range of 102-236 degrees C with the relative sensitivity (S-r) higher than 0.5% K-1 and the maximum temperature resolution attaining 0.39 K. Good reversibility and stability are also demonstrated in ambient atmosphere.

Abstract:The ability to precisely monitor temperature at a high resolution is an important task, particularly in terms of safety. Inspired by natural thermosensitive transient receptor potential cation channels, we developed a temperature sensor based on thermal-driven ionic charge separation. To mimic the function of nature, an ionic covalent organic framework-based nanofluidic membrane was fabricated. By engineering the membrane to separate two electrolyte solutions, the temperature difference across the membrane can synchronously induce a potential. The high charge density and narrow channel size render extraordinary permselectivity to the membrane, thus offering a thermosensation selectivity of up to 1.25 mV K-1, superior to that of any known natural system. Additionally, the generated potential is linearly related to the introduced temperature gradient, thus allowing for precise detection. With these attributes, an alarm device with high thermosensation sensitivity was constructed, demonstrating great promise for environmental temperature monitoring. [GRAPHICS] .

Abstract:Thermal sensation, which is the conversion of a temperature stimulus into a biological response, is the basis of the fundamental physiological processes that occur ubiquitously in all organisms from bacteria to mammals. Significant efforts have been devoted to fabricating artificial membranes that can mimic the delicate functions of nature; however, the design of a bionic thermometer remains in its infancy. Herein, we report a nanofluidic membrane based on an ionic covalent organic framework (COF) that is capable of intelligently monitoring temperature variations and expressing it in the form of continuous potential differences. The high density of the charged sites present in the sub-nanochannels renders superior permselectivity to the resulting nanofluidic system, leading to a high thermosensation sensitivity of 1.27 mV K −1 , thereby outperforming any known natural system. The potential applicability of the developed system is illustrated by its excellent tolerance toward a broad range of salt concentrations, wide working temperatures, synchronous response to temperature stimulation, and long-term ultrastability. Therefore, our study pioneers a way to explore COFs for mimicking the sophisticated signaling system observed in the nature.
Tracing the evolutionary emergence of the temperature sensing prion-like domain in EARLY FLOWERING 3 across the plant kingdom

Abstract:Plants have evolved to anticipate and adjust their growth and development in response to environmental changes. To mitigate the negative influence of global climate change on crop production, understanding the key regulators of plant performance is imperative. ( ) is such a regulator involved in the circadian clock and thermomorphogenesis. ELF3 contains a prion-like domain (PrLD) that functions as a thermosensor, enabling its liquid-liquid phase separation at high ambient temperatures. To understand the conservation of this function across the plant kingdom, we traced the evolutionary emergence of ELF3 with a focus on PrLD existence. We observed that the presence of the domain within ELF3, mainly contributed by the length of polyglutamine (polyQ) repeats, is most prominent . By analyzing 319 natural accessions, we detected a wide range of polyQ length variation in ELF3. However, it is only weakly associated with geographic origin, climate conditions and classic temperature-responsive phenotypes. Based on available prediction tools and limited experimental evidence, we conclude that although the emergence of PrLD is not likely to be a key driver of environmental adaptation, it adds an extra layer to ELF3’s role in thermomorphogenesis.

Abstract:Intelligent luminescent materials, characterized by optical switching in response to thermal stimulation, are highly attractive for cutting-edge temperature sensing technologies. However, the exploration of sensitive and reliable thermometric materials remains challenging. Herein, we establish a phase-transition-driven thermometric model that relies on the reversible transition between the monoclinic ( β ) and tetragonal ( α ) phase of LiYO 2 :Dy 3+ crystals under controlled thermal stimulations. Accordingly, the ratiometric emissions from hypersensitive electric diploe transitions ( 4 F 9/2 → 6 H 13/2 ) drastically varied across the phase-transition temperature range, rendering remarkable thermal sensitivity and resolution ( S r- max = 35.24% K −1 , δT min = 0.009 K). We further demonstrate the extended control of phase-transition temperature and thermal hysteresis by a partial substitution approach. These findings highlight an endogenic approach to constructing customized luminescence thermometers via local crystal-field engineering, which raises new possibilities for practical sensing applications.

Abstract:SUMMARY Circadian rhythms are generated by the cyclic transcription, translation and degradation of clock genes, including timeless ( tim ). Currently, little is known about the mechanisms by which the circadian clock senses and adapts to temperature changes. Here we show that temperature dramatically changes the splicing pattern of tim . We found that at 18°C TIM protein levels are diminished due to the induction of two cold-specific splicing isoforms ( tim-cold and tim-short&cold ). At 29°C, another isoform, tim-Medium is strongly upregulated. We found that this isoform switching mechanism allows flies to regulate the levels and activity of TIM by setting miRNA-dependent thresholds for expression as well as by expressing isoforms with specific functions. Flies in which the production of tim-short&cold is abrogated display altered patterns of locomotor activity and altered tim expression. Interestingly, the introns of tim carry the information for the temperature sensitivity, suggesting that tim splicing per se is the temperature sensor.

