Preliminary Study Of The Toxicity Of Chlorhexidine By Rats Intratracheal Instillation

Abstract:[Objective] To evaluate the toxic effects following the respiratory instillation of chlorhexidine in rats and organ distribution. [Methods] The rats were exposed following intracheal instillation of chlorhexidine with 100 and 1000 mu g / kg of chlorhexidine respectively in each group at one-time. The rats were executed in batches (one-day and seven days). The viscera index, hematology and serum biochemical and the concentrations of chlorhexidine in the blood, lungs, liver and kidney were detected. [Results] When rats were exposed to the 1000 mu g/kg chlorhexidine, the level of WBC increased rapidly and maintained at a high level for 7d (1d & 7d compared with the control group P<0.05), the level of TP and ALB that reflect the function of liver decreased, the level of BUN and CRE that reflect the function of kidney increased, the level of LDH that reflect the damage of tissue increased. When rats were exposed to the 100 mu g/kg of chlorhexidine, there was no significant effect in serum parameters except that a few parameters (WBC, TP, and ALB) decreased. The distribution of chlorhexidine in organ showed that most of the chlorhexidine were stored in lung following intratracheal instillation of chlorhexidine in rats, and maintained a high level more than 7d, which is 30 times higher than in kidney. [Conclusion] Intratracheal instillation of high-dose chlorhexidine in rats would lead to local damages in lung and some damages in liver and kidney, which suggested that we should pay attention to the self-protection work in daily life to avoid chlorhexidine poisoning following respiratory inhalation.