Abstract:Thermosensation, the ability to detect environmental temperature change, is one of most ancient and crucial processes for the survival of all living organisms. Mammals use temperature-sensitive transient receptor potential (thermoTRP) cation channels as thermometers to convert the temperature change into electrical signals that are finally received as action potentials by nerve endings. In this work, we report the bionic thermosensation by solid-state hybrid nanochannels based on the principle of thermally sensitive permselective ion transport. The hybrid nanochannels possess an asymmetric structure, consisting of ultrasmall silica nanochannels (∼2.3 nm in diameter, ∼100 nm in length) supported by large-sized track-etched poly(ethylene terephthalate) conical nanochannels. When the hybrid nanochannels are engineered to separate two electrolyte solutions, the temperature change in one solution can be directly converted into trans-nanochannel diffusion potential, akin to the natural thermosensation process. Two bionic modes, namely, in the absence and presence of a concentration gradient, were studied to imitate the natural thermosensation of thermoTRP ion channels and shark, respectively. In both cases, real-time thermoelectric response was captured with a fast relative response speed (electrical response time versus temperature change time) of higher than 98%, as well as excellent stability and reversibility. Moreover, the nanochannels are highly sensitive to thermal stimulus, showing a sensitivity of 0.71 mV/K comparable to the natural thermosensation. The experimental results coincide well with the theoretical relationship between electrical response and temperature change derived in terms of a quasi-steady-state ion transport model. Finite element simulations based on coupled Poisson-Nernst-Planck (PNP) and Einstein-Stokes equations were also performed, confirming that the sensitive thermoelectric response originates from the highly cationic selectivity of nanochannels.
A Sensitive and Reliable Organic Fluorescent Nanothermometer for Noninvasive Temperature Sensing

Abstract:Sensing temperature at the subcellular level is of great importance for the understanding of miscellaneous biological processes. However, the development of sensitive and reliable organic fluorescent nanothermometers remains challenging. In this study, we report the fabrication of a novel organic fluorescent nanothermometer and study its application in temperature sensing. First of all, we synthesize a dual-responsive organic luminogen that can respond to the molecular state of aggregation and environmental polarity. Next, natural saturated fatty acids with sharp melting points as well as reversible and rapid phase transition are employed as the encapsulation matrix to correlate external heat information with the fluorescence properties of the luminogen. To apply the composite materials for biological application, we formulate them into colloidally dispersed nanoparticles by a technique that combines in situ surface polymerization and nanoprecipitation. As anticipated, the resultant zwitterionic nanothermometer exhibits sensitive, reversible, reliable, and multiparametric responses to temperature variation within a narrow range around the physiological temperature (i.e., 37 °C). Taking spectral position, fluorescence intensity, and fluorescence lifetime as the correlation parameters, the maximum relative thermal sensitivities are determined to be 2.15% °C–1, 17.06% °C–1, and 17.72% °C–1, respectively, which are much higher than most fluorescent nanothermometers. Furthermore, we achieve the multimodal temperature sensing of bacterial biofilms using these three complementary fluorescence parameters. Abstract:Abstract The circadian clock regulates animal physiological activities. How temperature reorganizes circadian-dependent physiological activities remains elusive. Here, using in-vivo two-photon imaging with the temperature control device, we investigated the response of the Drosophila central circadian circuit to temperature variation and identified that DN1as serves as the most sensitive temperature-sensing neurons. The circadian clock gate DN1a’s diurnal temperature response. Trans-synaptic tracing, connectome analysis, and functional imaging data reveal that DN1as bidirectionally targets two circadian neuronal subsets: activity-related E cells and sleep-promoting DN3s. Specifically, behavioral data demonstrate that the DN1a-E cell circuit modulates the evening locomotion peak in response to cold temperature, while the DN1a-DN3 circuit controls the warm temperature-induced nocturnal sleep reduction. Our findings systematically and comprehensively illustrate how the central circadian circuit dynamically integrates temperature and light signals to effectively coordinate wakefulness and sleep at different times of the day, shedding light on the conserved neural mechanisms underlying temperature-regulated circadian physiology in animals.

Controlling composition of coexisting phases via molecular transitions

Abstract:Phase separation and transitions among different molecular states are ubiquitous in living cells. Such transitions can be governed by local equilibrium thermodynamics or by active processes controlled by biological fuel. It remains largely unexplored how the behavior of phase-separating systems with molecular transitions differs between thermodynamic equilibrium and cases where detailed balance of the molecular transition rates is broken due to the presence of fuel. Here, we present a model of a phase-separating ternary mixture where two components can convert into each other. At thermodynamic equilibrium, we find that molecular transitions can give rise to a lower dissolution temperature and thus reentrant phase behavior. Moreover, we find a discontinuous thermodynamic phase transition in the composition of the droplet phase if both converting molecules attract themselves with similar interaction strength. Breaking detailed-balance of the molecular transition leads to quasi-discontinuous changes in droplet composition by varying the fuel amount for a larger range of inter-molecular interactions. Our findings showcase that phase separation with molecular transitions provides a versatile mechanism to control properties of intra-cellular and synthetic condensates via discontinuous switches in droplet composition.

Ultrasensitive Ratiometric Fluorescent Nanothermometer with Reverse Signal Changes for Intracellular Temperature Mapping

Abstract:Sensing temperature at the subcellular level is pivotal for gaining essential thermal insights into diverse biological processes. However, achieving sensitive and accurate sensing of the intracellular temperature remains a challenge. Herein, we develop a ratiometric organic fluorescent nanothermometer with reverse signal changes for the ultrasensitive mapping of intracellular temperature. The nanothermometer is fabricated from a binary mixture of saturated fatty acids with a noneutectic composition, a red-emissive aggregation-caused quenching luminogen, and a green-emissive aggregation-induced emission luminogen using a modified nanoprecipitation method. Different from the eutectic mixture with a single phase-transition point, the noneutectic mixture possesses two solid-liquid phase transitions, which not only allows for reversible regulation of the aggregation states of the encapsulated luminogens but also effectively broadens the temperature sensing range (25-48 °C) across the physiological temperature range. Remarkably, the nanothermometer exhibits reverse and sensitive signal changes, demonstrating maximum relative thermal sensitivities of up to 63.66% °C-1 in aqueous systems and 44.01% °C-1 in the intracellular environment, respectively. Taking advantage of these outstanding thermometric performances, the nanothermometer is further employed to intracellularly monitor minute temperature variations upon chemical stimulation. This study provides a powerful tool for the exploration of dynamic cellular thermal activities, holding great promise in unveiling intricate physiological processes.

Abstract:Precise sensing of cellular temperature is one significant yet challenge task for studying miscellaneous biological processes. Herein, we report a light-controlled DNA nanothermometer that allow for real-time thermal sensing in extracellular microscope with high spatiotemporal resolution. The light-controlled DNA nanothermometer three key elements: a thermal-sensitive molecular beacon (MB) labelled with fluorophore Cy5 and Cy3 at its 5′ and 3’ termini, an inhibitor strand containing two photocleavable linkers (pc-linker), and a biotin modified strand, which could modify this three-strand hybridization complex onto the cell surface. Upon exposing to UV light irradiation, the light-controlled DNA nanothermometer could be remotely activated and enable to perform highly sensitive and practical ratiometric temperature sensing. Meanwhile, the light-controlled DNA nanothermometer could conduct temperature sensing in the extracellular microscope and demonstrates desirable sensitivity, excellent reversibility, and quantitative ability for extracellular temperature measurement. Therefore, this light-controlled DNA can serve as a promising tool for elucidating thermal-related cell physiological and pathological processes.

Abstract:Luminescence thermometry usually suffer from cellular complexity of the biochemical environment (such as pH and ionic strength), and thus the accuracy and reliability of the determined intracellular temperature are directly affected. Herein, a photoluminescent nanothermometer composed of polymer encapsulated quantum dots (P-QD) has been developed. And the prepared nanothermometer exhibits some advantages: such as non-sensitivity to pH and ionic strength, as well as high detection sensitivity and ultrahigh reversibility. The intracellular temperature was accurately determined under physiological conditions with different pH and ionic strength, and direct measurement of thermogenesis in individual cells has been achieved.

Abstract:Ultrasensitive optical thermometry is realized over a broad temperature range through progressive phase transitions in LiYO2 crystals. Remarkably, a programmable luminescence color switching at prescribed temperatures is realized. Thus an ultra‐high thermal sensitivity and resolution that outperforms the existing luminescent thermometers is obtained. The superiority of this thermometer for intelligent wearable devices is substantiated. Luminescent materials that display quick spectral responses to thermal stimuli have attracted pervasive attention in sensing technologies. Herein, a programmable luminescence color switching in lanthanide‐doped LiYO2 under thermal stimuli, based on deliberate control of the monoclinic (β) to tetragonal (α) phase transition in the crystal lattice, is reported. Specifically, a lanthanide‐doping (Ln3+) approach to fine‐tune the phase‐transition temperature in a wide range from 294 to 359 K is developed. Accordingly, an array of Ln3+‐doped LiYO2 crystals that exhibit progressive phase transition, and thus sequential color switching at gradually increasing temperatures, is constructed. The tunable optical response to thermal stimuli is harnessed for colorimetric temperature indication and quantitative detection, demonstrating superior sensitivity and temperature resolution (Sr = 26.1% K−1, δT = 0.008 K). The advances in controlling the phase‐transition behavior of luminescent materials also offer exciting opportunities for high‐performance personalized health monitoring.
